Abstract
La présente invention concerne une technique permettant de générer de façon systématique des souches de mutant de levure d'haploïde et de diploïde obligatoires, dont les mutations létales d'un gène sont sensibles aux conditions, gène qui, par ailleurs, est essentiel à la viabilité cellulaire sous toutes les conditions de la croissance cellulaire. Les mutants conditionnels permettent l'analyse des changements physiologiques induits par l'inactivation d'un gène ou d'un produit génique et ils peuvent être utilisés pour solliciter la fonction d'un gène quelconque. Dans un mode de réalisation de l'invention, la technique consiste en général à: (i) prendre une souche de Candida hétérozygote en vue d'une mutation de perte de fonction d'un allèle de gène essentiel; (ii) et introduire un homologue conditionnellement sensible de ce gène, par exemple avec une protéine conditionnellement sensible ou une séquence de régulation de la transcription conditionnellement sensible, au niveau de l'allèle fonctionnel par transformation intégrative.
