auf eine Verbindung der Formel
EMI0001.0009
Man kann beispielsweise 1-(3'-Methyl-phe- nyl)-cyclopentan-l-carboneäure oder ein reak tionsfähiges Derivat derselben, z. B. ein Halo genid, Anhydrid oder einen Ester,. in An- oder Abwesenheit von Kondensationsmitteln und Verteilungsmitteln mit Diäthylaminoäthyl- methyl-amin oder eine Metallverbindung des 1-(3'-Methyl-phenyl) - cyclopen;
Process for the preparation of a basic amide of a 1-aryl-cycloalkyl-1-earbonic acid. The present patent relates to a process for the preparation of a basic amide of a 1-aryl-cycloalkyl-1-carboxylic acid. The process is characterized in that a compound of the formula

to a compound of the formula
EMI0001.0009
wherein X and Y are two radicals, with the exception of a CH3N group contained in one of them, which are split off during the reaction.
You can, for example, 1- (3'-methyl-phenyl) -cyclopentane-l-carboxylic acid or a reactive derivative thereof, z. B. a halide, anhydride or an ester. in the presence or absence of condensing agents and distributing agents with diethylaminoethylmethylamine or a metal compound of 1- (3'-methylphenyl) cyclopene;
tan - 1 - carboxylic acid methyl amide with a reactive ester of diethylamino-ethanol.
The new compound obtained, 1- (3'-methyl-phenyl) -cyclopentane-1- carboxylic acid diethylaminoethyl-methyl-amide, is a base from .Boiling point 147 to 152 under 0.30 mm pressure; their hydrochloride melts at 101 to 102. The compound is intended to find therapeutic use.
Example 1: 22.2 parts of 1- (3'-methyl-phenyl) -cyclopentane-1-carboxylic acid chloride, dissolved in 200 parts of absolute benzene, become a solution of 13 Parts of diethylaminoethyl methyl amine in 100 parts of absolute benzene are added dropwise. The mixture is heated to boiling under reflux for 2 hours. After cooling, the mixture is stirred with water, separated and the base is completely removed from the benzene solution by shaking with dilute hydrochloric acid.
The combined aqueous solutions are extracted with ether, the base is set free with potash and taken up in ether. The ethereal solution is washed with water, dried over potash and the solvent is distilled off. The basic amide obtained boils at 147 to 152 under 0.30 mm pressure.
Somewhat better yields are obtained when using an excess of diamine, that is from 1.2 to 2 mol to 1 mol of acid chloride. Instead of the acid chloride, other halides of 1- (3'-methylphenyl) -cyclopentane-1-carboxylic acid, the acid itself, its ester or its anhydride can also be used.
Example 2: 21.7 parts of 1- (3'-methyl-phenyl) -cyclopentane-l-carboxylic acid-methyl-amide are mixed with 100 parts of absolute toluene and 5 parts of finely powdered sodium amide and heated with stirring until the ammonia formation has ended.
After cooling, 13.5 parts of diethylaminoethyl chloride in 150 parts of absolute toluene are added, the mixture is stirred for some time at room temperature and then heated to 90 to 100 for 2 hours. The cooled reaction product is mixed with 100 parts of water, separated in a separating funnel and the toluene solution is washed a few times with water.
Then the basic components are extracted with dilute acid, the combined acidic solutions are made alkaline and extracted with ether. The ither solutions after drying with sodium sulfate and evaporation give an oil with a boiling point of 147 to 152 under 0.30 mm pressure.
