Die Umsetzung kann dadurch mit guter Ausbeute erreicht werden, dass man einen Überschuss von 1-Ephedrin mit 1-Phenyl-l- oxo-2-brompropan (a-Brompropiophenon) zu sammenbringt.
Statt einen Überachuss von 1-Ephedrin zu verwenden, kann man die Kondensation mit Alkali bewirken.
Das 1-Phenylpropanolphenylpropanonme- thylamin ist in den üblichen organischen Lösungsmitteln, wie Alkohol, Äther, Benzol, Chloroform, leicht löslich, in Wasser schwer löslich. Es schmilzt bei<B>1260.</B> Mit Säure bil det es in Wasser lösliche, meist gutkristal lisierende Salze. Das Hydrochlorid ist sehr leicht löslich in Wasser und schmilzt bei 192-1930. Die spezifische Drehung seiner wässerigen Lösung ist [a]D =+93"- Die neue Verbindung soll als solche the rapeutische Verwendung finden oder als Zwischenprodukt zur Herstellung pharma zeutischer Präparate dienen.
<I>Beispiel</I> 33 Teile 1-Ephedrinbase werden in 100 Teilen Benzol und 100 Teilen Wasser mit 43 Teilen 1-Phenyl-l-oxo-2-brompropan (a- Brompropiophenon) zum Sieden erhitzt und eine konzentrierte wässerige Lösung von 12 Teilen Kaliumhydroxyd zugegeben. Man kocht noch 3-4 Stunden weiter, nimmt die Basen in Säure auf und macht unter Kühlen mit Natronlauge alkalisch. Es scheidet sich ein Öl ab. Dieses wird in heissem Methyl alkohol gelöst; beim Erkalten fällt das 1 Phenylpropanolphenylproparionmethylamin in guter Ausbeute kristallinisch aus.
In den Mutterlaugen findet sich wenig vom Racem- körper des 1-Plienylpropanolphenylpropanon- methylamins F.<B>92-93'</B> und etwas unver ändertes Ephedrin, das durch Vakuumdestil lation zurückgewonnen wird. Es wurden erhalten 28 Teile des optisch aktiven Kon densationsproduktes, 3 Teile Racemkörper ; 7 Teile unverändertes 1-Ephedrin konnten zurückgewonnen werden.
<I>Beispiel 2:</I> 17 Teile 1-Ephedrinbase werden unter Zusatz von 6 Teilen Natriumcarbonat mit 22 Teilen a-Brompropiophenon in 300 Teilen Benzol 4 Stunden gekocht. Dann nimmt man die Basen in Säure auf, macht mit Natron lauge alkalisch und kristallisiert das abge schiedene Öl aus Methylalkohol um. Erhalten wird, wie im Beispiel 1, die optisch aktive Verbindung vom Schmelzpunkt 126 .

Cook for another 3-4 hours, take up the bases in acid and make alkaline with sodium hydroxide solution while cooling. An oil separates out. This is dissolved in hot methyl alcohol; on cooling, the 1-phenylpropanolphenylproparionmethylamine precipitates in crystalline form in good yield.
Little of the racemic body of 1-plienylpropanolphenylpropanonomethylamine F. 92-93 'and some unchanged ephedrine, which is recovered by vacuum distillation, is found in the mother liquors. 28 parts of the optically active condensation product, 3 parts of racemic body were obtained; 7 parts of unchanged 1-ephedrine could be recovered.
Example 2 17 parts of 1-ephedrine base are boiled for 4 hours with the addition of 6 parts of sodium carbonate with 22 parts of α-bromopropiophenone in 300 parts of benzene. The bases are then taken up in acid, made alkaline with sodium hydroxide solution and the separated oil is recrystallized from methyl alcohol. As in Example 1, the optically active compound having a melting point of 126 is obtained.
