Abstract
본 발명은 갈조류로부터 이타콘산을 생산하는 신규한 재조합 비브리오 속 DHG 균주 및 이를 이용한 이타콘산의 생산방법에 관한 것이다. 본 발명의 일 실시예에 따른 재조합 미생물은 알긴산 및 만니톨을 이용하여 이타콘산을 생산할 수 있을 뿐만 아니라, 갈조류의 전처리 공정 없이 직접 투입하여도 이타콘산을 생산할 수 있는바 친환경적이고 저비용으로 이타콘산 고생산 공정 개발이 가능하여 합성수지, 라텍스 및 식품첨가물 등의 산업 분야에서 다양하게 활용될 수 있다.
Description
본 발명은 이타콘산을 생산하는 재조합 미생물에 관한 것으로서, 더욱 상세하게는 갈조류로부터 이타콘산을 생산하는 신규한 재조합 비브리오 속 DHG 균주 및 이를 이용한 이타콘산의 생산방법에 관한 것이다.
플랫폼 화합물의 일종인 이타콘산(itaconic acid)은 탄소 5 개로 이루어진 디카르복실산이며, 구조적 특성 덕에 산업적으로는 플라스틱 및 라텍스 등과 같은 고분자 합성의 전구체로 이용될 수 있다. 이러한 잠재성으로 인하여, 2004년에는 미국 에너지부(United States Department of Energy)의 탑 12 바이오-기반 플랫폼 화합물(Top 12 bio-based platform chemicals)에도 선정되었다.
이타콘산의 고효율 생산을 위한 대사공학적 접근법이 이미 다양하게 알려져 있다. 이와 관련된 여러 연구에 따르면, 다양한 균주와 탄소원에 대하여 각각 대사 경로를 최적화하여 생산성을 최대화하고 있다. 현재 미생물 발효를 통한 이타콘산의 생산은 아스퍼질러스 테레우스(Aspergillus terreus) 라고 하는 곰팡이 균을 이용하여 포도당이나 육상 식물 (옥수수 등)을 수확하여 얻은 당으로부터 대부분 생산되고 있는 실정이다.
다만 상기 아스퍼질러스 테레우스를 이용한 이타콘산 생산의 경우 미생물의 유전자 조작이 쉽지 않아 효과적인 대사공학적 기법을 도입하는 것에 한계가 있다. 또한 대부분 식량 자원으로 사용되는 사탕수수나 옥수수로부터 얻어진 당으로부터 이타콘산을 생산하기 때문에 윤리적 문제에 대한 비판의 여지도 존재한다.
또한 이러한 한계를 극복하기 위하여 플랫폼 균주인 대장균이나 효모를 통해서 이타콘산을 생산하는 경우에도, 공급 원료로 대부분 포도당이나 시트르산 등을 이용하는 경우가 많기 때문에 공급 원료의 비용이 전반적인 공정 및 이타콘산의 생산 비용을 상승시키는 주요 원인으로 작용하여 경제성을 하락시킨다는 문제점이 있다.
이에 본 발명자들은 전술한 바와 같은 종래기술의 문제점을 해결하기 위하여, 갈조류로부터 이타콘산을 생산할 수 있는 미생물을 개발함으로써 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은, 시스-아코니트산 탈탄산효소(cis-aconitate decarboxylase, cad) 유전자를 포함하는 cad 유전자 발현 카세트를 포함하는 이타콘산(itaconic acid) 생산용 재조합 벡터를 제공하는 것이다.
본 발명의 다른 목적은, 상기 이타콘산 생산용 재조합 벡터가 도입된 이타콘산 생산용 재조합 미생물을 제공하는 것이다.
본 발명의 또 다른 목적은, 상기 이타콘산 생산용 재조합 미생물을 배양하는 단계를 포함하는 이타콘산의 생산방법을 제공하는 것이다.
상기와 같은 목적을 달성하기 위하여, 본 발명의 일 실시예에 따르면 시스-아코니트산 탈탄산효소(cis-aconitate decarboxylase, cad) 유전자를 포함하는 cad 유전자 발현 카세트를 포함하는 이타콘산(itaconic acid) 생산용 재조합 벡터를 제공한다.
본 발명의 일 실시예에 있어서, "이타콘산(itaconic acid)”은 탄소 5 개로 이루어진 디카르복실산으로, 구조적 특성으로 인하여 플라스틱 및 라텍스 등과 같은 고분자 합성의 전구체로 이용된다.
