Abstract

本発明は、食感が改良された繊維集合体を提供する。本発明の繊維集合体は、複数の繊維を含む。繊維集合体は、非動物性タンパク質及び多糖類Pを含む。繊維集合体は、試験1により測定された押し込み力が15.0N~80.0Nであり、試験2により測定された引き裂き力が0.15N/mm~1.5N/mmである。繊維集合体は、例えば、複数の繊維のうち、少なくとも2つの繊維を互いに結着させる結着部を有する。

Description

繊維集合体、及び繊維集合体の製造方法
 本発明は、繊維集合体、及び繊維集合体の製造方法に関する。
 近年では、世界の人口増加に伴い、食肉需要が増加することが予想されている。今後の食肉需要の増加に対応するためには、従来のタンパク源について、生産効率を上げて増産するだけでは十分ではなく、新たなタンパク源の開発が不可欠である。新たなタンパク源としては、植物から生産される植物肉、昆虫類から生産される肉、微生物や細胞そのものを培養して生産される培養肉などの代替食品が挙げられる。これらの代替食品は、動物愛護の観点からも注目されている。特許文献1は、代替食品の一例として、乾燥肉様蛋白加工食品を開示している。
 なお、「植物肉」は、大豆などの植物性タンパク質を原料として用い、これに添加剤を加えて成形した加工食品であり、「フェイクミート」とも呼ばれる。「培養肉」は、再生医療技術を用いて、筋肉細胞を培養することによって作製される肉を意味し、「cultured meat」又は「clean meat」とも呼ばれる。
特開2022-118655号公報
 近年、代替食品として、非動物性タンパク質を含む繊維の集合体(繊維集合体)が検討されている。繊維集合体は、ステーキ肉、刺身、切り身など、ある程度の大きさを有する塊状の肉を模倣しやすい利点がある。しかし、本発明者らの検討によると、繊維集合体は、食感の観点から改良の余地がある。
 そこで本発明は、食感が改良された繊維集合体を提供することを目的とする。
 本発明は、
 複数の繊維を含む繊維集合体であって、
 前記繊維集合体は、非動物性タンパク質及び多糖類Pを含み、
 下記試験1により測定された押し込み力が15.0N~80.0Nであり、
 下記試験2により測定された引き裂き力が0.15N/mm~1.5N/mmである、繊維集合体を提供する。
 試験1:水で飽和状態の前記繊維集合体から、厚さ10mmの板状の試験片T1を切り出す。縦10mm×横1.5mmの矩形の表面を有する圧子を用いて、押し込み速度2.0mm/secの条件で、前記試験片T1の厚さ方向に前記圧子を前記試験片T1に押し込む。このときの最大荷重(N)を押し込み力として特定する。
 試験2:水で飽和状態の前記繊維集合体から板状の試験片T2を切り出す。前記試験片T2に2つのフック(外径3.0mm)を突き刺し、前記試験片T2の厚さ方向に2つの前記フックを貫通させる。引張速度2.0mm/secの条件で、2つの前記フックを互いに引き離す方向に引っ張り、前記試験片T2を引き裂く。このときの最大荷重(N)を前記試験片T2の厚さ(mm)で除した値を引き裂き力として特定する。
 さらに、本発明は、
 上記の繊維集合体の製造方法であって、
 前記製造方法は、
 前記非動物性タンパク質及び多糖類Q1を含む混合液を凝固液に吐出して、前記繊維を形成する紡糸工程を含む、繊維集合体の製造方法を提供する。
 本発明によれば、食感が改良された繊維集合体を提供できる。

 本発明の第1態様にかかる繊維集合体は、

 以下、本発明の詳細を説明するが、以下の説明は、本発明を特定の実施形態に制限する趣旨ではない。
<繊維集合体の実施形態>
 本実施形態の繊維集合体は、複数の繊維を含む。繊維集合体は、非動物性タンパク質及び多糖類Pを含む。繊維集合体について、下記試験1により測定された押し込み力が15.0N~80.0Nであり、下記試験2により測定された引き裂き力が0.15N/mm~1.5N/mmである。本発明者らの検討によると、押し込み力及び引き裂き力が上記の範囲に調整されている繊維集合体は、食感が改良されている傾向がある。
 試験1:水で飽和状態の繊維集合体から、厚さ10mmの板状の試験片T1を切り出す。縦10mm×横1.5mmの矩形の表面を有する圧子を用いて、押し込み速度2.0mm/secの条件で、試験片T1の厚さ方向に圧子を試験片T1に押し込む。このときの最大荷重(N)を押し込み力として特定する。
 試験2:水で飽和状態の繊維集合体から板状の試験片T2を切り出す。試験片T2に2つのフック(外径3.0mm)を突き刺し、試験片T2の厚さ方向に2つのフックを貫通させる。引張速度2.0mm/secの条件で、2つのフックを互いに引き離す方向に引っ張り、試験片T2を引き裂く。このときの最大荷重(N)を試験片T2の厚さ(mm)で除した値を引き裂き力として特定する。
 試験1は、詳細には、次の方法によって行うことができる。まず、水で飽和状態の繊維集合体を準備する。本明細書において、「水で飽和状態」とは、25℃の水に繊維集合体を1分間浸漬させた場合に、繊維集合体の重量変化率が0.1%以下であり、好ましくは0.05%以下、より好ましくは0.01%以下であることを意味する。繊維集合体の重量変化率は、水に浸漬前の繊維集合体の重量W1(g)に対する、重量W1(g)と、水に浸漬後の繊維集合体の重量W2(g)との差の比率(100×|W2-W1|/W1)を意味する。
 次に、繊維集合体から、厚さ10mmの板状の試験片T1を切り出す。試験片T1の形状としては、円板状、楕円板状、矩形板状などが挙げられる。試験片T1について、厚さ以外のサイズは、押し込み力の測定結果に影響を与えない範囲に調整されることが好ましい。なお、繊維集合体が板状である場合、繊維集合体自体を試験片T1として用いてもよい。
 試験片T1において、複数の繊維は、配向していないことが好ましい。複数の繊維が配向している場合は、複数の繊維のそれぞれが、試験片T1の厚さ方向に直交する方向に延びていることが好ましい。
 次に、図1に示すように、試験片T1を支持部2の上に配置し、試験片T1を支持部2に固定する。次に、縦10mm×横1.5mmの矩形の表面1aを有する圧子1を用いて、試験片T1の厚さ方向に圧子1を試験片T1に押し込む押込試験を行う。圧子1の表面1aは、試験片T1と接触する表面であり、ヒトの歯を模したサイズ及び形状である。圧子1は、角柱状であることが好ましい。押込試験は、市販のテクスチャーアナライザーを利用して行うことができる。押込試験は、押し込み速度2.0mm/secの条件で、圧子1が試験片T1を厚さ方向に貫通するまで行う。なお、押込試験は、25℃の雰囲気下で行う。
 次に、押込試験の結果から、荷重と時間との関係を示すグラフを作成し、当該グラフから最大荷重(N)を特定する。この最大荷重(N)を押し込み力として特定することができる。
 繊維集合体の押し込み力は、上述のとおり15.0N~80.0Nである。この範囲の押し込み力を有する繊維集合体は、ほどよい噛み応え(噛みしめたときの硬さ)を有する傾向がある。繊維集合体の押し込み力は、好ましくは20.0N以上であり、25.0N以上、30.0N以上、35.0N以上、40.0N以上、45.0N以上、50.0N以上、55.0N以上、さらには60.0N以上であってもよい。繊維集合体の押し込み力の上限は、特に限定されず、例えば75.0N以下であり、70.0N以下、さらには65.0N以下であってもよい。繊維集合体の押し込み力は、場合によっては、60.0N以下であってもよく、50.0N以下であってもよい。
 試験2は、詳細には、次の方法によって行うことができる。まず、水で飽和状態の繊維集合体を準備する。次に、繊維集合体から板状の試験片T2を切り出す。試験片T2の形状としては、円板状、楕円板状、矩形板状などが挙げられる。試験片T2の厚さは、約10mmであることが好ましい。試験片T2について、厚さ以外のサイズは、引き裂き力の測定結果に影響を与えない範囲に調整されることが好ましい。なお、繊維集合体が板状である場合、繊維集合体自体を試験片として用いてもよい。
