Description

撮像装置の制御方法および撮像装置
 本発明は、検体を測定する撮像装置の制御方法および撮像装置に関する。
 基板上にタンパク質やペプチドなどのペプチド結合を有した生体物質を多数固定したタンパク質アレイプレートあるいはペプチドアレイプレートが知られている。これを用いると基板上に固定された多数の生体物質に対する相互作用を一度に行うことができる。本アレイプレートは、生体由来の液状検体、例えば血液、細胞抽出液、唾液、組織間液などと多数のタンパク質あるいはペプチドとの相互作用を網羅的に解析するのに有効である。このような解析により、検体の特性を測定することができる。
 以下、基板上のタンパク質あるいはペプチドの固定部位をスポットと表記する。
 検体との相互作用を受けたスポットを観測する方法として、例えば、蛍光プローブによってスポットを標識することで、どのスポットが相互作用を受けたかを識別する方法が知られている。蛍光プローブによって標識されたアレイプレートを観察する装置として、マイクロアレイスキャナが知られている(特許文献1)。
 特許文献1では、照射光学系、蛍光対物光学系、2次元走査系を有している。照射光学系は、アレイプレートに複数の波長のレーザ光を集光して照射する機能を有する。蛍光対物光学系は、蛍光プローブによって標識されたスポットからの蛍光の光量を検出する機能を有する。
 2次元走査系は、アレイプレートあるいは光学系を2次元走査することで、アレイプレート上のスポットの蛍光光量を反映した撮像画像を取得する機能を有する。また、蛍光対物光学系としては共焦点光学系を用いている。アレイプレートにはガラスの厚みや傾きに個体差があるため、ピントが合う深さが浅い共焦点光学系においては、アレイプレート全面で蛍光画像を得るのが難しい。
 これに対して、特許文献1では、アレイプレート全面でピントの合った蛍光画像を得るために、2次元走査と同時に、フォーカスセンサを用いた自動焦点調整を行っている。
 一方、光学系を2次元走査する際に、往路と復路でレーザ光の波長を切り替えて、走査方向の違いによる画像上のずれを低減させる技術が特許文献2に開示されている。

 複数の波長の蛍光検出を行うマイクロアレイスキャナにおいて、各波長のレーザ光のフォーカス位置をアレイプレート全面にわたって完全に合わせこむのは難しく、波長間でフォーカス位置にずれが生じるおそれがある。その結果、波長によって蛍光画像の輝度が低下したり、アレイプレート内で輝度ばらつきが大きくなったりするおそれがある。アレイプレートには個体間のタンパク質量の違いがあり、それを補正するために蛍光輝度の値やその比で補正することが行われているが、その場合には測定データの信頼性の低下につながるおそれがある。
 特許文献1の技術では、アレイプレートあるいは光学系を高速に2次元走査しながら、精度よく自動焦点調整を行うためには、高性能なフォーカスセンサや低振動なアクチュエータなどからなる高速フィードバック制御が必要になり、装置構成が複雑で高コストになるという課題がある。
 特許文献2の技術では、複数のレーザ波長を往路と復路で切り替える点には記載があるが、複数の波長間のフォーカス位置ずれの対策については記載がない。
 そこで、本発明は、簡便な構成で、複数の波長間のフォーカス位置ずれを低減し、アレイプレート全面でピントの合った複数の波長の蛍光画像を取得することが可能な方法および装置を提供することを目的とする。
 本発明は、長手方向と短手方向を有し一方の面にスポットアレイを有するアレイプレートに対して、一次光を照射し二次光を検出する対物光学系を走査し、蛍光像を取得する、撮像装置の制御方法であって、第1の波長の一次光を出射する前記対物光学系を用いて前記一方の面を測距し、前記長手方向における複数の位置に対応する前記第1の波長の一次光における複数の第1の距離情報を取得する、第1の測距工程と、前記複数の第1の距離情報を用いて、前記長手方向における主走査位置に応じた前記第1の波長の一次光に対応する第1の走査条件を取得する、第1の走査条件取得工程と、前記第1の走査条件および前記第1の距離情報の少なくともいずれかを用いて、前記主走査位置に応じた前記第1の波長の一次光とは光の波長が異なる第2の波長の一次光に対応する第2の走査条件を取得する、第2の走査条件取得工程と、前記第1の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第1の走査工程と、前記第2の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第2の走査工程と、前記主走査位置を前記長手方向に沿って変更する、副走査工程とを含む、撮像装置の制御方法である。

 本発明に係る検体測定装置によれば、高速なフィードバック制御によらず、複数の波長間のフォーカス位置ずれをアレイプレート全面にわたって補正することができる。これにより、簡便な構成で、複数の波長間のフォーカス位置ずれを低減し、アレイプレート全面でピントの合った複数の波長の蛍光画像を取得することが可能となる。その結果、アレイプレート個体間のタンパク質量の違いを蛍光輝度の比で補正したりする場合に、より信頼性を向上させることができる。

<第1の実施形態>
 第1の実施形態は、撮像装置の制御方法についてである。
 本発明の撮像装置の制御方法は、長手方向と短手方向を有し一方の面にスポットアレイを有するアレイプレートに対して、一次光を照射し二次光を検出する対物光学系を走査し、蛍光像を取得する、撮像装置の制御方法であって、第1の波長の一次光を出射する前記対物光学系を用いて前記一方の面を測距し、前記長手方向における複数の位置に対応する前記第1の波長の一次光における複数の第1の距離情報を取得する、第1の測距工程と、前記複数の第1の距離情報を用いて、前記長手方向における主走査位置に応じた前記第1の波長の一次光に対応する第1の走査条件を取得する、第1の走査条件取得工程と、前記第1の走査条件および前記第1の距離情報の少なくともいずれかを用いて、前記主走査位置に応じた前記第1の波長の一次光とは光の波長が異なる第2の波長の一次光に対応する第2の走査条件を取得する、第2の走査条件取得工程と、前記第1の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第1の走査工程と、前記第2の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第2の走査工程と、前記主走査位置を前記長手方向に沿って変更する、副走査工程とを含む。
 本発明の撮像装置の制御方法は、長手方向と短手方向を有し一方の面にスポットアレイを有するアレイプレートに対して、一次光を照射し二次光を検出する対物光学系を走査し、蛍光像を取得する、撮像装置の制御方法である。スポットアレイに検体を配置し、これに対し、1次抗体と2次抗体を結合させる。対物光学系が、アレイプレート上を走査しながら、例えば、2次抗体にある蛍光標識に対して、一次光を照射することで得られる、二次光を検出することで、検体測定や検査に用いることができる蛍光像を取得することができる。以下に、各工程について、説明する。一部の用語については、第2の実施形態で説明されているので、参照されたい。
[第1の測距工程]
 本発明の撮像装置の制御方法は、第1の波長の一次光を出射する前記対物光学系を用いて前記一方の面を測距し、前記長手方向における複数の位置に対応する前記第1の波長の一次光における複数の第1の距離情報を取得する、第1の測距工程を含む。
 第1の波長の一次光は、例えば、スポットアレイの蛍光標識を有する分子用の照射光である。この第1の波長の一次光を、検体測定などの正確な測定に用いるためには、第1の波長の一次光をアレイプレートの一方の面にあるスポットアレイ上で合焦させることが必要である。そこで、測距を行い、第1の波長の一次光が合焦するときの、対物光学系の光軸の方向に沿っての対物光学系からスポットアレイまでの距離の情報(第1の距離情報)を取得しておく。
 ところで、アレイプレートは、観察用の試料台に固定され、対物光学系が走査する二次元方向と、なるべく平行になるように配置されるが、スポットアレイの並びと対物光学系が走査する二次元方向との位置関係は、完全な平行とはならず、僅かな傾きができることがある。そのため、特に、長手方向では、対物光学系の光軸に沿っての、アレイプレートと対物光学系との距離が、変わることがある。そこで、アレイプレートの長手方向に異なる1つ以上の位置で、第1の波長の一次光が合焦するときの、対物光学系の光軸の方向に沿っての対物光学系からスポットアレイまでの距離の情報(第1の距離情報)を取得しておく。
[第1の走査条件取得工程]
 本発明の撮像装置の制御方法は、前記複数の第1の距離情報を用いて、前記長手方向における主走査位置に応じた前記第1の波長の一次光に対応する第1の走査条件を取得する、第1の走査条件取得工程を含む。
