Abstract

PROBE ZAKLANJANJA PARTNER PROTEINA I NJIHOVA PRIMENA, opisan je brz, jednostavan i jeftin postupak zaklanjanja i klasifikacije proteina kao partnera heterodimernih proteina koji koristi konstrukcije fuzionog proteina sa područjem vezivanja DNA i dimerizacionim područjem iz različitog proteina. Prema postupku ovog pronalaska, heterodimerna formacija se detektuje sposobnošću proteinskog partnera da premesti homodimernu formaciju i tako ostvari fenotipsku promenu u bakterijskom domaćinu koji je zavisan od održavanja homodimerne konfiguracije. Postupak prema ovom pronalasku može da se koristi za identifikaciju jedinjenja od interesa koja inhibiraju takvu heterodimernu formaciju, a posebno za identifikaciju jedinjenja koja sprečavaju heterodimernu formaciju i aktivisanje onkogenih transkripcionih regulatornih proteina.
