Verfahren zur Herstellung eines Diphenylsulfonabkömmlings. Gegenstand des vorliegenden Patentes ist ein Verfahren zur gerstellung eines wasser löslichen Diphenylsulfonabkömmlings, das dadurch gekennzeichnet ist, dass man auf 1 Mol Diphenylsulfon-4,4'-bis-carbaminsäure- phenylester 2 Mol 2-(3'-Aminobenzoylamino)- naphthalin-3,6-disulfonsäure einwirken lässt.
Die so erhaltene neue Verbindung soll als Arzneimittel Verwendung finden. Die Um setzung erfolgt zweckmässig unter Erwärmen und in Gegenwart eines Löse- oder Verdünne mittels, wie Wasser oder einem zweckmässig mit Wasser mischbaren organischen Löse mittel, z. B. Aceton, Methyläthylketon, Diäthylketon, Diogan, Tetrahydrofuran und dergleichen.
<I>Beispiel:</I> 45,5 g des Mononatriumsalzes der 2-(3' Aminobenzoylamino) - naphthalin -<B>3,6</B> - disul - fonsäure werden in 300 ein' Wasser und 4 g Ätznatron gelöst, 25g DiphenyIsulfon- 4,4'-bis-carbaminsäurephenylester und 200 am' Diogan hinzugegeben, die Mischung unter Zusatz von primärem Natriumphosphat auf ein pH = 7,5 eingestellt und unter Rühren am Rüekfluss gekocht.
Nach 6stündigem Ko chen gibt eine kleine Probe mit Wasser ver dünnt keine Fällung mehr. Die klare Lösung wird mit Tierkohle heiss filtriert, das Filtrat nach dem Erkalten in 35%ige Natrium chloridIösung eingerührt. Dabei fällt das Tetranatriumsalz des neuen Diphenylsulfon- abkömmlings aus; es wird abgesaugt und bei 100 getrocknet.
Das als Ausgangsstoff verwendete Mono: natriumsalz der 2-(3'-Amino-benzoylamino)- naphthalin-3,6-disulfonsäure kann wie folgt hergestellt werden: 36,5g Mononatriumsalz der 2-Naphthyl- amin-3,6-disulfonsäure werden in 250 am' Wasser und 4 g Ätznatron gelöst, 30 g Na triumacetat hinzugegeben und bei einer Innentemperatur von 30 bis 35 36.5 g m- Nitrobenzoylchlorid eingetragen.
Nach kurzer Zeit kristallisiert das 2 - (3' -Nitrobenzoyl- amino)-nap.hthalin-3,6-disulfonsaure Natrium aus; es wird abgesaugt, in heissem Wasser gelöst und diese Lösung unter Rühren in eine siedende Mischung von 500g Eisen, 1000 cm" Wasser und 10 cm" Eisessig ein ; getropft. Nach 5stündigem Kochen wird die Reaktionslösung mit Kaliumcarbona,t alka lisch gemacht, mit Tierkohle heiss filtriert und das Filtrat nach dem Erkalten mit viel Salzsäure versetzt. Dabei fällt die 2-(3' Amino-benzoylamino)-na_phthalin-3,6-disul- fonsäure als Mononatriumsalz kristallinisch aus.
