Verfahren zur Darstellung eines Sulfonamides. Gegenstand der Erfindung ist ein eigen artiges Verfahren zur Darstellung eines Sul fonamides, nämlich des bekannten 2-(p- Aminobenzol-sulfonylamino-)pyrimidins, wel ches weisse Kristalle vom Schmelzpunkt 255 bis 256 bildet und als Heilmittel gegen In fektionen verwendbar ist.
Das Verfahren ist dadurch gekennzeich net, dass man ein p-Acylaminobenzol-sulfonyl- guanidin mit Malonsäure zum 2-(p-Acyl- aminGbenzol -sulfonylamino-)\4,6-diogy-pyri- midin kondensiert, letzteres mittels eines Ha- logenierungsmittels in das entsprechende 2-(p Acylaminobenzol -sulfonylamino-)4,6 - dihalo- gen-pyrimidin überführt,
dieses zum 2-(p- Acylaminobenzol - sulfonylamino -) pyrimidin reduziert und daraus durch Abspaltung des Acylrestes das 2-(p-Aminobenzol-sulfoDyl- amino-)pyrimidin .gewinnt.
Ausführungsbeispiel: 12,8 g p - Acetylaminobenzol - sulfonyl- guanidin werden in 100 cm' auf 0 abgekühlte rauchende Schwefelsäure eingetragen. Zu diesem Gemisch werden unter Rühren 5,2 g Malonsäure gegeben, wobei die Temperatur 0 nicht übersteigen darf. Hierauf lässt man einige Stunden bei Zimmertemperatur stehen und erwärmt schliesslich noch 1 Stunde unter Rühren auf 50-60 .
Das gut gekühlte Reak tionsgemisch wird auf Eis gegossen und bei 0 mit konzentriertem Ammoniak neutrali siert, worauf sich das 2-(p-Acetylaminoben= zol-sulfonylamino-) 4,6-diogy-pyrimidin aus scheidet. Ausbeute:<B>12,7</B> g oder 77,7 % d. Th.
10 g der letzteren Verbindung wer den mit 60 cm' Phosphorogychlorid am Rück flusskühler gekocht. Das überschüssige Phos- phorogychlorid wird im Vakuum abdestil- liert, der Rückstand auf Eis gegossen, die Lösung über Kohle filtriert und mit Ammo niak neutralisiert, wobei man dafür Sorge trägt, dass die Temperatur 5 nicht übersteigt.
Der farblose Niederschlag von 2-(p-Acetyl- aminobenzol-sulfonylamino-) 4,6-dichlor-pyri- midin wird mit Wasser gut gewaschen und bei 40-50 im Vakuum getrocknet. Aus beute: 9,9 g oder 88,4% d. Th. 10 g dieser Chlorverbindung werden, in i#lethylalkohol gelöst, in Gegenwart von auf Kalziumkarbonat niedergeschlagenem Palladium als Katalysator und von Ka,lzium- oxyd als säurebindendem Mittel mit Wasser stoff reduziert.
Hierauf wird der Methyl alkohol im Vakuum entfernt und das zurück bleibende 2-(p-Acetylaminobenzol-sulforiyl- amino-)pyrimidin in verdünnter Natronlauge gelöst und mit Säure wieder ausgefällt (pu 5-7). So gereinigt schmilzt es bei 258" unter Zersetzung. Ausbeute: 7,45 g oder <B>92%</B> d. Th.
7,45 g der letzteren Acetylverbindung werden zur Verseifung mit 3 Mol 10 prozentiger Salzsäure 1 Stunde gekocht. Hierauf wird das 2-(p-Aminobenzol-sulfony l- amino-) pyrimidin durch Neutralisieren der Lösung ausgefällt. Aus Wasser umkristalli siert zeigt es, in Übereinstimmung mit der Literatur, einen Schmelzpunkt von \354=255 (u.Zers.). Ausbeute: 5,86 g oder 92,3% d. Th.
Process for the preparation of a sulfonamide. this is reduced to 2- (p-acylaminobenzene-sulfonylamino-) pyrimidine and 2- (p-aminobenzene-sulfoDylamino-) pyrimidine is obtained therefrom by splitting off the acyl radical.
Exemplary embodiment: 12.8 g of p-acetylaminobenzene-sulfonylguanidine are introduced into 100 cm ′ of fuming sulfuric acid cooled to 0. 5.2 g of malonic acid are added to this mixture with stirring, the temperature not being allowed to exceed 0. It is then left to stand for a few hours at room temperature and then heated to 50-60 for a further hour while stirring.
The well-cooled reaction mixture is poured onto ice and neutralized at 0 with concentrated ammonia, whereupon the 2- (p-acetylaminobene = zol-sulfonylamino-) 4,6-diogy-pyrimidine separates out. Yield: 12.7 g or 77.7% of theory. Th.
10 g of the latter compound who boiled the reflux condenser with 60 cm 'phosphorogychloride. The excess phosphorogychloride is distilled off in vacuo, the residue is poured onto ice, the solution is filtered over charcoal and neutralized with ammonia, taking care that the temperature does not exceed 5.
The colorless precipitate of 2- (p-acetylaminobenzene sulfonylamino) 4,6-dichloropyrimidine is washed well with water and dried at 40-50 in a vacuum. From booty: 9.9 g or 88.4% d. Th. 10 g of this chlorine compound, dissolved in i # lethyl alcohol, reduced with hydrogen in the presence of palladium deposited on calcium carbonate as a catalyst and calcium oxide as an acid-binding agent.
The methyl alcohol is then removed in vacuo and the 2- (p-acetylaminobenzene-sulforiyl-amino) pyrimidine that remains is dissolved in dilute sodium hydroxide solution and reprecipitated with acid (pu 5-7). Purified in this way, it melts at 258 "with decomposition. Yield: 7.45 g or 92% of theory.
7.45 g of the latter acetyl compound are boiled for saponification with 3 mol of 10 percent hydrochloric acid for 1 hour. The 2- (p-aminobenzene-sulfony l-amino-) pyrimidine is then precipitated by neutralizing the solution. Recrystallized from water, it shows, in agreement with the literature, a melting point of \ 354 = 255 (u dec.). Yield: 5.86 g or 92.3% of theory. Th.
