La présente invention est relative aux phénacyl sulfones. Elle concerne, plus particulièrement,les composés phénacylsulfonyl-pyridiniques et leurs dérivés.
Les nouveaux composés sont représentés par la formule de structure suivante:

dans laquelle R désigne de l'hydrogène ou un radical 'alcoyle inférieur,et par les sels de ces composés. Dans la formule donnée' ci-dessus, le groupe "aryle" est un groupe aryle monocyclique, tel que le groupe phényle et les groupes phényle substitués dans le noyau, comme les groupes halo- phényle, hydroxyphényle, alcoxyphényle, acyloxyphényle, cyanophényle, nitrophényle aminophényle, etc.. Les sub- stituants nucléaires des groupes alcoxyphényle et acyloxy- Phényle sont, de préférence, des radicaux alcoyle inférieurs, tels que les radicaux méthoxyphényle et acétoxyphényle.
Les sels des.noyaux composés, tels que les sels d'addition avec des acides et les sels quaternaires, tombent particulièrement dans la portée de la présente invention . ' Parmi ces sels, on peut citer les sels d'addition avec des . acides, tels que les hydrohalogénures, comme-les chlorhydra- tes, sulfates et toluène suif ouates, et les sels quaternai- res, tels que les sels'quaternaires d'halogénures d'alcoyle inférieur d'halogénures d'aryle et d'halogénuresd'aracoyie.
Les nouveaux composés suivant la présente inven- tion sont obtenus par réaction d'un sel de métal alcalin de l'acide 3-pyridine suif inique avec une halogénure de phéna-

cyle dans un solvant' organique, tel que l'éthanol, la dimé- thylformamide, l'acétone, le nitrométhane, etc.. La base libre ainsi obtenue peut être convertie en un sel d'addition avec un acide, par réaction de la base, au sein d'un sol- vant, avec un acide fort, tel qu'un acide minéral.
Les . sels quaternaires sont obtenus par.réaction de la base li- bre, au sein d'un solvant organique, tel que le nitrométhane, l'acétone, l'acétonitrile, l'éthanol, etc.., avec un agent de quaternisation, tel qu'une halogénure d'alcoyle, une halo- génure d'aralcoyle, une halogénure de phénacyle, etc..
L'acide 3-pyridine sulfinique, qui constitue un des réactifs de départ pour la préparation des composés sui- vant la présente invention, se)prépare par condensation de chlorure de 3-pyridine sulfonyle avec de l'hydrazine,de ma- nière à obtenir de l'hydrazide d'acide 3-pyridine sulfonique et par réaction de cette hydrazide avec de l'acétone. Par neutralisation d'une solution aqueuse d'acide 3-pyridine ,suif inique avec de l'hydroxyde de sodium et par évaporation de la solution résult ante, le sel de sodium de l'acide 3-py- ridine sulfinique est obtenu .
Les aminophénacyl sulfones de cette classe peuvent être obtenus,,.par exemple, par désacétylation des acétami- dophénacyl sulfones correspondante à l'aide d'un acide fort aqueux. Les hydroxyphénacyl sulfones sont obtenues similai- rement à partir des acétoxyphénacyl sulfones correspondantes.
Les composés'suivant la présenté invention sont utilisables comme anthelmintiques; ainsi, ils sont actifs contre les vers de la famille des oxyures .
EXEMPLE 1.. -
19,9 g (0,12 mol.) du sel sodique anhydre d'acide 3-pyridine sulfinique et 18,6 g (0,12 mol.) de chlorure de phénacyle, dissous dans 200 cc d'éthanol absolu, ont été chauffés au reflux pendant 15 heures. Le produit de réaction;

