La proscillaridine A est un glycoside de la scille blanche que l'on emploie depuis quelque temps pour le traite- ment des maladies cardiaques. Dans le cas de nombreux glyco- sides du coeur et, par conséquent, également dans le cas de la proscillaridine A, le problème de l'activité orale n'est pas encore résolu d'une manière pleinement satisfaisante. Par suite d'une faible résorption entérale, on observe d'importan- tes variations individuelles de résorption, de sorte qu'il est cliniquement difficile de trouver la dose orale active dans un cas particulier et que l'intérêt thérapeutic est

fortement mis en doute, C'est pourquoi, la présente inven- tion a pourobjet de trouer de nouveaux dérives de la pros- cillaridine A, ces dérives ayant une haute activité par voie orale.

dans laquelle R1 et R2 ont les mêmes significations que celles indiquées à la formule I, , ou un dérivé réactif de ce composé, en présence d'un agent de condensation ou d'un catalyseur acide, en introduisant le radical alcoylidène dans le groupement cis-vic-glycol de la partie de rhamnose de la proscillaridine A et en isolant, du mélange de la réaction, l'alcoylidène-proscillaridine A obtenue.
On effectue la réaction de façon connue en soi.
Comme agents de condensation ou comme catalyseurs acides, on emploie les matières proposées dans la littérature pour ces réactions. On a obtenu des résultats particulièrement bons en ce qui concerne la réaction, c'est-à-dire, une courte du- rée réactionnelle, des températures réactionnelles modérées telles que la température ambiante et l'absence de formation de sous-produits non désirés, avec des sels tels que les halo- génures de lithium, de l'aluminium, du fer et, de préférence, de l'étain, tout particulièrement avec le chlorure d'étain II, même sous forme de son dihydrate. Les sels précités peuvent être employés en très faibles quantitiés, par exemple envi- ron 1/100 du poids de la proscillaridine A mise en oeuvre ; préférence, on emploie une quantitié d'environ 1/20 du poids de la proscillaridine A.
Le pentoxyde de phosphore accélère également très fortement la formation du dérivé d'alcoylidène.
Parmi les acides mis en oeuvre d'une manière préalablement connue pour ces réactions, les acides anhydres tels que, par exemple, l'acide sulfurique, l'acide chlorhydrique ou les acides sulfoniques organiques sont particulièrement appro-

priés, en particulier en quantitiés d'environ 1/100 à environ 1/1000 de la quantitié pondérale du glycoside mis en oeuvre ou des solutions de ces acides de 0,2% à 0,002t, de préféren- ce 0,02%, dans le solvant employé. Dans ce cas, on travaille également à des températures comprises entre-10 et +30 C. En une période de 1 à' 6 heures, on obtient les nouveaux dérivés désirés de la proscillaridine A avec de très bons rendements sans formation de sous-produits.
Bien qu'il soit préférable de travailler essentiel- lement en absence d'eau, on peut également effectuer la réac- tion en présence d'acides aqueux. Toutefois, la mise en oeu- vre d'acides aqueux dans les concentrations précitées ralentit déjà nettement la réaction.
On peut effectuer la réaction en absence ou en pré- sence de solvants organiques. Dans ce cas, on peut également employer un excès du composé de formule R1-CO-R2 comme sol- vant, avantageusement en une quantitié telle que la proscilla- ridine A soit complètement dissoute.
Au lieu des composés de la formule précitée R1-CO-R2, on peut également employer des dérivés réactifs de ces compo- sés, par exemple les acétals ou les cétals, ou encore les acé- tal-esters ou les cétal-esters, en particulier les acétals ou les cétals des alcools inférieurs de 1 à 6 atomes de carbone.
La réaction peut être arrêtée par l'addition d'une base telle que, par exemple, la pyridine ou une amine tertiaire, ou encore un sel tel que, par exemple, le carbonate de potassium.
Le dérivé d'alcoylidène de la proscillaridine A peut être isolé directement du mélange de la réaction, par exemple par distil- lation du solvant organique ou par précipitation avec ds l'eau et filtration. On peut le purifier par des méthodes de tra- vail habituelles telles que, par exemple, la cristallisation ou la chromatographie en colonne et l'on peut le séparer des autres impuretés éventuellement présentes ou de la matière

