Verfahren zur Darstellung Fon unsymmetrischen Thioharnstoffen. Nach De Clairmont (Berichte 9, Seite 446 und Berichte 10, Seite 493) werden Mono arylthioharnstoffe aus den entsprechenden Arylaminochlorhydraten durch Umsatz der wässerigen Lösungen mit Alkalirhodaniden erhalten.
Die Reaktion verläuft nach folgen dem Schema:

Dieses Verfahren ist jedoch für technische Zwecke wenig geeignet, einmal, da die Um setzung je nach der Art des angewandten Amins nicht einheitlich, sondern unter Bil dung von Nebenprodukten, insbesondere von symmetrischen Diarylthioharnstoffen (siehe Bertram, Berichte 25, Seite 48) verläuft, vor allem aber, da apparative Schwierigkeiten auftreten.
Es ist nämlich in den meisten Fällen nötig, das Reaktionsgemisch zwecks Umlagerung der als Zwischenprodukte ge- bildeten rhodanwasserstoffsauren Salze zur Trockne zu verdampfen und noch nachträg- lich in trockenem Zustande zu erhitzen, wo durch die Produkte in einer zur Weiterver arbeitung sehr ungünstigen Form anfallen und die Apparaturen sehr stark angegriffen werden.
Es wurde nun gefunden, daB man un symmetrische Thioharnstoffe der allgemeinen Formel

worin R = Aryl ist und an der durch den un- bezeichneten Strich gekennzeichneten freien Valenz Wasserstoff oder ein einwertiger Substituent vorhanden ist, in glatter Reaktion in vorzüglicher Ausbeute und Reinheit er halten kann, wenn man aromatische Amine der allgemeinen Formel:

