Verfahren zur Darstellung eines Thiophanabkömmlings. Das Biotin, welches für viele Organismen von lebenswichtiger Bedeutung ist, wurde als Abkömmling des 3,4-(2'-Oxo-tetrahydroimid- azol)-thiophans erkannt.
Es wurde nun gefunden, dass man für die Herstellung von in 2-Stellung einen Alkyl- rest oder einen co-substituierten Alkylrest tragenden Abkömmlingen Thiophan-on-(3)- carbonsäureester als Ausgangsverbindungen verwenden kann. Als Substituenten kommen neben Alkylresten auch z.
B. co-Oxyalkyl-, o)-Alkoxyalkyl-, co-Aryloxyalkyl- oder_co-Ha- logenalkylreste in Betracht.
Man führt die Thiophan-on-(3)-carbon- säureester (I), die in der 2-Stellung einen ge eigneten Substituenten tragen, zunächst z. B. mit Hilfe von Blausäure in die entsprechenden Cyanhydrine (II) über. Letztere werden, z. B. ohne weitere Reinigung, verseift und die ent stehenden Dicarbonsäuren (IH) verestert. Die in 2-Stellung substituierten - 3-Oxy-thiophan- 3,4-dica.rbonsäureester (IV) sind destillier- bare Öle.
Bei der Umsetzung mit Halogeniden des Phosphors und des Schwefels, z. B. Thio- nylchlorid, gehen sie in die 3-Chlorester (V) über. Durch reduzierende Mittel kann das Halogen eliminiert werden, wobei man die in der 2-Stellung substituierten Thiophan-3,4- dicarbonsäureester (VI) erhält.
Von diesen gelangt man zu den entsprechenden Säure aziden, indem man mit Hydrazinhydrat die Hydrazide (VII) herstellt und diese mit Na triumnitrit umsetzt. Aus den Aziden (VIII) erhält man durch Erwärmen mit Alkoholen die in der 2-Stellung substituierten 3,4-Di- (carbäthoxyamino)-thiophane (IX) und diese gehen beim Erwärmen mit Halogenwasser stoffsäuren, z.
B. Bromwasserstoffsäure, in die Salze der entsprechenden in der* 2-Stelle substituierten 3,4 - Diamino - thiophane (X) über. Durch Verrühren mit Phosgen in schwach alkalischer Lösung, erhält man aus ihnen die 3,4-(2'-Oxotetrahydroimidazol)-2- X-thiophane (XI).

Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung eines Thio- phanabkömmlings, welches dadurch gekenn zeichnet ist, dass man auf 2-Methyl-3-oxo- thiophan-4-carbonsäureäthylester ein Cyanid einwirken lässt, das gebildete Cyanhydrin verseift, die entstandene Carbonsäure ver- estert,
auf den Ester nacheinander ein Ha- logenierungsmittel und ein Reduktionsmittel einwirken lässt, den entstandenen Dicarbon- säureester mit Hydrazinhydrat umsetzt, auf das gebildete Hydrazid Natriumnitrit einwir ken lässt, das erhaltene Azid durch Einwir- kung von Alkohol zum Diurethan abbaut, dieses durch Säureeinwirkung spaltet und das Spaltprodukt mit Phosgen umsetzt.
Das bisher unbekannte 3,4-(2'-Ogo-tetra- hydro-imidazol)-2-methyl-thiophan ist eine farblose, kristalline Verbindung, welche bei längerem Erhitzen auf 350-360 verkohlt. Sie ist in Alkohol und Essigsäure leieht, in Aceton schwerer, in Äther; Benzol und Wasser schwer löslich. Sie soll als Zwischen- produkt zur Herstellung pharmazeutisch ver- tvendbarer Stoffe mit der Wirkung des Biotuns verwendet werden.
<I>Beispiel:</I> Zu einer Lösung von 188 Gewichtsteilen 2 - Methyl - 3 - oxo - thiophan - 4 - carbonsäure- äthylester in 500 Volumteilen Äther gibt man 80 Gewichtsteile fein gepulvertes .Ka- liumcyanid. Unter Rühren und gutem Kühlen tropft man nun 90 Volumteile konzentrierter Salzsäure innerhalb etwa 2 Stunden ein.
Hierauf wird der Äther verdampft, der zu rückbleibende 2-Methyl-3-oxy-3-cyan-thio- phan-4-carbonsäure-äthylester mit 300 Vo- lumteilen Äthylalkohol und 300 Volumteilen konzentrierter Salzsäure versetzt und dadurch verseift, dass man zunächst langsam den Al kohol abdestilliert und hierauf noch 2 Stun den im siedenden Wasserbad hält.
Durch Einengen im Vakuum gewinnt man die 2 - Methyl -3- oxy-thiophan -3,4- dicarbonsäure, die ohne weitere Reinigung mit Hilfe- von Äthylalkohol und Salzsäure in den 2-Methyl- 3 - ogy - thiophan - 3,4 - dicarbonsäure - diätlhyl- ester übergeführt wird. Er siedet unter 0,05 mm von 10l-102 als fast farbloses, dickflüssiges 01.
262 Gewichtsteile 2-Methyl-3-oxy-thio- phan-3,4-dicarbonsäure-diäthylester werden in 500 Volumteilen Chloroform gelöst- und nacheinander unter guter Eiskühlung mit 80 Gewichtsteilen Pyridin und 119 Gewichts teilen Thionylchlorid versetzt.