 第1の測距工程で得られた第1の距離情報とは、アレイプレートの長手方向に異なる少なくとも1つ以上の位置で、第1の波長の一次光が合焦するときの、対物光学系の光軸の方向に沿っての対物光学系からスポットアレイまでの距離の情報であった。実際の走査は長手方向の多地点で行うから、少なくとも1つ以上の位置に対する第1の距離情報から、データの内挿または外挿を行って、それぞれの主走査位置に対する対物光学系の位置の情報(多地点での情報)を得る。ここで、主走査位置とは、例えば、短手方向に走査を開始するときの始点であって、長手方向の多地点のどこかである。
 データの内挿または外挿の方法としては、例えば、2つの位置間で、直線の傾きを仮定することである。アレイプレート自体の反りなどを考慮して、3点以上の位置情報に対して、フィッティング関数を当てはめて、データ内挿または外挿を行ってもよい。この場合、フィッティング関数とは、例えば、2次以上の高次関数であることが好ましい。このようにすることで、本発明の撮像装置の制御方法を、より簡単な方法で、正確な検体測定を行うことができる。
[第2の走査条件取得工程]
 本発明の撮像装置の制御方法は、前記第1の走査条件および前記第1の距離情報の少なくともいずれかを用いて、前記主走査位置に応じた前記第1の波長の一次光とは光の波長が異なる第2の波長の一次光に対応する第2の走査条件を取得する、第2の走査条件取得工程を含む。
 本発明の撮像装置の制御方法は、前記第1の距離情報に基づいて、前記第2の波長の一次光における第2の距離情報を出力する、距離補正工程をさらに含み、前記第2の走査条件取得工程は、前記複数の第1の距離情報を用いて前記距離補正部より取得した前記第2の距離情報と、前記第1の走査条件とを用いて、前記第2の走査条件を取得してもよい。
 この場合、例えば、第2の距離情報は、第1の距離情報に定数を加減して求めることができる。このようにすることで、より簡便な方法で、測定結果が信頼できる蛍光像が得られる。これは、第1の波長の一次光の焦点距離と第2の波長の一次光の焦点距離との差が一定である仮定の下において成立しうる。得られた第2の距離情報と第1の走査条件を基に、第2の走査条件を求めることができるが、第2の距離情報の求め方は、定数の加減に限られない。
 本発明の撮像装置の制御方法は、前記第1の走査条件に対応する前記第2の波長の一次光における第2の走査条件を出力する、走査条件補正工程をさらに含み、前記第2の走査条件取得工程は、前記第1の走査条件に基づいて前記走査条件補正部から前記第2の走査条件を取得してもよい。
 この場合、例えば、第2の走査条件を第1の走査条件と同じにすることなどが考えられる。このようにすることで、より簡便な方法で、測定結果が信頼できる蛍光像が得られる。第2の走査条件の求め方は、第1の走査条件の求め方と同じにすることに限られない。
[第1の走査工程]
 本発明の撮像装置の制御方法は、前記第1の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第1の走査工程を含む。
 第1の走査工程では、第1の走査条件取得工程で得た、それぞれの主走査位置に対する対物光学系の位置の情報を基に、対物光学系を配置し、第1の波長の一次光をスポットアレイに向かって照射しながら、対物光学系をアレイプレートの短手方向に走査をする。
 本発明の撮像装置の制御方法は、第1の走査工程において、前記対物光学系が検出した蛍光強度情報と走査位置情報とに基づいて、前記第1の波長の一次光を前記スポットアレイに照射することで得られる第1の蛍光像を取得する工程を含むことが好ましい。このようにすることで、高速なフィードバック制御によらず、複数の波長間のフォーカス位置ずれをアレイプレート全面にわたって補正することができる。
[第2の走査工程]
 本発明の撮像装置の制御方法は、前記第2の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第2の走査工程を含む。
 第2の走査工程では、第1の走査工程と同様に、第2の走査条件取得工程で得た、それぞれの主走査位置に対する対物光学系の位置の情報を基に、対物光学系を配置し、第2の波長の光をスポットアレイに向かって照射しながら、対物光学系をアレイプレートの短手方向に走査をする。
 前記第2の走査工程において、前記対物光学系が検出した蛍光強度情報と走査位置情報とに基づいて、前記第2の波長の一次光を前記スポットアレイに照射することで得られる第2の蛍光像を取得する工程を含むことが好ましい。このようにすることで、高速なフィードバック制御によらず、複数の波長間のフォーカス位置ずれをアレイプレート全面にわたって補正することができる。
[副走査工程]
 本発明の撮像装置の制御方法は、前記主走査位置を前記長手方向に沿って変更する、副走査工程を含む。
 第1の走査工程と第2の走査工程とが一旦終わったら、アレイプレートの長手方向の位置を変更し、再び、第1の走査工程と第2の走査工程とを行う。前記第1の走査条件取得工程は、前記副走査工程毎に、前記第1走査条件を取得し、前記第2の走査条件取得工程は、前記副走査工程毎に、前記第2走査条件を取得することが好ましい。このようにすることで、より簡単な方法で、正確な検体測定を行うことができる。
 本発明の撮像装置の制御方法において、前記第1の走査工程と前記第2の走査工程は、前記第1の波長の一次光と前記第2の波長の一次光とのそれぞれが、前記長手方向において共通の主走査位置に対応する共通の走査経路を走査されるように行われることが好ましい。このようにすることで、第1の波長の一次光と第2の波長の一次光とで、同じ条件で、二次光を検出することができる。
 本発明の撮像装置の制御方法において、前記第1の走査工程と前記第2の走査工程は、前記第1の波長の一次光と前記第2の波長の一次光とが、前記共通の走査経路を逆方向に走査するように行われることが好ましい。特に、第1の波長の一次光による走査と第2の波長の一次光による走査とが、1ラインごとに交互に行われる場合、熱ドリフトによる位置ずれを最小限に抑えることができ、得られる蛍光像についてより信頼性を向上させることができる。
 本発明の撮像装置の制御方法は、前記第1の走査工程と前記第2の走査工程とをそれぞれ1回以上行う毎に、前記副走査工程を1回行うことが好ましく、前記第1の走査工程と前記第2の走査工程とをそれぞれ1回行う毎に、前記副走査工程を1回行うことがより好ましい。このようにすることで、熱ドリフトによる位置ずれを低減することができる。
[第2の測距工程]
 本発明の撮像装置の制御方法は、前記対物光学系を用いて前記一方の面を前記第2の波長の一次光で測距し、前記第2の波長の一次光における第2の距離情報を取得する、第2の測距工程をさらに含むことが好ましい。
 第2の波長の一次光は、第1の波長の一次光とは異なる光の波長を有する照射光である。それ故に、たとえ合焦のために同じレンズを使ったとしても、第2の波長の一次光と第1の波長の一次光とでは、焦点距離が異なることがある。すなわち、第2の波長の一次光と第1の波長の一次光とでは、アレイプレート上で合焦させるためには、対物光学系の光軸に沿っての、アレイプレートと対物光学系との距離が、異なってよい。
 そこで、第2の測距工程は、第1の測距工程とは別に、第1の測距工程と同様のことを行う。第2の測距工程では、第1の測距工程と同様の理由で、アレイプレートの長手方向に異なる少なくとも1つ以上の位置で、第2の波長の一次光が合焦するときの、対物光学系の光軸の方向に沿っての対物光学系からスポットアレイまでの距離の情報を取得しておく。本発明の撮像装置の制御方法は、第2の測距工程が、前記長手方向における複数の位置で行われてもよい。この情報を第2の距離情報としてもよい。
 第2の測距工程を行う場合の、第2の走査条件取得工程では、例えば、第1の走査条件取得工程と同様に、第2の波長の一次光に対して、走査条件を取得する。第1の走査条件取得工程と同様に、データの内挿または外挿の方法としては、例えば、2つの位置間で、直線の傾きを仮定することである。3点以上の位置情報に対して、フィッティング関数を当てはめて、データ内挿または外挿を行ってもよい。この場合、フィッティング関数とは、例えば、2次以上の多次関数であることが好ましい。このようにすることで、本発明の撮像の制御方法を、より簡単な方法で、正確な検体測定を行うことができる。
 本発明の撮像装置の制御方法は、前記第2の走査条件取得工程が、前記第2の測距工程で取得した前記第2の距離情報と、前記第1の走査条件とを用いて、前記第2の走査条件を取得してもよい。このようにすることで、アレイプレート全面でピントの合った複数の波長の蛍光画像を取得することができる。第2の走査条件の取得方法は1つに限られず、このように様々な方法がある。
[その他]
 本発明の撮像装置の制御方法は、前記スポットアレイの蛍光像を撮像するための一次光は、前記第1の波長の一次光および前記第2の波長の一次光のいずれか一方を含み、前記スポットアレイ上の反応生成物を標識する二次抗体の蛍光像を撮像するための一次光は、前記第1の波長の一次光および前記第2の波長の一次光のいずれか他方を含むことが好ましい。
 配列された複数のスポットが設けられたアレイプレートに対して、第1の波長の一次光を照射することで撮像された第1の蛍光像と、前記スポットの機能情報を取得するために前記第1の波長の一次光と光の波長が異なる第2の波長の一次光を照射することで撮像された第2の蛍光像を取得することができる。そして、第1の蛍光像と第2の蛍光像は、画像解析PCを用いて取得することができる。
