Description
Verfahren zur Herstellung eines Insulinpräparates. Gegentand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines Insulin- präparates. Das Verfahren ist dadurch ge- k .ennzeichnet,
da ein Insulinsalz und Percin in saurer Lösung miteinander in Reaktion ge bracht wenden, wodurch Percininsulinat ge bildet wind.
Bei .der Durchführung der Reaktion ist es vorteilhaft, die Gewiehtsmenge .des Protamins so zu bemessen, @da-ss sie etwa '/,> bis '/1o der Gewichtsmenge,des Insulinsalzes beträgt.
Zu bemerken ist, @dass die Reaktion, wie sich zeigte, bereits bei gewöhnlicher Tem peratur stattfindet und sehr schnell vor sich geht. Dabei wird der saure Rest des Insulin <B>2</B> abgespalten.
<I>Beispiel:</I> 10 Gewichtsteile Insulin und 1,2 Ge wichtsteile Percin werden in saurer Lösung miteinander in Reaktion gebracht.
Der Säure grad der Lösung wird grösser gewählt als einem pH-Wert von ca. 4 entspricht, und die Reaktion wird bei gewöhnlicher Temperatur ausgeführt.
Infolge des genannten Säure- graJes der Lösung verbleibt die bei der Re aktion gebildete neue chemische Verbindung (Percininsulinat) in Lösung, sie kann aber durch Erhölhung .des pH-Wertes ausgefällt werden. Das Produkt ist ein in Wasser schwer lösliches, weisses und amorphes:
Pul ver, welches. sich beim Erwärmen bei ca. <B>100</B> bis 120' C zu zersetzen anfängt.
Die gebildete chemische Verbindung Prot- amininsulinat ist im p,1-Wertgebiet 7 bis 7,5 sehr sohwer löslich, während im Gegensatz hierzu beide Reaktionskomponenten bei die sen pH-Werten leicht löslich sind.
Die ganze Reakkonsmasse besteht nach Beendigung der .rasoh verlaufenden Reaktion aus einer Lösung von Protamininsulinat. Dieses- Präparat wird zweckmässig in Ampul len eingefüllt, .die je einen einer gewissen An zahl internationaler Insulineinheiten entspre chenden Inhalt aufnehmen.
Die Lösung sollte nicht als solche, sondern erst nach Fällung durch Zusatz eines alkalisierenden Stoffes be nutzt werden.
Die durch Benutzung eines solchen In sulinpräparates entstandene Wirkung in dem menschlichen Organismus wird bedeutend langsamer verlaufen und infolgedessen auch bedeutend milder sein als es. bei Verwendung von Insulinhydrochlorid der Fall ist, und dieser langsamere Wirkungsverlauf bewirkt zugleich, dass, die Wiederholung einer Ein spritzung erst nach einer verhältnismässig langen Zeitperiode stattzufinden braucht.

As a result of the acidity of the solution mentioned, the new chemical compound formed during the reaction (percine insulinate) remains in the solution, but it can be precipitated by increasing the pH. The product is a poorly water-soluble, white and amorphous one:
Powder, which. begins to decompose when heated at approx. <B> 100 </B> to 120 ° C.
The chemical compound formed, protamine insulinate, is very soluble in the p.1 value range from 7 to 7.5, while in contrast to this, both reaction components are easily soluble at these pH values.
After the end of the reaction, the whole reaction mass consists of a solution of protamine insulinate. This preparation is expediently filled into ampoules, each of which contains a certain number of international insulin units.
The solution should not be used as such, but only after precipitation by adding an alkalizing substance.
The effect in the human organism resulting from the use of such an insulin preparation will proceed considerably more slowly and consequently also be considerably milder than it. is the case when using insulin hydrochloride, and this slower course of action also means that the repetition of an injection need only take place after a comparatively long period of time.
