Verfahren zur Darstellung von Nebennierenrindenpräparaten.
In dem Patent Nr. 142710 ist ein Verfahren zur Darstellung von Nebennierenrindenpräparaten, die frei von 1- (3. 4-Dioxyphenyl)-2-methylaminoäthanol-l sind, beschrieben, das darin besteht, dass man die Nebennierenrinde oder beliebige aus ihr gewonnene Präparate oder auch die gesamte Nebenniere oder ihre Präparate mit Formaldehyd oder Formaldehyd abspaltenden Verbindungen behandelt und gegebenenfalls nach erfolgter Einwirkung den überschüssigen Formaldehyd entfernt.
Bei weiterer Bearbeitung dieses Gebietes wurde gefunden, dass es besonders für die Technik zweckmässig ist, bei der Darstellung von kein 1-(3,4-Dioxyphenyl)-2-methylaminoäthanol-1 enthaltenden Lösungen des Nebennierenrindenhormons gemäss dem Verfahren des Stammpatentes Nr. 142710 zunächst die Hauptmenge des in den Extrakten enthaltenen 1- (3. 4-Dioxyphenyl) -2-methyl- aminoäthanol-t durch Zusatz von Ammoniak zur Abscheidung zu bringen und das nach einigem Stehen ausgefällte 1-(3,4-Dioxyphenyl)-2-methylaminoäthanol-1 durch Filtration zu entfernen.

Handlung der so vorgereinigten Extrakttösungen mit Formaldehyd oder Formaldehyd abspaltenden
Mitteln gemäss dem Verfahren des Stammpatentes Nr. 142710 werden die noch in den Extrakten ver- bliebenen geringen Mengen 1-(3,4-Dioxyphenyl)-2-methylaminoäthanol-1 unwirksam gemacht. Zur
Vermeidung von Zersetzungen des Nebennierenrindenhormons führt man die Ausfällung des 1- (3'4- Dioxyphenyl) -2-methylaminoäthanol-l mit Ammoniak in der Kälte aus und verwendet zweckmässig das Ammoniak unter Vermeidung eines grösseren Überschusses.
Man kann das Verfahren beispielsweise so ausführen, dass man nach beliebigen Verfahren gewonnene Nebennierenrindenextrakte nach und nach mit einer verdünnten, wässrigen Ammoniaklösung unter guter Kühlung versetzt, bis die Lösung deutlich ammoniakalisch reagiert. Man lässt das
Ganze in Eis stehen, saugt oder schleudert von ausgefallenem 1- (3. 4-Dioxyphenyl) -2-methylamino- äthanol-t ab. Für die weitere Behandlung des Filtrates gemäss Patent Nr. 142710 ist es nicht erforderlich, den Überschuss an Ammoniak vorher zu neutralisieren, da das übersehüssige Ammoniak mit Formaldehyd in Hexamethylentetramin übergeführt wird.
Mit gleichem Erfolge lässt sich das Verfahren auch so ausführen, dass man die Entfernung des 1-(3,4-Dioxyphenyl)-2-methylaminoäthanol-1 mit Hilfe von Ammoniak bereits bei der Darstellung der Extrakte vornimmt ; z. B. wird die gesamte Nebenniere bzw. die Nebennierenrinde mit schwach ammoniakalischem Wasser extrahiert, der erhaltene wässrige Extrakt von ungelösten wässrigen Bestandteilen befreit und darauf dem Verfahren gemäss dem Stammpatente unterworfen.
Das Verfahren hat den Vorzug, dass es gleichzeitig die technische Gewinnung des Nebennierenmarkhormons und des Nebennierenrindenhormons gestattet, und dass die gewonnenen Extraktlösungen des Nebennierenrindenhormons nur wenig unspezifische Ballaststoffe enthalten.

Ein nach beliebigem Verfahren hergestellter Nebennierenextrakt, der einen starken Gehalt an 1- (3. 4-Dioxyphenyl)-2-methylaminoäthanol-l aufweist, wird soweit im Vakuum unter CO2-Atmosphäre eingeengt, dass 1 cm3 Lösung = etwa 10-15 g Drüsen entspricht. Unter guter Kühlung wird so lange konzentrierte Ammoniaklösung zugesetzt, bis Curcumapapier deutlich gebräunt wird. Reibt man jetzt mit dem Glasstab ab, so beginnt das 1- (3. 4-Dioxyphenyl)-2-methylaminoäthanol-1 auszukristallisieren. Sobald die Fällung vollständig ist, wird abgesaugt und das Filtrat zur Entfernung letzter, in Lösung verbliebener Reste des 1- (3. 4-Dioxyphenyl)-2-methylaminoäthanol-1 gemäss dem Verfahren des Stammpatentes mit Formaldehyd behandelt.
Sodann wird die Lösung mit Salzsäure auf schwach saure Reaktion eingestellt.
Da in manchen Fällen die bei obigem Verfahren entstehenden Ammoniaksalze zweckmässiger weise vermieden werden sollen, können die Extrakte bis zum Neutralpunkt mit verdünnter Natronlauge versetzt und erst dann mit Ammoniaklösung behandelt werden. Mit sehr geringen Mengen Ammoniaklösung kommt man aus, wenn man dem meist sauren oder neutralen Nebennierenextrakt eine kleine Menge Ammoniaklösung oder einer wässrigen Lösung eines Ammoniumsalzes, wie z. B.
Ammoniumchlorids, zusetzt und dann so lange verdünnte Natronlauge zugibt, bis das gebundene Ammoniak frei wird und das 1- (3. 4-Dioxyphenyl) -2-methylaminoäthanol ausfällt.