a été séparé par filtration et extrait dans un appareil
Soxhlet avec du méthanol. L'extrait méthanolique a été concentré jusqu'à un faible volume, puis refroidi, après quoi les cristaux formés ont été séparés par filtracion et séchés . La phénacyl-3-pyridyl sulfone a été recristal- lisée dans du benzène et fondait à 106-107 C.
EXEMPLE 2. -
9,1 g (0,035 mol. ) de phénacyl 3-pyridyl sulfone ont été dissous dans 200 cc d'acétone glacée contenant 40% de bromure de méthyle en poids. La solution, placée dans un flacon hermétiquement clos, a été admise à se réchauffer lentement jusqu'à température ambiante et en@@ite laissée au repos pendant une nuit. Les cristaux formés ont été sé- parés par filtration, lavés à l'acétone et ensuite séchés.
Le bromure de 1-méthyl-3-phénacylsulfonyl-pyridinium fond à 124-127 C, après recristallisation dans un(mélange d'acé- tonitrile, d'acétone et d'éther .
EXEMPLE 3, -
A 12 g (0,046 mol. ) de phénacyl 3 pyridyl sulfone dissous dans 75 ce de nitrométhane, on a ajouté 25 g (0,23 mol.) de bromure d'éthyle. La solution a été chauffée dans un tube scellé,à 65 pendant 16 heures, puis refroidie.
Les matières volatiles'ont ensuite été chassées par distil- lation. Le,résidu a été cristallisé dansun mélange dtétha- nol et d'éther. Le bromure de 1-éthyl-3-phénacylsulfonyl- pyridinium fondait à 163-164 C en se décomposant .
EXEMPLE 4.- une suspension de 12 g (0,040 mol.) de phénacyl 3-pyridyl sulfone dans 75 ce de nitrométhane, on a ajouté 13e7 ce (0,15 mol.) de bromure de n-propyle. La suspension a été chauffée dans un tube scellé à 65 C pendant 15 heures.

La solution résultante a été refroidie et les matières volatiles ont été chassées par'distillation. La matière solide résiduelle, constituée par du bromure de 1-propyl- 3-phénacylsulfonylpyridinium, a été recristallisée dans un mélange à 95% d'éthanol et d'éther, P.F.:181-182 C (décom- position)
EXEMPLE 5.-
A une suspension de 12 g (0,046 mol.) de phénacyl 3-pyridyl sulfone dans 75 cc de nitrométhane, on a ajouté 16 cc (0,15 mol. ) de bromure de n-butyle. La suspension a été chauffée dans un tube scellé à 65 C pendant 15 heures.
La solution résultante a été refroidie et les matières vola... tiles ont été chassées par distillation. La matière rési- duelle, constituée par du bromure de 1-butyl-3-phénacyl- sulfonylpyridinium hémihydraté, a été recristallisée dans un mélange d'éthanol et de méthyléthylcétone ; P.F.: 164- 166 C (décomposition).
EXEMPLE 6.-
A une.suspension de 12 g (0,046 mol. ) de phénacyl 3-pyridyl sulfone dans 75 cc de nitrométhane, on a ajouté 16 cc de bromure de n-amyle. La suspension a été chauffée dans un tube scellé à 65 pendant 15 heures. La solution résultante a été refroidie et les matières volatiles ont ensuite été chassées par distillation. La matière solide a été recristallisée dans un mélange dtéthanol, de méthyl- éthylcétone et d'éther. Le bromure de 1-amyl-3-phénacylsul- fonylpyridinium fondait à 120-122 C.
EXEMPLE 7.-
10,2 g (0,038 mol.)de phénacyl 3-pyridyl sylfone ont été dissous dans 150 cc d'acétonitrile. A la solution on a ajouté 8,3 g (0,038 mol.) de bromure de p-nitrobenzyle.

Le mélange a été agité et chauffé au reflux pendant @ heures et refroidi. Les cristaux ont été séparés par filtration, lavés à l'acétonitrile et séchés. Le bromure p-nitrobenzy- lique de phénacyl 3-pyridyl sulfone hémihydraté a ét re- cristallisé dans un mélange d'eau, d'acétonitrile et d'é- ther; P.F. : 178-180 C (décomposition).
EXEMPLE 8.-
A une suspension de 17 gr (0,065 pol.) de phéna- cyl 3-pyridyl sulfone dans 75 cc de nitrométhane, on aajou- té 12 gr (0,078 mol. ) de chlorure de phénacyle. La suspen- sion a été chauffée dans un tube scellé à 65 C pendant 16 heures. La solution résultante a été refroidie et les matiè- res volatiles ont été chassées par distillation. La matière solide résiduelle a été recristallisée dans un mélange d'é- thanol et d'éther et le chlorure de 1-phénacyl-3-phénacyl- sulfonylpyridinium obtenu fondait à 195-197 C en se décompo- sant.
EXEMPLE 9. une. suspension bien agitée de 28,6 gr (0,2 mol.) d'acide 3-pyridine sulfinique dans 250 cc d'éthanol absolu, on a ajouté 12,gr de méthoxyde de dodium (85). A la solu- tion neutre résultante, on a ajouté 42,6 gr (0,2 mol.) de ' ' c( -bromopropiophénone.' Après 5 minutes de chauffage au re- flux, la solution s'est troublée. Le mélange a été agité et chauffé au reflux pendant 15 heures, puis'filtré encore à chaud. Le filtrat a .été',concentré sous vide pour chasser les matières volatiles. La gomme résiduelle a été lavée à l'eau, puis recristallisée dans un mélange d'éthanol et d'- isopropanol. La [alpha]-méthylphénacyl 3-pyridyl sulfone fondait à 81-82 C.