 試験片T2において、複数の繊維は、配向していないことが好ましい。複数の繊維が配向している場合は、複数の繊維のそれぞれが、試験片T2の厚さ方向、及び、後述する引張試験で2つのフック5A及び5Bを互いに引き離す方向のそれぞれに直交する方向に延びていることが好ましい。
 次に、図2に示すように、試験片T2に2つのフック5A及び5Bを突き刺し、試験片T2の厚さ方向に2つのフック5A及び5Bを貫通させる。図2は、楕円板状の試験片T2が、2つのフック5A及び5Bに突き刺された状態を示している。フック5A及び5Bは、例えば、試験片T2の中央(重心)付近で、試験片T2を突き刺している。引張試験の前において、フック5Aとフック5Bは、互いに接触している(初期のフック間距離が0mmである)ことが好ましい。フック5A及び5Bは、いずれも外径(特に、試験片T2と接触している部分の外径)が3.0mmである。
 次に、2つのフック5A及び5Bを互いに引き離す方向に引っ張る引張試験を行う。引張試験は、市販の引張試験機やテクスチャーアナライザーを利用して行うことができる。一例として、引張試験は、試験片T2の下方に位置する固定部4に、一方のフック5Bを固定し、他方のフック5Aを上方に引っ張ることによって行ってもよい(図2)。引張試験は、引張速度2.0mm/secの条件で、試験片T2が引き裂かれるまで行う。なお、引張試験は、25℃の雰囲気下で行う。
 次に、引張試験の結果から、荷重と時間との関係を示すグラフを作成し、当該グラフから最大荷重(N)を特定する。最大荷重(N)を試験片T2の厚さ(mm)で除した値を引き裂き力として特定することができる。
 繊維集合体の引き裂き力は、上述のとおり0.15N/mm~1.5N/mmである。この範囲の引き裂き力を有する繊維集合体は、ほどよい凝集性(咀嚼時のまとまり感、ほぐれにくさ)を有する傾向がある。繊維集合体の引き裂き力は、好ましくは0.20N/mm以上、0.30N/mm以上、0.40N/mm以上、0.50N/mm以上、0.60N/mm以上、0.70N/mm以上、さらには0.80N/mm以上であってもよい。繊維集合体の引き裂き力の上限は、特に限定されず、例えば1.2N/mm以下であり、1.0N/mm以下、さらには0.90N/mm以下であってもよい。繊維集合体の引き裂き力は、場合によっては、0.80N/mm以下であってもよく、0.70N/mm以下であってもよい。
 なお、発明者らの検討によれば、繊維集合体の押し込み力及び引き裂き力の両方を大きい値に調整することは難しい。一例として、繊維集合体の押し込み力をx(N)とし、引き裂き力をy(N/mm)とした場合、x及びyは、下記関係式を満たしていてもよい。

 さらに、繊維集合体では、食感をより改良する観点から、上記のx及びyが下記関係式を満たしていてもよい。

(繊維)
 上述のとおり、繊維集合体は、複数の繊維を含む。繊維集合体は、複数の繊維が集合した集合体であることが好ましい。繊維集合体において、繊維自体が非動物性タンパク質を含むことが好ましく、多糖類Pとして多糖類P1をさらに含むことがより好ましい。繊維において、多糖類P1の重量に対する非動物性タンパク質の重量の比は、特に限定されず、例えば0.5~20.0である。なお、繊維集合体は、上記の組成を満たす繊維以外の他の繊維を含んでいてもよいが、他の繊維を含まなくてもよい。繊維集合体は、上記の組成を満たす複数種類の繊維を含んでいてもよい。
 繊維の形状は、特に限定されない。繊維は、分岐構造を有していないことが好ましい。繊維は、長繊維であることが好ましいが、短繊維であってもよい。繊維の長さは、例えば0.5cm以上であり、1cm以上、3cm以上、5cm以上、さらには10cm以上であってもよい。繊維の長さは、例えば100cm以下であり、50cm以下、さらには30cm以下であってもよい。繊維の長さは、ノギス等のスケールで測定することができる。繊維の長さは、光学顕微鏡又はマイクロスコープにより繊維を観察し、得られた顕微鏡像から測定してもよい。
 繊維集合体において、複数の繊維の平均長さは、例えば0.5cm以上であり、1cm以上、3cm以上、5cm以上、さらには10cm以上であってもよい。複数の繊維の平均長さは、例えば100cm以下であり、50cm以下、さらには30cm以下であってもよい。複数の繊維の平均長さは、次の方法によって特定することができる。まず、任意の数(少なくとも5本)の繊維について、その長さをノギス等のスケールで測定する。繊維の長さは、光学顕微鏡又はマイクロスコープにより繊維を観察し、得られた顕微鏡像から測定してもよい。得られた測定値の平均値を平均長さとみなすことができる。
 繊維の繊維径は、例えば1cm以下であり、1mm以下、800μm以下、500μm以下、300μm以下、200μm以下、さらには100μm以下であってもよい。繊維径が小さい繊維によれば、緻密な構造を有する繊維集合体を作製しやすく、塊状の肉の食感を再現しやすい傾向がある。繊維の繊維径の下限は、例えば1μm以上であり、5μm以上、10μm以上、30μm以上、さらには50μm以上であってもよい。繊維の繊維径は、10μm~300μmであることが好ましく、50μm~200μmであることがより好ましい。繊維径は、次の方法によって特定することができる。まず、光学顕微鏡又はマイクロスコープにより繊維を観察する。得られた顕微鏡像において、繊維の直径をノギス等のスケールで測定する。任意の数の測定箇所(少なくとも3箇所)で繊維の直径を測定し、得られた測定値の平均値を繊維径とみなす。
 繊維集合体において、複数の繊維の平均繊維径は、例えば1cm以下であり、1mm以下、800μm以下、500μm以下、300μm以下、200μm以下、さらには100μm以下であってもよい。平均繊維径の下限は、例えば1μm以上であり、5μm以上、10μm以上、30μm以上、さらには50μm以上であってもよい。平均繊維径は、10μm~300μmであることが好ましく、50μm~200μmであることがより好ましい。平均繊維径は、任意の数(少なくとも5本)の繊維について、上述の方法で特定した直径の平均値を意味する。
 繊維集合体において、複数の繊維は、配向していないことが好ましい。ただし、複数の繊維は、場合によっては、配向していてもよい。この場合、複数の繊維は、単一の方向に配向していてもよく、2つ以上の方向に配向していてもよい。複数の繊維の配向分布の半値全幅(FWHM)は、例えば10°以上であり、20°以上、30°以上、40°以上、50°以上、60°以上、70°以上、さらには80°以上であってもよい。特に、複数の繊維は、模倣対象の肉に含まれる筋線維と同じ方向に配向していてもよい。
 繊維集合体における繊維(非動物性タンパク質及び多糖類P1を含む繊維)の含有率は、例えば30wt%以上であり、50wt%以上、60wt%以上、さらには70wt%以上であってもよい。繊維集合体は、繊維を主成分として含んでもよい。本明細書において、「主成分」は、繊維集合体に重量比で最も多く含まれた成分を意味する。繊維の含有率の上限は、例えば99.8wt%以下であり、99.0wt%以下、95wt%以下、90wt%以下、さらには80wt%以下であってもよい。
(結着部)
 繊維集合体は、複数の繊維のうち、少なくとも2つの繊維を互いに結着させる結着部を有することが好ましい。結着部は、繊維集合体の押し込み力や引き裂き力を調整することに適している。繊維集合体は、上記の結着部を1つ又は2つ以上有していてもよい。繊維集合体において、結着部は、複数の繊維の間の空隙を埋めるバインダーであってもよい。
 繊維集合体では、複数の繊維のうち、10%以上の繊維が隣接する繊維と結着部を介して結着していることが好ましく、50%以上の繊維が隣接する繊維と結着部を介して結着していることがより好ましく、実質的に全ての繊維が隣接する繊維と結着部を介して結着していることが特に好ましい。
 結着部は、多糖類Pとして多糖類P2を含むことが好ましく、多糖類P2を主成分として含むことが好ましい。結着部は、ゲル化剤をさらに含んでいてもよい。ゲル化剤の詳細については後述する。なお、結着部は、非動物性タンパク質を実質的に含まないことが好ましい。
