Verfahren zur Gewinnung von trans-Oestradioldiacetat aus einem Gemisch von Dihydroequilin und trans-Oestradiol. Im Harn trächtiger Stuten sind von Gi- xard und Mitarbeitern neben Oestron noch weitere oestrogene Wirkstoffe, wie das Equi- lin und Equilenin isoliert worden.
Zu Be ginn der Trächtigkeit tritt, wie Girard zeigen konnte, in der Hauptsache Oestron auf, im weiteren Verlauf nimmt die Menge der un- gesättigteren Verbindungen, der Dehydrie- rungsprodukte des Oestrons, zu, so dass gegen Ende der Trächtigkeit das Equilenin in gro sser Menge im Harn vorhanden ist.
Da das Equilenin einen stärker sauren Charakter be sitzt als die andern Verbindungen, lässt es sich verhältnismässig leicht von diesen ab trennen, dagegen bereitet die Trennung von Oestron und Equilin grosse Schwierigkeiten.
Es wurde nun gefunden, dass man aus einem Gemisch von Dihydroequilin und traps Oestra- diod das trans-Oestradiol als Diacetat, wie sieh zeigte, leicht gewinnen kann, wenn man auf ein Gemisch das Dihydroequil.in und trarns- Oestradiol enthält und das zum Beispiel aus einem Oestron-Equilin-Gemisch, z.
B. einem solchen aus Stutenharn, durch eine an sich bekannte Behandlung mit Natrium und Al kohol erhalten worden sein kann, ein Saponin einwirken lässt, den nicht mit dem Saponin in Reaktion getretenen Anteil des Reaktions gemisches von dem mit dem Saponin in Re aktion getretenen Anteil abtrennt, auf letz teren Essigsäureanhydrid einwirken lässt und das so erhaltene trans-Oestradioldiacetat iso liert.
Diese Trennung ist technisch von grösster Bedeutung. Das Eqiülin und seine Derivate sind physiologisch schwächer wirksam als dass an sich bekannte trans-Oestradiol.diacetat. Letzteres findet als Arzneimittel, sowie als Zwischenprodukte zur Herstellung von Arzneimitteln Verwendung.
Beispiel: 5 g einer nach dem Beispiel des Schwei zer Patentes Nr. 213055 hergestellten, vom DihS-droequilin a.bgetrerinten trans-Oestradiol- Digitonin-@'erbindting werden aus einer Ex traktionshülse mit siedendem Essigsäure anhydrid extrahiert.
Nach dein Verdampfen des Essibsäureanhvdrids wird (1,#r Rückstand aus verdünntem Methanol unikristallisiert. wobei etwa 1 g reines trans-Oestr < idioldiacet@:(t vom Schmelzpunkt 127 bis 1\_i6" erhalten wird. Aus den Mutterlaugen lassen sich noch weitere 3lenben isolieren.
Aus der Mutterlaube von der Digitonin- fällung kann man das @ilivcli-oeciuilin ge winnen, indem man diese Mutterlauge mit #Vher versetzt, um ülxrachüssiges Dinitonin auszufällen, filtriert und das Filtrat mehr fach mit Wasser iväseht. Wird die trockene ätherische Lösung verdampft und der Rück stand aus verdünntem Methanol umkristalli siert,
so erhält man reines Dilivdroequilin vom Smp. <B>175</B> .
Anstatt, wie im Beispiel des Patentes Nr. 213055 beschrieben, die mit Digitonin in wässriger, 90%iger alkoholischer Lösung durchzuführen, kann man auch Al kohol anderer Konzentraiion verwenden oder auch andere Alkohole oder sogar andere wäss- izge, mit )Vasser mischbare organische Lö sungsmittel.
Man kann auch ch(s Saponin in fein verteilter Form der Reaktionsmischung zusetzen, ohne es vorher aufzulösen. Selbst verständlich ist es in diesem Fall notwendig, die Reaktionsmischung kräftig, zu rühren. Die besten Resultate werden jedoch erhalten, wenn man entsprechend dem oben genannten Beispiel eine Lösung des Ausgangsmaterials verwendet und das Saponin in alkoholischer Lösung zusetzt.
