On fait réagir un dérivé hydroxy-indolique répondant à la formule 2 annexée, sous forme de son sel alcalin, avec une molé- cule de chlorophosphate de dibenzyle dans un solvant inerte, comme le toluène, le 1.2-diméthoxy-éthane ou l'alcool tertio-amylique., puis on secoue ou laisse reposer la solution pendant quelques heures à la température ambiante, éventuellement sous atmosphère d'azote.
On évapore ensuite à siccité, on reprend le résidu dans un solvant organique approprié tel que l'alcool ou un mélange d'alcool et de chloroforme, on élimine les insolubles par filtration, et on peut généralement faire cristalliser directement le phosphate de diben- zyle et du dérivé 4-hydroxy-indolique. En cas d'échec, il est re- commandé de parfaire la purification par une filtration sur une colonne d'alumine.
Sur le phosphate de dibenzyle et du dérivé 4-hydroxy- indolique obtenu, on effectue la scission d es groupes benzyliques en secouant le corps en solution méthanolique avec un catalyseur au palladium et de l'hydrogène puis, ayant éliminé le catalyseur, on fait cristalliser l'ester phosphorique formé dans un solvant conve- nable, de préférence dans du méthanol ou dans de l'eau.
Les dérivés indoliques de formule 2 utilisés comme matiè- res premières dans le procédé de l'invention se préparent de préfé- rence à partir des composés 4-benzyloxy-indoliques correspondants ne portant pas de substituant en position 1, par alcoylation ou aralcoylation selon des méthodes connues. On enlève ensuite le groupe benzyle, qui est le groupe protecteur préféré, par bydrogé- nation catalytique. La préparation des dérivés 4-benzyloxy-indoli- ques nécessaires s'effectue selon la demande de brevet français du 10 septembre 1959 pour "Procédé de préparation d e nouveaux déri- vés de l'indole" au nom de la demanderesse.
Les nouveaux composés sont des substances cristallisées, solides à la température ambiante. Ils sont très difficilement so- lubles dans presque tous les solvants organiques; par contre ils se dissolvent relativement facilement dans l'eau. Avec le réactif de

Keller (acide acétique glacial contenant du chlorure ferrique et acide sulfurique concentré) ils donnent une réaction colorée posi- tive dont la nuance dépend de la nature du substituant R3.
Les nouveaux dérivés phosphorés de l'indole se distinguent par d'intéressantes propriétés phamaco-dynamiques que l'on peut mettre à profit en thérapeutique. Ils provoquent en particulier une stimulation du système nerveux sympathique central qui se traduit par une mydriase, une élévation de la pression sanguine, une éléva- tion de la température et une augmentation de la glycémie, ainsi que par une inhibition de l'activité intestinale. De plus, ils font preuve de nettes propriétés antagonistes de la sérotonine et ils stimulent les réflexes spinaux. Ils exercent en même temps une fai- ble action tranquillisante et inhibitrice des états d'impulsions.
Ils permettent en outre de réprimer l'action sédative et convulsi- vante de la réserpine. En raison de leurs propriétés sur le sys- tème nerveux végétatif central et de leur action inhibitrice des états d'impulsions, et également grâce à leurs propriétés antagonis- tes de la réserpine, ils peuvent être utilisés pour le traitement des maladies psychiques les plus variées, principalement pour le traitement des névroses obsessionnelles ainsi que des dépressions, des maussaderies et des états d'angoisse d'origine neurotique ou psychosique. Les dérivés phosphorés de l'indole répondant à la formule 1 définie ci-dessus se distinguent par une très faible toxi- cité.
Ils sont résorbés pratiquement quantitativement par l'organisme et c'est pourquoi ils sont appliqués de préférence par voie buccale; mais l'on peut tout aussi bien les administrer par voie sous-cutanée, intramusculaire ou intraveineuse. Ils doivent être utilisés en thérapeutique, mais ils constituent aussi d'intéressants produits intermédiaires pour la préparation de médicaments.
Le présent brevet assure la protection d es nouveaux composés et des compositions pharmaceutiques qui les renferment au titre de substances actives.

On évapore à siccité 910 mg de 1-méthyl-4=hydrozy-N- diméthyltryptamine avec 4,35 cm3 de lessive de soude caustique 0,96-n et on sèche à 70 pendant 2 heures sous vide poussé. On dissout le résidu sec dans 30 cm3 de 1.2-diméthoxy-éthane et on ajoute une solution de chlorophosphate de dibenzyle dans du tétra- chlorure de carbone, solution qui a été préparée à partir de 1,13 g de phosphite de dibenzyle.
On secoue pendant 15 heures à la tempé- rature ambiante, on sépare par filtration le chlorure de sodium qui a précipité, on évapore le filtrat à siccité et on c hromato- graphie le résidu sur 30 fois sa quantité d'alumine : le phosphate
