Description
Verfahren zur Darstellung eines N-substituierten Amides einer Pyridincarbonsäure. Es wurde gefunden, dass man zu einem N--substituierten Amid einer Pyridinearbon- säure dadurch gelangen kann, dass man auf Pyridin-3-carbonsäure Methylanilin bei Ge genwart von Phosphorpentoxyd, gegebenen falls unter Anwendung eines geeigneten Ver- dünnungs- bezw. Lösungsmittels einwirken lässt.
Das aus Pyridin-3-carbonsäur e (Nikotin säure) auf diese Weise erhaltene Konden sationsprodukt stellt ein dickflüssiges Öl dar, das im Vakuum bei 206 bis 207 /18 mm siedet. Es ist leicht löslich in Alkohol und Äther, kaum löslich dagegen in Wasser.
Die neue Verbindung soll für therapeu tische Zwecke Verwendung finden. Beispiel: Einer Lösung von 36 Teilen Nikotinsäure in 6-1 Teilen Methylanilin werden unter Rüh ren und geringer Kühlung etwa 26 Teile Phosphorpentoxyd, das vorher zweckmässig mit etwa der bleichen Menge Quarzpulver oder feinem Sand vermischt wurde, allmäh lich zugesetzt. Nachdem die Selbsterwär mung nachgelassen hat, wird das Gemisch mehrere Stunden im Ölbad auf etwa 180 erhitzt.
Das Reaktionsgemisch wird alsdann in wenig Wasser gelöst und mit starker Natron lauge im Überschuss versetzt. Das entstan dene Pyridin-3-carbonsäuremethylanilid wird in Äther aufgenommen und durch Vakuum destillation gereinigt.

The reaction mixture is then dissolved in a little water and an excess of strong sodium hydroxide solution is added. The resulting pyridine-3-carboxylic acid methylanilide is taken up in ether and purified by vacuum distillation.
