Process for the preparation of low-electrolyte, aqueous solutions of silk fibroin. The main patent describes a process for the production of low-electrolyte, aqueous solutions of silk fibroin, according to which silk fibroin is dissolved in anhydrous ammonia, this solution is brought into water and the ammonia is expelled from the aqueous-ammoniacal solution.
Useful solutions can be obtained by adding the solution of the fibroin in liquid ammonia to the water or, conversely, by adding the water to the solution of the fibroin in liquid ammonia. In both cases, the excess ammonia is removed after mixing.
It has now been shown that it is advantageous for the stability of the aqueous fibroin solutions as well as for the quality of the products made from them if the occurrence of high ammonia concentrations is avoided in the stage where an aqueous-ammoniacal fibroin solution is present. It is also advantageous to remove the ammonia as quickly as possible.
The removal of the ammonia can be accelerated while mixing by letting the anhydrous ammoniacal solution of fibroin step onto the surface of the water in a finely atomized state and at the same time removing the excess ammonia by means of a strong air stream directed at the water surface, or also using an air stream, both liquids can act on each other in an atomized state. In both cases, the removal of the ammonia during mixing is expediently supported by vigorous stirring of the mixture and by supplying heat. After mixing is complete, the remainder of the ammonia is advantageously removed in vacuo.
The process can be made even more effective by allowing the solution of the fibroin in anhydrous ammonia to come together with the water in a vacuum with vigorous stirring from the outset. Since the solubility of ammonia in water depends on the pressure, if ammonia and water come together in a vacuum, higher concentrations of ammonia may temporarily not occur. It is also useful in this embodiment of the method; vigorously stir the liquid.
The ammoniacal fibroin solution can enter the vacuum vessel above or below the surface of the water. Foam formation is expediently combated in a known manner by adding foam-destroying agents, such as, for example, higher alcohols.
Example: Cocoons, raw silk waste or other fibroin-containing raw materials are removed by boiling with soap for six hours and then treated warm for a short time with 1-3% acetic acid or another weak acid. After drying, one part of Seiderrfibroin is added to 8-10 parts of liquid ammonia at -75 °, stirred more often and filtered after about 20-30 minutes.
The yellowish-colored, thin, completely clear, liquid-ammoniacal silk fibroin solution is now sucked through a pipe of suitable cross-section into a high container connected to a well-functioning vacuum pump, on the bottom of which there are 61 / 2-7 parts of water so that the ammoniacal solution enters below the surface of the water. The liquid, which is under a good vacuum, is constantly stirred vigorously by a scrambled egg reaching to the bottom of the vessel.
The temperature of the aqueous solution is kept at 5-10 through a warm water jacket and by regulating the ammonia inflow. To avoid foaming, add a little octyl alcohol.
The addition of the ammoniacal fibroin solution ends after about 12-1 hour. In order to largely remove the ammonia, the mixture is then left to stir in vacuo for a short time, and the temperature can be increased to 15-20 °. The preparation of the solution is therefore, calculated from the entry of the fibroin, in 1-2 hours.
The solution prepared in this way contains about 14% fibroin, 0.1-0.2 ammonia and traces of octyl alcohol. It is clear, slightly yellow and easily filterable.
