Verfabren zur Darstellung von 6-Amino-dihydrocinehonidin. Die Besetzung der 6-Stellung des Chino- linkernes im Chininmolekül gibt dem Gesamt- molel@ül eine entscheidende Wirkung. Der Er satz der Methogygruppe durch eine Hydro- xylgruppe, zum Beispiel im Dihydrocupr ein, macht das Molekül wirkungslos.
Ein in 6-Stellung nicht substituiertes Molekül, wie das Cinchonin und Cinchonidin, verringert die antimalarische Wirkung auf einen Bruch teil gegenüber der Wirkung des Chinins. Es ist deshalb sehr interessant, eine Aminover- bindungeines Chinaalkaloids kennenzulernen, die in 6-Stellung des Chinolinkernes durch eine Aminogruppe substituiert ist.
Obwohl man seit Jahrzehnten damit be schäftigt ist, Aminoderivate des Chinins und des Cinchonins bezw. Cinchonidins herzustel len, war es bisher noch nicht gelungen, die Aminogruppe in 6-Stellung einzuführen. Durch direkte Nitrierung werden 5- und 8- Nitroverbindungen erhalten, die in die ent sprechenden Aminoverbindungen übergeführt werden können. Um zu den 6-Aminokörpern zu gelangen, müssen daher andere Wege ein geschlagen werden.
Es zeigte sich nun, dass der Austausch der 6-Hydroxylgruppe am Hydrocuprein bezw. Hydrocupreidin gegen die Amino- gruppe durch die Verwendung von Ammo- niumsulfit möglich ist, eine Methode, die von der Herstellung des ,B-Naphthylamins aus ss-Naphthol und von 6-Aminochinolin aus 6-Oxychinolin an sich bekannt ist.
Ob das Chininmolekül einen derartigen Eingriff ohne Schädigung, zum Beispiel Aufspaltung in die Toxine, überstehen würde, war ohne weiteres nicht zu erwarten. Besonders über raschend ist aber vor allem, dass die 6-Amino- verbindungen überhaupt abgetrennt werden können, obwohl die Umsetzung bei Tempe raturen von über 100 erfolgt; denn es ist nämlich bekannt, dass Aminoverbindungen des Chinins, zum Beispiel das 5-Aminohydro- chinin, ein Erhitzen auf 100 , wie dies ins besondere beim Sterilisieren von Lösungen üblich ist, nicht aushalten.
Die Aminogruppe wird dabei unter Austausch gegen eine HY- droxylgruppe abgespalten.
Das vorliegende Patent betrifft nun die Herstellung von 6-Amino-dihydrocinchonidin aus Hy drocuprein durch Behandeln der Oxy- verbindung mit Ammoniumsulfit in Gegen wart von wässrigem Ammoniak bei erhöhter Temperatur.
Für die Durchführung des Verfahrens kann man das Hydrocuprein mit Arnmonium- sulfit in Gegenwart von wässrigem Ammo niak auf etwa. 12(l bis 160 erhitzen. Die Ausbeute ist aber eine wesentlich bessere, wenn man in Gegenwart eines wasserlöslichen organischen Lösungsmittels, zum Beispiel eines niedermolekularen Alkohols, arbeitet und die Temperatur niedriger, vornehmliissh zwischen etwa 110 und 120 , wählt.
Es emp fiehlt sich weiter, die Umsetzung nicht bis zu Ende durchzuführen, weil sonst das Ver hältnis zwischen der gewünschten Verbin dung und den bei der Reaktion entstehenden Nebenprodukten sich zugunsten dieser ver schiebt. Bei Einhaltung dieser Massregeln ge lingt es, bis zu 80 % der Oxyverbindung in die Aminoverbindung überzuführen.
Das Verfahrensprodukt stellt eine farb lose oder schwach gelb gefärbte kristalline Verbindung dar, welche, ebenso wie die Aus gangsstoffe, 2 Aquiva.lente Säure bindet und trotz der 3 vorhandenen N-Atome nur mit 1 und 2 Äquivalenten Säure besstän- di,g.,e Salze bilidet. Die Salze mit 1 Äqui valent Säure sind meist echwa@ch,
die mit 2 Äquivalenten Säure stark gelb gefärbt; beide Salzreihen sind wasserlöslich. Die Ver bindung gibt mit Formaldehyd eine ll:ethylol- verbindung. Sie besitzt antiparasitäre Wir kung und soll als Heilmittel und als Zwi schenprodukt für die Herstellung von sol chen verwendet werden.