 アレイプレートは、複数種類のタンパク質がアレイ状にスポッティングされている。一つのスポットは直径が100μm程度で、スポットの間隔は約40μmである。スポットは9×9のブロックを単位として構成されており、ブロックは3×6のマトリクス状に配列されている。
 本実施形態のアレイプレートのスポットは、例えば、Alexa Flour 680で蛍光標識することができる。一部のタンパク質スポットはリン酸化しており、リン酸化したタンパク質スポットのみ、例えば、Alexa Flour 790で蛍光標識することができる。各スポットがタンパク質BCAR1を含んでいる場合は、タンパク質BCAR1にGST(グルタチオンS-トランスフェラーゼ)をタグ付けしておき、そのGSTタグを波長670nmの光に対して蛍光を発する蛍光プローブで標識することもできる。
 Alexa Flour 680またはGSTタグで蛍光標識されたタンパク質スポットを励起するために、第1の波長の一次光の波長が、670nmであることが好ましい。また、Alexa Flour 790で蛍光標識されたタンパク質スポットを励起するために、第2の波長の一次光の波長が、780nmであることが好ましい。このようにするとき、アレイプレートを波長670nmの光で励起して取得した蛍光画像が第1の蛍光像であり、アレイプレートを波長780nmの光で励起して取得した蛍光画像が第2の蛍光像である。
<第2の実施形態>
 第2の実施形態は、撮像装置についてである。
 本発明の撮像装置は、長手方向と短手方向を有し、一方の面にスポットアレイを有するアレイプレートを、光学的に二次元に撮像する、撮像装置であって、前記アレイプレートが載置される、載置部と、異なる波長の一次光を切り替えて射出する光源と光学的に結合し、前記アレイプレートに一次光を出射する光射出部と、前記アレイプレートからの二次光を検出する採光部とを含む、対物光学系と、前記載置部と前記対物光学系との前記一次光の光軸方向における作動距離を規定する、距離規定部と、前記載置部と前記対物光学系との、前記光軸方向と交差する面内における相対位置を変更する、走査部と、前記一次光の波長毎の前記面内の複数位置における前記作動距離を走査条件として取得する走査条件取得部と、前記一次光の波長に関する情報と、前記採光部からの受光強度に関する情報と、前記相対位置に関する情報とに基づいて、前記一次光に対応する蛍光像を取得する画像化部と、を有する。
(撮像装置)
 第2の実施形態に係る撮像装置100について、図1~図3および図7を用いて説明する。
 撮像装置100は、長手方向と短手方向を有し、一方の面にスポット202を有するアレイプレート101を、光学的に二次元に撮像する撮像装置であるため、スキャナ100、蛍光スキャナ100と換言される場合がある。
(載置部)
 載置部109は、Y方向リニアステージ109上に設けられた被載置部(不図示)を備えている。
(対物光学系)
 撮像装置100は、アレイプレート101が載置される載置部109と、異なる波長λ1、λ2の一次光Ip(Ip1、Ip2)を切り替えて射出する光源と光学的に結合し、アレイプレート101に一次光Ipを出射する光射出部104iと、アレイプレート101からの二次光Isを採光する光採光部104cとを含む、対物光学系104と、を含む。
 対物光学系104は、共焦点光学系103と光学的に結合され、対象物であるアレイプレート101に対して、一次光Ipを照射し、アレイプレート101からの二次光Isを採光する。
(距離規定部)
 また、撮像装置100は、さらに、載置部109と対物光学系104との一次光lpの光軸方向における作動距離WDを規定する距離規定部316を備える。距離規定部316は、高さ走査モータ113、モータドライバ114、モータ制御回路309、を介して、載置部109と対物光学系104との一次光Ipの光軸方向における作動距離WDを規定する。距離規定部316は、高さ走査モータ113、モータドライバ114、モータ制御回路309、を介して、載置部109と対物光学系104との一次光Ipの光軸方向における作動距離WDを制御すると換言される。
(走査部)
 また、撮像装置100は、載置部109と対物光学系104との光軸方向と交差する面内における相対位置を変更する走査部170(主走査部190、副走査部180)、とを備える。副走査部180は、対物光学系104を、不図示のベースに対して、相対的にY軸方向に走査するように構成され、Y方向リニアステージ109、副走査モータ110と、備える。主走査部190は、対物光学系104を、不図示のベースに対して、相対的にX軸方向に走査するように構成され、モータドライバ115、主走査モータ107、ピストンクランク機構120、エンコーダ108、で構成される。ピストンクランク機構120は、クランク118、コネクティングロッド119で構成される。

(画像化部)
 また、撮像装置100は、さらに、一次光の波長λ1に関する情報Iwpと、採光部724、725からの二次光lsの受光強度に関する情報Iisと、相対位置に関する情報Irpとに基づいて、画像再構成を行う画像化部317を備える。画像化部317は、一次光lp(lp1またはlp2)に対応する蛍光像を形成し取得する。画像化部317は、一次光の波長λ1に関する情報Iwpを光源制御回路305より取得し、データ取得制御回路312より二次光lsの受光強度に関する情報Iisを取得し、相対位置に関する情報Irpを座標算出回路310より取得する。走査条件取得部315、距離規定部316、画像化部317は、いずれも、CPU301、メモリ303、不図示の記憶部、通信回路313、メモリ制御回路304、等によりその構成の一部または全部を置換することが可能である。
 このとき、本発明の撮像装置は、光源をさらに有し、距離補正部をさらに有し、走査条件補正部をさらに有することが好ましい。ここで、光源は、下記の光源102に適用でき、距離補正部は、下記のモータドライバ114と高さ走査モータ113とに適用でき、走査条件補正部は、下記のモータドライバ115と主走査モータ107とモータドライバ111と副走査モータ110とに適用できる。
 また、本発明は、長手方向と短手方向を有し、一方の面にスポットアレイを有するアレイプレートを、光学的に二次元に撮像する、撮像装置であって、前記アレイプレートが載置される、載置部と、異なる波長の一次光を切り替えて射出する光源と光学的に結合し、前記アレイプレートに一次光を出射する光射出部と、前記アレイプレートからの二次光を検出する採光部とを含む、対物光学系と、前記載置部と前記対物光学系との前記光軸方向と交差する面内における相対位置を変更するために、前記短手方向と前記長手方向に沿って、前記相対位置を変更し、主走査を行う主走査部と、副走査を行う副走査部とをそれぞれ含む、走査部と、前記短手方向における副走査位置と、前記長手方向における主走査方向と、主走査回数とに応じて、前記光射出部から射出される光の波長を変更するように、前記光源と前記走査部とを制御する、射出波長制御部と、前記一次光の波長に関する情報と、前記採光部からの受光強度に関する情報と、前記相対位置に関する情報とに基づいて、前記一次光に対応する蛍光像を取得する、画像化部とを有する。

(射出波長制御部)
 撮像装置100は、アレイプレート101の長手方向における副走査位置Psと、短手方向における主走査方向(+X方向か、―X方向か)と、主走査回数次、とに応じて、光射出部104iから射出される光の波長λp(λp1かλp2)を変更するように、光源102と走査部170(主走査部190、副走査部180)とを制御する射出波長制御部318を備える。射出波長制御部318は、走査条件取得部315、距離規定部316、画像化部317と同様に、CPU301、メモリ303、不図示の記憶部、通信回路313、メモリ制御回路304、等によりその構成の一部または全部を置換することが可能である。
 このとき、本発明の撮像装置は、光源をさらに有することが好ましい。本発明の撮像装置は、前記載置部と前記対物光学系との前記一次光の光軸方向における作動距離を規定する、距離規定部をさらに有し、前記射出波長制御部は、前記距離規定部が規定する前記作動距離を前記一次光の波長に応じて制御することが好ましい。本発明の撮像装置は、前記一次光の波長毎の前記面内の複数位置における前記作動距離を走査条件として取得する走査条件取得部をさらに有することが好ましい。
 ここで、距離規定部は、下記のCPU301と座標算出回路310とに適用でき、走査条件取得部は、下記のCPU301と座標算出回路310とに適用できる。以下に説明をする。なお、第1の実施形態と重複する部分は、説明を省略することがある。
 本発明の撮像装置は、長手方向と短手方向を有し、一方の面にスポットアレイを有するアレイプレートを、光学的に二次元に撮像する。
 図1を用いて本発明に係る撮像装置について説明する。
 アレイプレート101は、測定対象である。アレイプレート上はアレイプレート101上に多数の生体物質のスポットが固定化され、蛍光標識されたものである。