본 발명의 일 실시예에 있어서, “cad 유전자”는 아스퍼질러스 테레우스 유래의 시스-아코니트산 탈탄산효소(cis-aconite decarboxylase)를 발현하는 유전자를 의미하며, 상기 시스-아코니트산 탈탄산효소는 미생물 내의 시스-아코니트산을 탈카복실화하여 이타콘산으로 전환시킨다.
본 발명의 일 실시예에 있어서, 상기 cad 유전자는 아스퍼질러스 테레우스 유래의 cad 유전자(Shin, et al, 2008)를 비브리오 속 DHG 균주에 맞게 코돈 최적화한 cad 유전자일 수 있다. 상기 코돈 최적화한 cad 유전자는 서열번호 1의 염기서열로 표시되는 것이 바람직하나, 이에 제한되지 않는다.
본 발명의 일 실시예에 있어서, "발현 카세트"는 프로모터와 목적 단백질을 코딩하는 유전자를 포함하고 있어서, 프로모터 하류에 작동 가능하게 연결되어 있는 목적 단백질을 생산하기 위해 발현시킬 수 있는 단위 카세트를 의미한다. 이와 같은 발현 카세트의 내부 또는 외부에는 상기 목적 단백질의 효율적인 생산을 도울 수 있는 다양한 인자가 포함될 수 있다. 상기 목적 단백질 발현 카세트는 구체적으로, 프로모터 서열 하류에 목적 단백질을 코딩하는 유전자가 작동 가능하게 연결된 것일 수 있다.
상기 "작동 가능하게 연결"이란 본 발명의 프로모터 활성을 갖는 핵산 서열이 목적 단백질을 코딩하는 유전자의 전사를 개시 및 매개하도록, 상기 유전자 서열과 프로모터 서열이 기능적으로 연결되어 있는 것을 의미한다. 작동 가능한 연결은 당업계의 공지된 유전자 재조합 기술을 이용하여 제조할 수 있으며, 부위-특이적 DNA 절단 및 연결은 당업계의 절단 및 연결 효소 등을 사용하여 제작할 수 있으나, 이에 제한되지 않는다.
본 발명의 일 실시예에 있어서, “재조합 유전자 발현 카세트”는 숙주세포의 염색체 내에 삽입되어 재조합 미생물을 제조하는 데 사용될 수 있으며, 본 발명이 속하는 기술 분야의 당업자에게 있어 상기 재조합 유전자 발현 카세트를 숙주세포의 게놈 염색체에 삽입하여도 상기와 같이 재조합 벡터를 숙주세포에 도입한 경우와 동일한 효과를 가질 것은 자명하다.
재조합 유전자 발현 카세트를 숙주세포의 염색체상에 삽입하는 방법으로는 통상적으로 알려진 유전자조작방법을 사용할 수 있으며, 일 예로는 레트로바이러스 벡터, 아데노바이러스 벡터, 아데노-연관 바이러스 벡터, 헤르페스 심플렉스 바이러스 벡터, 폭스바이러스 벡터, 렌티바이러스 벡터 또는 비바이러스성 벡터를 이용하는 방법이있다.
본 발명의 일 실시예에 있어서, "벡터"는 적합한 숙주 내에서 목적 유전자를 발현시킬 수 있도록 적합한 조절 서열에 작동 가능하게 연결된 유전자의 염기서열을 포함하는 유전자 제작물을 의미하는 것으로, 상기 조절 서열은 전사를 개시할 수 있는 프로모터, 그러한 전사를 조절하기 위한 임의의 오퍼레이터 서열, 및 전사 및 해독의 종결을 조절하는 서열을 포함할 수 있다. 본 발명의 벡터는 세포 내에서 복제 가능한 것이면 특별히 한정되지 않고 당업계에 알려진 임의의 벡터를 이용할 수 있으며, 예컨대 플라스미드, 코즈미드, 파지 입자, 바이러스 벡터일 수 있다.
