Abstract
Método para analizar una region genomica de un organismo, que comprende cuatro partes principales. La primera parte involucra el aislamiento de ARNm de un organismo seleccionado que se utiliza para la preparacion de pequenos fragmentos de ADN de unica hebra con un adaptador que contiene una etiqueta de afinidad. Estos fragmentos de ADN se utilizan en la parte tres. En la segunda parte, se aísla ADN genomico del mismo organismo o de un organismo relacionado. Este ADN genomico es fragmentado y ligado a moléculas adaptadoras. En la tercera parte, estos fragmentos genomicos son hibridados con fragmentos de ADN de unica hebra de la parte uno y los híbridos formados en este proceso son utilizados para la síntesis de fragmentos de ADN. Estos fragmentos serán utilizados en la parte cuatro, la cual involucra la secuenciacion de estos fragmentos utilizando uno de los métodos de secuenciacion de alto rendimiento disponibles. El método descripto hace posible determinar secuencias en una parte seleccionada del ADN genomico que representa las regiones codificantes de la mayoría de genes expresados. La comparacion de dichas partes seleccionadas entre diferentes individuos permite la identificacion de sitios polimorficos. Esto puede dar como resultado el descubrimiento de marcadores por cada característica, siendo la obtencion de marcadores util en tecnologías de mejoramiento genético.
