Verfahren zur Gewinnung von Morphin und Codein. Immer und immer wieder hat das Pro blem, die Opiumalkaloide statt aus dem ein getrockneten Milchsaft des Mohns unmittel bar aus der unreifen oder reifen Mohnpflanze zu gewinnen, die Forscher beschäftigt. Es sind auch bereits auf einige Verfahren zur Herstellung der Opiumalkaloide unmittelbar aus der Pflanze Patente ausgefolgt worden (Deutsche Patentschriften Nr. 232126, 524946, franz. Patentschrift Nr. 748308).
Wenn die fabrikmässige Morphingewin- nung unmittelbar aus der Pflanze bis jetzt nur vereinzelt durchgeführt wurde, erklärt sich dies aus den grossen Schwierigkeiten, welche die Verarbeitung der sehr geringe Mengen Alkaloid, dafür aber sehr grosse Men gen Verunreinigungen enthaltenden Auszüge verursacht. Bisher hat man die sehr ver dünnten Alkaloidlösungen stets eingedampft und die Verunreinigungen durch Ausfällung beseitigt. Die Umkehrung :
dieser Arbeits weise, nämlich die Extraktion der Alkaloide aus den verdünnten Lösungen, erschien un durchführbar und wurde noch von keiner Seite versucht, da man lästige Emulsions- bildungen und einen gleichzeitigen Übergang der Verunreinigungen mit .den Alkaloiden in das zur Extraktion verwendete organische Lösungsmittel annehmen musste.
Es wurde nun gefunden, dass die Extrak- tion der Alkaloide aus dem in .gewaltigem Überschuss vorhandenen wässerigen Lösungs mittel und die Abtrennung von den grossen Mengen Verunreinigungen mit geeigneten organischen Lösungsmitteln in technisch sehr ökonomischer und leicht durchführbarer 'Weise, gelingt, und dass, dabei Ausbeuten er zielt werden, welche diejenigen, die bisher mit technischen Verfahren erhalten worden sind, um ein Mehrfaches übertreffen.
Die wässerigen, wässerig-sauren oder wässerig-alkalischen Auszüge von unreifen oder reifen Mohnpflanzen oder Teilender selben werden :durch Zugabe von schwachen Alkalien, wie Natrium- oder Ammonium karbonat, oder von Säuren auf ein pH von ungefähr 9 eingestellt. Dadurch werden die als Salze in :der Lösung vorhandenen Alka- loide in die freien Basen übergeführt.
Aus der so vorbereiteten Lösung werden die Al kaloide mit organischen Lösungsmitteln, die Morphin und Codein lösen und leicht wieder freigeben, wie Chloroform, Butanol, Amyl- alkohol und dergleichen oder durch Mischun gen solcher Lösungsmittel extrahiert. Mor phin und Codein werden von den organischen Lösungsmitteln aufgenommen, ohne dass da bei eine lästige Emulsionsbildung entsteht oder wesentliche Mengen von Verunreinigun gen mitgehen.
Aus dem organischen Lösungs mittel können die Alkaloide in üblicher Weise, zum Beispiel durch Überführung in konzentrierte wässerige Alkaloidsalzlösung durch Ausrühren mit verdünnten Säuren und Fällung der Alkaloide leicht gewonnen wer den. Nach,dem Entzug der Alkaloide ist das organische Lösungsmittel nicht wesentlich verunreinigt, es kann ohne Destillation zur weiteren Extraktion Verwendung finden.
Da durch ist es möglich, die Alkaloide, trotzdem sie in sehr grosser Verdünnung vorliegen und trotzdem der Verteilungskoeffizient für Mor phin zwischen Wasser und organischem Lö- sungsmittel allgemein nicht besonders günstig ist, mit kleinen Mengen organischen Lösungs mitteln vollständig und in sehr wirtschaft licher Weise zu extrahieren.
Die viel höheren Ausbeuten, die mit dem neuen Verfahren erzielt werden, erklären sich dadurch, dass bei den bisher gebräuch lichen Methoden .durch das lange Eindamp fen und besonders durch Okklusion und Ad sorption bei der Fällung der Verunreinigun gen grosse Mengen von Alkaloiden verloren gehen, während beim vorliegenden Verfahren diese Verlustquellen vermieden werden.
<I>Beispiel Z: .</I>
6 Tonnen zerkleinertes, lufttrockenes Mohnstroh, das zu einem guten Teil aus Köp fen besteht, werden in .6 Ansätzen zu je einer Tonne mit Wasser im Gegenstrom, also nach dem Anreicherungsprinzip, ausgezogen. Die ungefähr 20 m3 betragende Lösung wird nach Einstellung mit Soda auf ein p$ von unge fähr 9 in einer Flüssigkeitsextraktionskolonne mit einer Mischung gleicher Volumen Bu- tanol und Benzol extrahiert.