본 발명의 일 실시예에 있어서, "재조합 벡터"는 발현시키고자 하는 목적 폴리펩타이드의 암호화 유전자가 작동 가능하게 연결될 경우, 적절한 숙주 세포에서 상기 목적 폴리펩타이드를 높은 효율로 발현시킬 수 있는 목적 폴리펩타이드의 발현 벡터로 사용될 수 있으며, 상기 재조합 벡터는 숙주 세포에서 발현 가능할 수 있다. 숙주 세포는 바람직하게는 진핵세포일 수 있으며, 숙주세포의 종류에 따라 프로모터(promoter), 종결자(terminator), 인핸서(enhancer) 등과 같은 발현 조절 서열, 막 표적화 또는 분비를 위한 서열 등을 적절히 선택하고 목적에 따라 다양하게 조합할 수 있다.
또한 본 발명의 일 실시예에 따르면 상기 이타콘산 생산용 재조합 벡터가 도입된 이타콘산 생산용 재조합 미생물을 제공한다.
본 발명의 일 실시예에 있어서, 상기 “재조합 미생물”은 본 발명의 재조합 벡터로 형질전환된 것을 말한다. 본 발명의 일 실시예에서 "형질전환"은 본 발명에 따른 프로모터, 또는 추가적으로 목적 단백질을 코딩하는 유전자를 포함하는 벡터를 숙주세포 내에 도입하는 것을 의미한다. 또한, 형질전환 된 목적 단백질을 코딩하는 유전자는 숙주세포 내에 발현될 수 있기만 한다면, 숙주세포의 염색체 내에 삽입되어 위치하거나 염색체 외에 위치할 수 있다.
본 발명의 일 실시예에 있어서, 상기 이타콘산 재조합 미생물은 본 발명의 재조합 벡터를 포함하는 하나 또는 복수 개의 재조합 벡터가 도입될 수 있으며, 상기 하나 또는 복수 개의 재조합 벡터가 각각 미생물에 도입될 수 있다. 또한, 상기 재조합 벡터는 미생물에 순차적으로 도입될 수 있고, 상호 간 순서가 바뀌어 도입될 수도 있다.
본 발명의 일 실시예에 있어서, 상기 이타콘산 생산용 재조합 미생물은 이소시트르산 탈수소효소(isocitrate dehydrogenase, icd) 유전자가 추가로 결실된 것일 수 있다. 상기 icd 유전자는 서열번호 2의 염기서열로 표시되는 것이 바람직하나, 이에 제한되지 않는다.
본 발명의 일 실시예에 있어서, “icd 유전자”는 이소시트르산 탈수소효소(isocitrate dehydrogenase)를 발현하는 유전자를 의미하며, 상기 이소시트르산 탈수소효소는 TCA 회로 내에서 이소시트르산(isocitrate)을 알파-케토글루타르산(alpha-ketoglutarate)으로 전환시킨다.
본 발명의 일 실시예에 있어서, 상기 이타콘산 생산용 재조합 미생물은 박테리아, 효모, 곰팡이로 구성된 군에서 선택되는 것을 특징으로 할 수 있고, 바람직하게는 비브리오 속 균주일 수 있고, 더욱 바람직하게는 수탁번호 KCTC 19196P로 기탁된 재조합 비브리오 속 DHG 균주일 수 있다.
본 발명의 일 실시예에 있어서, 상기 이타콘산 생산용 재조합 미생물은 알긴신(alginic acid) 및 만니톨(mannitol)을 이용하여 이타콘산을 생산하는 것일 수 있다. 알긴산은 갈조류 세포벽에 함유된 다당류 산이며, 만니톨은 D-만노오스의 환원 형태인 당알코올로 자연계에 널리 분포하며, 고구마, 버섯, 셀러리 외에, 다시마 등의 갈조류에 많이 함유되어 있다.
본 발명의 일 실시예에 있어서, 상기 알긴산 및 만니톨은 원재료로부터 추출되어 정제된 것을 포함할 뿐만 아니라, 추출 공정을 거치지 않아 원재료에 비정제 상태로 함유된 알긴산 및 만니톨을 포함한다.
본 발명의 일 실시예에 있어서, 상기 이타콘산 생산용 재조합 미생물은 해조류를 이용하여 이타콘산을 생산하는 것일 수 있고, 바람직하게는 갈조류를 이용하여 이타콘산을 생산하는 것일 수 있다.
* 본 발명의 일 실시예에 있어서, "해조류"는 일반적으로 바다에 사는 광합성 생물 중, 바위나 펄 등에 부착하여 자라는 눈으로 볼 수 있을 정도로 큰 다세포의 무리를 말하고, 광합성 색소에 따라 크게 녹조류, 갈조류, 홍조류로 구분한다.
