Process for the preparation of a special phenyl-carbamate.
In the published Swedish patents 149,650, 151,103, 151,104, 151,105 and 151,204 a general formula has been given for a phenyl-carbamate and in addition, in the examples of these patents, certain compounds have been mentioned which are taken up by this general formula. Intensive and extensive research work in the chemical and pharmacological field has led to a compound, diethyl-aminoethyl 2-butoxy-4,6-dimethyl-carbanilate, designated by the applicant by LAC-1803 and having the following formula:
EMI1.1
<Desc / Clms Page number 2>
which has properties making this compound, not described, superior to the compounds mentioned in these patents.
The new compound can be used as a surface anesthetic as well as an infiltration anesthetic. The base itself is used as a surface anesthetic. If the thesic anes is in a gel, a salt of the anesthetic is used that is compatible with the tissue. A suitable salt is then the hydrochloride. For infiltration anesthesia a salt of the anesthetic which is compatible with the tissue is also used, suitably the hydrochloride.
In the following, the new anesthetic LAC-1803 will be compared to tetracaine, a summary of the results obtained during the comparison being given in the following table

As can be seen from the table above, the new anesthetic LAC-1803 is therefore considerably less toxic than tetracaine and in addition three times as compatible with
EMI2.6
the tissue as the tetracatne. Moreover it causes less indiepoei- tion than tetracaine
In addition to the advantages mentioned, the new anesthetic LAC-1803 offers the advantage that in an aqueous phase it is very stable in solutions with a pH between 3 and 6.
In this pH range the disintegration is only 0, C2%, which means that the new anesthetic LAC-1803 is 100 times more
<Desc / Clms Page number 3>
stable than tetracaene.
Nerve blocking tests were done with the new local anesthetic LAC-1803 in both mice and rats and 40% longer duration was obtained. long with) the new local anesthetic than with retracaine. The effect factor with the new local anesthetic LAC-1803 is 1.75 compared to tetracaine 1.75 compared to tetracaine
As a surface anesthetic, the new local anesthetic has a duration which is as long as that of tetra- caine. Despite this the new local anesthetic, as a surface anesthetic, has the advantage over tetracaine of being less toxic and less perishable. These results were obtained in the anesthesia of the cornea of a human being.
The therapeutic index for the new local anesthetic as a surface anesthetic, compared to tetracaine, is 1.3.
When mixed with polyethoxy-decane, it has been found that a molecular complex is formed which can be determined by means of differentiation thin-layer chromatography.
This molecular complex produces a surface anesthesia which is synergistic, namely that the duration of the molecular complex is greater than the sum of the duration of its constituents,
The novel local anesthetic LAC-1803 can be prepared by the methods which have been described in the aforementioned patents.
However, in the following, the process for preparing the most advantageous compound will be described.
Example 1.
2.3 g of metallic sodium are dissolved while stirring in a glass retort at 80 ° C. in 470 g of diethylaminoethanol. When the metal has dissolved, 265 g of ethyl 2-butoxy-4,6-dimethyl-carbanilate are added, the temperature is raised to 90 ° C. and the components are allowed to react with each other for 5 hours under a pressure of 200 mm Hg. When the

re-esterification has been carried out, the excess amino alcohol is removed empty. The residue from the distillation is dissolved in 800 cm3 of toluene and filtered.
The toluene solution is then extracted twice with 500 cm3 of cold 10% hydrochloric acid. Ammonia is added to the mixed acidic saline solutions until pH 4 and the solutions are extracted three times with 500 cm3 of toluene. The aqueous solution containing the

Diethylasinoethyl 2-butoxy-496-dimethyl-carbanilate is treated with activated carbon and filtered. Ammonia is slowly added to the mother liquid while constantly cooling at 15 ° C to pH 10 and the free carbanil ester in toluene is absorbed by double extraction with 400 cm3 of toluene.
The toluene solution is dried with anhydrous sodium sulfate and evaporated in vacuo. The anhydrous base is dissolved in 1200 cm3 of dried methyl ethyl ketone and gaseous hydrochloric acid is passed through the solution until a pH value of 2. 2-butoxy-4 hydrochloride is obtained. Diethylaminoethyl 6-dimethyl-carbanilate is then crystallized, obtaining white crystals. A total yield of 90% relative to the base is obtained. Melting point of