 繊維集合体における結着部の含有率は、例えば0.2wt%以上であり、1.0wt%以上、5wt%以上、10wt%以上、さらには20wt%以上であってもよい。結着部の含有率の上限は、例えば70wt%以下であり、50wt%以下、40wt%以下、さらには30wt%以下であってもよい。
(非動物性タンパク質)
 上述のとおり、繊維集合体(特に、繊維集合体に含まれる繊維)は、非動物性タンパク質を含む。非動物性タンパク質は、動物を原料とする動物性タンパク質以外の他のタンパク質を意味する。非動物性タンパク質は、繊維集合体の栄養価を調整することに適した成分である。
 非動物性タンパク質は、植物性タンパク質を含むことが好ましい。植物性タンパク質としては、大豆タンパク質、エンドウ豆タンパク質、レンズ豆タンパク質、ひよこ豆タンパク質、そら豆タンパク質などの豆タンパク質;ジャガイモタンパク質;セイタンタンパク質;アマランサスタンパク質;キノアタンパク質;小麦タンパク質;米タンパク質;トウモロコシタンパク質などが挙げられる。非動物性タンパク質は、植物性タンパク質として、大豆タンパク質、エンドウ豆タンパク質、レンズ豆タンパク質、ひよこ豆タンパク質、そら豆タンパク質、ジャガイモタンパク質、セイタンタンパク質、アマランサスタンパク質、キノアタンパク質、小麦タンパク質、米タンパク質及びトウモロコシタンパク質からなる群より選ばれる少なくとも1つを含むことが好ましく、大豆タンパク質を含むことが特に好ましい。なお、非動物性タンパク質は、植物性タンパク質に限定されない。非動物性タンパク質は、菌類に由来するタンパク質を含んでいてもよい。
 水で飽和状態の繊維集合体において、非動物性タンパク質の含有率R1は、例えば5.0wt%以上であり、8.0wt%以上、10wt%以上、13wt%以上、15wt%以上、18wt%以上、さらには20wt%以上であってもよい。非動物性タンパク質の含有率R1は、後述する多糖類Pの含有率R2より高くてもよい。非動物性タンパク質の含有率R1は、例えば35wt%以下であり、33wt%以下、30wt%以下、27wt%以下、25wt%以下、さらには24wt%以下であってもよい。非動物性タンパク質の含有率R1は、5.0wt%~27wt%であることが好ましい。なお、繊維集合体の固形分に対する非動物性タンパク質の重量比率は、例えば35wt%~95wt%である。なお、非動物性タンパク質の含有率R1は、繊維集合体を製造するときに用いられる材料の組成などから算出することができる。
(多糖類P)
 上述のとおり、繊維集合体は、多糖類Pを含む。詳細には、繊維集合体は、多糖類Pとして、繊維に含まれる多糖類P1と、結着部に含まれる多糖類P2とを含むことが好ましい。多糖類P1と多糖類P2は、互いに同じであってもよく、異なっていてもよい。
 繊維に含まれる多糖類P1は、アルギン酸の二価金属塩を含むアルギン酸類を有することが好ましい。なお、アルギン酸の二価金属塩は、典型的には、アルギン酸と二価の金属イオンとの塩である。本明細書では、「アルギン酸の二価金属塩」を単に「アルギン酸塩」と呼ぶことがある。アルギン酸類は、アルギン酸塩とともに、アルギン酸をさらに含むことが好ましい。この場合、繊維集合体の食感がより改良される傾向がある。
 アルギン酸は、海藻類などに含まれる多糖類であり、β-D-マンヌロン酸に由来する構造単位(Mブロック)とα-L-グルロン酸に由来する構造単位(Gブロック)とを有する。アルギン酸において、各構造単位は、1,4-グリコシド結合を介して結合している。アルギン酸におけるGブロックの含有率は、特に限定されず、例えば30モル%以上であり、好ましくは40モル%以上であり、より好ましくは50モル%以上である。Gブロックの含有率の上限値は、90モル%であってもよく、80モル%であってもよい。
 一例として、アルギン酸塩では、アルギン酸に含まれる少なくとも1つのGブロックが二価の金属イオンとイオン結合を形成している。言い換えると、アルギン酸塩では、アルギン酸が二価の金属イオンと少なくとも部分的に塩を形成している。アルギン酸塩は、典型的には、二価の金属イオンを介した架橋構造を有している。二価の金属イオンとしては、カルシウムイオン、マグネシウムイオン、バリウムイオン、鉄イオン、亜鉛イオン、銅イオンなどが挙げられ、カルシウムイオンが好ましい。
 繊維は、多糖類P1としてアルギン酸類のみを含むことが好ましい。ただし、繊維は、アルギン酸類とともにアルギン酸類以外の他の多糖類を含んでいてもよい。他の多糖類としては、例えば、繊維集合体の保水性を十分に向上させる観点から、キシログルカン、メチルセルロース、カルボキシメチルセルロース、キサンタンガム及びこれらの誘導体が挙げられる。特に、キシログルカンは、繊維集合体の含水率を上げすぎずに、繊維集合体の保水性を改善することに適している。また、メチルセルロースやその誘導体については、健康上の課題が生じる可能性があると報告されている。これに対して、キシログルカンは、人体への安全性が高く、代替食品としての繊維集合体の成分に適している。
 キシログルカンは、グルコースに由来する構成単位U1と、キシロースに由来する構成単位U2とを含む多糖類である。キシログルカンは、複数の構成単位U1がβ-1,4-グリコシド結合を介して結合した主鎖と、当該主鎖に対して、構成単位U2がα-1,6-グリコシド結合を介して結合した側鎖とを有する。キシログルカンは、構成単位U1及びU2以外に、ガラクトースに由来する構成単位U3などの他の構成単位をさらに含んでいてもよい。キシログルカンは、重量平均分子量が5万~100万であることが好ましい。
 キシログルカンは、例えば、エンドウ豆、大豆、ポプラ、イネ、タケノコなどの高等植物の細胞壁や、タマリンド種子などから抽出することができる。キシログルカンは、タマリンド種子に由来するタマリンドシードガムであることが好ましい。すなわち、他の多糖類は、キシログルカンとして、タマリンドシードガムを含むことが好ましい。
 結着部に含まれる多糖類P2は、アルギン酸の二価金属塩を含むアルギン酸類を有することが好ましい。アルギン酸類としては、多糖類P1について上述したものが挙げられる。結着部は、多糖類P2としてアルギン酸類のみを含むことが好ましく、アルギン酸類以外の他の多糖類(特にメチルセルロース)を含まないことが好ましい。ただし、結着部は、他の多糖類を少量含んでいてもよい。
 上述のとおり、本実施形態では、多糖類Pが、アルギン酸の二価金属塩を含むアルギン酸類を有することが好ましく、特に、多糖類P1及び多糖類P2の両方がアルギン酸類を有することが好ましい。
 水で飽和状態の繊維集合体において、多糖類P(多糖類P1及びP2)の含有率R2は、例えば0.1wt%以上であり、0.3wt%以上、0.5wt%以上、1.0wt%以上、1.5wt%以上、1.8wt%以上、2.0wt%以上、さらには2.1wt%以上であってもよい。多糖類Pの含有率R2が高ければ高いほど、繊維集合体の弾力が向上し、これにより、繊維集合体の食感が改良される傾向がある。多糖類Pの含有率R2の上限は、例えば10wt%以下であり、8.0wt%以下、5.0wt%以下、さらには4.0wt%以下であってもよい。繊維集合体の固形分に対する、多糖類Pの重量比率は、例えば5wt%~50wt%である。なお、多糖類Pの含有率R2は、繊維集合体を製造するときに用いられる材料の組成などから算出することができる。
(水)
 水で飽和状態の繊維集合体において、含水率R3は、例えば50wt%以上であり、55wt%以上、60wt%以上、65wt%以上、70wt%以上、71wt%以上、75wt%以上、78wt%以上、80wt%以上、81wt%以上、82wt%以上、さらには83wt%以上であってもよい。含水率R3が高ければ高いほど、繊維集合体のジューシー感やしっとり感が向上し、これにより、繊維集合体の食感が改良される傾向がある。含水率R3の上限は、例えば93wt%以下であり、90wt%以下、88wt%以下、さらには85wt%以下であってもよく、場合によっては50wt%未満であってもよい。なお、水で飽和状態の繊維集合体における固形分の重量比率は、例えば12wt%~40wt%であり、12wt%~29wt%であってもよい。
