Verfahren zur Herstellung einer neuen Verbindung der Cyelopentanopolyhydrophenanthr en-Reihe. Es wurde gefunden, dass man zu einer im Ring C ungesättigten Verbindung der Cyclo- pentanopolyhydrophenanthren - Reihe gelan gen kann, wenn man einen 3-Keto-ätioeholan- säure-methylester,
der im Ring C in 12-Stel- lung einen zusammen mit einem benachbarten Wasserstoffatom abspaltbaren Substituenten aufweist, mit diesen Substituenten unter Bil dung einer Doppelbindung abspaltenden Mit- teln behandelt.
Der 12ständige, zusammen mit einem be nachbarten Wasserstoffatom abspaltbare Sub- stituentdes Ausgangsstoffes kann eine freie Hydroxylgruppe oder eine beispielsweise durch Carbonsäuren, wie Essig-, Propion- oder Benzoesäure, durch Sulfonsäuren,
Ha logenwasserstoffsäuren oder Xanthogensäuren veresterte Hydroxylgruppe sein.
Die Abspaltung dieses Substituenten unter Bildung einer Doppelbindung kann mit den für .diese Reaktion an sich bekannten Mitteln erfolgen. Beispielsweise lässt sich eine freie Hydroxylgruppe unter der Ein wirkung von Mineralsäuren, vorzugsweise in Lösungsmitteln, wie Eisessig, Alkohol, Dioxan und dergleichen, von Phosphoroxy- chlorid, Bisulfaten, von Ameisensäure,
Oxal- säure, von Säureanhydriden, wie Aeetanhy- drid oder Phosphorpentoxyd, oder durch die Einwirkung von Katalysatoren, wie Jod- oder Carbonsäure-Salzen, abspalten.
Eine ver- esterte Hydroxylgruppe wird ausser durch die genannten Mittel vorzugsweise auch mit Al- kalien, Erdalkalien., Carbonaten, organischen Basen, wie Pyridin, Dimethylanilin usw., ab gespalten.
An Stelle oder in Kombination mit den genannten Mitteln lässt sich auch erhöhte Temperatur und/oder verminderter Druck an wenden. Gegebenenfalls arbeitet man auch in Gegenwart indifferenter Gase. Statt aus Ila- logenwasserstoffsäureestern direkt SaJogen- wasserstoff abzuspalten,
kann man das Ha logen auch in bekannter Weise durch einen quaternären Ammoniumrest ersetzen und die sen abspalten.
Das neue Verfahrensprodukt, der A11,12 3 - Keto - ätiochoiensäure - methylester vom F. 133 bis 135 , soll als Zwischenprodukt zur Herstellung therapeutisch wertvoller Pro dukte dienen.
<I>Beispiel 1:</I> 8 - Keto -12 - benzoyloxy - ätioaholansäure- methyIester (erhalten aus 8-Oxy-12-acetoxy- ätiocholansäuremethylester durch Oxydation mit Chromsäure in Eisessig bei Zimmertem peratur, Verseifung des so entstandenen Pro duktes zur freien 3-Keto-12-axy-ätiocholan- säure, Behandlung mit Diazomethan und an schliessende Benzoylierung)
wird in einem Claisen-Kolben mit Wurstansatz im Wasser strahlvakuum (12 mm) auf 380 bis 340 (Metallbadtemperatur) erhitzt, während durch die feine Kapillare Kohlendioxyd geleitet wird. Den Kolben senkt man dabei nur so weit ins Metallbad, dass die Benzoesäure ab destillieren kann, das höher siedende Ma terial jedoch zurückfliesst. Nach etwa l1/2 Stunden wird der Kolben tiefer eingetaucht; und die Badtemperatur zum Schluss auf 380 bis 400 gesteigert, wobei die ganze Menge bis auf einen sehr geringen Rückstand destil liert.
Das Destillat wird mit Lauge in neu trale und alkalilösliche Anteile zerlegt. Aus ersteren erhält man den d 11,12-3_Keto-ätio- chalensäure-methylester der Formel

Er kristallisiert aus Äther-Petroläther in Körnern vom F. 138 bis 185 . Die Verbin dung gibt, mit wenig Chloroform verflüssigt, auf Zusatz von Tetranitromethan eine deut liche Gelbfärbung.
<I>Beispiel 2:</I> 5 Teile 3 - Keto -12 - tosyloxy - ätioeholan- säure-methylester (erhalten aus der im Bei spiel 1 erwähnten 3-Keto-12-oxy-ätiach(>lan- säure durch Behandlung mit Diazomethan und anschliessend mit Tosylchlorid in Pyridin) werden mit 50 Teilen Pyridin im Vakuum eingeschmolzen und 40 Stunden in siedendem Xylol erhitzt.
Das Pyridin wird im Vakuum abdestilliert, der Rückstand in Äther auf genommen, die ätherische Lösung mit ver dünnter Salzsäurelösung, Sodalösung und Wasser gewaschen und nach dem Trocknen über Natriumsulfat eingedampft.
Durch chro- matographisehe Reinigung über Aluminium- oxyd liefert der Rückstand den im Beispiel 1 beschriebenen d 11.l2 - 3_ Keto-ätiocholensäure- methylester vom F.133 bis 135 .
An Stelle von Pyridin kann z. B. auch Collidin verwendet werden.

<I> Example 2: </I> 5 parts 3-keto-12-tosyloxy-ethioeholanic acid methyl ester (obtained from the 3-keto-12-oxy-ethic acid mentioned in Example 1 (> lanic acid by treatment with diazomethane and then with tosyl chloride in pyridine) are melted with 50 parts of pyridine in vacuo and heated in boiling xylene for 40 hours.
The pyridine is distilled off in vacuo, the residue is taken up in ether, the ethereal solution is washed with dilute hydrochloric acid solution, soda solution and water and evaporated after drying over sodium sulfate.
