Composés narcotiques et procédé pour leur préparation
La présente invention comprend des composés narcotiques nouveaux ayant une activité hypnotique plus marquée et un taux , thérapeutique favorable.
Les nouveaux composés suivant l'invention sont des carbamates de carbinols de la formule:

où R1 et R2 sont des groupes alkyles. Dans chaque cas le groupe alkyle est de préférence un groupe alkyle inférieur, c'est-à-dire un group ne contenant pas plus de 4 atomes de carbone.

Un composé particulièrement utile de cette classe est le carbamate de 3-méthyl-1-pentynol-3, c'est-à-dire le carbinol de la formule .ci-dessus,' dans lequel R1 est méthyle' et R2 est éthyle.
L'activité de ce carbamate est très marquée même à de faibles doses, sa' période d'induction est courte et son action prolongée.
En outre il a un taux thérapeutique très favorable.
Les. composés suivant la formule ci-dessus peuvent être produits en mettant en contact un cyanate de métal alcalin ' et une solution comprenant le carbinol de départ, par exemple le 3-méthyl-l-pentynol-3, et l'acide trichloracétique.
Le solvant de cette solution peut être un solvant polaire comme Le dioxane, ou un solvant possédant des caractéristi- ques polaires intermédiaires, comme le dichlorure de méthylène, susceptible de dissoudre. la fois le carbinol et l'acide trichlor- acétique. Le rendement du carbamate est toutefois généralement moins élevé lorsqu'on utilise une telle solution.
Une autre caractéristique de la présente invention est que' de bons rendements du carbamate sont obtenus lorsque le solvant de la solution est constitué, entièrement, ou presqu'entièrement par un excès de carbinol suffisant pour dissoudre l'acide trichlor- acétique, ou lorsqu'on utilise un solvant non polaire comme le tétrachlorure de carbone en présence d'un faible excès du carbinol.
Toutefois, le solvant de la solution peut être constitué principalement par un excès du carbinol, en utilisant en outre un tiers solvant du carbinol et de l'acide.trichloracétique.
La réaction entre le cyânate de métal alcalin et la solu- tion comprenant l'acide trichloracétique et le carbinol peut se dérouler en un temps relativement long, c'est-à-dire quelques jours,à la température ordinaire.-!La réaction peut également s'effectuer plus.rapidement, c'est-à-dire en moins d'un jour, en appliquant de la chaleur,, La température du mélange de réaction, toutefois, ne peut dépasser 80 C et de préférence 50 C, parce que des températures élevées nuisent au rendement et à la pureté du produit finalement obtenu.

L'invention est illustrée par les Exemples qui suivent,
EXEMPLE 1.-
On ajoute 32,4 g (0,4 mole) de cyanate de potassium en poudre fine à une solution de 39,2 g (0,4 mole) de 3-méthyl- 1-pentynol-3 dans 40 cm3 de dioxane anhydre, et on refroidit le mélange dans un bain de glace. On ajoute àlors graduellement une solution de 65,2 g (0,4 mole) d'acide trichloracétique dans 40 cm3 de dioxane anhydre et on agite le mélange jusqu'au lendemain à la température ordinaire. On verse la suspension dans un litre d'eau puis on ajoute 100 cm3 d'éther. L'extrait par l'éther est lavé à l'aide d'une 'solution aqueuse de bicarbonate de sodium et à l'eau.
Après séchage avec du sulfate de sodium anhydre, l'éther est chassé par distillation et l'huile restante est distillée sous une pressio de 16 mm, Le produit brut est recueilli' dans la gamme de 120-121 C.
Ce produit, qui cristallise spontanément en donnant des aiguilles incolore, a un point de 49,5 - 52 C. On le purifie par chromato- graphie et recristallisation du cyclohexane. Le point de fusion du produit purifié est 53,5 - 55 C.
' On trouvera ci-dessous des exemples de modes d'exécution préférée.
EXEMPLE ?-
On ajoute par portions 32,5 g de cyanate de sodium anhydre en remuant, à unesolution de 81,7 g d'acide trichloracétique (préalablement séché dans le vide) dans 100 cm3 de 3-méthyl- l-pentynol-3. Pendant l'addition, la mélange de réaction est re- froidi dans de l'eau glacée, puis- agité à la température ambiante pendant sept jours en le protégeant de l'air humide.
Le mélange de réaction est versé dans l'eau, et l'huile est extraite par l'éther. L'extrait par l'éther est lavé avec une solution saturée de bicarbonate de sodium et finalement à l'eau.
Après avoir été séché sur du sulfate de sodium anhydre l'éther est évaporé et l'excès de 3-méthyl-1-pentynol-3 est séparé par distillation sous une pression de 10 mm dans un bain d'huile à 70 C.