 含水率R3は、次の方法によって特定することができる。まず、水で飽和状態の繊維集合体を準備し、その重量W3(g)を測定する。重量W3は、1gであることが好ましい。次に、この繊維集合体を加熱式水分計にセットし、105℃で加熱処理を行う。この加熱処理により、繊維集合体に含まれる水分が揮発する。加熱処理は、繊維集合体における水の減少速度が0.10wt%/minを下回るまで行う。重量W3と、加熱処理後の繊維集合体の重量W4(g)とに基づいて、以下の計算式により、含水率R3を算出することができる。
含水率R3(wt%)=100×(重量W3-重量W4)/重量W3
 なお、本実施形態の繊維集合体は、水で飽和していない場合であれば、含水率が50wt%未満であってもよい。特に、保管時や輸送時などでは、繊維集合体における含水率が低いことが好ましく、その含水率は、例えば30wt%以下であり、10wt%以下、5wt%以下、1wt%以下、さらには0.1wt%以下であってもよい。含水率が低い繊維集合体は、培養肉を作製するための足場材として利用することにも適している。一方、喫食者に提供される繊維集合体は、含水率が50wt%以上(特に65wt%以上や71wt%以上)であることが好ましい。
(他の成分)
 本実施形態の繊維集合体は、非動物性タンパク質、多糖類P及び水以外の他の成分をさらに含んでいてもよく、含んでいなくてもよい。他の成分としては、動物性タンパク質、脂質、アルカリ化合物、凝固剤、酸化合物、軟化剤、潤滑剤、ゲル化剤、香料、調味料、保水剤、乳化剤、油分などが挙げられる。
 脂質としては、蜜蝋、リン脂質、脂肪酸などが挙げられる。アルカリ化合物、凝固剤及び酸化合物は、繊維を作製するための材料に由来するものであり、詳細は後述する。潤滑剤としては、グリセリンなどが挙げられる。潤滑剤は、繊維を巻き取るときなどに、繊維がフリーローラやガイドなどと接触して擦り剥けることを抑制することに適している。
 ゲル化剤は、繊維集合体の製造方法について後述する結着工程に用いられるものであり、例えば、二価の金属イオンを含む金属塩、及び酸発生剤を含む。繊維集合体において、酸発生剤が分解していてもよい。言い換えると、繊維集合体は、酸発生剤の分解物を含んでいてもよい。金属塩や酸発生剤の詳細は後述する。
 繊維集合体は、油分を含むことにより、食感がより改善される傾向がある。油分としては、サラダ油、菜種油(例えばキャノーラ油)、ヤシ油(ココナッツ油)、大豆油、パーム油、こめ油、ごま油、ひまわり油、とうもろこし油(コーン油)、綿実油、パーム核油、オリーブ油、べに花油(サフラワー油)、えごま油、あまに油、ひまし油などが挙げられる。
 水で飽和状態の繊維集合体において、他の成分の含有率は、例えば45wt%以下であり、25wt%以下、20wt%以下、15wt%以下、10wt%以下、5wt%以下、1wt%以下、さらには0.1wt%以下であってもよい。
 水で飽和状態の繊維集合体において、油分を含む場合の油分含有率は、例えば1wt%以上が好ましく、5wt%以上、10wt%以上、15wt%以上、さらには20wt%以上であってもよい。含有率の上限は、特に限定されず、例えば40wt%以下である。なお、油分含有率は、例えば酸分解法やジエチルエーテルを抽出溶剤として用いたソックスレー法により測定できる。
 なお、本実施形態の繊維集合体は、典型的には、可食性を有する。本明細書において、「繊維集合体が可食性を有する」とは、繊維集合体が、各国法令等により食品又は食品添加物として認められている物質のみから構成されていることを意味する。繊維集合体は、食品又は食品添加物以外の成分、例えば可塑剤(軟化剤)や界面活性剤を実質的に含まなくてもよい。
(繊維集合体の形状)
 繊維集合体の形状は、特に限定されず、シート状、キューブ状、ディスク状などが挙げられる。繊維集合体は、模倣対象の肉と同様の形状を有していることが好ましい。模倣対象の肉としては、甲殻類、貝、魚、牛、豚、鳥(特に鶏)などの肉が挙げられる。繊維集合体は、牛肉(特に牛もも肉)の形状を有していることが好ましい。
(繊維集合体の製造方法)
 本実施形態の繊維集合体の製造方法は、非動物性タンパク質及び多糖類Q1を含む混合液を凝固液に吐出して、繊維を形成する紡糸工程を含むことが好ましい。紡糸工程によって得られた複数の繊維を集合させることにより繊維集合体を作製することができる。なお、多糖類Q1は、凝固液と接触することにより、上述した多糖類P1に変化するものであることが好ましい。
 紡糸工程は、例えば、図3に示す紡糸装置100を用いて作製することができる。紡糸装置100は、上記の混合液を吐出する吐出部10、吐出部10からの吐出物を凝固させる凝固液21を収容する凝固部20、及び凝固部20で形成された繊維6を巻き取る巻取部30を備えている。吐出部10、凝固部20及び巻取部30は、例えば、巻取部30によって繊維6を巻き取る方向(方向X)に沿ってこの順で並んでいる。
 吐出部10は、混合液を収容する収容部11と、混合液を吐出するノズル13とを有する。吐出部10としては、市販のギアポンプやシリンジポンプを備えた吐出装置などを利用できる。収容部11に収容された混合液は、上述のとおり、非動物性タンパク質及び多糖類Q1を含む。混合液は、アルカリ化合物をさらに含むことが好ましい。
 上記の混合液に含まれる非動物性タンパク質としては、繊維集合体について上述したものが挙げられる。なお、非動物性タンパク質は、通常、アミノ酸配列に基づいた固有の立体構造を有している。しかし、混合液がアルカリ化合物をさらに含む場合には、非動物性タンパク質の立体構造が破壊され、非動物性タンパク質がひも状の構造となる傾向がある。本明細書では、アルカリ化合物により非動物性タンパク質の立体構造が破壊されることを「アンフォールディング」と呼ぶことがある。
 本発明者らの検討によると、非動物性タンパク質をアルカリ化合物と混合し、非動物性タンパク質がアンフォールディングされた状態で紡糸工程を行うことによって、水で飽和状態の繊維集合体(特に繊維集合体に含まれる繊維)における多糖類Pの含有率R2や含水率R3を適切な値に調整しやすい。
 また、非動物性タンパク質をアンフォールディングすることによって、混合液に対する非動物性タンパク質の溶解性が向上し、繊維を形成しやすい傾向がある。言い換えると、混合液の紡糸性が向上する傾向がある。アンフォールディングした非動物性タンパク質を用いた場合、得られた繊維の強度が向上する傾向もある。
 混合液における非動物性タンパク質の含有率は、特に限定されず、例えば1.0wt%~30wt%である。
 多糖類Q1は、アルギン酸の一価金属塩を含むことが好ましい。なお、アルギン酸の一価金属塩は、典型的には、アルギン酸と一価の金属イオンとの塩である。本明細書では、「アルギン酸の一価金属塩」についても、単に「アルギン酸塩」と呼ぶことがある。
 混合液に含まれるアルギン酸塩(アルギン酸の一価金属塩)において、一価の金属イオンとしては、ナトリウムイオン、カリウムイオンなどのアルカリ金属イオンが挙げられ、ナトリウムイオンが好ましい。混合液において、アルギン酸塩は、典型的には、二価の金属イオンを実質的に含まない。
 混合液は、多糖類Q1としてアルギン酸の一価金属塩のみを含むことが好ましい。ただし、混合液は、上述した他の多糖類をさらに含んでいてもよい。
 混合液における多糖類Q1の含有率は、0.5wt%よりも大きいことが好ましく、0.8wt%以上、0.9wt%以上、1.0wt%以上、さらには1.1wt%以上であってもよい。多糖類Q1の含有率の上限は、特に限定されず、例えば5.0wt%以下である。なお、多糖類Q1の含有率が0.5wt%を大きく下回ると、繊維6の紡糸が難しい傾向がある。
 上述のとおり、アルカリ化合物は、混合液中で非動物性タンパク質をアンフォールディングするための成分である。アルカリ化合物は、可食性を有することが好ましい。アルカリ化合物としては、水酸化ナトリウムや炭酸水素ナトリウム(重曹)などが挙げられ、好ましくは水酸化ナトリウムである。混合液におけるアルカリ化合物の含有率は、例えば10wtppm~5000wtppmである。