Beispiel: 300 g Hydrocupreinbase werden in einer Lösung von 900 cm' 40 % iger wässriger Ammoniumsulfitlösung, 600 cm' '-)5%igem Ammoniak und 700 cm' 96 % igem Äthanol in einem eisernen Druckkessel 24 Stunden unter Rühren auf 110 bis 115 erhitzt.
Nach dem Erkalten wird das Ammonsulfit mit Alkohol a a usgefällt und von der Lösung abgetrennt.
Diese wird im Vakuum eingedampft. Der Abdampfrüekstand wird in 880 cm' etwa 20%iger Salzsäure unter Erwärmen gelöst und im Vakuum zur Entfernung der schwef ligen Säure eingeengt. Es hinterbleibt ein harzartiges Stoffgemisch, welches mit heisser Natriumehloridlösung mehrmals ausgezogen wird. Die Auszüge werden eingeengt, vom auskristallisierenden Natriumchlorid abge trennt und in 800 cm' doppelnormale Na tronlauge eingegossen.
Dabei scheidet sich (las gebildete 6-Amino-dihydrocinchonidin zusammen mit geringen Mengen eines Neben produktes aus, während die unveränderte Ausgangsbase und die bei der Umsetzung gebildeten phenolischen Verunreinigungen in der Mutterlauge verbleiben. Der bei dem Ausziehen des harzartigen Stoffgemisches mit Natriumchloridlösung verbliebene Rest wird in Wasser gelöst; diese Lösung wird eben falls in doppeltnormale Natronlauge einge tragen. Es fällt eine weitere Menge der Ver bindung aus, welche von der Mutterlauge ab getrennt wird. Aus den vereinigten alka lischen Mutterlaugen kann durch Ausziehen mit Äther noch eine weitere geringe Menge erhalten werden.
Die mit Natronlauge aus gefällten Fraktionen werden in so viel heisser verdünnter Schwefelsäure gelöst, dass eine gegen Kongopapier neutrale Lösung entsteht. Beim Erkalten kristallisiert ein Sulfat des 6-Amino-dihydrocinchonidins von der For inel Cl"H"Na0 . SO,HZ . 5 HZO aus. Die aus ihm mit Alkali freigesetzte Base kristallisiert. mit einem Mol Kristallwasser und schmilzt bei 137 unter Sintern.
Statt die neue Verbindung aus dem nach der Entfernung der schwefligen Säure hin terbleibenden Reaktionsgemisch mit Natrium- ehloridlösung auszuziehen, kann man auch für ihre Absonderung von der ausserordent lichen Wasserlöslichkeit des in dem Reak tionsgemisch vorhandenen Dihydrochlorids Gebrauch machen. Man geht in diesem Fall so vor, dass man die salzsaure Lösung stufen- weise eindampft. Dabei kristallisieren zu nächst die salzsauren Salze der Ausgangsbase und der bei der Umsetzung gebildeten Nebenprodukte aus.
Nach Abtrennung dieser Salze wird weiter eingeengt, wobei ein in der Hauptsache aus Chlorammonium bestehendes Salzgemisch auskristallisiert. Nach erneutem Abtrennen wird mit Lauge gefällt und der das 6-Amino-dihydrocinchonidin enthaltende Niederschlag, wie oben beschrieben, in Schwe felsäure gelöst und weiterbehandelt.

Direct nitration gives 5- and 8-nitro compounds which can be converted into the corresponding amino compounds. In order to get to the 6-amino bodies, other paths must therefore be taken.
It was now found that the exchange of the 6-hydroxyl group on Hydrocuprein BEZW. Hydrocupreidine against the amino group is possible through the use of ammonium sulfite, a method which is known per se from the production of, B-naphthylamine from ß-naphthol and of 6-aminoquinoline from 6-oxyquinoline.
Whether the quinine molecule would survive such an intervention without damage, for example splitting into toxins, was not to be expected without further ado. The fractions precipitated with sodium hydroxide solution are dissolved in so much hot dilute sulfuric acid that a solution that is neutral to Congo paper is created. On cooling, a sulfate of 6-amino-dihydrocinchonidine crystallizes from the formula Cl "H" Na0. SO, HZ. 5 HZO off. Instead of removing the new compound from the reaction mixture with sodium chloride solution that remains after the removal of the sulphurous acid, use can also be made of the extraordinary water solubility of the dihydrochloride present in the reaction mixture for its separation.
