Abstract

métodos para detecção de dna livre de células derivadas de doador em receptores de transplantes de múltiplos órgãos. a presente divulgação fornece métodos de amplificação e sequenciamento de dna, compreendendo: extrair dna livre de células de uma amostra de sangue, plasma, soro ou urina de um receptor de transplante que recebeu transplante de um ou mais órgãos, incluindo transplante simultâneo ou sequencial de múltiplos órgãos, em que o dna livre de células compreende dna livre de células derivado do doador e dna livre de células derivado do receptor; realizar amplificação direcionada em 200 a 50.000 loci-alvo em um único volume de reação usando 200 a 50.000 pares de iniciadores, em que os loci-alvo compreendem loci polimórficos e loci não polimórficos; sequenciar os produtos de amplificação por sequenciamento de alto rendimento para obter uma leitura de sequenciamento e quantificar a quantidade de dna livre de células derivado do doador e a quantidade de dna total livre de células com base nas leituras de sequenciamento; e determinar se a quantidade de dna livre de células derivado de doador ou uma função do mesmo excede um limite de corte indicando rejeição de transplante ou lesão de enxerto.
