Abstract
Se describen compuestos que de forma selectiva modulan negativamente los receptores de NMDA que contienen una subunidad NR2A, composiciones farmacéuticas que comprenden los compuestos y métodos para tratar una enfermedad utilizando los compuestos. Reivindicación 1: Un compuesto de fórmula (1) o una sal, profármaco, solvato, hidrato, tautómero o estereoisómero farmacéuticamente aceptable de este donde: A, B, C y D son independientemente CR o N siempre que al menos un A, B, C o D sea N; cada R se selecciona independientemente del grupo que consiste en H, halógeno, alquilo C₁₋₅, haloalquilo C₁₋₅, CN y O-alquilo; o cuando ambos A y B son CR, los grupos R en A y B pueden formar, con A y B, un grupo arilo bicíclico 5,6 ó 6,6 o grupo heteroarilo; R¹ es arilo o heteroarilo ambos están opcionalmente sustituidos con uno o más sustituyentes que se seleccionan del grupo que consiste en halógeno, alquilo C₁₋₅, alquilo -C(O)C₁₋₅, haloalquilo, CN, O-arilo, O-alquilo, arilo, y heteroarilo; o R¹ se selecciona del grupo que consiste en alquilo opcionalmente sustituido, cicloalquilo opcionalmente sustituido o heterocicloalquilo opcionalmente sustituido; R² y R² se seleccionan independientemente del grupo que consiste en H, y alquilo C₁₋₅; o R² y R² pueden formar, con el carbono con el que están conectados, un cicloalcano; X es C=O o SO₂; Y se selecciona del grupo que consiste en H, alquilo C₁₋₅, y un grupo -CHP¹P²; P¹ se selecciona del grupo que consiste en H, y alquilo C₁₋₅; P² es alquilo O(C₁₋₅) opcionalmente sustituido con uno o más grupos hidroxi y/o alcoxi; o P² se selecciona del grupo que consiste en OC(O)R⁴, OC(O)OR⁴, OC(O)NHR⁴, OC(O)NR⁴R⁵, OC(O)-alquilo-NR⁴R⁵, y O(PO)O₂⁻²; o P¹ y P², forman un cicloalquilo o un heterociclo; R⁴ y R⁵ se seleccionan independientemente del grupo que consiste en H, y alquilo C₁₋₅ con la condición de que cuando P² es OC(O)OR⁴, R⁴ no es hidrógeno; R³ es -L¹-Ar¹, L¹-N(alquilo C₁₋₆)₂, o un resto de fórmula (2); L¹ es alquilo C₁₋₅ lineal o ramificado, opcionalmente sustituido con -OH, alquilo -OC₁₋₆, o (=O); Z se selecciona del grupo que consiste en H y alquilo C₁₋₅; o Z y R³, con el nitrógeno al que están conectados pueden formar un heterociclo; Ar¹ es arilo opcionalmente sustituido con uno o más sustituyentes que se seleccionan del grupo que consiste en alquilo C₁₋₅, halógeno, haloalquilo C₁₋₆, CN, y O-alquilo; o Ar¹ es heteroarilo opcionalmente sustituido con uno o más sustituyentes que se seleccionan del grupo que consiste en alquilo C₁₋₅, halógeno, haloalquilo C₁₋₆, CN, y O-alquilo; y n es 1 ó 2.