본 발명의 일 실시예에 있어서, "갈조류"는 다시마(Saccharina japonica), 미역(Undaria pinnatifida) , 톳(Sargassum fusiforme), 감태(Ecklonia cava), 괭생이모자반(Sargassum horneri), 경단구슬모자반(Sargassum muticum), 지충이(Sargassum thunbergii) 및 곰피(Ecklonia stolonifera)로 이루어진 군에서 선택된 1종 이상일 수 있으며, 만니톨을 포함하는 갈조류라면 제한 없이 포함될 수 있다.
본 발명의 일 실시예에 있어서, 이타콘산을 생산하기 위한 해조류의 형태는 제한되어 있지 않으며, 과립, 분말과 같은 가루 및 즙 등의 형태를 포함한다.
또한 본 발명의 일 실시예에 따르면 상기 이타콘산 생산용 재조합 미생물을 배양하는 단계를 포함하는 이타콘산의 생산방법을 제공한다.
본 발명의 일 실시예에 있어서, 미생물의 배양에 사용되는 배지 및 기타 배양조건은 통상의 에스케리키아 속 미생물의 배양에 사용되는 배지이면 어느 것이나 사용될 수 있으나, 본 발명의 미생물의 요구 조건을 적절하게 만족시켜야 한다. 바람직하게는, 본 발명의 미생물을 적당한 탄소원, 질소원, 아미노산, 비타민 등을 함유한 통상의 배지 내에서 호기성 조건 하에서 온도, pH 등을 조절하면서 배양한다.
본 발명의 일 실시예에 있어서, 상기 배지는 탄소원으로서 아세트산을 포함할 수 있다. 무기화합물로는 염화나트륨, 염화칼슘, 염화철, 황산마그네슘, 황산철, 황산망간 및 탄산칼슘 등이 사용될 수 있으며, 그 외에 아미노산, 비타민 및 적절한 전구체 등이 포함될 수 있다. 이들 배지 또는 전구체는 배양물에 회분식 또는 연속식으로 첨가될 수 있다.
배양 중에 수산화암모늄, 수산화칼륨, 암모니아, 인산 및 황산과 같은 화합물을 배양물에 적절한 방식으로 첨가하여, 배양물의 pH를 조정할 수 있다. 또한, 배양 중에는 지방산 폴리글리콜 에스테르와 같은 소포제를 사용하여 기포 생성을 억제할 수 있다. 또한, 배양물의 호기 상태를 유지하기 위하여, 배양물 내로 산소 또는 산소 함유 기체를 주입하거나 혐기 및 호기 상태를 유지하기 위해 기체의 주입 없이 혹은 질소, 수소 또는 이산화탄소 가스를 주입할 수 있다.
배양물의 온도는 보통 27℃ 내지 37℃, 바람직하게는 30℃ 내지 35℃로 설정할 수 있다. 배양 기간은 원하는 유용 물질의 생성량이 수득될 때까지 계속될 수 있으며, 바람직하게는 10 내지 100 시간 동안 배양할 수 있다.
본 발명의 상기 배양 단계에서 생산된 이타콘산은 추가로 정제 또는 회수하는 단계를 포함할 수 있으며, 미생물 또는 배양물로부터 이타콘산을 회수하는 방법은 당업계에 알려진 방법, 예컨대 원심분리, 여과, 음이온 교환 크로마토그래피, 결정화 및 HPLC 등이 사용될 수 있으나, 이들 예에 한정되는 것은 아니다.
상기 회수 단계는 정제 공정을 포함할 수 있으며, 당업자는 공지된 여러 정제 공정 중 필요에 따라 선택하여 활용할 수 있다.
본 발명에 따른 이타콘산 생산용 재조합 미생물은 갈조류에 함유된 알긴산 및 만니톨을 이용하여 이타콘산을 생산할 수 있을 뿐만 아니라, 갈조류의 전처리 공정 없이 직접 투입하여도 이타콘산을 생산할 수 있는바, 친환경적이고 저비용으로 이타콘산 고생산 공정 개발이 가능하여 이타콘산이 활용되는 합성수지, 라텍스 및 식품첨가물 등의 산업 분야에서 다양하게 활용될 수 있다.
도 1은 본 발명의 일 실시예에 따른 재조합 균주가 이타콘산을 생산하는 전체적인 과정 및 구성을 나타낸 것이다.

