Verfahren zur Herstellung von 3-Amino-4-oxy-5-methoxybenzol-l-arsinsäure. Es wurde gefunden, dass man amino- sabstituierte l.-Oxy-2'-methoxybenzolarsinsäu- ren durch Reduktion der entsprechenden 1-Oxy-2-methoxynitrobenzolarsinsäuren, die nach bekannten Methoden dargestellt werden, erhalten kann. Die Verfahrensprodukte sind zur Bekämpfung von Protozoenkrankheiten ausserordentlich geeignet. Die neuen Ver bindungen sind, peroral gegeben, verhältnis mässig wenig giftig, "haben dabei aber recht gute Heilwirkung.
So ist zum Beispiel die Dos. tol. der 3-Amino-4-oxy-5-methoxyben- zol-1-arsinsäure je kg Kaninchen mehr als 500 mgr per os. Schon 25 mgr heilen Ka ninchensyphilis glatt, so dass sich ein Index von mehr als '/" ergibt.
Dies war nicht vorauszusehen, da nach Berichte 47, 1914, S. 996, die CH,0-Gruppe distherapeutisch ist und auch die in Berichte 48, 1915, S.<B>513</B> und<B>521</B> erwähnte 2 Methoxy-4-oxy-5-amino- benzol-l-arsinsäure keine brauchbare Wir kung zeigt. Die Arsinsäuren können zu Arsinoxyden, Arsenobenzolen oder Arsinen reduziert wer den.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Darstellung von 3- Amino -4- oxy- 5- methoxybenzol -l- arsinsäure, welches dadurch gekennzeichnet ist, dass man 3- Nitro -4- oxy-5- methoxybenzol -l- arsinsäure der Reduktion unterwirft.
Die neue Verbindung soll zur Bekämp fung von Protozoenkrankheiten verwendet werden; sie löst sich in Alkalien und Mi neralsäuren, ferner ist sie leicht löslich in Wasser, Methyl- und Äthylalkohol. Sie lässt sich nach bekannten Methoden zum Arseno- benzol, Arsinoxyd oder Arsin reduzieren.
<I>Beispiel:</I> 15 gr 3-Nitro-4-oxy-5-methoxybenzol-l- arsinsäure, erhalten aus 5-Nitro-2-acetyl- aminoanisol durch Austausch der Acetyl- aminogruppe gegen Hydorxyl, Reduktion der Nitrogruppe zur Aminogruppe und Ersatz letzterer durch den Arsinsäurerest und dar auffolgendes Nitrieren der so erhaltenen 4- Oxy-5-methoxybenzol-l-arsinsäure,
werden in 150 cm' Wasser und 50 em3 10 n Natron lauge gelöst. Zur Lösung gibt man unter lebhaftem Rühren 35 gr Natriumhydrosulfit. Die filtrierte Lösung wird mit 31 cm' Eis essig angesäuert, wobei die 3-Amino-4-oxy- 5-methoxybenzol-l-arsinsäure in Form ihres Natriumsalzes ausfällt. Durch Auflösen in Salzsäure und Abstumpfen der so erhaltenen Lösung auf kongoneutral erhält man die freie Säure.
Die Ausbeute beträgt 70 bis 75%. Die 3-Amino-4-oxy-5-methoxybenzol- 1-arsisäure löst sich in Alkalien und Mineral säuren, ferner leicht in Wasser, Methyl- und Äthylalkohol. Sie lässt sich zum Arsenoben- zol, Arsinoxyd oder Arsin reduzieren.

are dissolved in 150 cm 'water and 50 em3 10 N sodium hydroxide solution. 35 g of sodium hydrosulphite are added to the solution with vigorous stirring. The filtered solution is acidified with 31 cm 'glacial acetic acid, the 3-amino-4-oxy-5-methoxybenzene-1-arsinic acid precipitating in the form of its sodium salt. The free acid is obtained by dissolving it in hydrochloric acid and blunting the resulting solution to Congo-neutral.