p-méthoxyphénacyle. Le mélange a été agité et chauffé au re- flux pendant 15 heures puis filtré à chaud et le résidu cris- tallin a été lavé vigoureusement à l'eau et séché à l'air.
La p-méthoxyphénacyl 3-pyridyl sulfone ainsi obtenus fondait à 106-108 C.
Une suspension dans l'eau glacée de 20,6 gr du produit obtenu dans le paragraphe précédent a été traitée à l'aide de 200 cc d'acétone, contenant 40% de bromure de mé- thyle en poids. Le mélange a été chauffé dans un tube scellé à 100 pendant 8 heures, puis refroidi. Les cristaux obtenus ont été séparés par filtration, lavés à l'acétone et séchés.
Le bromure de 1-méthyl-3-(p-méthoxyphénacylsulfonyl)pyri- dinium fondait à 183-185 C, après recristallisation dans un mélange de méthanol et d'éther.
EXEMPLE 14. -
On a préparé du bromure de p-acétoxyphénacyle en ajoutant lentement 22 gr de brome à une solution de 24,7 gr de p-acétoxyacétophénone dans 100 cc d'éther absolu conte- nant 0,2 gr de chlorure d'aluminium anhydre. Les matières volatiles ont ensuite été chassées par distillation sous vide et le résidu aété triture avec de l'eau et de l'éther de pé- trole. Le produit a ensuite été.séparé par filtration et cris- tallisé dans un mélange'd'éther et d'éther de pétrole; P.F.
65-67 C.
Il,5 gr (0,0 mol.) d'acide 3-pyridine sulfinique et 20 gr (0,078 mol.) de bromure de p-acétoxyphénacyle ont été mis en réaction selon le mode opératoire décrit dans l'ex- emple 9. 3-Pyridine sulfinic acid, which constitutes one of the starting reagents for the preparation of the compounds according to the present invention, is prepared by condensation of 3-pyridine sulfonyl chloride with hydrazine, as follows: obtaining hydrazide of 3-pyridine sulfonic acid and by reacting this hydrazide with acetone. By neutralization of an aqueous solution of 3-pyridine acid tallow with sodium hydroxide and evaporation of the resulting solution, the sodium salt of 3-pyridine sulfinic acid is obtained.
The aminophenacyl sulfones of this class can be obtained, for example, by deacetylation of the corresponding acetamidophenacyl sulfones with a strong aqueous acid. The hydroxyphenacyl sulfones are obtained similarly from the corresponding acetoxyphenacyl sulfones.
The compounds following the present invention can be used as anthelmintics; thus, they are active against worms of the pinworm family.
EXAMPLE 1 .. -

EXAMPLE 3, -

To a suspension of 12 g (0.046 mol) of phenacyl 3-pyridyl sulfone in 75 cc of nitromethane was added 16 cc of n-amyl bromide. The suspension was heated in a sealed tube at 65 for 15 hours. The resulting solution was cooled and the volatiles were then removed by distillation. The solid material was recrystallized from a mixture of ethanol, methyl ethyl ketone and ether. 1-Amyl-3-phenacylsulfonylpyridinium bromide melted at 120-122 C.

To a suspension of 17 g (0.065 pol.) Of phenacyl 3-pyridyl sulfone in 75 cc of nitromethane, 12 g (0.078 mol) of phenacyl chloride was added. The suspension was heated in a sealed tube at 65 ° C for 16 hours. The resulting solution was cooled and the volatiles were removed by distillation.