달리 정의되지 않는 한, 기술적이거나 과학적인 용어를 포함해서 본 발명에서 사용되는 모든 용어들은 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자에 의해 일반적으로 이해되는 것과 동일한 의미를 갖고 있다. 일반적으로 사용되는 사전에 정의되어 있는 것과 같은 용어들은 관련 기술의 문맥상 갖는 의미와 일치하는 의미를 갖는 것으로 해석되어야 하며, 본 출원에서 명백하게 정의하지 않는 한, 이상적이거나 과도하게 형식적인 의미로 해석되지 않는다.
또한, 본 발명을 설명함에 있어서 관련된 공지 기술에 대한 구체적인 설명이 본 발명의 요지를 불필요하게 흐릴 수 있다고 판단되는 경우 그 상세한 설명을 생략한다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1. 이타콘산 생산용 재조합 비브리오 속 DHG 균주의 제조
1-1. 시스-아코니트산 탈탄산효소(cis-aconitate decarboxylase, cad) 유전자 도입을 위한 플라스미드 벡터 제작
이타콘산 생산을 위하여, 시스-아코니트산 탈탄산효소(cis-aconitate decarboxylase)를 발현하는 cad 유전자를 비브리오 속 DHG 균주(Vibrio sp. dhg)에 도입하였다. 상기 시스-아코니트산 탈탄산효소는 시스-아코니트산(cis-aconitate)을 이타콘산으로 전환하는 효소이다.
구체적으로, 서열번호 1의 염기서열로 표시되는 아스퍼질러스 테레우스(Aspergillus terreus) 유래 cad 유전자를 비브리오 속 DHG 균주에 도입하기 위하여, cad 유전자를 포함하는 cad 발현 카세트를 포함하는 재조합 벡터를 제작하였다.
상기 cad 발현 카세트를 포함하는 재조합 벡터를 제작하기 위하여, 벡터 플라스미드는 pACYC_duet 플라스미드를 주형으로 pACYC_cad FWD (1) 및 pACYC_cad REV (1) 프라이머를 사용하여 증폭되었다. cad 유전자는 pCDF_CAD 플라스미드를 주형으로 pACYC_cad FWD (2) 프라이머와 pACYC_cad REV (2) 프라이머를 사용하여 증폭되었다. 또한 유전자의 합성 과발현을 위해 Vibrio sp. dhg에서 강한 것으로 알려진 tac 프로모터 하에서 발현시켰으며, 높은 번역 효율을 갖는 합성 5‘ UTR을 설계하여 단백질 발현이 원활하게 일어나게 하였다. 증폭한 벡터 플라스미드와 cad 유전자를 결합하여 재조합 벡터인 pACYC_cad 플라스미드를 생성하였다. 이렇게 생성한 pACYC_cad 플라스미드를 비브리오 속 DHG 균주에 도입하여 VC 균주를 제작하였다. pACYC_cad 플라스미드를 제작하기 위해 사용된 프라이머의 염기서열을 표 1에 나타내었다.

1-2. 이소시트르산 탈수소효소(isocitrate dehydrogenase, icd) 유전자 삭제를 위한 플라스미드 벡터 제작 및 재조합 균주의 제조
서열번호 2의 염기서열로 표시되는 이소시트르산 탈수소효소(isocitrate dehydrogenase, icd)를 코딩하는 유전자를 삭제하기 위하여, 선행문헌에 기재된 SXT 재조합 시스템을 활용하였다. icd 유전자를 삭제하기 위해 사용된 pCDF_icd_del 플라스미드는 표적 서열 및 cat 유전자 조각이 포함된 3kb의 상동성 팔을 가지도록 설계되었다.
