Verfahren zur Herstellung eines Derivates des 3,5-Dioxo-pyrazolidins. Gegenstand vorliegender Erfindung sind Verfahren zur Herstellung neuer Derivate des 3,5-Dioxo-pyrazolidins, welche der allge meinen Formel R-CH-C -= 0 I 0 = C@N-Aryl (I) N I Aryl oder ihrer tautomeren Form entsprechen, so wie ihrer Salze mit anorganischen und orga nischen Basen.
In dieser Formel und im fol genden bedeuten R ein Kohlenstoffgerüst mit 2 bis 10 C-Atomen, welches dureh ein primäres oder sekundäres C-Atom mit dem Pyrazolidinkern verbunden ist, insbesondere einen gesättigten oder ungesättigten aliphatischen oder cyelo- aliphatisehen oder einen aryl-substituierteri aliphatisehen Kohlenwasserstoffrest,
und Aryl einen unsubstituierten oder einen durch Halogenatome, Alkyl- und/oder Alkoxy- gruppen substituierten Phenylrest, wobei die Alkyl- und Alkoxygruppen 1. bis 3 C Atome enthalten.
Diese 3,5-Dioxo-pyrazolidine können her gestellt werden, indem man ein reaktions fähiges Derivat einer monosubstituierten Ma- lonsäure der Formel R-CH (C0-X). (1I) worin X den abspaltbaren Rest des Säure derivates, wie z. B.
Halo en S<U>äure</U>halo enid eine Alkoxygruppe (Säureester) usw., bedeu tet, mit einem 1,2-Diaryl-hydrazin der Formel Aryl-NH-N.'11-Ary 1 (III ) zweckmässig in Gegenwart eines Kondensa tionsmittels bzw. eines säurebindenden Mit tels, wie nachstehend näher eeläutert wird, zur Reaktion bringt.
Eine bevorzugte Ausführungsform besteht darin, einen Dialkylester, vor allem den Di- äth@lester, einer monosubstituierten Malon- r säure mit einem Diary lhydrazin in Gegen wart eines Metallalkoholats, z. B.

Diese Ausführungsformen bestehen darin, dass man statt eines Diesters der Malonsäure II andere reaktionsfähige Derivate derselben verwendet., wie z. B. ein Dihalogenid, ein Esterchlorid, ein Esteramid usw.
Die Kondensation des Diarylhydrazins III mit einem Halogenid der Malonsäure II, bei spielsweise dem Dichlorid, erfolgt zweck mässig in einem gegen das Säurehalogenid indifferenten Lösungsmittel, wie Äther oder Benzol, und, um die Umlagerung des Diaryl- hydrazins durch den entstehenden Halogen wasserstoff möglichst zurückzudrängen, zweck mässig bei Gegenwart eines säurebindenden Mittels, bevorzugterweise in Gegenwart einer tertiären organischen Base,
wie Pyridin oder Dimethylanilin. Die Umsetzung erfolgt in dieseln Fall schon bei niedrigen Temperatu ren, c.veckmässig üi der Nähe von 0 .
Besonders bei Verwendung von unsym- inetrischen reaktionsfähigen Derivaten der Malonsäure II gelingt es, die Kondensation stufenweise auszuführen.

Zur Herstellung der lösliehen Salze kann das freie Pyrazolidin der Formel I mit der äquimolekularen Menge einer das jewünsehte Kation enthaltenden Base, z. B. mit wässriger Natronlauge, Äthylendiamin, Kalilauge oder Natrirrmcarbonatlösung oder mit alkoholi scher Natronlauge, behandelt werden, am besten unter Rühren oder Schütteln.
Verwen det man eine wä.ssrige Lange, so kann man auch einen Überschuss des Pyrazolidins ver wenden, der, wenn die Lösung auf Phenol phthalein neutral geworden ist., durch Filtra tion entfernt werden kann.
Das feste Salz kann gewünsehtenfalls durch Eindampfen der filtrierten Lösung oder auch durch Zusatz eines Lösungsmit tels, in welchem das Salz nicht löslich ist, z. B. durch Zugabe von Äther zur alkoholi- sehen Lösung des Natriumsalzes, erhalten werden.
Es ist jedoch auch möglich, bei alkalischen Kondensationsansätzen, z. B. bei Verwendung von Natriumäthylat als Kondensationsmittel, die entsprechenden Salze direkt zu erhalten.
1)ie Alkalisalze werden als farblose, meist weitgehend luftbeständige Pulver erhalten, die in Wasser und Alkohol leicht löslich sind und deren Lösungen Plrenolphthalein nicht röten.
Schwerer lösliche Salze können auch durch doppelte Umsetzung eines Alkali- oder Ammo- niumsalzes des Pyrazolidins der Formel I z. B. mit einem löslichen Erdalkali- bzw. Schwer metallsalz, wie Caleiumehlorid oder Silber nitrat, erhalten werden. Die erhaltene neue Verbindung, das 1,2- Diplrenyl-3,5-dioxo-4-n-brrtyl-pyrazolidin, kri stallisiert in farblosen Nadeln vom Schmelz- punkt 105". Sie soll therapeutische Verwen dung finden.
Beispiel <I>1:</I> Man-löst 7,6 Teile Natrium in<B>190</B> Volurn- teilen absolutem Alkohol, gibt 65 Teile n-Bu- tyl-malorrsäure-diätlryiester und 55 Teile Hy- drazobenzol zrr, destilliert den Alkohol lang sam ab und erhitzt dann 12 Stunden bei einer Badtemper atur von 150 , am Schluss im Vakuum, bis kein Alkohol mehr abdestilliert.
Man löst in Wasser, klärt mit etwas Kohle und versetzt langsam mit 15 /oiger Salzsäure bis zur sauren Reaktion auf Kongopapier. Das 1,2-Diplrerryl-3,5-dioxo-4-n-butyl-pyrazol- idin fällt dabei als rasch kristallisierendes Öl ans. Aus Alkohöl kristallisiert es in farb losen Nadeln vom Schmelzpunkt l05 .
Das gleiche 1,2-Diplrenyl-3,5-dioxo-l-n- butyl-pyrazolidin erhält man auch, wenn man das 1-Tydrazobenzol durch 68 Teile N-Acetyl- lr\#drazobenzol ersetzt und im übrigen gleich verfährt. Bei dieser Ausführungsform bilden sieh etwas weniger gefärbte Nebenprodukte. Die Ausbeute ist ebenfalls gut.
<I>Beispiel 2:</I> Zu einer Mischung von 50 Teilen absolu tem Pyridin und 200 Volumteilen absolutem Äther tropft man bei etwa 0 19 Teile n-Bu- tyl-malonsäure-dichlorid vom Siedepunkt 8511 bei 14 mm Druck, gibt dann 18 Teile Hy- drazobenzol, gelöst in 100 Volumteilen Äther, hinzu und rührt zwei Stunden bei Zimmer temperatur.
Die Lösung wird zunächst zur Entfernung des Pyridins mit 2n-Salzsäure ausgeschüttelt, hierauf mit 2n-Natriumcarbo- natlösung.