 なお、混合液は、アルカリ化合物により、pHが8以上に調整されていることが好ましい。混合液のpHが8以上である場合、混合液中で非動物性タンパク質がアンフォールディングされている傾向がある。混合液のpHは、8~13であることが好ましく、9~12であることがより好ましい。
 混合液は、水をさらに含むことが好ましい。混合液における水の含有率は、特に限定されず、例えば50wt%~98.5wt%である。
 混合液は、繊維集合体に含まれる他の成分として上述したもの(軟化剤、潤滑剤、乳化剤、油分など)をさらに含んでいてもよい。なお、混合液が油分及び乳化剤を含む場合、混合液中で油分がエマルションを形成する傾向がある。この場合、混合液中により多くの油分を添加できる傾向がある。
 吐出部10において、混合液は、収容部11からノズル13に送られ、ノズル13の吐出口から吐出部10の外部に吐出される。ノズル13からの混合液の吐出速度は、特に限定されず、例えば0.1~10000mL/minである。ノズル13の吐出口のサイズは、目的とする繊維6の繊維径に応じて適宜調整することができる。ノズル13は、複数の吐出口を有していてもよい。この場合、ノズル13から吐出された吐出物を凝固させることによって、複数の繊維6を一度に作製することができる。ノズル13における吐出口の数は、特に限定されず、例えば1~20000個である。吐出口の数が多いノズル13を用いて、さらに、混合液の吐出速度を大きく設定すれば、繊維6を効率的に作製でき、生産性が向上する。ノズル13は、例えば、凝固部20に収容された凝固液21中に浸漬されている。
 凝固部20に収容された凝固液21は、ノズル13から吐出された吐出物(混合液)と接触することによって、吐出物を凝固させることができる。ノズル13からの吐出物が凝固することによって繊維6が形成される。凝固液21は、典型的には、凝固剤を含む水溶液である。
 凝固液21は、凝固剤として、二価の金属イオンを含む金属塩を含むことが好ましい。この金属塩としては、塩化カルシウムなどの塩化物や、炭酸カルシウムなどの炭酸塩が挙げられる。二価の金属イオンを含む金属塩は、例えば、凝固液21に溶解することによって、二価の金属イオンを発生させる。多糖類Q1がアルギン酸塩を含む場合、二価の金属イオンは、アルギン酸塩のGブロックとイオン結合を形成することができる。詳細には、混合液中でアルギン酸塩に含まれる金属イオン(一価の金属イオン)が二価の金属イオンと交換され、これにより、複数のアルギン酸分子が二価の金属イオンを介して架橋する。この架橋反応によって、ノズル13から吐出された吐出物のゲル化が進行し、繊維6が得られる。凝固液21における凝固剤の含有率は、特に限定されず、例えば0.1wt%~10wt%である。
 凝固液21は、凝固剤(金属塩)とともに酸化合物をさらに含むことが好ましい。凝固液21が酸化合物を含む場合、ノズル13から吐出された吐出物が凝固液21と接触することによって、アンフォールディングされた非動物性タンパク質の立体構造を元に戻すことができる。本明細書では、アンフォールディングされた非動物性タンパク質の立体構造が元に戻ることを「リフォールディング」と呼ぶことがある。
 酸化合物は、可食性を有することが好ましい。酸化合物としては、特に限定されず、クエン酸や酢酸などが挙げられ、好ましくは酢酸である。凝固液21における酸化合物の含有率は、特に限定されず、例えば0.1wt%~10wt%である。
 凝固液21は、アルカリ化合物をさらに含むことが好ましい。言い換えると、凝固液21は、酸化合物及びアルカリ化合物を含むことが好ましい。この凝固液21によれば、形成された繊維6の含水率を上昇させやすい傾向がある。アルカリ化合物としては、特に限定されず、水酸化ナトリウム、炭酸水素ナトリウムなどが挙げられる。凝固液21におけるアルカリ化合物の含有率は、酸化合物の含有率よりも低いことが好ましい。凝固液21におけるアルカリ化合物の含有率は、例えば0.1wt%~10wt%である。なお、凝固液21において、アルカリ化合物は、酸化合物と塩を形成していてもよい。
 なお、凝固液21は、酸化合物、又は、酸化合物とアルカリ化合物の混合物により、pHが6以下に調整されていることが好ましい。凝固液21のpHは、1~6であることが特に好ましい。上記pHの範囲であれば、非動物性タンパク質をリフォールディングできる。
 凝固液21は、アルコール類をさらに含むことが好ましい。アルコール類は、例えば、防腐剤として機能することができる。一例として、アルコール類を配合すると、凝固液21中の細菌類を殺菌できる傾向があり、また、細菌類の増殖を抑制できる傾向もある。アルコール類は、特に限定されず、例えば炭素数が5以下の低級アルコールであり、好ましくはエタノールである。凝固液21におけるアルコール類の含有率は、特に限定されず、例えば0.1wt%~90wt%である。
 凝固部20で形成された繊維6は、巻取部30に送られる。紡糸装置100は、繊維6を巻取部30に送るためのフリーローラ40及び41をさらに備えていてもよい。フリーローラ40は、例えば、凝固液21中に配置されており、ノズル13から10cm~10m程度離れている。繊維6は、フリーローラ40を通過してから、凝固部20の外部に送られる。凝固部20の外部に送られた繊維6は、フリーローラ41を通過してから巻取部30に送られる。なお、紡糸装置100は、フリーローラ40及び41を備えていなくてもよい。
 巻取部30は、凝固部20で形成された繊維6を巻き取るボビン31と、繊維6を巻き取るときに、繊維6をトラバースするためのガイド35とを有する。ボビン31の回転数は、特に限定されず、例えば0.1~500rpsである。ボビン31による繊維6の巻き取り速度は、特に限定されず、例えば0.1~500m/minである。1秒当たりのトラバースの回数は、特に限定されず、例えば0.1~10である。トラバースの速度は、特に限定されず、例えば10~500mm/secである。なお、トラバースの速度は、方向Xに直交する方向におけるガイド35の往復運動の平均速度を意味する。巻取部30は、ガイド35を有していなくてもよい。
 本実施形態の製造方法は、凝固液21から取り出した繊維6を洗浄する洗浄工程をさらに含むことが好ましい。洗浄工程によれば、繊維6の表面に付着した余分な凝固剤や酸化合物などを除去することができる。洗浄工程に用いられる洗浄液の温度は、例えば室温(25℃)である。繊維6を洗浄液と接触させる時間は、特に限定されず、例えば1秒間~1時間である。
 洗浄工程では、アルカリ水溶液又は水を洗浄液として使用することが好ましい。アルカリ水溶液は、例えば、アルカリ化合物を含む水溶液であり、アルカリ化合物としては上述したものを用いることができる。アルカリ水溶液におけるアルカリ化合物の含有率は、特に限定されず、例えば0.05wt%~10wt%である。アルカリ水溶液は、pHが8以上に調整されていることが好ましく、pHが8~11であることが特に好ましい。
 洗浄工程は、繊維6がボビン31に巻き取られる前に行ってもよく、繊維6がボビン31に巻き取られた後に行ってもよい。一例として、洗浄工程は、凝固部20と巻取部30との間に配置され、かつ洗浄液を収容する洗浄槽(図示せず)を利用して行ってもよい。洗浄槽によれば、凝固部20から送られた繊維6を洗浄液と接触させることによって、繊維6を洗浄することができる。洗浄処理が行われた繊維6は、洗浄槽から巻取部30に送られ、ボビン31に巻き取られる。別の例として、洗浄工程は、繊維6をボビン31に巻き取ることにより作製した巻回体に洗浄液をかける、又は、ボビン31ごと巻回体を洗浄液に浸漬させることによって行ってもよい。さらに、洗浄工程は、ボビン31から取り外した繊維6を洗浄液と接触させることによって行ってもよい。
 本実施形態の製造方法は、繊維6を乾燥させる乾燥工程をさらに含んでいてもよい。乾燥工程によれば、繊維6の含水率を所望の値に調節することができる。乾燥工程の条件は、特に限定されず、例えば室温(25℃)下で繊維6を放置することによって、繊維6を乾燥させてもよい。乾燥工程では、凍結乾燥や加熱乾燥を行ってもよい。加熱乾燥を行う場合、繊維6の加熱温度は、例えば50℃~120℃である。加熱乾燥を行う前には、繊維6をエタノールなどのアルコールに浸漬して、繊維6に含まれる水をアルコールと置換してもよい。