 光源102は、波長670nmおよび波長780nm付近の光を発する半導体レーザである。それぞれの波長の発光および停止は独立して制御可能なものとする。
 共焦点光学系103は、光源102からの一次光をアレイプレートまで導くとともに、アレイプレート101上のスポットからの蛍光およびスライドガラス表面からの反射光を光センサまで導く共焦点光学系である。共焦点光学系103は、ピンホール、フィルタ、ダイクロイックミラー、1/4波長板、偏光ビームスプリッタ、レンズから構成される。共焦点光学系を用いることにより、スライドガラスからの自家蛍光成分の影響を低減し、スポット由来の蛍光成分測定の信号対雑音比を高めることができる。
 対物光学系104は、一次光をアレイプレート101上のスポットに照射させるための対物光学系であり、一次光をアレイプレート101の方向に向けるためのミラーおよび、一次光をアレイプレート101上のスポットに集光させるためのレンズから構成される。
 光センサ105は、光を電気信号に変換するための光センサであり、蛍光を取得するための光電子増倍管と反射光を取得するためのフォトダイオードなどで構成される。光センサ105はアレイプレート上のスポットからの蛍光とアレイプレート表面からの反射光を取得できるものとする。また、光センサ105は光源102の複数の波長に対応する複数の光電子増倍管、フォトダイオードなどで構成されているものとする。
 受信回路106は、光センサ105からの電気信号を増幅し、デジタル値に変換するための受信回路であり、増幅回路、フィルタ回路、ADコンバータなどから構成される。
 ピストンクランク機構120は、対物光学系104をアレイプレート101の短手方向に往復運動させるためのピストンクランク機構である。対物光学系104の往復運動により、光源102からの一次光はアレイプレート短手方向に走査される。なお、本発明において、アレイプレートの短手方向を主走査方向といい、ピストンクランク機構120による往復走査を主走査という。主走査のストロークは30mmとする。対物光学系104は主走査方向のガイド上で、主走査方向に動作方向が限定されている。
 主走査モータ107は、ピストンクランク機構120を高速回転させるためのパルスモータである。主走査モータ107の回転速度は1200rpmとする。
 エンコーダ108は、ピストンクランク機構120に設置されたリニアエンコーダであり、対物光学系104の主走査方向の位置を測定することができる。エンコーダ108は主走査方向の位置に応じてA相、B相、Z相からなる位相差パルス電圧を出力する。
 Y方向リニアステージ109は、アレイプレート101を水平面内で主走査と直交する方向に移動させるためのリニアステージであり、ボールねじ、原点センサなどから構成される。水平面内で主走査と直交する方向の走査を副走査という。リニアステージ上にはアレイプレートを載置するための載置台があり、使用者は事前に測定対象のアレイプレートを載置台の上に乗せる。
 副走査モータ110は、Y方向リニアステージ109に接続されるパルスモータである。パルスモータの回転運動は、Y方向リニアステージ109のボールねじにより直線運動に変換される。
 モータドライバ111は、副走査モータ110を回転させるためのモータドライバ回路である。モータドライバ111に信号を1パルス入力すると、パルスモータが0.72°回転し、アレイプレート101は副走査方向に2μm移動する。
 Z方向リニアステージ112は、アレイプレート101を垂直方向に移動させるためのリニアステージであり、ボールねじ、原点センサなどから構成される距離規定部316を構成する。したがって、Z方向リニアステージ112は、アレイプレート101と対物光学系104との間の距離を規定する距離規定部316により制御可能に構成されている。垂直方向の走査を高さ走査とよぶ。
 高さ走査モータ113は、Z方向リニアステージ112に接続されるパルスモータである。高さ走査モータ113の回転運動は、Z方向リニアステージ112のボールねじにより直線運動に変換される。
 モータドライバ114は、高さ走査モータ113を回転させるためのモータドライバ回路である。モータドライバ114に信号を1パルス入力すると、パルスモータが0.72°回転し、アレイプレート101は垂直方向上方に1μm移動する。
 モータドライバ115は、主走査モータ107を回転させるためのモータドライバ回路である。モータドライバ115に信号を1パルス入力すると、主走査モータ107が0.72°回転し、対物光学系104が主走査方向に沿って移動する。
 コントローラ116は、全体を制御するコントローラ回路であり、FPGA、CPU、メモリ、組込ソフトウェアなどで構成される。光源102、モータドライバ111、モータドライバ114、モータドライバ115を制御して、アレイプレート101上で一次光を主走査、副走査、高さ方向に走査する。これと同時に、エンコーダ108で測定される対物光学系104の位置情報と、光センサ105からの光信号データを取得して内部のメモリに保存する。
 コントローラ116は、内部にアレイプレートの高さ情報、傾き情報の取得手段、主走査、副走査および高さ走査のパルス列生成手段、ADコンバータ制御手段、同期制御手段、データ取得制御手段、メモリを備える。ADコンバータ制御手段は、受信回路106のADコンバータへ変換指示を行い、光センサ105の光量に対応した電気信号をAD変換し、デジタル値である光信号データを取得する。
 同期制御手段は、対物光学系104の位置情報に同期して副走査、高さ走査、データ取得のタイミングを制御する。
 これにより、対物光学系104がアレイプレート上の撮像範囲外にいる時にアレイプレートの厚みおよび傾きを補正し、アレイプレート101の全体でスポットが固定化されている面に光が合焦するように高さ走査を行う。
 さらに同期制御手段は、光源102の発光、波長の切り替えタイミングを制御する。また、対物光学系104がアレイプレート端部に近い所定の位置に到達したところで、副走査、高さ走査の開始および光源102の各波長の半導体レーザの発光および停止を制御する。
 具体的には、往路では780nmの波長のレーザを発光させ、670nmの波長のレーザの発光を停止する。復路では往路では670nmの波長のレーザを発光させ、780nmの波長のレーザの発光を停止する。高さ走査においては、往路では780nmのフォーカス位置に合うように高さを調整し、復路では670nmのフォーカス位置に合うように高さを調整する。それぞれの波長のフォーカス位置はアレイプレートごとの事前の測定で求め、アレイプレート全面での目標高さを算出しておく。
 ユーザインタフェース117は、使用者からの指示を受け付けたり、結果を表示したりするためのユーザインタフェースであり、キーボードやマウス、ディスプレイで構成される。
 コントローラ116は、ユーザインタフェース117を介して、アレイプレート撮像指示を受け付けたり、光信号データに基づく画像データを使用者に提示したりすることができる。また、使用者はアレイプレート撮像時に、ユーザインタフェース117上のGUIを介して撮像範囲、主走査方向および副走査方向の画素ピッチを指定することができる。
 クランク118は、ピストンクランク機構を構成するクランクであり、主走査モータ107の回転軸およびコネクティングロッド119とそれぞれジョイントを介して接続されている。クランクの長さはrとする。
 コネクティングロッド119は、ピストンクランク機構を構成するコネクティングロッドであり、クランク118および対物光学系104とそれぞれジョイントを介して接続されている。コネクティングロッドの長さをlとする。図16に、ピストンクランク機構120のクランク118、コネクティングロッド119、対物光学系104の位置関係を示す。
 続いて、図2A~図2Cを用いて本発明に係る検体について説明する。
 図2Aは、アレイプレート101を上から見た図であり、図2Bは、アレイプレート101を側面からみた図である。図2Cは、アレイプレートに枠を取り付けて観察液で満たした状態である。
 スライドガラス201上のスポット202の各々にはペプチド結合を含む生体物質が固定化されている。スライドガラス201と多数のスポット202でアレイプレート101が構成される。一つのスポットは1種類の生体物質が固定化されている。スポットの直径は約100μm、スポット間隔は200μmとする。また、アレイプレートの短手方向の長さは25mm、長手方向の長さは75mmである。
 アレイプレートの左上の点203を原点、短手方向右向きをX軸の正方向、長手方向下向きをY軸方向とする。XおよびY座標の単位をμmとするとアレイプレートの4隅の座標は(0,0)、(25000,0)、(0,75000)、(25000,75000)となる。
 ピストンクランク機構120のストロークは30mmであり、アレイプレート101の長さよりも5mm長い。すなわち、主走査では、アレイプレート左右に2.5mm長い範囲を走査され、走査範囲のX座標は-2500から27500の間である。