worin R und der unbezeichnete Strich die oben angegebene Bedeutung haben, in Form ihrer mineralsauren Salze mit Alkalirhoda- niden in Gegenwart von inerten organischen Verdünnungsmitteln erhitzt.
Zur Durchführung der' Reaktion ist es nicht erforderlich, von mineralsauren Aminen auszugehen. Man kann auch so verfahren, dass man die als Ausgangskörper dienenden Basen zum Beispiel durch Zusatz der be rechneten Menge Schwefelsäure oder durch Einleiten von Salzsäuregas in Gegenwart von inerten Lösungsmitteln in die mineral sauren Salze überführt.
Die glatte 'Bildung der Thioharnstoffe ist überraschend, da es sich bei der vor liegenden Reaktion in der oben angeführten ersten Phase (Umwandlung der mineral sauren Salze in rhodanwasserstoffsaure Salze) um eine Ionenreaktion handelt, die bei der vorliegenden Arbeitsweise, auch im Falle der völligen Abwesenheit von Wasser und der völligen Unlöslichkeit der beiden Komponen ten in dem Verdünnungsmittel, sich glatt vollzieht.
Die nach der neuen Arbeitsweise gebil deten Thioharnstoffe fallen in kristallisierter, fein verteilter Form an und sind nach dem Entfernen der bei der Reaktion gebildeten anorganischen Salze sofort schmelzpunktsrein. Dieser Umstand stellt einen besonderen Vor teil des vorliegenden Verfahrens dar, da in folgedessen bei der weiteren Verarbeitung eine Isolierung der Reaktionsprodukte über haupt nicht erforderlich ist, sondern sofort in einer Operation weitergearbeitet werden kann.
So lässt sich beispielsweise die Dar stellung von 2-Amino-benzothiazolen gemäss schweizer. Patent Nr. 147 453 und lesen Zu sätzen, beziehungsweise nach Hugershoff (Berichte 36, Seite 3121) mit der Bildung der für diese Reaktion als Ausgangskörper dienenden Thioharnstoffe in einem Arbeits gang ausführen, so dass man, ausgehend von Arylaminen, in einem Reaktionsgefäss ohne Isolierung von Zwischenprodukten zu den entsprechenden 2-Aminobenzothiazolen ge langen kann.
<I>Beispiel 1:</I> Zur Suspension von 13 Teilen Anilin chlorhydrat in etwa 130 Teilen Chlorbenzol gibt man 8 Teile Ammoniumrhodanid oder die entsprechende Menge Na-rhodanid und rührt bei 95 bis<B>100'</B> C sechs bis acht Stun den nach. Nach dem Erkalten saugt man ab, wäscht den Rückstand mit Wasser aus und trocknet. Er besteht aus reinem Phenyl- thioharnstoff vom Schmelzpunkt 152 bis 154 C.
<I>Beispiel 2:</I> Zur Suspension von 17,3 Teilen p-Phene- tidinchlorhydrat in 70 Teilen Benzin oder Butylalkohol gibt man 9 Teile Ammonium- rhodanid und rührt sieben bis acht Stunden bei 95 bis<B>100'</B> C. Nach dem Aufarbeiten wie in Beispiel 1 erhält man den 4-Äthogy- phenylthioharnstoff vom Schmelzpunkt 171 bis 173 C in nahezu theoretischer Ausbeute.
Das gleiche Ergebnis erzielt man, wenn znan wie folgt arbeitet: In die Lösung von 13,7 Teilen Phene- tidinbase in 70 Teilen Chlorbenzol lässt man unter gutem Rühren 5,4 Teile Schwefelsäure <B>60B6</B> B6 einlaufen, setzt hierauf 9 Teile Am- moniumrhodanid zu und erhitzt noch drei Stunden auf 90 C.
Lässt man in die so erhaltene Suspension des 4-Äthoxyphenylthioharnstoffes gemäss schweizer. Patent Nr. 151138 20,8 Teile Chlorschwefel einlaufen und arbeitet wie dort auf, so erhält man das 2-Amino-6-äth- ogybenzothiazol in reiner Form.
<I>Beispiel 3:</I> In die Lösung von 10;8 Teilen p-Toluidin in etwa 70 Teilen Chlorbenzol lässt man un ier gutem Rühren 5,4 Teile Schwefelsäure 66 B. einlaufen, setzt hierauf 9 Teile Na- triumrhodanid zu und erhitzt noch drei Stun den auf<B>110'</B> C. Nach dem Abkühlen wird das Reaktionsprodukt abgesaugt, mit Wasser .gewaschen und getrocknet. Es liegt der reine p-Tolylthioharnstoff vom Schmelzpunkt 185 bis 186 C vor.
In gleicher Weise lassen sich p-Amino- diphenyl und Benzidin in die entsprechen den Thioharnstoffderivate überführen.
Zur Darstellung des 6-Methyl-2-amino- benzthiazols nach Hugershoff (Berichte 36, Seite 3121) oder dem schweizer. Patent Nr. 1511.37 wird der p-Tolylthioharnstoff nicht isoliert, sondern man lässt direkt zur Suspension in Chlorbenzol 15 Teile Brom, oder gemäss schweizer. Patent Nr. 151137 16 bis 18 Teile Sulfurylchlorid einfliessen und arbeitet wie dort auf.
<I>Beispiel</I> Zu einer Suspension von 17 Teilen Di- phenylaminchlorhydrat in etwa 120 Teilen o-Dichlorbenzol werden 10 Teile Ammonium- rhodanid zugefügt und die Mischung längere Zeit auf Temperaturen von 110 bis 130 C erhitzt. Das Reaktionsprodukt wird in übli cher Weise isoliert, nachdem man die Re aktionsmasse mit Wasser und kochendem Benzol extrahiert hat.
Der so erhaltene Di- phenylthioharnstoff schmilzt nach Umkri- stallisieren aus Butylalkohol bei etwa<B>212'C.</B> <I>Beispiel 5:</I> Zur Suspension von 20,7 Teilen N-äthyl- a-naphthylaminchlorhydrat in etwa 70 Tei len Chlorbenzol gibt man 8 bis 9 Teile Am moniumrhodanid und erhitzt drei bis vier Stunden unter Rühren auf 90 bis 100 C, dann noch etwa zwei Stunden zum Sieden. Nach dem Erkalten wird das Reaktions produkt abgesaugt, mit Wasser gewaschen und getrocknet.
Es kristallisiert aus Sprit in derben, farblosen Prismen vom Schmelz punkt 156 bis 158 C und sielt reinen un symmetrischen a-Naphthyl-äthyl-thioharn- stoff dar.
<I>Beispiel 6:</I> Zur Suspension von 21 Teilen 8-Methoxy- 1-naphthyla-min-chlorhydrat in etwa 180 Tei len Chlorbenzol gibt man 8 Teile Ammonium- rhodanid und rührt bei 95 bis 100 C sechs bis acht Stunden nach. Nach dem Aufarbeiten wie in Beispiel 1 erhält man reinen 8-Meth- oxy-l-naphthyl-thioharnstoff vom Schmelz punkt 185 bis 187 C.
In gleicher Weise erhält man aus 4-Amino- indazol (Berichte 53, Seite 12:23) den 4 Indazylthioharnstoff, der aus Butylalkohol in farblosen Blättchen vom Schmelzpunkt 225 bis 226 C erhalten wird.
<I>Beispiel 7:</I> Zur Suspension von 23 Teilen 1.5 Naphthylendiaminchlorhydrat in etwa 140 Teilen Chlorbenzol gibt man 18 bis 20 Teile Ammoniumrhodanid, erhitzt unter Rühren zirka drei Stunden auf 70 .bis 80 C, dann noch 10 bis 15 Stunden auf 95 bis 100 C. Nach dem Erkalten saugt man ab, wäscht den Rückstand mit -sehr verdünnter heisser Salzsäure aus und trocknet. Das Produkt ist reiner Thioharnstoff der Formel:

