Verfahren zur Darstellung eines Barbitursäureabkömmlings. Die C,C-Isopropy 1-allyl-N-m-ethyl-barbi- tursäure -ist bereits als Narcotieum und Schlafmittel empfohlen worden.
Es wurde nun gefunden, dass die Bar- bitursäure, welche an Stelle des Isopropyl- restes den sek. Amylrest enthält, in ihrer Wirkung der erwähnten Säure überraschend ätark überlegen ist.
Während die den Iso- propylrest enthaltende Säure bei intravenöser Darreichung am Kaninchen eine minimal hypnotische Dosis (Bauch-Seitenlage) von <B>0,015</B> g und eine minimalnarkotische Dosis (Rückenlage) von 0,03 g pro kg Tier besitzt, beträgt die minimalhypnotische Dosis für die 5-Propylmethylcarbinyl-5-allyl-N-me- thyl-barbitursäure (sek. 5-Amyl-5-allyl-N-me- thyl-barbitursäure) nur 0,004 g pro kg und die minimalnarkotische Dosis nur 0,0125 g bei praktisch gleicher hypnotischer und nar kotischer Breite.
Infolge der hohen Wirk samkeit der Säure, welche gestattet, das zu injizierende Lösungsvolumen auf ein Mini- mum zu reduzieren, ist es möglich, mit ihrer Hilfe auch eine Grosstiernarkose unter Be nutzung der in der Allgemeinpraxis üblichen Methoden durchzuführen. Auch bei peroraler Verabreichung erweist sich die neue Säure der Isopropyl-allyl-N-methyl-barbitursäure als stark überlegen.
So zeigt sie zum Beispiel, per os verabreicht, beim Kaninchen bereits in der Dosis von 0,05 g/kg eine hypnotische Wirkung, während die eben wirksame Dosis der Isopropylallyl - N - methyl-barbitursäure bei gleicher Verabreichung 0,25 g/kg ist.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung der 5-Propyl methyl-carbinyl-5-allyl-N-methyl-barbitur- säure, welches dadurch gekennzeichnet ist, dass man eine Iminogruppe der ,5-Propyl- methylcarbinyl-5-allyl-baxbitursäure methy- liert.
Die erhaltene Säure lässt sich in ihre Al kali- oder Erdalkalisalze überführen. Diese stellen kristalline Pulver dar. <I>Beispiel:</I> 66,2 Gewichtsteile 5-Propylmethylcar- binyl-5-allylbarbitursäure (F. 94 bis 96') werden in 826 Raumteilen einer 4 öigen Na tronlauge gelöst und unter allmählichem Zu satz von 43,2 Gewichtsteilen Dimethvlsulfat Stunden unter Kühlung lebhaft gerührt.
Die alkalische Lösung wird dann von dem im Laufe der Umsetzung abgeschiedenen ge ringfügigen Beiprodukt abgetrennt und unter Kühlung vorsichtig angesäuert. Hierbei schei det sich die 5-Propylmethylecirbinyl-5-allyl- N-methyl-barbitursäure in öliger Form ab. Sie wird von der Fällauge getrennt. und mit Äther aufgenommen. die ätherische Lösung mit Wasser gewaschen und über Chlor calcium getrocknet.
Nach Abdunsten des .Äthers und Destillation des dabei erhaltenen Rückstandes (Kp. 172 bis<B>176</B> bei 2 mm) wird die 5-Propylmethylca.rbinyl-5-allyl-N- methyl-barbitursäure als klares, farbloses, in üblichen organischen Lösungsmitteln leicht lösliches, zähes 01 gewonnen.
Durch Zusammengeben einer alkoholi schen Lösung von 100 Gewichtsteilen der Propylmethylcarbinyl - allyl - N - methyl - bar - bitursäure mit einer aus 9,2 Gewichtsteilen Natrium und absolutem Alkohol hergestell ten Athylatlösung und anschliessendem Ab- dunsten des Alkohols unter vermindertem Druck erhält man das kristalline Natrium salz des neuen Barbitursäureabkömmlings. Es ist im Verhältnis 1 : 2 in Wasser löslich.
Aus einer solchen Lösung kann durch Fällung mit einer 65 % igen wässerigen Caleiumchlo- ridlösung das propylmethylcarbinyl-allyl-N- methyl-barbitursaure Calcium gewonnen wer den.