 一方、液体204は、アレイプレート101の一方の面アレイ状に配置されたスポット202と接するようにスライドガラス201と枠203とで規定される貯留部に貯留される。アレイプレート101上には、スポット作成上の都合や、使用者が把持する都合により、スポットが存在する領域と存在しない領域がある。液体204のある領域の4隅の座標は、本実施例では(2000,2000)、(23000,2000)、(2000,65000)、(23000,65000)とする。副走査のY座標の範囲は使用者が指定可能とする。
 枠203は、アレイプレートに取り付けられた枠であり、4辺を囲むことで内部に液体を保持することができる。液体204は枠203内部の保持された液体であり、スポット202を乾燥させずに保持する役割および蛍光測定時のバックグラウンドノイズを削減する役割を持つ。液体204を観察液という。観察液としては、アレイプレートと屈折率が近くアレイプレート上の物質を保護し、蛍光を発しないものが望ましい。
[実施例1]
 図3は本発明の実施例1のコントローラ116の内部構成を示すブロック図である。CPU301は、全体を制御する制御ソフトウェアを動作させるためのCPUであり、マイクロプロセッサ、キャッシュメモリなどで構成される。
 バスインタフェース302は、CPU301と各種周辺回路をつなぐためのバスインタフェースである。
 メモリ303は、使用者から入力される撮像条件や検体検査装置のパラメータ、光信号データを保存するためのメモリであり、DDR4-SDRAMやSSDなどで構成される。
 メモリ制御回路304は、バスインタフェース302を介したメモリへのアクセスコマンドに基づいてメモリ303を制御するためのメモリ制御回路である。
 光源制御回路305は、CPU301が光源102を制御するための光源制御回路であり、インタフェース変換回路やDAコンバータなどから構成される。CPU301は光源制御回路305を介して光源102のレーザ照射のオンオフおよび光量を制御することが可能である。
 ADコンバータ制御回路(ADC制御回路)306は、CPU301からの指示に基づき、受信回路106のADコンバータを制御し、光信号データを取得するADコンバータ制御回路である。ADコンバータ制御回路306は同期回路311からのデータ取得トリガ信号を受信すると、受信回路106のADコンバータに対し、変換開始信号を出力する。ADコンバータは変換開始信号が入力されたタイミングで受信回路内のアナログ信号のサンプリングを行い、光信号データに変換してADコンバータ制御回路306に出力する。ADコンバータ制御回路306はADコンバータから入力された光信号データをデータ取得制御回路312に転送する。
 モータ制御回路307は、CPU301からの指示に基づき、主走査モータ用のモータドライバ115への制御信号を生成する回路である。CPU301から主走査モータ107の回転速度、加速度、移動量、回転方向、回転開始タイミングを指示すると、それに応じた駆動パルス電圧を生成する。
 モータ制御回路308はCPU301からの指示に基づき、副走査モータ用のモータドライバ111への制御信号を生成する回路である。CPU301から副走査モータ110の回転速度、加速度、移動量、回転方向、回転開始タイミングを指示すると、それに応じた駆動パルス電圧を生成する。
 モータ制御回路309は、CPU301からの指示に基づき、高さ走査モータ用のモータドライバ114への制御信号を生成する回路である。CPU301から高さ走査モータ113の回転速度、加速度、移動量、回転方向、回転開始タイミングを指示すると、それに応じた駆動パルス電圧を生成する。
 座標算出回路310は、エンコーダ108からのA相とB相の2つの位相差パルス信号を計数し、対物光学系104の位置を算出する座標算出回路である。エンコーダ108の分解能を1μmとする。A相信号またはB相信号のレベルが変化し、かつA相信号のほうがB相信号よりも位相が進んでいる時に対物光学系104の座標を1μm増加させる。A相信号またはB相信号のレベルが変化し、かつB相信号の方がA相信号よりも位相が進んでいる時に対物光学系104の座標を1μm減少させる。
 同期回路311は、座標算出回路310から得られた座標情報をもとにデータ取得制御回路312、モータ制御回路308、モータ制御回路309、光源制御回路305へトリガ信号を生成する同期回路である。本発明において、ADコンバータ制御回路306へのトリガ信号をデータ取得トリガ信号という。モータ制御回路308へのトリガ信号を副走査トリガ信号という。モータ制御回路309へのトリガ信号を高さ走査トリガ信号という。光源制御回路305へのトリガ信号を波長切り替えトリガ信号という。
 モータ制御回路308は、副走査トリガ信号を受信すると、副走査方向の画素ピッチ分の移動量にあたる駆動パルス列をモータドライバ111へ出力する。副走査方向の画素ピッチは使用者により撮像開始時に指定され、メモリ303に保存されている。
 モータ制御回路309は高さ走査トリガ信号を受信すると、高さ走査方向の移動量にあたる駆動パルス列をモータドライバ114へ出力する。高さ走査方向の移動量はCPU301で算出される。算出方法は後述する。
 データ取得制御回路312は、ADコンバータ制御回路306から出力された光信号データを、メモリ制御回路304を介してメモリ303に転送する回路である。バッファメモリ、DMAコントローラなどから構成される。
 通信回路313は、撮像装置を外部のネットワークに接続するための通信回路である。通信方法はEthernetの規格に沿った通信プロトコルを用いる。撮像装置を外部のPCやサーバに接続し、遠隔で検体撮像を制御したり、外部の大容量ストレージにデータを保存したりすることができる。
 UI回路314は、撮像装置をユーザインタフェース117と接続するためのUI回路である。キーボードやマウスからの入力回路およびディスプレイを制御するための画像表示回路から構成される。
 コントローラ116を構成する周辺回路はFPGAやASICなどの半導体チップ上に実装され、クロックに同期して動作する。クロック周波数は100MHzとする。
 続いて図4、図5A~図5C、図6Aおよび図6Bを用いて本実施例のアレイプレートの撮像動作について説明する。
 図4は本実施例の撮像装置でアレイプレートの撮像を行う際の動作フローである。
 図5A~図5Cは撮像動作におけるアレイプレート上の副走査の位置関係を説明する図である。図5Aおよび図5Bは、アレイプレート101を上面からみた図である。
 図6Aおよび図6Bは撮像動作におけるアレイプレートの高さ走査の位置関係を説明する図である。図6Aおよび図6Bは、アレイプレート101を側面からみた例を示す図である。図6Aと図6Bとでは、各波長のフォーカス位置が異なる。
 本発明の撮像装置において、前記射出波長制御部は、第1の波長の一次光と、前記第1の波長の一次光とは光の波長が異なる第2の波長の一次光と、を切り替えるように前記光射出部から射出される光の波長を変更するように、前記光源と前記走査部とを制御することが好ましい。
 本発明の撮像装置において、前記射出波長制御部は、共通の走査経路上を前記第1の波長の一次光と前記第2の波長の一次光が主走査するように、前記第1の波長の一次光および前記第2の波長の一次光の一方を主走査した後で、前記第1の波長の一次光および前記第2の波長の一次光の他方に切り替えるように、前記光源と前記走査部とを制御することが好ましい。
 本発明の撮像装置において、前記射出波長制御部は、共通の走査経路上を前記第1の光で走査する第1の走査ステップと前記第2の光で走査する第2の走査ステップとをそれぞれ1回以上行う毎に、前記副走査するステップを1回行うように、前記光源と前記走査部とを制御することが好ましく、共通の走査経路上を前記第1の光で走査する第1の走査ステップと前記第2の光で走査する第2の走査ステップとをそれぞれ1回行う毎に、前記副走査するステップを1回行うように、前記光源と前記走査部とを制御することがより好ましい。
 これらのようにすることで、簡便な構成で、複数の波長間のフォーカス位置ずれを低減し、アレイプレート全面でピントの合った複数の波長の蛍光画像を取得することが可能となる。以下に、これらの好適な例について、図4のフローチャートを用いて説明する。
 ステップS401にて、使用者により撮像開始指示が行われると、CPU301は使用者がユーザインタフェース117を介して指定した撮像条件を読み込み、メモリ303に保存する。また、CPU301はコントローラ116内の各種回路に撮像条件に基づく情報を設定する。撮像条件として、アレイプレート101上の撮像範囲を示す点501(X1,Y1)、点502(X2,Y2)および主走査方向の画素ピッチXp、副走査方向の画素ピッチYp、主走査方向の回転速度Xsが入力される。
 本実施例ではX1=500、X2=22500、Y1=500、Y2=64500、Xp=10μm、Yp=10μm、Xs=1200rpmとする。本発明において、アレイプレート101上の点501、点502を対角とする矩形の領域を撮像領域503という。
 また、一点鎖線で示した枠内領域508は枠203の内壁を示す。本発明において、枠内領域508の内側は測定時に観察液で満たされている領域である。観察液としてはグリセロール溶液を用いる。枠内領域508ではアレイプレートと観察液との界面での反射光が少なく、蛍光測定におけるバックグラウンドノイズを少なくすることができる。
