Abstract
Reivindicación 1: Un método para manufacturar semilla de maíz transgénica con un aumento del nivel de un nutriente, caracterizado porque comprende: (a) seleccionar una primera planta de maíz transgénica que comprende la construcción de DNA recombinante que comprende: un primer promotor ligado operativamente a por lo menos un primer elemento de supresión génica para silenciar al menos un primer gen objetivo, y un segundo promotor que está en la orientación opuesta a dicho primer promotor y está situado 3’ a dicho al menos un primer elemento de supresión génica, donde dichos primer y segundo promotores comprenden un promotor vegetal específico de embrión y un promotor vegetal específico de endosperma y donde la transcripción de dicha construcción de ADN recombinante en una célula vegetal da como resultado el silenciamiento de dicho al menos un primer gen objetivo, caracterizada porque dicho por lo menos un primer gen objetivo es un gen del catabolismo de un nutriente seleccionado entre un aminoácido, un lípido o un carbohidrato; (b) efectuar la introgresión de dicha construcción de DNA recombinante en una segunda planta de maíz; (c) cultivar una semilla de la segunda planta de maíz para producir una población de plantas de maíz de progenie; (d) evaluar la población de plantas de maíz de progenie para seleccionar las plantas de maíz de progenie que producen semilla de maíz con un nivel aumentado de dicho nutriente, con relación a las plantas de maíz no transgénicas; (e) seleccionar de dicha población una o más plantas de maíz de progenie que producen semilla de maíz con un aumento del nivel de dicho nutriente, con relación a las plantas de maíz no transgénicas; (f) verificar que dicha construcción de DNA recombinante esté integrada de manera estable en dichas plantas de maíz de progenie seleccionadas; (g) verificar que dicho gen del catabolismo de dicho nutriente esté silenciado en dichas plantas de maíz de progenie seleccionadas, con relación a las plantas de maíz que carecen de dicha construcción de DNA recombinante; (h) recolectar una semilla de maíz transgénica de dichas plantas de maíz de progenie seleccionadas.Reivindicación 3: Un núcleo de una célula de una planta, caracterizado porque dicho núcleo tiene por lo menos un cromosoma que comprende la construcción de DNA recombinante que comprende: un primer promotor ligado operativamente a por lo menos un primer elemento de supresión génica para silenciar al menos un primer gen objetivo, y un segundo promotor que está en la orientación opuesta a dicho primer promotor y está situado 3’ a dicho al menos un primer elemento de supresión génica, donde dichos primer y segundo promotores comprenden un promotor vegetal específico de embrión y un promotor vegetal específico de endosperma y donde la transcripción de dicha construcción de ADN recombinante en una célula vegetal da como resultado el silenciamiento de dicho al menos un primer gen objetivo, caracterizada porque dicho por lo menos un primer gen objetivo es un gen del catabolismo de un nutriente seleccionado entre un aminoácido, un lípido o un carbohidrato.
