<Desc/Clms Page number 2>
<Desc/Clms Page number 3>
EMI3.1
<Desc/Clms Page number 4>

EXEMPLE I
<Desc/Clms Page number 6>
<Desc/Clms Page number 7>

"Quaternary ammonium and quaternary ammonium-dialkyl xanthine compounds, and methods of preparation."
The invention relates to quaternary ammonium compounds, to quaternary ammonium-dialkyl xanthine compounds and to processes for their preparation. The invention relates more particularly to alkoxy choline compounds, choline-alkoxy xanthine compounds, and a process for the production of these compounds; it also includes discoveries and improvements by which the preparation of these compounds is facilitated.
Heretofore, choline-dialkyl xanthine compounds, such as choline-theophylline, have been produced by
<Desc / Clms Page number 2>
a condensation of a choline base with theophylline, in which the choline base molecule reacts with a theophylline molecule, with the formation of a cho molecule. line theophylline and a water molecule. The presence of water is disadvantageous because it greatly increases the solubility of the choline theophylline product and, therefore, retards the separation by crystallization of the product from the reaction mixture, and also causes a poor yield and obtaining a relatively impure product.
The removal of water could be accomplished by azeotropic distillation or other known method. However, such processes tend to cause decomposition of the reaction product, which results in soiling and discoloration of this product.
A main object of the invention is to provide a process for the preparation of dialkyl xanthine products, in which the disadvantages inherent in the old processes are avoided.
.
Another object of the invention is to provide a process for the preparation of dialkyl xanthine products which does not lead to the release of water.
Another object of the invention is to provide a process for the preparation of choline dialkyl xanthine products, which leads to the production of products in high yields and of great purity, and which can be carried out easily, economically. and effectively.
A further object of the invention is to provide a process for the preparation of dialkyl xanthine products, in which an alkoxycholine is reacted with a dialkyl xanthine, for example a reaction between methoxy choline and - theophylline-1,3-dimethyl xanthine.
A particular aim of the invention is to provide, as a new compound, an alkoxy choline having the formula:
<Desc / Clms Page number 3>
EMI3.1
in which R represents an alkyl radical containing 1 to 5 carbon atoms.
Other objects of the invention, either will be obvious, or will appear from the description below.
The invention relates to the different phases of the process and the relationship of one or more of these phases with each of the others; it also relates to compounds which exhibit the characteristics, properties and ratio of constituents, which are given in the examples of the following description.
In practicing the invention, dialkyl xanthine products can be prepared by first preparing a quaternary alkoxy ammonium compound by reacting a quaternary ammonium chloride in an anhydrous medium with a metal alkoxide. alkali metal of the formula ROM, in which R denotes an alkyl radical containing 1 to 5 carbon atoms and M represents sodium and potassium, with formation of a quaternary alkoxy ammonium compound, and then separation of the liquid and the Precipitated alkali metal chloride.
The quaternary ammonium alkoxy compound is then reacted with a dialkyl xanthine in which the carbon content of the alkyl is not more than two, thereby forming a quaternary ammonium-dialkyl xanthine product, and the product thus formed is cooled and separated.
The alkyl radicals in the quaternary ammonium compound can be methyl, ethyl, propyl, butyl and amyl, and in the dialkyl xanthine the alkyl radicals can be methyl and l. 'ethyl. It will be understood that the compounds containing the alkyl radicals can be either single or mixed, ie, the radicals can be the same or different. Thus, the xanthine can be a dimethyl, a diethyl or a methyl ethyl xanthine.
<Desc / Clms Page number 4>
In particular, choline chloride is reacted in a low molecular weight saturated monovalent aliphatic alcohol, such as methanol, ethanol and isopropanol, with an alkali metal alcoholate, such as as sodium and potassium methylate and ethoxide, with formation of alkoxy, for example, Choline, / methoxy chemins, and alkoxy inclined! is then reacted with a dialkyl xanthine, with subsequent separation of the choline-dialkyl xanthine product.
In particular, choline chloride is reacted in methanol with sodium methoxide, with the formation of methoxy choline which in turn is reacted with theophylline, thus forming the choline product theophylline.
