Abstract

"CONSTRUTO, MéTODO DE GERAçãO DE PLANTA TRANSGêNICA OU PROGENIA DA MESMA RESISTENTE A VìRUS ssDNA, ELEMENTO GENéTICO LINEAR E ESPéCIE AVIáRIA E PLANTA GENETICAMENTE MODIFICADAS OU PARTE DA MESMA". A presente invenção relaciona-se, em geral, a construtos e, em particular, a construtos genéticos que compreendem seq³ências de polinucleotídeos capazes de liberar, em forma circular, covalentemente fechada, a partir de uma seq³ência de nucleotídeos maior tal como, mas, sem limitação, um genoma de uma célula eucariótica. De preferência, uma vez liberada, uma seq³ência de polinucleotídeos é reconstituída numa forma que permite a expressão da seq³ência de polinucleotídeos. Numa modalidade, a seq³ência de polinucleotídeos reconstituída compreende uma seq³ência de codificação com todo ou parte de um nucleotídeo estranho tal como, mas, sem limitação, uma seq³ência intrónica ou outro sinal de junção nela inserido. A expressão e, em particular, a transcrição de seq³ência da codificação envolve junção ã seq³ência estranha. A liberação e circularização é geralmente em resposta a um estímulo tal como um estímulo mediado por proteína. Mais particularmente, a proteína é uma proteína derivada de vírus ou de célula procariótica ou eucariótica ou proteína desenvolvimentalmente e/ou regulada especifica de tecido. O construto da presente invenção é particularmente útil para conferir resistência contra patógenos ou indução de apoptose ou outros mecanismos de morte celular úteis, por exemplo, no tratamento do câncer ou indução da esterilidade masculina ou feminina em plantas. Os construtos permitem, portanto, a expressão específica do sítio e/ou expressão desenvolvimentalmente regulada de seq³ências genéticas selecionadas.
