Abstract

6-Aminopenicillanic acid (6-APA) is produced by (a) treating an aqs. soln. of benzylpenicillin (Ia) or phenoxymethylpenicillin (Ib) or a salt thereof at a pH of 6.0-9.0 with a water-soluble enzyme prepn. comprising a penicillin desacylase and a solid, water-soluble absorbent, (b) separating the enzyme prepn. from the reaction mixt. by ultrafiltration, (c) isolating the resulting 6-APA; and (d) reacting the enzyme prepn. with more (Ia) or (Ib). The absorbent is pref. (i) a maleic anhydride polymer, esp. a copolymer with methyl vinyl ether, or (ii) a modified polysaccharide polymer esp. a sucrose/epichlorohydrin copolymer. The water-soluble enzyme prepns. allow more concd. solns. to be used than do water-insoluble prepns, and the reaction mixt. is homogeneous, thus facilitating continuous operation.
