Description
A stand used for assisting flax hybridization
Technical Field The invention relates to plant hybrid breeding technology, in particular to a stand used for assisting flax hybridization. Background Flax (Linum usitatissimum) , it is an important oil crop, suitable for growing in areas with cold, dry and long sunshine hours and distribute in Shanxi, Gansu, Ningxia, Inner Mongolia and Xinjiang in China. Artificial hybridization is the main method for obtaining flax hybrid materials. By artificial hybridization, theoretically the target agronomic traits can be integrated to obtain high-performing hybrid plants and achieve different breeding goals. However, due to unclear descriptions of detasseling time and pollination time, and unclear/untold the relationship between the cross-seed rate and the elongation length of the female stigma in conventional flax hybrid breeding methods, according to the teach from these prior arts, the success rate of cross breeding is extremely low. It is necessary to study and improve flax hybrid technology.
Summary In order to solve the needs in the above-mentioned fields, the inventor of the present invention proposes a technical scheme of a hybridization method, which is simple in operation and can significantly improve the success rate of flax hybridization as follows: A hybridization method for flax, it includes artificial hybridization steps as follow: detassel: in the flax bloom period, after 2-3 hours of sunrise time in the flax cultivation area, on the plants selected as the female parent, select flax flowers whose petals extend 2-4mm unopened and peel off their petals and stamens, bag and set aside; cross-pollinate: 12-24 hours after the detasseling, when the stigmas of the demaleized flax flowers are 3-5mm longer than that before detasseling, collect the stamens of flax flowers just blooming on the plant selected as the male parent, and make them touch the stigma of the demaleized flax flowers, then bag to prevent mixing; wait for natural maturity, harvest hybrid seeds produced by the flowers of the female parent undergoing above artificial hybridization steps.
According to above said hybridization method, wherein each demaleized flax flowers on female parent is pollinated with pollens from 3-5 male parent flax flowers.
A stand used for assisting flax hybridization, it has rigid support unit(1), flexible support unit(2), clamping unit(3), storage unit(4)and ground fixing structure; wherein the lower end of the rigid pillar unit(1) is used to connect to the upper end of the ground fixing structure; the upper end of the rigid pillar unit(1) is configured to be able to install at least one of the flexible support unit(2), the free end of each flexible support unit(2) is provided with a clamping unit(3) which is used to fix the tender shoots of the plants to be hybridized; the storage unit(4) is set on the rigid support unit(1).
According to above said stand used for assisting flax hybridization, wherein the flexible support unit (2) can be arbitrarily straightened, bent and/or folded and keep the shape, and be made of flexible shaped hose or aluminum alloy universal hose material.
According to above said hybridization method, wherein the clamping unit(3) are configured to has two concave clamping surface and has soft layers on the surfaces that used to contact to the plant, wherein the two concave clamping surface do not contact each other.
According to above said stand used for assisting flax hybridization, wherein the length of the rigid pillar unit (1) is adjustable according to the height of the plant, thereby controlling the height of the stand to adapt to the plant' height.
According to above said hybridization method, wherein the ground fixing structure are used to insert into ground and fix the stand, and has a base(5) which is perpendicular to the rigid pillar unit(1)and a plugin unit(6) at the bottom of the rigid pillar unit(1); the storage unit(4)is a box or a tray, its bottom is provided with a magnetic component, which makes the storage unit(4) be able to fix on any level of the rigid pillar unit(1).
The flax hybridization method provided by the present invention is easy to operate. By selecting the time of detasseling and pollination, the success rate of hybridization and the purity of hybridization are improved. Field statistics show that the method of the present invention can achieve a hybridization rate of more than 80%, that is, 80% or more of the hybrid flowers selected produce hybrid seeds.

Embodiments The embodiments of the present invention will be described in detail below in conjunction with examples and figures, but those skilled in the art will understand that the following examples are only for illustrating the present invention and should not be considered as limiting the scope of the present invention. If no specific conditions are indicated in the examples, the conventional conditions or the conditions recommended by the manufacturer shall be used. The reagents or instruments used do not indicate the manufacturer, are all conventional products that can be purchased through the market.
