Verfahren zur Herstellung eines neuen therapeutisch wertvollen
Calciumsalzes
Die hohe tuberkulostatische Wirksamkeit des Isonicotinsäurehydrazids und seiner Hydrazone ist bekannt. Während das Isonicotinsäurehydrazid hinreichend wasserlöslich ist, um injektionsfähigelösungen zu liefern, sind die meisten seiner Hydrazone mit Oxoverbindungen nicht löslich genug, um zu Injektionen und Infusionen verwendet zu werden. Anderseits zeichnen sich die Hydrazone zum Teil durch eine bessere Verträglichkeit aus.
Es besteht ein Bedürfnis danach, die wertvollen Eigenschaften des Isonicotinsäurehydrazids und seiner Hydrazone - eine hohe tuberkulostatische Aktivität, beste Verträglichkeit und ausreichende Wasser- löslichkeit-in einer Verbindung zu vereinigen.
Es wurde nun gefunden, dass das Calciumsalz des Hydrazons aus Isonicotinsäurehydrazid und Lävu- linsäure in besonderem Masse diesen Anforderungen entspricht. Diese neue Verbindung und ihre Hydrate, wie z. B. das Dihydrat, die durch nachstehende Formel wiedergegeben wird :

lässt sich erfindungsgemäss dadurch herstellen, dass man Calciumlävulinat mit Isonicotinsäurehydrazid umsetzt.
Die tuberkulostatische Wirkung, auf die Gewichtseinheit bezogen, entspricht überraschenderweise durchaus derjenigen des Isonicotinsäurehydrazids, obwohl nach dem Vergleich der Molekulargewichte (544 : 137) eine wesentlich geringere Wirksamkeit, etwa entsprechend der Menge des durch Hydrolyse abspaltbaren Isonicotinsäurehydrazids, zu erwarten gewesen wäre. In vitro-Versuche an Mycobacterium tuberculosis Typus humanus von normaler Empfindlichkeit zeigten eine totale Hemmwirkung der neuen Ca-Verbindung noch in Verdünnungen bis zu 1 : 50000000. Bei einem andern Tuberkelbazillen-Stamm war volle Hemmwirkung bei Anwendung von Isonicotinsäurehydrazid sowie vom Verfahrensprodukt bei Verdünnungen von l : Z5000000 zu beobachten, während beide Verbindungen bei einer Verdünnung von 1 : 50000000 das Wachstum dieser Bazillen nicht mehr hemmten.
Erwartungsgemäss entsprechen diesen in vitro erhobenen Befunden die Beobachtungen Im Tierexperiment. Bei Meerschweinchen, die mit Tuberkelbazillen des Typus humanus in der Leistenbeuge infiziert waren, wurde durch 20-malige Gabe von 5 bzw. 10 mg/kg der neuen Ca-Verbindung eine Generalisierung der Tuberkulose verhindert. In einem weiteren Versuch wurden Kaninchen intravenös mit 1 mg Tuberkelbazillen des Typus humanus R H 37 infiziert. Je 2 Tiere erhielten 2, 5. 5,0 bzw. 10,0 mg/kg dieses Präparates paranteral injiziert. Schon eines der mit 2,5 mg/kg behandelten Tiere zeigte bei der Sektion völliges Freisein der Organe von Tuberkeln, während die Kontrollen inder Lunge zahlreiche miliare Herde aufwiesen und zum Teil vor der Sektion an Tuberkulose verstorben waren.

Bei gleichwertiger tuberkulostatischer Aktivität in vitro und in vivo übertrifft die neue Verbindung dasIsonicotinsäurehydrazid an Verträglichkeit, wie aus dem in beifolgender Tabelle gegebenen Vergleich der mittleren tödliche Dosen (der sogenannten LD 50) hervorgeht.