 また、主走査方向の画素数Nx=(X2-X1)/Xp、副走査方向の画素数Ny=(Y2-Y1)/Ypを算出しておく。なお、本実施例ではNx=2200、Ny=6400となる。
 ステップS402にて、CPU301はアレイプレート101の高さ情報および傾き情報を波長ごとに取得する。本発明において、アレイプレート101を構成する、スライドガラス201の上面、すなわちスポット202がある側の面に一次光を合焦させるための、Z方向リニアステージ112の移動量を高さ情報という。アレイプレートの副走査方向の傾きを傾き情報という。
 ステップS402にて、2つのY座標Y3、Y4に対する各波長の高さ情報Z3_670、Z3_780、Z4_670、Z4_780が測定により取得される。Z3_670はY3の位置で680nmの光を合焦させるための高さ情報、Z3_780はY3の位置で780nmの光を合焦させるための高さ情報である。同様にZ4_670はY4の位置で680nmの光を合焦させるための高さ情報、Z4_780はY4の位置で780nmの光を合焦させるための高さ情報である。
 本実施例ではY3=750、Y4=65000とする。仮に図6Aのような傾きの場合にはZ4_670>Z3_670、Z4_780>Z3_780となり、図6Bのような傾きの場合にはZ3_670<Z4_670、Z3_780<Z4_780となる。
 図5Aで示すように、Y3とY4は枠内領域508の外にあり、アレイプレート上には観察液でなく空気が存在する。そのため、Y3とY4では、アレイプレートと空気の界面の反射光からアレイプレートの高さを測定することができる。
 高さ情報の取得方法は後述する。
 また波長ごとの傾き情報K_670およびK_780は以下の式で算出される。K_670は670nmの一次光に対する傾き情報、K_780は780nmの一次光に対する傾き情報である。

 ステップS403にて、CPU301は、撮像条件および各波長の高さ情報、傾き情報K_670m、K_780に応じて各行の目標高さを波長ごとに算出する。任意の副走査位置座標Yにおける波長670nmの目標高さZ_670(Y)は以下の式で与えられる。波長670nmに対する目標高さ601は、図6Aおよび図6Bでアレイプレート101の表面に波長の670nmの一次光を合焦させるためのZ座標に相当する。

 同様に、任意の副走査位置座標Yにおける波長780nmの目標高さZ_780(Y)は以下の式で与えられる。波長780nmの目標高さ602は図6Aおよび図6Bでアレイプレート101の表面に波長の780nmの一次光を合焦させるためのZ座標に相当する。

 ステップS404にて、CPU301は、モータ制御回路307、308、309に指示を出し、アレイプレート101および対物光学系104を撮像開始位置に移動させる。
 撮像開始位置のX座標はピストンクランク機構120の走査範囲の端部であり、X座標値は-2500とする。撮像開始位置のY座標は撮像条件で指定されたY1とする。また、撮像開始位置の波長670nmの一次光に対するZ座標Z1_670は式3においてZ_670(Y1)であらわされる。同様に撮像開始位置の波長780nmの一次光に対するZ座標Z1_780は式4においてZ_780(Y1)であらわされる。
 ステップS405にて、CPU301は、モータ制御回路307に指示を出し、主走査モータ107を回転速度Xsで回転させる。これにより、対物光学系104がX方向に往復運動を開始する。エンコーダ108および座標算出回路310により対物光学系104のX座標はクロックごとに算出され、同期回路311に出力される。
 ステップS406にて、CPU301は光源制御回路305に指示を出し、光源102の発光を開始させる。これにより、対物光学系104を介してアレイプレート101へ光照射が開始される。この際に、同期回路311を用いて670nmの光と780nmの2種類の波長の光を主走査の往路と復路で交互に切り替えながらアレイプレート101に照射する。これにより波長間のクロストークを低減し、光信号データの測定精度を向上させる。
 ステップS407にて、同期回路311は対物光学系104が波長切り替え位置に到達したか否かを判定する。座標算出回路310から出力される対物光学系104のX座標が撮像領域外の所定の位置X3およびX4に移行した時に波長切り替え位置に到達したと判断する。
 X3およびX4の座標はあらかじめ設定され同期回路311に保存されている。X3、X4とも波長切り替え位置は改行位置X1、X2よりも対物光学系104のストローク端に近い位置に設定される。これは、波長を切り替えるのにもある程度の時間が必要であり、対物光学系が撮像範囲外にいる間に波長の切り替えを完了させるためである。本実施例ではX3=200、X4=22800とする。
 波長切り替え位置に到達した場合には、同期回路311は光源制御回路305に対し波長切り替えトリガ信号を出力するとともに、モータ制御回路309に対し高さ走査信号を出力しステップS421とステップS418を並行して動作させる。波長切り替え位置に到達していない場合には同期回路311は波長切り替えトリガ信号を出力せずにステップS408に進む。
 ステップS408にて、同期回路311は対物光学系104が改行位置に到達したか否かを判定する。座標算出回路310から出力される対物光学系104のX座標が撮像領域内から撮像領域外に移行した時に改行位置に到達したと判断する。本実施例では現在の主走査方向が復路方向(X座標が減少する方向)の場合に、対物光学系104のX座標がX1よりも小さくなったところで改行位置に到達したと判断する。本実施例では対物光学系104のX座標がX2よりも大きくなったところでは改行位置に到達したと判断しない。
 往路方向の走査は、図5Bの軌跡504であらわされる。復路方向の走査は、図5Bの軌跡506であらわされ往路方向の軌跡と同じ軌跡をたどり改行位置に戻る。主走査方向の初期値は往路方向であり、その後ストローク端に到達するたびに復路方向、往路方向を交互に繰り返す。
 改行位置に到達した場合には、同期回路311は副走査トリガ信号を出力し、ステップS417に進む。改行位置に到達していない場合には同期回路311は副走査トリガ信号を出力せずにステップS409に進む。
 ステップS409にて同期回路311は対物光学系104が目標位置に到達したか否かを判定する。図5Bの点507はアレイプレート101上で画像化される画素の位置を示す。
 目標位置とは、画素を生成するためにアレイプレート上で光信号データを取得する点であり、N番目の目標位置のX座標P(N)は、以下の式であらわされる。

 目標位置は軌跡上の複数の点であり、X方向のピッチがXp、Y方向のピッチはYpであり、本実施例では画素の位置507と一致している。また、目標位置の初期値はP(0)であり、同期回路311内部に保存されている。最初の目標位置の判定においては、座標算出回路310から出力される対物光学系104のX座標が往路方向(X座標が増加する方向)でP(0)を通過した場合に目標位置に到達したと判断する。
 2回目以降の目標位置の判定においては、座標算出回路310から出力される対物光学系104のX座標が、後述するステップS411で更新された目標位置を通過した時に目標位置に到達したと判断する。目標位置に到達した場合には、同期回路311はデータ取得トリガ信号を出力し、ステップS410に進む。まだ目標位置に到達していない場合には、同期回路311はデータ取得トリガ信号を出力せずにステップS412に進む。
 ステップS410にてADコンバータ制御回路306はデータ取得トリガ信号を受信すると、変換開始信号を出力し、光センサ105からの出力電圧を受信回路106で増幅、フィルタリングしたのちにAD変換する。AD変換された光信号データはデータ取得制御回路312に転送される。本実施例ではそれぞれの光源の波長に対応するADコンバータを備え、それぞれの光源の波長で励起された蛍光光量を反映した光信号データを保存する。
 データ取得制御回路312はADコンバータ制御回路306からの光信号データを内部のDMAコントローラおよびバスインタフェース302を介してメモリ制御回路304へ出力する。メモリ制御回路304はメモリ303内の指定されたアドレスに光信号データを保存する。メモリ303のアドレスはデータを出力するたびにインクリメントされる。
 データ取得制御回路312は、それぞれの光源の波長に対応するNx×Ny個のデータをメモリ303に保存した後には内部のデータ取得完了レジスタを1にする。データ取得制御回路312はそれぞれの光源の波長に対応するNx×Ny個の光信号データをメモリ303に保存していない時には内部のデータ取得完了レジスタを0にする。
 続いてステップS411にて同期回路311は内部に保持する目標位置を更新する。現在の主走査方向が往路方向(X座標が増加する方向)の場合には、目標位置P(N)をP(N+1)に更新する。現在の主走査方向が復路方向(X座標が減少する方向)に場合には目標位置P(N)をP(N―1)に更新する。