The dialkyl xanthines can be theophylline -1,3-dimethyl xanthine, theobromine-3,7-dimethyl xanthine-, and paraxanthine-1,7-dimethyl xanthine.
According to the publication titled "The Chemistry of Organic Medicinal Products" by Jenkins-Hartung, 3rd Edition, published by John Wiley & Sons, 1949, pages 625-626, the formula for xanthine is:
EMI4.1
Hence, the formula for theophylline is:
EMI4.2
and the formula for theobromine is:
EMI4.3
<Desc / Clms Page number 5>
and that for paraxanthin would be:
EMI5.1
The formation of methoxy choline by reaction between choline chloride and sodium methoxide can be represented by the following equation:
EMI5.2
and the reaction between methoxy inclined and atheophylline can draw represented by the equation! :
EMI5.3
Choline theophylline
The following examples are given by way of illustration of one embodiment of the invention.
EXAMPLE I
1 mole (139.6 g) of choline chloride in solid form is added to 1 mole + 10% excess (59 g) of sodium methoxide in 230 cc of methanol; it is stirred for two hours at room temperature. The sodium chloride formed is collected on a filter and washed with small amounts of cold methanol. A sufficient quantity of CO2 is introduced into the filtrate until there is no more precipitate. Sodium carbonate
<Desc / Clms Page number 6>
formed is removed by filtration. To the filtrate containing methoxy choline is then added 0.9 mole (162.7 g) of anhydrous theophylline and the mixture is heated to about 50 ° C. that all the theophylline is in solution.
Then 100 cc of methanol are removed by vacuum distillation. The reaction mass is allowed to crystallize under cooling. The next day crystals were filtered off under reduced pressure, washed with small amounts of isopropyl alcohol until quite white, and dried at about 100 ° C. The yield was 242 g of choline theophylline or 95% of theory, with a melting point of 188 * -190 * Ce. The mother liquor yielded about 8.5 gr upon subsequent evaporation in vacuum. The melting point of this second product was 180 -181 C.
EXAMPLE II
A theobromine choline product can be obtained by the procedure of Example I, using 1 mole of anhydrous theobromine instead of theophylline.
EXAMPLE III
A theophylline choline product can be produced following the procedure of Example I, using 75 gr. of sodium ethoxide instead of 59 g of sodium methoxide.
The above procedures lead to the preparation of quaternary alkoxy ammonium compounds, such as methoxy chloride, which when reacted with a dialkyl xanthine, such as theophylline, yields products of Quaternary ammonium-dialkyl xanthine, for example, choline-theophylline. These products, as indicated above, are obtained in high yield * and with high purity. In addition, xanthine products are characterized in that they are soluble in water, somewhat soluble in alcohol and chloroform, and either slightly soluble or insoluble in ether, ether. petroleum, acetone and benzene.
<Desc / Clms Page number 7>
In addition, the xanthine compounds, because of their solubility in water, can be readily used in therapy and more especially for oral and parenteral administration.
An advantage of xanthine compounds is that in addition to providing the effect of the xanthine component, such as theophylline, the effect of the quaternary ammonium component, such as choline is also provided.
Further, it should be understood that the term "a dialkyl kanthine," as used hereinafter in the claims, includes derivatives also, containing a substituent in position, such a substituent possibly being a. a chlorine atom or a bromine atom or the nitro group.
The quaternary ammonium chlorune used can be of the general formula:
EMI7.1
in which X, T and Z represent alkyl radicals containing 1 to 5 carbon atoms, and W represents alkyl radicals and hydroxy alkyl radicals containing 2 to 5 carbon atoms,
The quaternary alkoxy ammonium compounds formed can be represented by the general formula:
EMI7.2
in which Y, Z and R represent alkyl radicals containing 1 to 5 carbon atoms; and W represents alkyl radicals and hydroxy alkyl radicals containing 2 to 5 carbon atoms.
<Desc / Clms Page number 8>
EMI8.1
It can be seen that the objects developed above are effectively achieved and, therefore, certain changes can be made in carrying out the above process and in the composition without departing. of the spirit of the invention, it is understood that any matter contained in the above description should be interpreted as illustrative and not in a limiting sense.

It should also be understood that the following rsvondicat3.on are intended to cover all generic and specific characteristics of the invention, described above.