Die Löslichkeit der neuen Calciumverbindung in Wasser ist mit zirka 110 bei 200C ausreichend, um das Präparat für Injektionen und besonders für Dauertropfinfusionen zu verwenden. Derartige Lösungen sind nach bekannten Methoden sterilisierbar, ohne Veränderungen zu erleiden.
Die Darstellung des Calciumsalzes des Hydrazons aus Lävulinsäure und Isonicotinsäurehydrazid kann so erfolgen, dass man eine Lösung von Calciumlävulinat mit einer Lösung von Isonicotinsäurehydrazid vereinigt. Eine günstige Ausführungsform des Verfahrens wird in folgendem Beispiel beschrieben.

lende Niederschlag, der hauptsächlich aus dem Hydrazon der Lävulinsäure mit Isonicotinsäurehydrazid besteht, wird abgesaugt. Aus dem Filtrat kristallisieren nach einigem Stehen in der Kälte 255 g des gewünschten Calciumsalzes, die abgesaugt und getrocknet werden.
Der abgetrennte, aus dem Hydrazon der Lävulinsäure bestehende Niederschlag kann auf folgende Weise zur Überführung in das gewünschte Endprodukt weiter verarbeitet werden :
Der Niederschlag wird in siedendem Wasser gelöst und diese Lösung auf Calciumcarbonat gegossen.
Man rührt bis zum Ende der Kohlensäureentwicklung, saugt heiss vom überschüssigen Calciumcarbonat ab und engt das Filtrat im Vakuum ein. Nach gutem Kühlen saugt man das gewünschte Calciumsalz ab.

The high tuberculostatic effectiveness of isonicotinic acid hydrazide and its hydrazones is known. While the isonicotinic hydrazide is sufficiently water soluble to provide injectable solutions, most of its hydrazones with oxo compounds are not soluble enough to be used for injections and infusions. On the other hand, some of the hydrazones are characterized by better compatibility.
There is a need to combine the valuable properties of isonicotinic hydrazide and its hydrazones - high tuberculostatic activity, best tolerability and adequate water solubility - in one compound.
It has now been found that the calcium salt of the hydrazone made from isonicotinic acid hydrazide and levulic acid meets these requirements to a particular extent. This new compound and its hydrates, such as B. the dihydrate, which is represented by the following formula:

can be produced according to the invention by reacting calcium levulinate with isonicotinic acid hydrazide.
The tuberculostatic effect, based on the weight unit, surprisingly corresponds to that of isonicotinic acid hydrazide, although a comparison of the molecular weights (544: 137) would have expected a significantly lower effectiveness, roughly corresponding to the amount of isonicotinic acid hydrazide that can be split off by hydrolysis. In vitro tests on Mycobacterium tuberculosis type humanus of normal sensitivity showed a total inhibitory effect of the new Ca compound even in dilutions of up to 1: 50,000,000 : Z5000000 can be observed, while both compounds no longer inhibited the growth of these bacilli at a dilution of 1: 50000000.
As expected, these in vitro findings correspond to the observations in animal experiments. In guinea pigs infected with tubercle bacilli of the humanus type in the groin, 20 doses of 5 and 10 mg / kg of the new Ca compound prevented generalization of tuberculosis. In a further experiment, rabbits were infected intravenously with 1 mg of tubercle bacilli of the humanus R H 37 type. 2 animals each received 2, 5, 5.0 and 10.0 mg / kg of this preparation by paranteral injection. Even one of the animals treated with 2.5 mg / kg showed that the organs were completely free of tuberculosis during the dissection, while the controls had numerous miliary foci in the lungs and some had died of tuberculosis before the dissection.

With equivalent tuberculostatic activity in vitro and in vivo, the new compound is more tolerable than isonicotinic acid hydrazide, as can be seen from the comparison of the mean lethal doses (the so-called LD 50) given in the table below.

The solubility of the new calcium compound in water is around 110 at 200C, sufficient to use the preparation for injections and especially for continuous drip infusions. Such solutions can be sterilized by known methods without suffering any changes.
The preparation of the calcium salt of the hydrazone from levulinic acid and isonicotinic acid hydrazide can be carried out by combining a solution of calcium levulinate with a solution of isonicotinic acid hydrazide. A favorable embodiment of the method is described in the following example.
Example: 250 g of crystallized levulinic acid are dissolved in 1000 cm3 of 750C warm water and 110 g of calcium carbonate are gradually added while stirring. The liquid is stirred on the steam bath until the vigorous foaming has stopped, with 308 g of isonicotinic acid hydrazide, in 600 ems of sieve.