Man lässt zu nächst 2 Stunden stehen und erwärmt hierauf im kochenden Wasserbad etwa 40 Minuten bis zum Aufhören der Gasentwicklung. Darauf wird auf Eis gegossen, im Scheide trichter getrennt und mit Wasser und ver- dürinter Södalösung bis zum Eintritt neutra ler Reaktion gewaschen. Der vom Chloroform getrennte 2-Methyl-3-chlor-thiophan-3,4-di- carbonsäure-diäthylester siedet unter 0,1 mm von 100-102 .
Zu einer Auflösung von 280 Gewichts teilen 2 - Methyl - 3 - chlor - thiophan - 3,4 - di- carbonsäure-diäthylester in 4000 Volumteilen 80%iger Essigsäure werden<B>186</B> Gewichts teile Natriumjodid-hydrat gefügt und unter Rühren innerhalb etwa 4 Stunden insgesamt 1400 Gewichtsteile' Zinkstaub in kleinen Portionen eingetragen. Jedes Mal tritt schwache Erwärmung ein.
Nach weiterem Verrühren während etwa 12 Stunden wird vom Zinkacetat getrennt, die Essigsäure durch Destillation entfernt und der Rück stand nach gründlichem Waschen mit Wasser und Sodalösung im Vakuum destilliert. Der 2-Methyl-thiophan-3,4-dicarbonsäure-diäthyl- ester siedet unter 0,02 mm von 88-90 als farbloses 01.
246 Gewichtsteile 2-Methyl-thiophan-3,4- dicarbonsäure-diäthylester werden in 250 Vo- lumteilen abs. Alkohol gelöst und die Lösung mit 50 Gewichtsteilen Hydrazinhydrat etwa 10 Stunden am Rückfluss erwärmt. Nach einiger Zeit beginnt das Hydrazid auszufal len.
Der Niederschläg vermehrt sich im Ver laufe des Erhitzens. Nach dem Abkühlen; wird das 2-Methyl-thiophan-3,4-dicarbon- säure-dihydrazid abgetrennt und aus heissem Wasser umgelöst. Es schmilzt bei 241-242 unter vorherigem Sintern.
Man überschichtet unter Rühren und Kühlen eine Lösung von 218 Gewichtsteilen 2 - Methyl - thiophan - 3,4 - dicarbonsäure - di- hydrazid in 670 Volumteilen 3 n Salzsäure mit 2000 Volumteilen Äther und tropft eine Lösung von 140 Gewichtsteilen Natrium nitrit in .800 Volumteilen Wasser zu. Das entstehende Säureazid ist ätherlöslich.
Die Ätherlösung wird nach beendeter Umsetzung von der Salzlösung getrennt, der Äther ver jagt und das als leicht gelbliches 01 zurück bleibende 2 - Methyl - thiophan - 3,4 - dicarbon- säure-diazid mit 2000 Volumteilen abs. Al kohol versetzt. Die klare Lösung wird auf etwa 65 erwärmt, bei welcher Temperatur starke Stickstoffentwicklung einsetzt. Diese ist nach etwa 30 Minuten beendet.
Die klare, fast farblose Lösung wird mit Wasser bis zur beginnenden Trübung versetzt und ab- gekühlt. Durch Nutschen wird das farblose, in feinen Nädelchen kristallisierende 2-Me- thyl-3,4-äi-(carbäthoxyamino)-thiophan vom Schmelzpunkt 182-183 gewonnen.

Man erwärmt 276 Gewichtsteile dieses Urethans zusammen mit 2760 Volumteilen 45 %igei Bromwasserstoffsäure bis zum Auf hören der Kohlensäureentwicklüng etwa 1 Stunde auf 105-1l0 : Beim Einengen im Vakuum bleibt das 2-Methyl-3,4-diamino- thiophan-dihydrobromid als feste Masse zu rück: Es kann aus verdünntem Aceton oder verdünntem Alkohol umgelöst werden.
Die farblosen, 2 Kristallwasser einschliessenden Kristalle schmelzen unter Zersetzung bei 225-227 . Das Diamin ist durch folgende Derivate charakterisiert.
Das 2 - Methyl - 3,4 - diureido - thiophan schmilzt bei 262 unter Zersetzung. Das 2 - Methyl - 3,4 - diamino - thiophan - dipikrat schmilzt bei 243 unter Zersetzung. Das 2 -Methyl - 3,4 - di- (benzoylamino) -thiophan schmilzt bei 271-272 . Das 2-Methyl-3,4-di- (acetylamino)-thiophan schmilzt bei 260 bis 261 .
330 Gewichtsteile 2-Methyl-3,4-diamino- thiophan-dihydrobromid werden in 2000 Vo- lumteilen n-Natronlauge gelöst und unter Kühlen gut verrührt.
Zu der Lösung tropft man gleichzeitig so, dass immer schwach alkalische Reaktion vorherrscht, einesteils 2000 Volumteile n-Natronlauge und andern teils 100 Gewichtsteile Phosgen in Form einer 20 % igen Lösung in Tolüol. Sofort beginnen farblose Kristalle -- auszufallen. Die Umsetzung ist bald nach Beendigung des Eintropfens vollständig. Der Kristallbrei wird genutscht und mit Wasser gewaschen.
Das 3,4 - (2' - Oxo - tetrahydroimidazol) - 2 - methyl-thiophan kann aus verdünnter Essig säure oder verdünntem Alkohol umgelöst wer den. Es ist in. Alkoholen und Essigsäure leicht, in Aceton schwerer, in Äther, Benzol und Wasser schwer löslich. Es verkohlt bei längerem Erhitzen auf 350-360 .