 本実施形態の製造方法は、複数の繊維6のうち、少なくとも2つの繊維6を互いに結着させる結着工程をさらに含むことが好ましい。繊維6同士の結着は、例えば、次の方法によって行うことができる。まず、多糖類Q2を複数の繊維6と接触させる。詳細には、多糖類Q2を水に分散させた分散液を複数の繊維6と接触させる。分散液は、後述するゲル化剤に含まれる一部の成分を含んでいてもよい。さらに、分散液は、繊維集合体に含まれる他の成分として上述したもの(乳化剤、油分など)を含んでいてもよい。なお、分散液が油分及び乳化剤を含む場合、分散液中で油分がエマルションを形成する傾向がある。この場合、分散液中により多くの油分を添加できる傾向がある。分散液が油分を含む場合、分散液における油分の含有率は、例えば0.5wt%~60wt%であり、好ましくは20wt%~40wt%である。
 多糖類Q2は、後述するゲル化剤と接触することにより、上述した多糖類P2に変化するものであることが好ましい。多糖類Q2は、アルギン酸の一価金属塩(アルギン酸塩)を含むことが好ましい。アルギン酸塩としては、多糖類Q1について上述したものが挙げられる。本実施形態の製造方法では、多糖類Q2としてアルギン酸塩のみを用いることが好ましく、アルギン酸塩以外の他の多糖類(特にメチルセルロース)を含まないことが好ましい。
 次に、多糖類Q2を複数の繊維と接触させた状態で、多糖類Q2をゲル化剤と接触させる。これにより、多糖類Q2(詳細には、多糖類Q2を含む分散液)がゲル化し、結着部が形成される。結着部を介して、繊維6同士が結着する。
 ゲル化剤は、二価の金属イオンを含む金属塩、及び酸発生剤を含むことが好ましい。金属塩としては、凝固剤について上述したものが挙げられ、炭酸カルシウムなどの炭酸塩であることが好ましい。酸発生剤は、例えば、加水分解によってカルボキシル基などの酸性基が形成される化合物である。酸発生剤は、グルコノデルタラクトン(GDL)を含むことが好ましい。多糖類Q2としてのアルギン酸塩と、酸発生剤としてのGDLとの組み合わせは、繊維集合体の押し込み力や引き裂き力を向上させることに適している。なお、酸発生剤としてGDLを用いた場合、繊維集合体は、酸発生剤の分解物としてグルコン酸を含んでいてもよい。なお、酸発生剤は、縮合リン酸、縮合リン酸塩などの縮合リン酸化合物を含まないことが好ましい。
 多糖類Q2をゲル化剤と接触させる方法は、特に限定されない。一例として、ゲル化剤が上記の金属塩及び酸発生剤を含む場合は、次の方法によって、多糖類Q2をゲル化剤と接触させることができる。まず、多糖類Q2及び金属塩を水に分散させた分散液を準備し、この分散液を複数の繊維6と接触させる。このとき、分散液と複数の繊維6とをよく混合することが好ましい。この態様において、分散液における多糖類Q2の含有率は、例えば0.5wt%~10.0wt%であり、好ましくは2.0wt%~6.0wt%である。分散液における金属塩の含有率は、例えば0.01wt%~5.0wt%であり、好ましくは0.1wt%~3.0wt%である。複数の繊維6の重量と、分散液の重量との比S1は、例えば5:5~9:1である。
 次に、酸発生剤をさらに添加する。酸発生剤の添加量は、全体の重量(複数の繊維6、分散液及び酸発生剤の合計重量)に対する酸発生剤の重量の比率S2が例えば0.1wt%~10.0wt%(好ましくは0.5wt%~3.0wt%)となるように調整することが好ましい。酸発生剤を添加することにより、分散液中の金属塩が酸発生剤と混合され、ゲル化剤として機能する。これにより、多糖類Q2をゲル化剤と接触させることができる。
 なお、酸発生剤は、上記の分散液と接触することにより酸を発生させる。詳細には、分散液中で酸発生剤が加水分解されて、酸性基が形成される。酸性基が形成されることによって、加水分解された酸発生剤が酸として機能する。この酸が金属塩と反応することによって、金属塩から二価の金属イオンが発生する。多糖類Q2がアルギン酸塩を含む場合、二価の金属イオンは、アルギン酸塩のGブロックとイオン結合を形成することができる。詳細には、アルギン酸塩に含まれる金属イオン(一価の金属イオン)が二価の金属イオンと交換され、これにより、複数のアルギン酸分子が二価の金属イオンを介して架橋する。この架橋反応によって、多糖類Q2(詳細には、多糖類Q2を含む分散液)のゲル化が進行し、結着部が形成される。
 結着工程では、酸発生剤の加水分解を促進させるために、多糖類Q2をゲル化剤と接触させた状態でこれらを加熱してもよい。この場合、加熱温度が例えば30℃~70℃であり、加熱時間が例えば1分間~5時間である。ただし、多糖類Q2をゲル化剤と接触させた状態で、これらを室温(25℃)下で1時間~72時間程度放置してもよい。
 結着工程では、成形用型を利用してもよく、利用しなくてもよい。成形用型は、例えば、模倣対象の肉と同様の形状の収容部を有する。一例として、複数の繊維6を多糖類Q2及びゲル化剤と接触させた状態で、複数の繊維6を成形用型に収容して、結着工程を行ってもよい。
 結着工程では、成形用型の内部を加圧してもよい。結着工程において、成形用型内は、例えば、周囲環境の大気圧から0.01kPa~100kPa加圧されていてもよい。加圧時間は、例えば、1秒間~10時間である。加圧下では、繊維6同士が接触しやすいため、繊維6同士の結着が促進される傾向がある。さらに、加圧下で結着工程を行うことにより、繊維集合体の空隙が低減する傾向もある。
 本実施形態の製造方法は、結着工程でゲル化剤と接触させた多糖類Q2(詳細には多糖類Q2がゲル化して形成された結着部)を冷凍する冷凍工程をさらに含んでいてもよい。冷凍工程によれば、結着部が脱水されて硬くなり、これにより繊維集合体の押し込み力が向上する傾向がある。本発明者らの検討によれば、この傾向は、結着部が、多糖類P2としてアルギン酸類のみを含み、かつ、非動物性タンパク質を実質的に含まない場合に特に顕著である。なお、非動物性タンパク質は、保湿性が高い傾向があるため、非動物性タンパク質を含む繊維は、冷凍工程で脱水されにくく、硬くなりにくい。
 冷凍工程において、結着部の冷凍温度は、例えば-80℃~0℃であり、好ましくは-20℃~0℃である。結着部の冷凍時間は、例えば10分間~24時間である。
 本発明は、その別の側面から、
 非動物性タンパク質及び多糖類Q1を含む混合液を凝固液に吐出して、繊維を形成する紡糸工程と、
 多糖類Q2を複数の繊維と接触させた状態で、多糖類Q2をゲル化剤と接触させることによって、複数の繊維のうち、少なくとも2つの繊維を互いに結着させる結着工程と、
 ゲル化剤と接触させた上記の多糖類Q2を冷凍する冷凍工程と、
を含む、繊維集合体の製造方法を提供する。
 本実施形態の製造方法は、冷凍工程の前又は冷凍工程の後に、繊維集合体を加熱する加熱工程をさらに含んでいてもよい。加熱工程によれば、繊維6に含まれる非動物性タンパク質が会合し、繊維集合体の食感がさらに改良されることがある。加熱工程は、繊維集合体の滅菌処理を兼ねることもできる。加熱工程の条件は、特に限定されず、例えば、温度が50℃~120℃であり、時間が1分間~1時間である。加熱工程は、50℃以上の水蒸気を繊維集合体に接触させて行ってもよい。
(繊維集合体の用途)
 本実施形態の繊維集合体は、食感が改良されている傾向がある。繊維集合体は、特に、甲殻類、貝、魚、牛、豚、鳥(特に鶏)などの肉の代替食品の用途に適している。繊維集合体は、場合によっては、培養肉の足場材に用いることもできる。すなわち、繊維集合体に、甲殻類、貝、魚、牛、豚、鳥(特に鶏)などの細胞を付着させ、当該細胞を培養することによって培養肉を作製することもできる。なお、繊維集合体を培養肉の足場材として用いる場合、繊維集合体には、細胞の接着性を向上させる接着向上剤が付与されてもよい。繊維集合体は、上記以外の用途、例えば化成品、薬品などの用途、にも利用することができる。
 以下に、実施例及び比較例により本発明をさらに詳細に説明するが、本発明はこれに限定されるものではない。
(例1)