Les exemples suivants servent à illustrer davantage le procédé suivant l'invention.
Exemple 1 Isopropylidène-proscillaridine A
Tout en agitant à environ 20 C, on dissout 10 g de proscillaridine A dans 500 ml d'acétone anhydre, puis on les fait réagir avec 500 mg de chlorure d'étain II. Après une heure, on interrompt la réaction par addition de 1,5 mlde pyri- dine. On filtre et distille l'acétone sous vide. On àbsorbe le résidu dans environ 200 ml de chloroforme et l'on intro- duit cette solution dahs une colonne de gel de silice. On pro- cède à une élution avec un mélange de chloroforme et d'acétone dans le rapport de 2 :1. proscillaridine n'ayant pas réagi peut être séparée ultérieurement du gel de silice au moyen de méthanol. On a obtenu 9,66 g de produit (soit un rendement de 90%, calculés sur la proscillaridine mise en oeuvre).
Point de fusion: 140 C (Kofler).
Exemple 2 Isopropylidène-proscillaridine A
On dissout 10 g de proscillaridine A dans 500 ml d'acétone anhydre contenant 0,7 ml d'une solution éthérée de HCl (140 mg de HCl/ml) et l'on agite pendant 2 heures à la température ambiante. Par addition de 1,5 ml de pyridine, on interrompt la réaction, puis on extrait l'acétone sous

vide. On absorbe le résidu dans du chloroforme, on lave la solution avec de l'eau, on la sèche, puis on la soumet à une chromatographie dans une colonne de gel de silice. On procè- de à une élution avec un mélange d'ester acétique et de chloo- forme dans le rapport de 1:1 et l'on obtient 10,2 g d'isopropy- lidène-proscillaridine A (rendement: 95%) d'un point de fusion de 137 - 140 C (Kofler).
Exemple 3
Isopropylidène-proscillaridine A
On dissout 15 g de proscillaridine A dans 750 ml d'acétone anhydreet, tout en refroidissant à environ 0 C, on introduit 1,5 g de pentoxyde de phosphore. On agite convena- blement la solution pendant 1 heure, on la laisse décanter, on la traite avec environ 2 g de carbonate de potassium anhydre, on la concentre sous vide jusqu'à environ 50 ml et on l'intro- duit dans de l'eau glacée. On essore l'isopropylidène-proscilla- ridine A qui précipite, on la lave avec de l'eau et on la sèche.
Le rendement est de 13,7 g.
Les substances obtenues dans les exemples 1, 2 et 3 peuvent être recristallisées dans du méthanol aqueux. P.F.

Exemple 4 Isobutylidène-proscillaridine A
On dissout, à la température ambiante, 3 g de proscilla- ridine A dans 170 ml de méthyl-éthyl-cétone. Aprs addition de
0,2 ml d'acide chlorhydrique éthéré, contenant 140 mg de HCl/ml, on agite pendant 2 heures, puis on interrompt la réaction par addition de 0,5 ml de pyridine. On extrait la méthyl-éthyl-cé- tone on excès sous le vide de la trompe, on absorbe le résidu dans du chloroforme et on l'élue dans une colonne de gel de silice avec un mélange de chloroforme et d'acétone dans le

Exemple 5 Ethylidène-proscillaridine A
On dissout 2 g de proscillaridine A dans 100 ml' d'acétaldéhyde contenant 20 mg de HCl et l'on agite pendant 1 1/2 heure à la température ambiante. Après avoir interrom- pu la réaction par addition de 300 mg de pyridine et après extraction du solvant, on purifie le mélange de la réaction dans une colonne de gel de silice comme décrit à l'exemple 1.
On obtient 1,8 g d'un produit d'éthylidène d'un point de fu- sion de 68 - 70 C[aD22] = = -53 (dans du méthanol). Par re- cristallisation dans du méthanol aqueux, la substance a un point de fusion de 142 C.
Exemple 6 Cyclohexylidène-proseillaridine A
On dissout 3 g de proscillaridine A dans 150 ml de cyclohexanone en présence de 300 mg de HCl. Après une durée réactionnelle de 3 heures, on traite comme décrit à l'exemple 5. et l'on obtient 2,5 g d'une huile visqueuse claire se soli- difiant lorsqu'on la 'laisse reposer. Point de fusion: 88-90 C.
Exemple Isopropylidène-proscillaridine A
On met 20 g de proscillaridine A en suspension dans
500 ml d'acétone pure: contenant 0,02% de HCl anhydre. On agi- te la suspension pendant environ 40 minutes à la température ambiante jusqu'à ce que tout le produit soit entré en solution.
Ensuite, on continue à agiter pendant 1/2 heure, on fait réa- gir la solution légèrement colorée en brun foncé avec envi-

ron 10 ml d'eau et l'on ajoute du carbonate de calcium, jus-' qu'à ce que la solution n'ait plus de réaction acide. On sé- pare par filtration et l'on concentre l'acétone sous vide. On dissout le résidu dans 100 ml de chloroforme et on le filtre sur du gel de silice. On procède à un lavage avec du chloro- forme, on extrait le solvant et l'on obtient environ 15 g d'isopropylidène-proscillaridine A que l'on peut recristalli- ser dans du toluène ou du méthanol aqueux.
P.F. : 140 - 143 C (Kofler).
Suivant les méthodes classiques et en ajoutant des substances auxiliaires habituelles, les nouveaux dérivés de la proscillaridine A peuvent être employés en comprimés, en dragées et en solutions pour injection.