Das aus der Kolonne abfliessende, mit .den Alkaloiden be ladene Extraktionsmittel leitet man durch wässerige Säure, welche die Alkoloide auf- nimmt, worauf die Butanolbenzolmischung zur Fortsetzung der Extraktion der wäs serigen Lösung benützt wird.
Die vollstän dige Überführung der Alkaloide aus dem wässerigen Pflanzenextrakt in die Säure lösung gelingt in weniger als 48 Stunden. Die saure Alkaloidlösung wird bei 50 mit Soda pulver auf ein p$ von ungefähr 9 gebracht und das dabei ausfallende Morphin vom Lö sungsmittel abgetrennt. Dem Filtrat entzieht man das Codein mit Benzol und reinigt es über das Sulfat.
Es werden auf diese Weise 34,6 kg trockenes Rohmorphin mit einem Ge halt von 83,87 % reiner wasserfreier Morphin base entsprechend 4,8.1 /oo der lufttrockenen Mohnpflanzen, sowie 1,2 kg wasserfreie Co deinbase erhalten. <I>Beispiel 2:</I> 1000 kg trockene, zerkleinerte Mohn pflanzen werden mit 7000 Liter Wasser ver rührt. Nach einigen Stunden setzt man 100 kg gelöschten Kalk zu und sorgt für gründliche Mischung.
Die Brühe reagiert sehr stark phenolphthalein-alkalisch. Es folgt die Filtration und anschliessend das Aus waschen .des Rückstandes, wobei die Filtrate getrennt aufgefangen und zum Ansetzen und Auswaschen einer zweiten Menge von 1000 kg verwendet werden.
Die getrennt gesammelten Filtrate werden, besonders wenn sie nicht so fort zur Aufarbeitung gelangen, mit ver dünnter Salz- oder Schwefelsäure auf Lack mus neutral oder schwach sauer gestellt. 6 solche 1000 kg-Ansätze geben eine Flüssig keitsmenge von etwa 25 m?. Durch Zusatz von wenig Salzsäure wird,die Alkaloidlösung schwach angesäuert und dann mit Sodapulver das pH der Lösung auf etwa 9 eingestellt.
Nach dem Abfiltrieren einer geringfügigen Calciumkarbonatausscheidung werden die Al kaloide durch Behandlung mit einer Mi schung von etwa je 1000 Liter Butanol und Benzol herausgelöst und wie in Beispiel 1 ge wonnen.
<I>Beispiel 3:</I> 10 kg Mohnköpfe werden nach dem im Deutschen Arzneibuch, 3. Ausgabe, Seite 277, beschriebenen Verfahren ausgezogen und die im Filtrate vorhandenen geringen Alkohol mengen im Vakuum abgedampft. Das pn der wässerigen Lösung wird hernach durch Zusatz von Ammoniumkarbonat auf etwa 9 eingestellt und die Morphin- und Codeinbase mit Amylalkohol ausgezogen. Die Überfüh rung des Morphins und Codeins aus .dem organischen Lösungsmittel in verdünnte wäs serige Säurelösung und die Isolierung der reinen Alkaloide geschieht in bekannter Weise.
<I>Beispiel</I> .4: 1000 kg zerkleinerte, naturfeuchte, un reife Mohnpflanzen werden mit angesäuertem Wasser erschöpfend ausgezogen und die etwa 9000 Liter betragende wässerige Lösung im Vakuum auf 6000 Litereingeengt. Die wei tere Verarbeitung des wässerigen alkaloid- haltigen Auszuges erfolgt wie in Beispiel 3.
Process for the production of morphine and codeine. Time and again, researchers have been concerned with the problem of extracting opium alkaloids directly from the immature or ripe poppy seed instead of the dried milky sap of the poppy. Patents have also already been issued for some processes for the production of opium alkaloids directly from the plant (German patent specifications No. 232126, 524946, French patent specification No. 748308).
If the factory production of morphine directly from the plant has so far only been carried out in isolated cases, this is explained by the great difficulties caused by the processing of the extracts, which contain very small amounts of alkaloid, but very large amounts of impurities. So far, the very dilute alkaloid solutions have always been evaporated and the impurities removed by precipitation. The reverse:
This way of working, namely the extraction of the alkaloids from the dilute solutions, seemed impracticable and has not yet been attempted, since annoying emulsion formations and a simultaneous transfer of the impurities with the alkaloids into the organic solvent used for the extraction had to be assumed .