 まず、ディスポカップに蒸留水を加え、ホモジナイザー(IKA社製、T18)で撹拌した。蒸留水の添加量は、カップに添加される材料の総重量が100重量部となるように調整した。次に、大豆タンパク質(不二製油社製、フジプロ748)7.5重量部と、粉末のアルギン酸ナトリウム(キミカ社製、キミカアルギンI-1G)1.1重量部をこの順番で少しずつカップに加えて、ダマにならないように水に溶解させた。次に、水酸化ナトリウム(富士フイルム和光純薬社製、食品添加物グレード)を水と混合することにより得られた濃度5wt%の水酸化ナトリウム水溶液を、カップに添加される材料の総重量100重量部に対して3重量部添加した。この混合液を、あわとり練太郎(シンキー社製、ARV-310)で回転数2000rpm、圧力0.6kPa、運転時間5分で、攪拌脱泡の処理を行った。これにより、非動物性タンパク質、アルギン酸塩(アルギン酸の一価金属塩)及びアルカリ化合物を含む混合液を調製した。混合液における水酸化ナトリウムの含有率は、1500wtppmであり、混合液のpHは10.2であった。なお、混合液において、大豆タンパク質は、水酸化ナトリウムによって、アンフォールディングされていた。
 次に、上述の紡糸装置100を用いて、以下の方法によって繊維を作製した。まず、紡糸装置の吐出部として、ギアポンプを備えた吐出装置を準備した。吐出部のノズルとしては、多孔ノズルを用いた。多孔ノズルには、直径0.2mmの吐出口が50個形成されていた。次に、上記の混合液を吐出部の収容部に注入し、ノズルから当該混合液を吐出させた。ノズルからの混合液の吐出速度は、21mL/minに設定した。ノズルから混合液が吐出されたことを確認してから、ノズルを凝固部に収容された凝固液に浸漬させた。凝固液としては、凝固剤としての塩化カルシウム(カナダ薬品工業社製)と、酸化合物としての氷酢酸(小堺製薬社製)と、アルカリ化合物としての水酸化ナトリウム(富士フイルム和光純薬社製、食品添加物グレード)とを含む水溶液を用いた。凝固液において、塩化カルシウムの含有率が0.5wt%であり、酢酸の含有率が2wt%であり、水酸化ナトリウムの含有率が0.4wt%であった。
 ノズルからの吐出物(混合液)は、凝固液と接触することによって凝固した。これにより、繊維が形成された。この繊維を手でつかみ、巻取部のボビン(シリコン製、直径7.5cm)にセットした。次に、ボビンを駆動させて、繊維を巻き取った。このとき、ボビンによる巻き取り速度を21m/minに設定した。ボビンの巻き取りによる繊維の延伸倍率は1.55であった。
 十分な量の繊維を巻き取った段階で、ノズルからの混合液の吐出を停止した。次に、ボビンから取り外した繊維をハサミで切断した。得られた繊維に25℃の水をかけて、数分間洗浄した。キムタオルを用いて、繊維から余分な水をふき取る操作を3回繰り返した。次に、作製した繊維を長さ9.5cmとなるように切断した。
 次に、粉末のアルギン酸ナトリウム(キミカ社製、キミカアルギンI-1G)、炭酸カルシウム及び水を混合し、これらを攪拌することにより分散液(ゾル)を調製した。分散液において、アルギン酸ナトリウムの含有率が4.0wt%であり、炭酸カルシウムの含有率が1.0wt%であった。次に、複数の繊維の重量と分散液の重量との比S1が5:5となるように、これらを混合した。
 次に、得られた混合物にグルコノデルタラクトン(GDL)を添加した。GDLの添加量は、全体の重量(複数の繊維、分散液及びGDLの合計重量)に対するGDLの重量の比率S2が2.5wt%となるように調整した。GDLの添加により、アルギン酸ナトリウムに、ゲル化剤(炭酸カルシウム及びGDL)を接触させた。GDLの添加後に、これらを速やかに混ぜて、シリコン製の成形用型に収容した。成形用型は、縦9.5cm、横9.5cm、深さ3cmのサイズを有する直方体状の収容部を有していた。
 次に、成形用型の収容部の上方をラップフィルムで封をして、その上に押し子(縦9.5cm、横9.5cm、高さ3cm)を配置した。さらに、押し子の上には、重さ2kgの分銅を1分間配置した。次に、分銅を取り外してから、成形用型を加熱乾燥機(エスペック社製、LC-114)に入れて、50℃で45分間加熱した。これにより、結着部が形成され、結着部を介して繊維同士が結着した(結着工程)。
 次に、得られた繊維集合体を容器(蒸し器)に収容し、当該容器内に100℃の水蒸気を30分間送り続けることにより、繊維集合体に対して加熱工程を実施した。加熱工程の後に、繊維集合体を-4℃の環境下に一晩放置して、結着部を冷凍する冷凍工程を行った。これを室温下で解凍することにより、例1の繊維集合体を得た。
(例2~8)
 結着工程において、分散液の組成や、複数の繊維の重量と分散液の重量との比S1、及び、全体の重量に対するGDLの重量の比率S2を表1に示すように変更したことを除き、例1と同じ方法によって例2~8の繊維集合体を得た。
(例9)
 例9では、市販の大豆タンパク食品(不二製油社製、商品名「大豆ミート(ブロック)」)を準備した。この大豆タンパク食品は、乾燥品であり、繊維を含んでいなかった。
(例10)
 まず、例1と同じ方法によって繊維を作製した。次に、キレート剤であるメタリン酸ナトリウム(関東化学社製、製品番号58023-17)を少量の水に分散させた分散液を調製した。複数の繊維に対して、分散液を添加し、これらを1分間混錬した。これにより、複数の繊維とキレート剤とが接触し、繊維の表面がゾル化した。キレート剤の添加量は、繊維の重量に対する、繊維に添加したキレート剤の重量の比率が2.0wt%となるように調整した。
 次に、キレート剤が繊維に接触した状態で、繊維を静置した。このとき、繊維に含まれる水によって、キレート剤の加水分解が徐々に進行した。次に、硫酸カルシウム(富士フイルム和光純薬社製、製品番号031-00935)を少量の水に分散させた分散液を調製した。繊維をキレート剤と接触させてから3分後に、上記の分散液を繊維に添加し、これらを約1分間混錬した。硫酸カルシウムの添加量は、繊維の重量に対する、繊維に添加した硫酸カルシウムの重量の比率が1.0wt%となるように調整した。
 複数の繊維と硫酸カルシウムとを混錬した後に、シリコン製の成形用型に収容し一晩静置した。成形用型は、縦9.5cm、横9.5cm、深さ3cmのサイズを有する直方体状の収容部を有していた。繊維を静置している間に、繊維の表面がゲル化した。これにより、接触した繊維同士が直接結着し、例10の繊維集合体が得られた。
<評価>
 上記の各例で準備した繊維集合体及び大豆タンパク食品を用いて、以下の評価を行った。なお、繊維集合体及び大豆タンパク食品は、いずれも常温の水に1時間浸漬させて、水で飽和状態とした。
[押し込み力]
 繊維集合体(及び大豆タンパク食品)について、次の方法によって、押し込み力を測定した。まず、繊維集合体から厚さ10mmの板状の試験片T1を切り出した。試験片T1をテクスチャーアナライザー(Stable Micro Systems社製、TA.XTplusC Texture Analyser)の支持部の上に配置した。次に、支持部の上部に位置する可動部に、縦10mm×横1.5mmの矩形の表面を有する圧子をセットした。このとき、圧子の表面が支持部から20mm離れるように圧子を配置した。次に、圧子を試験片T1に向かって移動させ、試験片T1の厚さ方向に圧子を試験片T1に押し込む押込試験を行った。押込試験は、押し込み速度2.0mm/secの条件で、圧子が試験片T1を厚さ方向に貫通するまで行った。押込試験の結果から、荷重と時間との関係を示すグラフを作成し、当該グラフから最大荷重(N)を読み取り、押し込み力として特定した。
 上記の試験は、各繊維集合体に対して少なくとも2回行った。表2及び図4は、複数回の試験で特定された押し込み力の平均値を示している。