벡터 조각은 pCDF_duet 플라스미드를 주형으로 pCDF_icd_Cm_3K FWD (1) 및 pCDF_icd_Cm_3K REV (1) 프라이머를 사용하여 증폭되었다. Icd 유전자의 상부 및 하부 상동성 서열은 정제된 비브리오 속 DHG 균주의 유전체 DNA를 사용하여 pCDF_icd_Cm_3K FWD/pCDF_icd_Cm_3K REV (2) 및 pCDF_icd_Cm_3K FWD (4)/pCDF_icd_Cm_3K REV (4) 프라이머로 증폭되었다. Cat 유전자는 FRTwt_cat 플라스미드를 주형으로 pCDF_icd_Cm_3K FWD (3) 및 pCDF_icd_Cm_3K REV (3) 프라이머를 사용하여 증폭되었다. Icd 영역에 pCDF_icd_del 플라스미드의 도입을 통해 FRT 서열이 둘러싸인 cat 유전자 조각을 통합한 후, pRSF_FLP 플라스미드를 도입하여 플리파제(flippase)를 발현시켜 VI 균주를 제작하였다. 이후 VI 균주에 pACYC_cad 플라스미드를 도입하여 최종적으로 VIC 균주를 제작하였다. 상기 pCDF_icd_del 플라스미드를 제작하기 위해 사용된 프라이머의 염기서열을 표 2에 나타내었다. 또한 상기와 같은 과정을 통해 제작한 VIC 균주의 이타콘산 생산 경로를 도 1에 나타내었다. 이와 같이 제작한 재조합 비브리오 속 DHG 균주인 VIC 균주를 한국생명공학연구원 생물자원센터에 2024년 5월 28일자로 기탁하여 기탁번호 KCTC19196P를 부여받았다.

실시예 2. 재조합 비브리오 속 DHG 균주를 이용한 이타콘산 생신 실험 준비
2-1. 배양 배지의 준비 및 균주 배양
E. coli Mach1-T1R은 10 g/L NaCl, 10 g/L 트립톤, 5 g/L 효모 추출물이 포함된 37°C의 LB 배지에서 배양되었고, 비브리오 속 DHG 균주 및 그 유래 균주는 선행문헌에 기재된 바와 같이 30°C의 LBv2 또는 변형된 완충 최소 배지에서 배양되었다. LBv2 배지는 10 g/L 트립톤, 5 g/L 효모 추출물, 21.9 g/L NaCl, 0.3 g/L KCl, 2.2 g/L MgCl2를 포함하며, 변형된 완충 최소 배지는 5 g/L (NH4)2SO4, 10 g/L NaCl, 10.7 g/L K2HPO4, 5.2 g/L KH2PO4, 0.5 g/L MgSO4·7H2O 및 2 mL/L 미량 금속 용액(ATCC MD-TMS)을 포함한다. 변형된 완충 최소 배지에는 알긴산과 만니톨의 혼합물 또는 원시 S. japonica와 다양한 농도의 글루탐산이 추가되었다. 모든 배지는 pH 7로 조정되고 적절한 항생제로 보충되었다. 한천 배지는 배지에 15 g/L의 한천을 포함하여 준비되었다.
2-2. 알긴산 및 만니톨로부터 이타콘산 생산을 위한 균주의 접종
이타콘산 생산을 위해, LBv2 한천 배지에서 콜로니를 선택하고, 15 mL 시험관에 10 g/L 알긴산, 10 g/L 만니톨, 1 g/L 글루탐산이 보충된 3 mL의 변형된 완충 최소 배지에 선택한 콜로니를 접종하였다. 하루 동안 배양한 후, 배양액을 OD600 (600 nm에서의 광학 밀도) 0.05로 새로운 배지에 희석하였다. OD600이 0.6에서 1.0에 도달하면, 세포를 OD600 0.05로 조정된 20 mL의 변형된 완충 최소 배지를 포함한 350 mL의 삼각 플라스크에 옮겼다. 탄소원으로 10 g/L 알긴산과 10 g/L 만니톨을 배지에 추가하였다. 정제된 알긴산 및 만니톨이 아닌 S. japonica를 직접 기질로 사용할 때는 UV로 멸균한 분말 40 g/L를 배지에 추가하였다. Cad 유전자의 발현을 위해, 배지에 0.5 mM의 이소프로필 β-D-1-티오갈락토피라노사이드(IPTG)를 추가하였다. 또한, 주 배양액에 다양한 농도의 글루탐산(0에서 3 g/L)을 추가하였다. 모든 배양은 30°C에서 220 rpm으로 회전 셰이커(Hanil Scientific, Gimpo, Korea)에서 3개의 생물학적 복제물로 수행되었다.
2-3. 세포 성장 정량화
세포 성장의 정량화를 위해 UV-1700 분광광도계(Shimadzu, Kyoto, Japan)를 사용하여 OD600을 측정하였다. 우선, 1 mL의 배양액을 수확하여 13,000 rpm에서 1분간 원심분리하였다. 그 후, 상층액 100 μL를 Sigma-Aldrich(St. Louis, MO, USA)의 알긴산 분해효소 100 mg/L가 포함된 용액에 희석하고, 37°C에서 1시간 동안 배양하였다. 그 후 샘플을 Sigma-Aldrich(St. Louis, MO, USA)의 0.22 μm 나일론 필터로 여과하였다.
