Abstract

Se estipulan métodos y composiciones que emplean microRNA (miARN) que, cuando se expresa en una célula vegetal, tiene la capacidad de reducir el nivel de mARN de una secuencia objetivo (es decir, secuencia endógena) sin reducir el nivel de mARN de una o más secuencias estrechamente relacionadas. Mientras que se pueden diseñar miARN con especificidad para una secuencia objetivo particular, la aplicación instantánea demuestra que un miARN puede silenciar específicamente una secuencia objetivo sin silenciar una secuencia estrechamente relacionada que tiene una alta identidad de secuencia con la secuencia objetivo. En ciertas modalidades, una secuencia objetivo endógena puede suprimirse con una construcción de expresión de miARN recombinante sin silenciar un polinucleótido de interés recombinante que tenga una secuencia estrechamente relacionada con la secuencia objetivo. Tales métodos y composiciones emplean construcciones de expresión de miARN recombinante que producen un miARN de 21 nt. Se estipulan, además, células de plantas transgénicas, plantas y semillas que incorporan construcciones de expresión de miARN y construcciones de polinucleótidos recombinantes que comprenden polinucleótidos de interés.
