Verfahren zur Darstellung eines Sulfonamides. Gegenstand der Erfindung ist ein eigen artiges Verfahren zur Darstellung eines Sulfonamides, nämlich des bekannten 2-(p Aminobenzol-sulfonylamino-)4-methyl-pyri midins, welches, weisse Kristalle vom Sehmelzi- punkt 235 bildet und als Heilmittel geben Infektionen verwendbar ist.
Das Verfahren ist dadurch gekennzeich net, :dass man ein p-Acylaminobenzol-sulfonyl- guanidin mit Acetessi.gsäure zum 2-(p-Acyl- aminübenzol -isulfonylamino -) 6-oxy-4-methyl- pyrimidin .kondensiert,
letzteres mittels eines Halogenierungsmiltels in das entsprechende 2-(p-Acylamm.inobenzol-sulfonylamino-) 6-hal.o- gen-4-metbyl-pyrimidin überführt, dieses zum 2-.(p-Acylaminobenzal-sulfonylamino-)
4- methyl-pyrimidin reduziert und daraus durch Abspaltung des Acylrestes das 2-(p-A.mino- benzol - sulfo@nylamino -)4 - methyl - pyrimidsn gewinnt.
<I>Ausführungsbeispiel:</I> In eine auf 0 abgekühlte Suspension von 25,7 g p-Acetylaminobenzol-sulfonyl-:guani- din in 100 cm, rauchender Schwefelsäure von 20-307o 803-Gehalt werden 11g Acetessig- säure eingerührt. Das Gemisch wird einige Stunden auf Zimmertemperatur gehalten.
dann unter Rühren 1 Stunde auf dem Wasser bade erwärmt. Nach dem Erkalten wird es. auf 500g Eis gegossen und mit 30 % igem Ammoniak neutralisiert, worauf das gebildiete 2 - (p - Acetylamin;obenzo@l - sulfonylamino -)6- oxy-4-m@ethyl-pyrimidin auskristallisiert. Sein Sehmelzpun'kt liegt über 300 . Ausbeute:
23,8 :g oder 73,1 % d. Th.
32,3 .g .der letzteren Verbindung werden mit 200 cm' Phasphoroxycblorid 5 Stunden am Rückflusskühler .gekocht. Das überschüs- sige Phosplioroxych@lorid wird im Vakuum abdestilliert, der Rückstand auf 200 g Eis gegossen, die Lösung über Kohle filtriert und mit Ammoniak auf ein p$ von 6-7 gebracht, wobei mau,dafür Sorge träg -t, dass,
die Tempe- ratur 5 nicht übersteigt. Der farblose Nie derschlag von 2-(p-Acetylaminobenzol-sul- fo@nylam.ino-)6-cblor-4-methyl-pyrimdin. wird mit Wasser .gut ausgewaschen und bei 50 ge trocknet.
Ausbeute 24,6 ,g oder 72,3 % d. T.h. 34,1 g dieser Chlorverbi.udung werden, in Methylalkohol gelöst, bei Gegenwart von auf Kalziumkarbonat niederbeschlagenem Palla dium als Katalysator und von Kalziumoxyd als säurebindendem Mittel mit Wasserstoff reduziert.
Hierauf wird der Methylalkohol im Vakuum entfernt und das zurückbleibende - (p - Acetylaminobenzol - sulfonylamino- )4 - methyl-pyrimidin in verdünnter Natronlauge gelöst und mit Säure wieder ausgefällt (pA 5-7). So gereinigt schmilzt es bei 248 . Ausbeute: 30,0 g oder 97,8 % d. Th.
30 g der letzteren Acetylverbindung wer- den zur Verseifung mit 5 Mop 10 % i--er Salz säure 3 Stunden auf dem @'GTasserbad erhitzt. Hierauf wird das 2-(p-Aminobenzol-sulfonyl- amino-)4-methyl-pyrimidin .durch Neutrali seren der Lösung ausgefällt.
Aus Wasser umkristallisiert zeigt es, in Übereinctimmung mit dem .Schrifttum, einen Schmelzpunkt von 235 (u. Zers.). Ausbeute: 24,9 g oder 96.6o' d. Th.
Die Kondensation des p-Acylamiiiobenzo@l- s.ulfonyl-guanidins mit der Acetessigsäiire lässt sich statt mit rauchender Schwefelsäure auch in Gegenwart von andern wasser entziehenden Mitteln. ausführen.
Die Reduk tion des 2 - (p - Acylaminabenzol -sulfonyl - amino-)6-h@a:l.o,en.-4-m,ethyl-pyrimidi.ns zum 2 - (p - Acylaminobenzol - sulfonylamiiro -) 4 - m-ethyl-pyrimidin kann auch mit Ziiilzstaub in wäeseriger Lösung vorgenommen werden.