<I>Beispiel 8:</I> Zur Suspension von 22 Teilen 2-Amino- carba.zolchlorhydrat in 200 Teilen Chlor benzol gibt man 8 bis 9 Teile Ammonium- 21 und erhitzt unter Rühren sechs bis acht Stunden auf 120 bis 125 C. Man ar beitet wie in Beispiel 2! auf und erhält so reinen 2-Ca-rbacyl-thioharnstoff vom Schmelz punkt 278! C.
In gleicher Weise erhält man aus 3-Amino- 9-äthyl-carbazol den 3-(9-Äthyl)-carbazol- thioharnstoff, der aus Butylalkohol in farb losen Blättchen vom .Schmelzpunkt 208 bis 210 C kristallisiert. <I>Beispiel<B>9:</B></I> Man löst 17,8 Teile 5-Amino-8-chlor- chinolin vom Schmelzpunkt 160 bis 162 o C in der etwa zehnfachen Menge kochendem Chlorbenzol und leitet Salzsäuregas ein.
Aus der Anschlämmung des ausfallenden Chlorhydrats wird dann die überschüssige Salzsäure durch Luftdurchblasen wieder ent fernt. Man gibt dann 12 Teile Ammonium rhodanid zu und erhitzt etwa 12 Stunden im kochenden Wasserbad. Nach dem Ab kühlen saugt man den Niederschlag ab und wäscht gründlich mit Wasser nach.
Der so gebildete 8-Chlor-5-cIiinolylthio- harnstoff stellt ein hellgelbes kristallines Pulver dar und schmilzt bei 210 bis 211 C unter Zersetzung.

One can also proceed in such a way that the bases used as starting bodies are converted into the mineral acid salts, for example by adding the calculated amount of sulfuric acid or by introducing hydrochloric acid gas in the presence of inert solvents.
The smooth 'formation of the thioureas is surprising, since the reaction in the first phase mentioned above (conversion of the mineral acid salts into rhodanic acid salts) is an ionic reaction, which in the present procedure, even in the case of complete absence of water and the complete insolubility of the two components in the diluent, takes place smoothly.
The thioureas formed by the new procedure are obtained in crystallized, finely divided form and, after removing the inorganic salts formed during the reaction, are immediately pure at their melting point. This fact represents a particular advantage of the present process, since as a result, isolation of the reaction products is not necessary at all during further processing, but can be continued immediately in one operation.