 ステップS412にて、CPU301は光信号データ取得が終了しているか否かを判定する。CPU301はデータ取得制御回路312のデータ取得完了レジスタを読み出し、値が1の場合には光信号データ取得が終了したと判断しステップS413に進む。値が0の場合には光信号データ取得が終了していないと判断し、ステップS407に進む。
 ステップS413にて、CPU301は光源制御回路305に指示を出し、光源102の発光を停止させる。これにより、対物光学系104を介してアレイプレート101へ光照射が停止される。
 ステップS414にて、CPU301は、モータ制御回路307に指示を出し、主走査モータ107の回転を停止させる。これにより、対物光学系104のX方向の往復運動が停止する。ステップS415にて、CPU301は、モータ制御回路307、308、309に指示を出し、アレイプレート101および対物光学系104を停止位置に移動させる。停止位置のX座標、Y座標およびZ座標は0である。停止位置への移動は、エンコーダ108のZ相パルス信号および、Y方向リニアステージ109内の原点センサ信号、Z方向リニアステージ112の原点センサ信号を用いて各軸を原点復帰させることで実施する。Y方向リニアステージ109は、アレイプレート101が載置される載置部を構成するため、載置部109を換言される場合がある。
 ステップS416にて、CPU301はそれぞれの光源の波長に対応するNx×Ny個の光信号データをメモリ303から読み出し、データ圧縮、フォーマット変換処理を行い、TIFF形式の撮像画像ファイルを作成する。撮像画像ファイルはメモリ303に保存され、UI回路314およびユーザインタフェース117を介して使用者に提示される。また、使用者からの指示により、通信回路313を介して外部のデータサーバに転送する。
 ステップS417では、CPU301はモータ制御回路308に指示を出し、アレイプレート101を副走査方向にYpだけ移動させる。モータドライバ111へ電圧パルス信号を1パルス送った際のアレイプレート101の移動量をMyとすると、モータ制御回路308がモータドライバ111へ出力するパルス数はYp/Myとなる。本実施例ではMy=2μmとする。副走査は図5Bの505であらわされ、Y方向の移動距離はYpである。
 モータ制御回路308は、CPU301からの指示に基づき、対物光学系104が主走査方向の撮像領域外にいる間に、副走査が完了するような速度でモータドライバ111にパルス信号を出力する。副走査が完了したらステップS420に進む。
 ステップS418では、CPU301はメモリ303から副走査前後の目標高さを読み出す。その際に波長切り替え後の波長に対応した目標高さを読み出す。本実施例では往路で780nm、復路で670nmのレーザ光を照射する場合の例を用いて説明する。すなわち、波長が670nmから780nmに切り替えられる際には、Z_780(Y+Yp)と、Z_670(Y)を読み出し、高さ走査の移動量を算出する。
 波長が780nmから670nmに切り替えられる際には、Z_670(Y)と、Z_780(Y)を読み出し、高さ走査の移動量を算出する。
 高さ走査の移動量は現在のY座標によって異なるが、移動後のZ方向の高さが、ステップS416の副走査およびステップS421の波長切り替え後のY座標における目標高さに最も近くなるように、出力パルス数およびモータの回転方向を算出する。
 すなわち、往路から復路に切り替える際には、780nmから670nmへの波長切り替えが行われるが副走査は行われないのでZ_670(Y)に近くなるようにする。
 すなわち、復路から往路に切り替える際には、670nmから780nmへの波長切り替えと副走査が行われるのでZ_780(Y+Yp)に近くなるようにする。
 算出方向を以下に述べる。モータドライバ114へ電圧パルス信号を1パルス送った際のアレイプレート101の移動量をMzとする。本実施例ではMz=1μmとする。
 ある数xに最も近いMzの倍数をRoundMz(x)とし、ある数の絶対値をABS(x)、ある数の符号をSign(x)とする。
 本発明において、目標高さに最も近いMzの倍数を目標パルス数という。目標パルス数は離散的な値を取り、図6Aおよび図6Bの矩形603、矩形604に相当する。ハッチングされた矩形603の高さは波長670nmの一次光に対する目標パルス数、白で示す矩形604の高さは波長780nmの一次光に対する目標パルス数をあらわす。
 往路から復路に切り替える際にモータ制御回路309がモータドライバ114に出力するパルス数は以下の式であらわされる。

 アレイプレートの傾き方が図6Aのような時はDirの値は往路から復路に切り替える際に1、復路から往路に切り替える時に-1となる。図6Bのような時はDirの値は往路から復路に切り替える際に-1、復路から往路に切り替える時に1となる。
 なお、図6Aでは、全部のY座標の範囲にわたってZ_780(Y)<Z_670(Y)の例を、図6Bでは、全部のY座標の範囲にわたってZ_670(Y)<Z_780(Y)の例を示したが、Z_670(Y)とZ_780(Y)の大小関係はこれに限定されず、入れ替わっていてもよい。
 ステップS419において、CPU301はモータ制御回路309に指示を出し、アレイプレート101をZp分だけ、傾き方向Dirを打ち消す方向に移動させる。Dirの値が正の時はアレイプレートを下方に移動させ、Dirの値が負の時はアレイプレートを上方に移動させる。
 モータ制御回路309がモータドライバ111へ出力するパルス数はZp/Mzとなる。モータ制御回路309は、CPU301からの指示に基づき、対物光学系104が主走査方向の撮像領域外にいる間に、高さ走査が完了するような速度でモータドライバ114にパルス信号を出力する。
 ステップS420において、同期回路311は現在のY座標をそれまでのY座標からYpだけインクリメントして、ステップS412に進む。
 ステップS421において、光源制御回路305は、同期回路311からの波長切り替えトリガ信号の情報に基づいて、670nmおよび780nmの半導体レーザの発光、停止を切り替える。ステップS419の高さ走査とステップS421の光源制御への指示が完了したらステップS412に進む。
 現在の主走査方向が往路方向(X座標が増加する方向)の場合には、対物光学系104のX座標がX3を超えたところで780nmのレーザを発光させ、X4を超えたところで780nmのレーザを停止させる。主走査方向が往路方向(X座標が増加する方向)の場合には、670nmのレーザは発光を停止させる。
 現在の主走査方向が復路方向(X座標が減少する方向)の場合には、対物光学系104のX座標がX4よりも小さくなったところで670nmのレーザを発光させ、X3よりも小さくなったところで670nmのレーザを停止させる。主走査方向が往路方向(X座標が増加する方向)の場合には、780nmのレーザは発光を停止させる。
 図7は本実施例の撮像装置の光源102、共焦点光学系103、対物光学系104、光センサ105および受信回路106の詳細を示す図である。
 光源102について説明する。
 光源102は波長670nmと波長780nmの光を独立したタイミングで出力可能な光源であり、レーザダイオード701およびレーザダイオード714から構成される。
 レーザダイオード701は、波長780nmの光を発生させるレーザダイオードであり、外部からの電気信号により発光、停止、変調、光量制御が可能である。
 レーザダイオード714は、波長670nmの光を発生させるレーザダイオードであり、外部からの電気信号により発光、停止、変調、光量制御が可能である。
 続いて共焦点光学系103について説明する。共焦点光学系103は出射光、反射光および蛍光を分離して取得可能であり、波長780nm用の共焦点光学系、波長670nm用共焦点光学系、合波部の3つから構成されている。
 波長780nm用の共焦点光学系について以下に説明する。
 コリメートレンズ702は、レーザダイオード701から出射された780nmの光を平行光にするコリメートレンズである。
 バンドパスフィルタ703は、波長780nm付近の光を通過させるバンドパスフィルタである。
 偏光ビームスプリッタ704は、出射光と反射光を分離するための偏光ビームスプリッタである。
 1/4λ波長板705は、偏光ビームスプリッタ704の偏光方向に対して遅軸が45°傾いた1/4λ波長板である。
 レーザダイオード701からの出射光は偏光ビームスプリッタ704を透過し、1/4λ波長板705にて直線偏光から円偏光に変換され、ダイクロイックミラー706に至る。
 一方、アレイプレート101からの反射光は、1/4λ波長板705にて円偏光から出射光と90°直交する直線偏光に変換され、偏光ビームスプリッタ704で反射され、集光レンズ707に至る。
 ダイクロイックミラー706は、蛍光と反射光を分離するためのダイクロイックミラーである。
 レーザダイオード701からの出射光はダイクロイックミラー706で反射され、ダイクロイックミラー719へ至る。
 アレイプレート101からの反射光はダイクロイックミラー706で反射され、1/4λ波長板705へ至る。