 繊維集合体(及び大豆タンパク食品)について、次の方法によって、引き裂き力を測定した。まず、繊維集合体から板状の試験片(厚さ約10mm)を切り出した。図2について上述した態様で、この試験片に2つのフック(外径3.0mm)を突き刺し、試験片の厚さ方向に2つのフックを貫通させた。これらのフックをテクスチャーアナライザー(Stable Micro Systems社製、TA.XTplusC Texture Analyser)にセットした。詳細には、試験片の下方に位置する固定部に、一方のフックを固定し、試験片の上方に位置する可動部に他方のフックを取り付けた。次に、可動部により、他方のフックを上方に引っ張ることによって引張試験を行った。引張試験は、引張速度2.0mm/secの条件で、試験片が引き裂かれるまで行った。引張試験の結果から、荷重と時間との関係を示すグラフを作成し、当該グラフから最大荷重(N)を特定し、さらに、最大荷重(N)を試験片の厚さ(mm)で除した値を引き裂き力として特定した。
 上記の試験は、各繊維集合体に対して少なくとも2回行った。表2及び図4は、複数回の試験で特定された引き裂き力の平均値を示している。
[食感の評価]
 各例で準備した繊維集合体(及び大豆タンパク食品)について、次の方法によって食感を評価した。まず、繊維集合体を厚さ5mm程度にスライスし、その両面を油を引いたフライパンで1分程度ずつ(合計2分程度)焼いて調理した。なお、例10の繊維集合体については、調理前に、容器(蒸し器)に収容し、当該容器内に100℃の水蒸気を30分間送り続けることにより、加熱工程を実施した。
 次に、調理した繊維集合体を食感の評価に熟練した5名のパネラーが実際に喫食した。模倣対象となりうる肉の一例である牛もも肉(繊維集合体と同様の調理を行ったもの)を比較対象として、繊維集合体の噛み応え(噛みしめたときの硬さ)、凝集性(咀嚼時のまとまり感、ほぐれにくさ)、繊維感、ジューシー感について、官能評価を行った。詳細には、下記の評価基準によりパネラーごとに評価し、その平均値を求めた。
<評価基準>
・噛み応え
5点:牛もも肉と同等の硬さである。
4点:牛もも肉より少し柔らかい、又は、牛もも肉より少し硬い。
3点:牛もも肉より柔らかい、又は、牛もも肉より硬い。
2点:牛もも肉よりかなり柔らかい、又は、牛もも肉よりかなり硬い。
1点:牛もも肉と比べ物にならない程度に柔らかい、又は、牛もも肉と比べ物にならない程度に硬い。
・凝集性
5点:牛もも肉と同等のほぐれやすさである。
4点:牛もも肉より少しほぐれやすい、又は、牛もも肉より少しほぐれにくい。
3点:牛もも肉よりほぐれやすい、又は、牛もも肉よりほぐれにくい。
2点:牛もも肉よりかなりほぐれやすい、又は、牛もも肉よりかなりほぐれにくい。
1点:牛もも肉と比べ物にならない程度にほぐれやすい、又は、牛もも肉と比べ物にならない程度にほぐれにくい。
・繊維感
5点:牛もも肉と同等の繊維感である。
4点:牛もも肉より少し繊維感を感じにくい、又は、牛もも肉より少し繊維感を感じる。
3点:牛もも肉より繊維感を感じにくい、又は、牛もも肉より繊維感を感じる。
2点:牛もも肉よりかなり繊維感を感じにくい、又は、牛もも肉よりかなり繊維感を感じる。
1点:牛もも肉と比べ物にならない程度に繊維感を感じにくい、又は、牛もも肉と比べ物にならない程度に繊維感を感じる。
・ジューシー感
5点:牛もも肉と同等のみずみずしさである。
4点:牛もも肉より少しパサつきを感じる、又は、牛もも肉より少しみずみずしい。
3点:牛もも肉よりパサつきを感じる、又は、牛もも肉よりみずみずしい。
2点:牛もも肉よりかなりパサつきを感じる、又は、牛もも肉よりかなりみずみずしい。
1点:牛もも肉と比べ物にならない程度にパサつきを感じる、又は、牛もも肉と比べ物にならない程度にみずみずしい。
 さらに、繊維集合体の総合的な食感を9点法の嗜好尺度で評価した。詳細には、下記の評価基準によりパネラーごとに評価し、その平均値を求めた。
<評価基準>
9点:牛もも肉と同じ食感であり、牛もも肉と区別がつかない。
6点:牛もも肉として許容できる食感である。
1点:牛もも肉の食感からは程遠く、食品として相応しくない食感である。
 表1に記載した材料の詳細は以下のとおりである。
 I-1G:アルギン酸ナトリウム(キミカ社製、キミカアルギンI-1G)
 フジプロ748:大豆タンパク質(不二製油社製、フジプロ748)
 油分:サラダ油(イオン、サラッとこくうまサラダ油)
 HPMC:ヒドロキシプロピルメチルセルロース(ユニテックフーズ社製、E19)
 表2及び図4からわかるとおり、押し込み力が15.0N~80.0Nであり、引き裂き力が0.15N/mm~1.5N/mmである例1~8の繊維集合体は、例9及び10に比べて、食感が改良されていた。
 本実施形態の繊維集合体は、代替食品の用途に適している。

Claims (20)

  1.  複数の繊維を含む繊維集合体であって、
     前記繊維集合体は、非動物性タンパク質及び多糖類Pを含み、
     下記試験1により測定された押し込み力が15.0N~80.0Nであり、
     下記試験2により測定された引き裂き力が0.15N/mm~1.5N/mmである、繊維集合体。
     試験1:水で飽和状態の前記繊維集合体から、厚さ10mmの板状の試験片T1を切り出す。縦10mm×横1.5mmの矩形の表面を有する圧子を用いて、押し込み速度2.0mm/secの条件で、前記試験片T1の厚さ方向に前記圧子を前記試験片T1に押し込む。このときの最大荷重(N)を押し込み力として特定する。
     試験2:水で飽和状態の前記繊維集合体から板状の試験片T2を切り出す。前記試験片T2に2つのフック(外径3.0mm)を突き刺し、前記試験片T2の厚さ方向に2つの前記フックを貫通させる。引張速度2.0mm/secの条件で、2つの前記フックを互いに引き離す方向に引っ張り、前記試験片T2を引き裂く。このときの最大荷重(N)を前記試験片T2の厚さ(mm)で除した値を引き裂き力として特定する。
  2.  複数の前記繊維のうち、少なくとも2つの前記繊維を互いに結着させる結着部を有する、請求項1に記載の繊維集合体。
  3.  前記多糖類Pは、アルギン酸の二価金属塩を含むアルギン酸類を有する、請求項1に記載の繊維集合体。
  4.  前記アルギン酸類は、アルギン酸をさらに含む、請求項3に記載の繊維集合体。
  5.  前記非動物性タンパク質は、大豆タンパク質、エンドウ豆タンパク質、レンズ豆タンパク質、ひよこ豆タンパク質、そら豆タンパク質、ジャガイモタンパク質、セイタンタンパク質、アマランサスタンパク質、キノアタンパク質、小麦タンパク質、米タンパク質及びトウモロコシタンパク質からなる群より選ばれる少なくとも1つを含む、請求項1に記載の繊維集合体。
  6.  油分をさらに含む、請求項1に記載の繊維集合体。
  7.  可食性を有する、請求項1に記載の繊維集合体。
  8.  複数の前記繊維の平均繊維径が1cm以下である、請求項1に記載の繊維集合体。
  9.  複数の前記繊維の平均長さが1cm以上である、請求項1に記載の繊維集合体。
  10.  請求項1~9のいずれか1項に記載の繊維集合体の製造方法であって、
     前記製造方法は、
     前記非動物性タンパク質及び多糖類Q1を含む混合液を凝固液に吐出して、前記繊維を形成する紡糸工程を含む、繊維集合体の製造方法。
  11.  前記多糖類Q1は、アルギン酸の一価金属塩を含む、請求項10に記載の製造方法。
  12.  前記混合液は、アルカリ化合物をさらに含む、請求項10に記載の製造方法。
  13.  前記凝固液は、酸化合物を含む、請求項10に記載の製造方法。
  14.  前記凝固液は、アルコール類を含む、請求項10に記載の製造方法。
  15.  複数の前記繊維のうち、少なくとも2つの前記繊維を互いに結着させる結着工程をさらに含む、請求項10に記載の製造方法。
  16.  前記結着工程は、多糖類Q2を複数の前記繊維と接触させた状態で、前記多糖類Q2をゲル化剤と接触させることによって行う、請求項15に記載の製造方法。
  17.  前記多糖類Q2は、アルギン酸の一価金属塩を含む、請求項16に記載の製造方法。
  18.  前記ゲル化剤は、二価の金属イオンを含む金属塩、及び酸発生剤を含む、請求項16に記載の製造方法。
  19.  前記酸発生剤がグルコノデルタラクトンを含む、請求項18に記載の製造方法。
  20.  前記ゲル化剤と接触させた前記多糖類Q2を冷凍する冷凍工程をさらに含む、請求項16に記載の製造方法。