Le résidu est recristallisé d'un mélange de 100 cm3 de cyclohexane et-de 200 cm3 d'éther de pétrole (40-60 C). Le produit final pèse 46,1 g (65% du rendement théorique sur la base de l'acide trichloracétique utilisé) et a un point de fusion de 52-53,5 C.
EXEMPLE 3. -
On ajoute à 1634 g d'acide trichloracétique, séché dans le vide dans le récipient de réaction, 2500 cm3 (2175 g) de 3- méthyl-1-pentynol-3, et 650 g de cyanate de sodium également soigneusement séché. Là suspension, protégée de l'air humide et agitée mécaniquement, est chauffée à 45-50 C pendant 20 heures, puis neutralisée au :pH 7 environ par addition de carbonate de sodium anhydre par'petites portions, Après avoir été refroidi à 30 C environ le mélange est filtré en utilisant un auxiliaire de filtration et le récipient est rincé au tétrachlorure de carbone: ce solvant (environ 200 cm3 en tout) est utilisé pour laver le gâteau de filtration principalement formé de trichloracétate de sodium.
Le tétrachlorure'de carbone'est éliminé du filtrat sous une pression légèrement.'réduite* On sépare alors le méthyl- pentynol n'ayant pas réagi par.distillation sous une pression de 6 mm en chauffant légèrement; la, distillation est arrêtée lorsque la température du résidu a atteint 50 C. On sépare environ 1240 g de méthyl-pentynol. Le 'résidu'brut est alors traité en le versant dans deux volumes d'eau et en agitant à 45 C. Une huile se sépare et se solidifie lorsqu'on refroidit à la température ordinaire en agitant énergiquement. La poudre jaunâtre est recueillie et soigneusement séchée. Rendement 1138 g, point de fusion environ 40 C. La recristallisation du cyclohexane donne un produit presque blanc (940 g), point de fusion 53-55 C.
Le milieu aqueux qui reste lorsque l'huile solidifiée a été recueillie, refroidi dans la glace pendant 24 heures, donne encore 22 g de matière pure, de tell sorte que le rendement totan atteint 962 g (68%).

32.4 g (0.4 mole) of fine powdered potassium cyanate are added to a solution of 39.2 g (0.4 moles) of 3-methyl-1-pentynol-3 in 40 cm3 of anhydrous dioxane, and the mixture is cooled in an ice bath. A solution of 65.2 g (0.4 mol) of trichloroacetic acid in 40 cm3 of dry dioxane is then gradually added and the mixture is stirred overnight at room temperature. The suspension is poured into a liter of water and then 100 cm3 of ether are added. The ether extract is washed with an aqueous solution of sodium bicarbonate and with water.
After drying with anhydrous sodium sulfate, the ether is removed by distillation and the remaining oil is distilled off under a pressure of 16 mm. The crude product is collected in the range of 120-121 C.
This product, which crystallizes spontaneously giving colorless needles, has a point of 49.5-52 C. It is purified by chromatography and recrystallization from cyclohexane. The melting point of the purified product is 53.5 - 55 C.

The residue is recrystallized from a mixture of 100 cm3 of cyclohexane and 200 cm3 of petroleum ether (40-60 C). The final product weighs 46.1 g (65% of theoretical yield based on the trichloroacetic acid used) and has a melting point of 52-53.5 C.
EXAMPLE 3. -
To 1634 g of trichloroacetic acid, dried under vacuum in the reaction vessel, are added 2500 cm3 (2175 g) of 3-methyl-1-pentynol-3, and 650 g of also thoroughly dried sodium cyanate.
