Abstract

Un método para detectar la presencia de una molécula objetivo en una muestra y determinar su peso molecular y concentracion. El método involucrala determinacion de velocidades electrocinéticas midiendo el tiempo requerido para que moléculasindividuales rotuladas con por lo menos dos sondasfluorescentes se desplacen a través de una distancia fija entre dos haces láser. La comparacion de la velocidad electroforética conocida comocaracterística para los estándares de peso molecularparticulares con la velocidad electroforética de las especies objetivo permiten la determinaciondel peso molecular del objetivo. Alternativamente, un método con una unica zona deteccion y dos sondas que utiliza flujo mecánico, electrosmotico oelectrocinético de una muestra a través de un tubo de transporte que permite la identificacion de una molécula objetivo en una muestra de ensayo compleja. Elmétodo es sumamente sensible y permite detectar y determinar en cuestion de segundos el peso molecular de una molécula objetivo presente en unaconcentracion de tan solo un femtomolar.
