Verfahren zur Darstellung eines Sulfonamides. Gegenstand der. Erfindung ist ein eigen artig-es Verfahren' zur Darstellung eines Sulfonamides, nämlich dles bekannten 24p- Aminobenzol-.sulfonylamino-)pyrimidins, wel ches weisse Kristalle vom Schmelzpunkt 255 bis 2,56 bildet und als Heilmittel gegen In fektionen verwendbar ist.
Das Verfahren ist dadurch gekennzeich net, dass man p-Aminobenzol-.sulfonyl-guani- din mit Formylessigsäure zum 2-(p-Amiuo- benzol-sulfonylamino-)6-oxy-pyrimidin, ko@n- - ,densiert, letzteres mittels eines Hal'ogenie- rungsmittels in das 2-(p-Aminobenzol-sul- fonylamino-)
6-halogen-pyrimidin überführt und dieses zum 2-(p-Aminobenzol-sulfonyl- amino-)pyrimidin reddziert.
Ausführungsbeispiel: Zu einer .gekühlten Lösung von 35,0 .g Formylessi:gsäure in 280 cm' rauchender Schwefelsäure von 20-30 % S03-Gelialt gibt man langsam und unter Rühren 44,0 g p-Aminobenzol-s.ulfonyl-guanidin. Während des Eintragens wird die Temperatur auf -5 biss 0 gehalten, hierauf lässt man .sie auf Zimmertemperatur ansteigen, und .schliesslich ; wird das Gemisch noch 1 Stunde unter ;stän- digem Rühren auf dem Wasserbade erhitzt.
Das erkaltete Reaktionsgemisch wird auf 1000.g Eis gegossen, mit 30 %igem Ammoniak neutralisiert, worauf das gebildete <B>2-(p-:</B> Amino#benzol-sulfonylamino-)6-ogy-pyrimidin auskristallisiert. Ausbeute: 39,8 g oder 72,8 d. Th.
50 Gramm: der letzteren Verbindung wer den mit 250 cm' Phos@phoroxychlorid am Rückflusskühler gekocht. Das überschüssige Phosphorogychlorid wird im Vakuum ab- idestil'liert, der Rückstand auf Eis gegossen, die Lösung über Kohle filtriert und mit Am maniakauf ein pe von 6-7 gebracht, wobei man dafür Sorge trägt, .dass de Temperatur 15 nicht übersteigt.
Der farblose Nieder schlag von 2-(p-Aminobenzol-@sulfo-ylamino-)6.- chlor-pyrimidin wird mit Wasser gut aus,- gewaschen und bei 50-60 getrocknet. Aus beute: 41,7 .g oder 78,1 % d. Th.
40 Gramm dieser Chlorverbindung wer den, in blethylalkoh ol gelöst, bei Gegenwart von auf Kalziumkarbonat niedergeschlage nem Palladium als Katalysator und von Ka-1- ziumoxyd als säurebindendem Mittel mit Wasserstoff reduziert. Hierauf wird der Methylalkohol im Vakuum entfernt und das zurückbleibende 2-(p-Aminobenzol-sulfonyl- amino-)pyrimidin in verdünnter Natronlauge gelöst und mit Säure wieder ausgefällt (p$ 5-7).
So gereinigt schmilzt es bei 254 bis 255 unter Zersetzung. Ausbeute: 33,8 g oder 96 % d. Th.
Die Kondensation des p-Aminobenzol- sulfonyl-.g-uanidins mit der Formylessigsäure lässt sich statt mit rauchender Schwefelsäure auch in Gegenwart von andern wasser entziehenden Mitteln ausführen. Die Reduk tion des 2-(p-Aminobenzol-sulfonylamino-)6- chlor-pyrimidins zum 2-(p-Aminobenzol--sul,- fonyl.amino-)pyrimidin kann auch mit Zink staub in wässeriger Lösung vorgenommen werden.
Process for the preparation of a sulfonamide. Subject of. Invention is a peculiar-es method for the preparation of a sulfonamide, namely the well-known 24p-aminobenzene-.sulfonylamino-) pyrimidine, which forms white crystals with a melting point of 255 to 2.56 and can be used as a remedy against infections.
The process is characterized in that p-aminobenzene-sulfonyl-guanidine is condensed with formylacetic acid to give 2- (p-aminobenzene-sulfonylamino) 6-oxy-pyrimidine, ko @ n-, the latter by means of a halogenating agent into the 2- (p-aminobenzene-sulphonylamino-)
6-halo-pyrimidine transferred and this reduced to 2- (p-aminobenzene-sulfonyl-amino) pyrimidine.
Exemplary embodiment: 44.0 g of p-aminobenzene-s-sulfonyl-guanidine are added slowly and with stirring to a cooled solution of 35.0 g of formylessi: g acid in 280 cm 'of fuming sulfuric acid of 20-30% SO 3 gel. During the entry, the temperature is kept at -5 to 0, then allowed to rise to room temperature, and finally; the mixture is heated on the water bath for a further 1 hour, with constant stirring.
The cooled reaction mixture is poured onto 1000 g of ice, neutralized with 30% ammonia, whereupon the 2- (p-: Amino-benzene-sulfonylamino-) 6-ogy-pyrimidine formed crystallizes out. Yield: 39.8 g or 72.8 d. Th.
50 grams: the latter compound who boiled the reflux condenser with 250 cm 'phosphorus oxychloride. The excess phosphorogychloride is removed by distillation in vacuo, the residue is poured onto ice, the solution is filtered over charcoal and a pe of 6-7 is brought with ammania purchase, taking care that the temperature does not exceed 15.
The colorless precipitate of 2- (p-aminobenzene- @ sulfo-ylamino-) 6-chloro-pyrimidine is thoroughly washed out with water and dried at 50-60. From prey: 41.7 g or 78.1% d. Th.
40 grams of this chlorine compound, dissolved in methyl alcohol, reduced with hydrogen in the presence of palladium precipitated on calcium carbonate as a catalyst and calcium oxide as an acid-binding agent. The methyl alcohol is then removed in vacuo and the 2- (p-aminobenzene-sulfonylamino) pyrimidine that remains is dissolved in dilute sodium hydroxide solution and reprecipitated with acid (p $ 5-7).
Purified in this way, it melts at 254 to 255 with decomposition. Yield: 33.8 g or 96% of theory. Th.
The condensation of p-aminobenzenesulfonyl-g-uanidine with formylacetic acid can be carried out in the presence of other dehydrating agents instead of fuming sulfuric acid. The reduction of 2- (p-aminobenzene-sulfonylamino-) 6- chloro-pyrimidine to 2- (p-aminobenzene-sul-, -phonyl.amino-) pyrimidine can also be carried out with zinc dust in an aqueous solution.
