Abstract
Cette invention concerne un procédé d'amplification d'une sonde d'acide nucléique circulaire qui est produite suite à une interaction entre une sonde d'acide nucléique et une séquence cible, ladite sonde d'acide nucléique circulaire étant enrichie avant son amplification. L'enrichissement permet de réduire le niveau d'amplification de fond en éliminant toutes les sondes d'acide nucléiques linéaires, et peut en outre être enzymatique ou non enzymatique. L'amplification peut consister en une amplification de cercle rotative, tandis que la sonde peut consister en une sonde cadenas. Les séquences terminales de la sonde peuvent former des duplex non contigus, la sonde étant circularisée par ligature d'un ligand de capture ou d'une molécule d'acide nucléique d'espacement entre les deux séquences terminales. Le ligand de capture ou la molécule d'acide nucléique d'espacement peuvent en outre être marqués, par exemple par de la biotine. Ce procédé peut être utilisé lors d'applications visant à détecter des séquences d'acide nucléique spécifiques.