2-4. 반응물 및 생성물 농도 정량화
만니톨, 이타콘산, 아세트산의 농도는 Shimadzu(Kyoto, Japan)의 UltiMate™ 3000 분석용 고성능 액체 크로마토그래피(HPLC) 시스템을 사용하여 정량화하였다. HPLC는 Bio-Rad Laboratories(Richmond, CA, USA)의 Aminex HPX-87H 컬럼을 장착하여 30°C에서 수행하였다. 5 mM 황산이 이동상으로 사용되었으며, 유속은 0.6 mL/min였다. 굴절률 신호는 Shodex(Klokkerfaldet, Denmark)의 Shodex RI-101 검출기를 사용하여 모니터링되었다.
알긴산 정량화를 위해, 0에서 2 g/L의 농도 범위로 희석된 여과된 샘플 50 μL를 Bio-Rad Laboratories(Richmond, CA, USA)의 마이크로타이터 플레이트에 투입하였다. 그 후, 0.8 M 설파믹산 시약 5 μL와 75 mM 이나트륨 테트라보레이트가 포함된 98% 황산 200 μL를 추가하고, 철저히 혼합한 후 65°C에서 2시간 동안 배양하였다. 샘플을 실온에서 20분 동안 냉각한 후, 무수에탄올에 용해한 0.125% 카바졸 50μL를 추가하였다. 각 웰의 흡광도는 BioTek(Winooski, VT, USA)의 Synergy™ LX Multi-Mode Reader를 사용하여 550 nm에서 측정하였다. 이후, 0, 0.1, 0.5, 1, 2 g/L의 농도로 준비된 알긴산 용액을 사용하여 작성한 표준 곡선으로 보정하였다.
실시예 3. 재조합 비브리오 속 DHG 균주를 이용한 이타콘산 생산능 확인
3-1. Icd 유전자를 삭제한 균주의 생장 및 이타콘산 생산능 확인
Icd 유전자를 삭제한 균주의 이타콘산 생산능을 확인하기 위해, 상기 실시예 1에서 제작한 VC 균주 및 VIC 균주를 10 g/L의 알긴산과 10 g/L이 보충된 변형된 완충 최소 배지에 접종하였고, 그 결과를 도 2에 나타내었다.
도 2에 나타난 바와 같이, VC 균주의 경우 세포 성장, 알긴산 및 만니톨 소비가 활발하며 아세트산 생산량이 증대된 반면에, VIC 균주의 경우 세포 성장, 알긴산 및 만니톨 소비가 미미하며, 아세트산 생산량 또한 미비한 것을 확인할 수 있었다. 그러나 VC 균주 및 VIC 균주의 이타콘산 수율은 비슷하게 나타났으므로, icd 유전자의 삭제가 알파-케토글루타르산 생산으로의 탄소 흐름을 줄여 이타콘산 생산에 효과적임을 확인할 수 있었다.
3-2. 글루탐산 보충 시 VC 균주, VIC 균주의 생장 및 이타콘산 생산능 확인
재조합 비브리오 속 균주의 생체량을 증가시키기 위해, TCA 회로 중간체인 글루탐산을 보충하여 VIC 균주의 성장을 회복시키는 방법을 시도하였다. 실시예 3-1의 배지에서 글루탐산의 양을 0g/L 부터 3g/L까지 조절하면서 VC 균주 대비 VIC 균주의 생체량, 당 소비량, 이타콘산 생산량, 이타콘산 수율을 측정하여 도 3a 내지 도 3d에 나타내었다.
도 3a에 나타난 바와 같이, 글루탐산이 없는 조건에서 VIC 균주는 거의 성장하지 않으나, 글루탐산이 추가될수록 성장이 회복되는 것을 확인할 수 있었다.
도 3b에 나타난 바와 같이, 글루탐산이 없는 조건에서 VIC 균주는 알긴산 및 만니톨을 거의 소비하지 않으나, 글루탐산이 추가될수록 알긴산 및 만니톨의 소비량이 증가하는 것을 확인할 수 있었다.