Example 1. Flax hybridization Specific implementation time: June 2015; Implementation area: Yining City, Xinjiang Uygur Autonomous Region Yili Kazakh Autonomous Prefecture Male parent variety: Longya No.8. Female parent variety: Yia No.4. 1. Detasseling: During 2-3 hours after sunrise, on the plant selected as the female parent, select the petals to protrude the sepals 2-4mm unopened flax flower for detasseling. Detasseling operation: peel off petals and stamens with pointed tweezers, wrap the demaleized flax flower with a sulfuric acid paper bag to prevent the female parent from being polluted by pollen from non-target individual plants.
2. Pollination hybridization: 24 hours after detasseling, when the stigma of the demaleized flax flower is 3-5mm longer than that before detasseling, the target single flax plant as male parent is blooming and used to pollinate the the demaleized flax flower artificially. The operation of artificial pollination: Open the sulfuric acid paper bag and touch the stigma of the demaleized flax flower with the stamens of the target single flax plant. Each demaleized flax flower is pollinated with 3-5 flax flower from male parents. After that, the pollinated female parent is bagged with sulfuric acid paper bags to prevent pollen contamination of non-target individual plants. During the bagging step, the head of the female parent cannot be touched. The pollinated female parents are naturally matured and harvested to obtain hybrids. Example 2. Hybridization assisted by the hybridization assisted stand Specific implementation time: June 2016; Implementation area: Yining City, Xinjiang Uygur Autonomous Region Yili Kazakh Autonomous Prefecture Male parent variety: Longya No.10. Female parent variety: Yia No.4.
1. Detasseling: During 2-3 hours after sunrise, on the plant selected as the female parent, select the petals to protrude the sepals 2-4mm unopened flax flower for detasseling. Detasseling operation: peel off petals and stamens with pointed tweezers, wrap the demaleized flax flower with a sulfuric acid paper bag to prevent the female parent from being polluted by pollen from non-target individual plants.

The operation of artificial pollination: Open the sulfuric acid paper bag and touch the stigma of the demaleized flax flower with the stamens of the target single flax plant. Each demaleized flax
flower is pollinated with 3-5 flax flower from male parents. After that, the pollinated female parent is bagged with sulfuric acid paper bags to prevent pollen contamination of non-target individual
plants. During the bagging step, the head of the female parent cannot be touched. In the step 1 detasseling and/or step 2 pollination step, a stand with the following structure is used to assist hybridization, the stand has rigid support unit(1), flexible support unit(2), clamping
unit(3), storage unit(4)and ground fixing structure; wherein the lower end of the rigid pillar unit(1)
is used to connect to the upper end of the ground fixing structure; the upper end of the rigid pillar
unit(1) is configured to be able to install at least one of the flexible support unit(2), the free end of each flexible support unit(2) is provided with a clamping unit(3) which is used to fix the tender shoots of the plants to be hybridized; the storage unit(4) is set on the rigid support unit(1). When in use, the stand is fixed beside the single plant to be hybridized through the ground fixing structure, and the height and angle of the clamping unit are adjusted to be close to the plant to be hybridized and fix the tender shoots of the plant. If there are multiple flexible supporting unit(2) and clamping unit(3), multiple plants can be fixed at the same time, improving the operation efficiency.
In some embodiments, wherein the clamping unit(3) are configured to has two concave clamping surface and has soft layers on the surfaces that used to contact to the plant, wherein the two concave clamping surface do not contact each other.
In some embodiments, wherein the ground fixing structure has a base(5) which is perpendicular to the rigid pillar unit(1)and a plugin unit(6) at the bottom of the rigid pillar unit(1); the storage unit(4)is a box or a tray, its bottom is provided with a magnetic component, which makes the storage unit(4) be able to fix on any level of the rigid pillar unit(1). The success rate of hybridization is comparable to the Example 1. In this example, the number of hybridized flowers is about twice that of Example 1. The required hybridization operation time is less than the hybridization time in Example 1. That is, the use of the above-mentioned auxiliary hybridization scaffold ensures that Hybridization efficiency, the efficiency of the operation process has more than doubled.