Anstatt das Saponid durch längeres Stehenlassen ans dir Reaktions- mischung auszufällen, kann man auch die Lösung teilweise oder v ollstä ndi- zur Trockne eindampfen.
wobei im ersteren Fall die Ausfällung des Saponids erleichtert wird, während im letzteren Fall der trockne Ver- dampfungsrückstand in das Saponid und die andern Bestandteile durch Extraktion mit einem Lösungsmittel getrennt wird, welches fähig ist, lediglich die andern Bestandteile der Reaktionsmischung zu extrahieren, nicht:
aber das Saponid. Hierzu eignet sich zum Beispiel Äther, doch kann man auch andere geeignete Lösungsmittel für den gleichen Zweck benutzen.
Eine weitere Methode der Abtrennung des Saponi(is aus der Reaktionsmischung i)esteht in der Ausfällung des Saponids durch Ver dünnung der Reaktionsmischung mit einer geeigneten Flüssigkeit, die fähig ist, das Sa- ponid zu fällen. Wasser eignet sich zum Bei spiel als Fällmittel hierfür.
Dibitonin hat sich als besonders hervor ragendes Mittel znr Abtrennung des trans- Oestradiols erwiesen, doch kann man auch andere Saponine, wie z. B. Solanin, Cyclamin, Diosein und dergleichen hierfür benutzen.
Die Reduktion des Oestron-Equilin-CTe- misches zii den entsprechenden Diolen kann auch auf andere Art und Weise bewirkt erden als mit Natrium und Alkohol:

This separation is of the greatest technical importance. The Eqiülin and its derivatives are physiologically less effective than that known per se trans-Oestradiol.diacetat. The latter is used as a drug and as an intermediate product in the manufacture of drugs.
Example: 5 g of a trans-estradiol-digitonin - @ 'erbindting prepared according to the example of the Swiss patent no. 213055 and separated from DihS-droequilin a.bindting are extracted from an extraction sleeve with boiling acetic anhydride.
After evaporation of the acetic acid anhydride, (1, # r residue is unicrystallized from dilute methanol. Approximately 1 g of pure trans-ester <idioldiacet @ :( t with a melting point of 127 to 1 \ _i6 "is obtained. Further 3 levs can be obtained from the mother liquors isolate.
The @ilivcli-oeciuilin can be obtained from the mother's arbor from the digitonin precipitation by adding #Vher to this mother liquor in order to precipitate ulcerous dinitonin, filtering it and washing the filtrate several times with water. If the dry ethereal solution is evaporated and the residue is recrystallized from dilute methanol,
pure dilivdroequilin with melting point <B> 175 </B> is thus obtained.
Instead of, as described in the example of patent no. 213055, carry out the digitonin in an aqueous 90% alcoholic solution, one can also use alcohol of other concentration or other alcohols or even other aqueous organic solutions that are miscible with water solvent.
It is also possible to add finely divided ch (s saponin to the reaction mixture without dissolving it beforehand. Of course, in this case it is necessary to stir the reaction mixture vigorously. However, the best results are obtained if one follows the example given above a solution of the starting material is used and the saponin is added in alcoholic solution.

Another method of separating the saponide (is from the reaction mixture i) consists in the precipitation of the saponide by diluting the reaction mixture with a suitable liquid which is capable of precipitating the saponide. For example, water is suitable as a precipitant for this.
Dibitonin has proven to be a particularly excellent means of separating trans-estradiol, but other saponins, such as e.g. B. use solanine, cyclamine, diosein and the like for this purpose.
The reduction of the oestron-equilin-C mixture to the corresponding diols can also be effected in other ways than with sodium and alcohol:
All such reduction methods are suitable for this, in which only the ketone groups of the starting materials are reduced to the secondary alcohol groups, but in which the double bonds in the molecule remain unaffected.