도 3c에 나타난 바와 같이, 글루탐산이 없는 조건에서도 VIC 균주는 VC 균주와 유사한 이타콘산 생산능을 보이며, 글루탐산이 추가될수록 이타콘산 생산량이 증가하여 3g/L의 글루탐산이 추가된 배지에서는 이타콘산 생산 농도가 약 5배까지 증가한 것을 확인할 수 있었다.
도 3d에 나타난 바와 같이, 글루탐산이 추가된 경우 VIC 균주의 당 소비량 대비 이타콘산 전환량이 감소하지만, 여전히 VC 균주보다 높은 이타콘산 수율을 보이는 것을 확인할 수 있었다.
3-3. 재조합 균주의 갈조류로부터 직접적인 이타콘산 생산능 확인
본 발명의 재조합 VIC 균주가 가공되지 않은 갈조류를 기질로 사용하여 직접 이타콘산을 생산할 수 있는지를 확인하기 위해, 40 g/L의 다시마(S. japonica)를 포함하는 최소 배지에 추가적으로 3 g/L의 글루탐산을 보충하여 VIC 균주를 배양한 후, 이타콘산 생산능을 측정한 결과를 도 4에 나타내었다.
도 4에 나타난 바와 같이, 40 g/L의 다시마에서 총 20.1 g/L의 당(알긴산 11.8 g/L 및 만니톨 8.3 g/L)이 정량되었다. 48시간 동안 측정한 결과 1.5 g/L의 이타콘산이 0.08 g/g의 수율로 생산되었으며, 11 g/L의 알긴산과 8.3 g/L의 만니톨이 소비되었다. 따라서 재조합 균주를 사용하여 정제되지 않은 알긴산 및 만니톨을 포함하고 있는 다시마를 이용하여 직접 이타콘산을 생산할 수 있음을 확인할 수 있었다.
이상에서 본 발명의 바람직한 실시예들에 대하여 상세하게 설명하였지만, 본 발명의 권리 범위는 이에 한정되는 것은 아니고 다음의 청구범위에서 정의하고 있는 본 발명의 기본 개념을 이용한 당해 기술분야의 통상의 기술자가 실시할 수 있는 여러 변형 및 개량 형태 또한 본 발명의 권리범위에 속하는 것이다.

본 발명에 따른 이타콘산 생산용 재조합 미생물은 갈조류에 함유된 알긴산 및 만니톨을 이용하여 이타콘산을 생산할 수 있을 뿐만 아니라, 갈조류의 전처리 공정 없이 직접 투입하여도 이타콘산을 생산할 수 있는바, 친환경적이고 저비용으로 이타콘산 고생산 공정 개발이 가능하여 이타콘산이 활용되는 합성수지, 라텍스 및 식품첨가물 등의 산업 분야에서 다양하게 활용될 수 있어, 산업상 이용가능성이 있다.
Claims (10)
- 시스-아코니트산 탈탄산효소(cis-aconitate decarboxylase, cad)를 코딩하는 유전자를 포함하는 cad 유전자 발현 카세트를 포함하는 이타콘산(itaconic acid) 생산용 재조합 벡터.
- 제1항에 있어서,상기 cad 유전자는 서열번호 1의 염기서열로 표시되는 것을 특징으로 하는, 이타콘산 생산용 재조합 벡터.
- 제1항의 재조합 벡터가 도입된 이타콘산 생산용 재조합 미생물.
- 제3항에 있어서,상기 이타콘산 생산용 재조합 미생물은 서열번호 2의 염기서열로 표시되는 이소시트르산 탈수소효소(isocitrate dehydrogenase, icd) 유전자가 결실된 것을 특징으로 하는 이타콘산 생산용 재조합 미생물.
- 제3항에 있어서,상기 이타콘산 생산용 재조합 미생물은 비브리오 속 DHG 균주(Vibrio sp. dhg)인 것을 특징으로 하는 이타콘산 생산용 재조합 미생물.
- 제5항에 있어서,상기 이타콘산 생산용 재조합 미생물은 수탁번호 KCTC 19196P로 기탁된 재조합 비브리오 속 DHG 균주인 것을 특징으로 하는 이타콘산 생산용 재조합 미생물.
- 제6항에 있어서,상기 이타콘산 생산용 재조합 미생물은 알긴산 및 만니톨을 이용하여 이타콘산을 생산하는 것을 특징으로 하는 이타콘산 생산용 재조합 미생물.