It has now been found that the extraction of the alkaloids from the aqueous solvent present in .gewaltigem excess and the separation of the large amounts of impurities with suitable organic solvents succeeds in a technically very economical and easily feasible manner, and that in doing so Yields he aims to be which exceed those that have been obtained by technical processes many times over.
The aqueous, aqueous-acidic or aqueous-alkaline extracts of immature or ripe poppy plants or parts of the same are: adjusted to a pH of approximately 9 by adding weak alkalis, such as sodium or ammonium carbonate, or acids. As a result, the alkaloids present as salts in the solution are converted into the free bases.
From the solution prepared in this way, the alkaloids are extracted with organic solvents that dissolve morphine and codeine and easily release them again, such as chloroform, butanol, amyl alcohol and the like or by means of mixtures of such solvents. Morphine and codeine are absorbed by the organic solvents without causing an annoying emulsion formation or significant amounts of impurities.
The alkaloids can easily be obtained from the organic solvent in the usual way, for example by converting them into concentrated aqueous alkaloid salt solution by stirring with dilute acids and precipitating the alkaloids. After removing the alkaloids, the organic solvent is not significantly contaminated; it can be used for further extraction without distillation.
As a result, it is possible to use the alkaloids completely and in a very economical manner with small amounts of organic solvents, despite the fact that they are very dilute and the distribution coefficient for morphine between water and organic solvent is generally not particularly favorable extract.
The much higher yields that can be achieved with the new process can be explained by the fact that with the methods commonly used up to now, large amounts of alkaloids are lost due to the long evaporation and especially due to occlusion and adsorption during the precipitation of the impurities. while in the present method these sources of loss are avoided.
<I> Example Z:. </I>
6 tons of shredded, air-dry poppy straw, a good part of which consists of heads, are extracted in .6 batches of one ton each with water in counter-current, i.e. according to the enrichment principle. The solution, which amounts to about 20 m3, is extracted after adjusting with soda to a p $ of approximately 9 in a liquid extraction column with a mixture of equal volumes of butanol and benzene.
The extractant which flows out of the column and is loaded with the alkaloids is passed through aqueous acid which absorbs the alkoloids, whereupon the butanolbenzene mixture is used to continue the extraction of the aqueous solution.
The complete transfer of the alkaloids from the aqueous plant extract into the acid solution takes less than 48 hours. The acidic alkaloid solution is brought to a p $ of approximately 9 with soda powder at 50 and the morphine which precipitates is separated from the solvent. The codeine is removed from the filtrate with benzene and purified using the sulfate.
In this way, 34.6 kg of dry raw morphine with a Ge content of 83.87% pure anhydrous morphine base, corresponding to 4.8.1 / oo of the air-dry poppy plants, and 1.2 kg of anhydrous co-deinbase are obtained. <I> Example 2: </I> 1000 kg of dry, chopped up poppy seeds are mixed with 7000 liters of water. After a few hours, add 100 kg of slaked lime and ensure thorough mixing.
The broth has a very strong phenolphthalein-alkaline reaction. This is followed by filtration and then washing out of the residue, the filtrates being collected separately and used for preparing and washing out a second amount of 1000 kg.
The separately collected filtrates are, especially if they cannot be processed immediately, made neutral or slightly acidic with dilute hydrochloric or sulfuric acid on lacquer. 6 such 1000 kg approaches give a liquid amount of about 25 m ?. The alkaloid solution is weakly acidified by adding a little hydrochloric acid and then the pH of the solution is adjusted to about 9 with soda powder.
After a slight calcium carbonate excretion has been filtered off, the alkaloids are dissolved out by treatment with a mixture of about 1000 liters of butanol and benzene each and, as in Example 1, are obtained.
<I> Example 3: </I> 10 kg poppy heads are extracted according to the method described in the German Pharmacopoeia, 3rd edition, page 277, and the small amounts of alcohol present in the filtrate are evaporated in vacuo. The pn of the aqueous solution is then adjusted to about 9 by adding ammonium carbonate and the morphine and codeine bases are extracted with amyl alcohol. The transfer of the morphine and codeine from .dem organic solvent in dilute aqueous acid solution and the isolation of the pure alkaloids is done in a known manner.
<I> Example </I> .4: 1000 kg of chopped, naturally moist, unripe poppy plants are exhausted with acidified water and the approximately 9000 liters of aqueous solution is reduced to 6000 liters in a vacuum. The further processing of the aqueous alkaloid-containing extract is carried out as in Example 3.