 アレイプレート101にて、波長780nmの光に励起された蛍光は反射光よりも波長が長く、ダイクロイックミラー706を透過し、蛍光フィルタ708へ至る。
 集光レンズ707は、波長780nmの反射光に対する集光レンズであり、反射光は集光されピンホール710に至る。
 蛍光フィルタ708は、波長780nmの光に励起された蛍光を透過させるバンドパスフィルタである。
 集光レンズ709は、波長780nmの光に励起された蛍光に対する集光レンズであり、蛍光は集光されピンホール711に至る。
 ピンホール710は、アレイプレート101の表面近傍からの反射光を通過させる。通過した光は光センサ105のフォトダイオード712に至る。
 ピンホール711は、アレイプレート101の表面近傍からの蛍光を通過させる。通過した光は光センサ105の光電子増倍管713に至る。
続いて波長670nmの共焦点光学系103について以下に説明する。
 レーザダイオード714は、波長670nmの光を発生させるレーザダイオードである。
 コリメートレンズ715は、670nmの光を平行光にするコリメートレンズである。
 バンドパスフィルタ716は、波長670nm付近の光を通過させるバンドパスフィルタである。
 偏光ビームスプリッタ717は、出射光と反射光を分離するための偏光ビームスプリッタである。
 1/4λ波長板718は、偏光ビームスプリッタ717の偏光方向に対して遅軸が45°傾いた1/4λ波長板である。
 レーザダイオード714からの出射光は偏光ビームスプリッタ717を透過し、1/4λ波長板718にて直線偏光から円偏光に変換され、ダイクロイックミラー719に至る。
 一方、アレイプレート101からの反射光は、1/4λ波長板718にて円偏光から出射光と90°直交する直線偏光に変換され、偏光ビームスプリッタ717で反射され、集光レンズ720に至る。
 ダイクロイックミラー719は、蛍光と反射光を分離するためのダイクロイックミラーである。
 レーザダイオード714からの出射光はダイクロイックミラー719で反射され、ダイクロイックミラー727へ至る。
 アレイプレート101からの反射光はダイクロイックミラー719で反射され、1/4λ波長板718へ至る。
 アレイプレート101にて、波長780nmの光に励起された蛍光は反射光よりも波長が長く、ダイクロイックミラー719を透過し、蛍光フィルタ721へ至る。
 集光レンズ720は、波長780nmの反射光に対する集光レンズであり、反射光は集光されピンホール723に至る。
 蛍光フィルタ721は、波長780nmの光に励起された蛍光を透過させるバンドパスフィルタである。
 集光レンズ722は、波長780nmの光に励起された蛍光に対する集光レンズであり、蛍光は集光されピンホール724に至る。
 ピンホール723は、アレイプレート101の表面近傍からの反射光を通過させる。通過した光は光センサ105のフォトダイオード725に至る。
 ピンホール724は、アレイプレート101の表面近傍からの蛍光を通過させる。
 通過した光は光センサ105の光電子増倍管726に至る。
 続いて合波部について説明する。合波部は波長670nmの出射光と波長780nmの出射光を合波するとともに、波長670nm由来の反射光、蛍光と、波長780nm由来の反射光、蛍光と波長670nm由来の反射光、蛍光とを分離する役割を担う。
 ダイクロイックミラー727は、波長670nm由来の光と、波長780nm由来の光を分離するためのダイクロイックミラーである。波長670nmの出射光、反射光および波長670nmの出射光によって発生した蛍光は、波長が短いためダイクロイックミラー727で反射される。
 一方、波長780nmの出射光、反射光および波長780nmの出射光によって発生した蛍光は、波長が長いためダイクロイックミラー727を透過する。
 ダイクロイックミラー727にて反射された波長670nmの出射光と、ダイクロイックミラー727にて透過された波長780nmの出射光は同一光路を進み、ミラー728に至る。
 アレイプレート101からの波長670nmの反射光および波長670nmの出射光によって発生した蛍光はダイクロイックミラーにより反射されダイクロイックミラー719に至る。
 アレイプレート101からの波長780nmの反射光および波長780nmの出射光によって発生した蛍光はダイクロイックミラーを透過しダイクロイックミラー706に至る。
 続いて対物光学系104について説明する。
 ミラー728は、水平方向の光を垂直方向に跳ね上げるミラーである。
 対物レンズ729は、波長670nmおよび波長780nmの光をアレイプレート101上に集光させる。
 主走査機構730は、クランク118、コネクティングロッド119、ピストンクランク機構120、主走査モータ107により構成される。
 主走査機構730により、ミラー728および対物レンズ729を主走査方向に往復走査され、集光された波長670nmおよび波長780nmの光がアレイプレート101上を走査される。
 続いて光センサ105について詳細を説明する。
 光センサ105は波長670nmの反射光、波長670nmの光によって発生した蛍光、波長780nmの反射光、波長780nmの光によって発生した蛍光それぞれの光量を検出可能な光センサであり、フォトダイオード712、725および光電子増倍管713、726から構成される。
 フォトダイオード712は、波長780nmの反射光を検知し、その光量に応じた電流を出力する。
 光電子増倍管713は、波長780nmの光によって発生した蛍光を検知し、その光量に応じた電流を出力する。
 フォトダイオード725は、波長670nmの反射光を検知し、その光量に応じた電流を出力する。
 光電子増倍管726は、波長670nmの光によって発生した蛍光を検知し、その光量に応じた電流を出力する。
 続いて受信回路106について詳細を説明する。受信回路106は光センサ105から出力された電気信号から光量に応じたデジタル値を得るための回路である。波長670nmの反射光、波長670nmの光によって発生した蛍光、波長780nmの反射光、波長780nmの光によって発生した蛍光それぞれの光量に応じたデジタル値を取得可能であり、電流電圧変換アンプ731、732、733、734およびADコンバータ735、736、737、738から構成される。
 電流電圧変換アンプ731は、光電子増倍管713からの電流を電圧に変換し適切な電圧まで増幅する電子回路である。
 電流電圧変換アンプ732は、フォトダイオード712からの電流を電圧に変換し適切な電圧まで増幅する電子回路である。
 電流電圧変換アンプ733は、光電子増倍管726からの電流を電圧に変換し適切な電圧まで増幅する電子回路である。
 電流電圧変換アンプ734は、フォトダイオード725からの電流を電圧に変換し適切な電圧まで増幅する電子回路である。
 ADコンバータ735は、電流電圧変換アンプ731からの出力電圧をデジタル値に変換する電子回路である。ADコンバータ735により波長780nmの光によって発生された蛍光の光量に応じたデジタル値を得ることができる。
 ADコンバータ736は、電流電圧変換アンプ732からの出力電圧をデジタル値に変換する電子回路である。ADコンバータ736により波長780nmの反射光の光量に応じたデジタル値を得ることができる。
 ADコンバータ737は、電流電圧変換アンプ733からの出力電圧をデジタル値に変換する電子回路である。ADコンバータ737により波長670nmの光によって発生された蛍光の光量に応じたデジタル値を得ることができる。
 ADコンバータ738は、電流電圧変換アンプ734からの出力電圧をデジタル値に変換する電子回路である。ADコンバータ738により波長670nmの反射光の光量に応じたデジタル値を得ることができる。
 ADコンバータ735、736、737、738は外部からの信号により独立したタイミングで入力信号をサンプリングしデジタル値に変換することができる。
 図8は本実施例の撮像装置での主走査、副走査、高さ走査、光照射、光信号データ取得の動作タイミングを示すタイミングチャートである。これらのタイミングチャートの横軸は時間である。タイミングチャート801は、主走査の動作タイミングを示すタイミングチャートであり、6角形で示した時刻でピストンクランク機構120が回転し、主走査が行われていることを示す。タイミングチャート802は、縦軸が対物光学系104のX軸方向の位置、横軸は時刻である。ピストンクランク機構120により対物光学系104は往復動作をするが、その速度はX軸位置のストローク中央付近で速く、ストローク端付近では遅くなることが示されている。タイミングチャート803は、副走査の動作タイミングを示すタイミングチャートである。6角形で示した時刻で副走査モータ110が動作し、副走査が行われていることを示す。
 タイミングチャート804およびタイミングチャート805は、高さ走査の動作タイミングを示すタイミングチャートである。タイミングチャート804は、波長780nmの目標高さへの高さ走査のタイミングを示し、タイミングチャート805は、波長670nmの目標高さへの高さ走査のタイミングを示す。6角形で示した時刻で高さ走査モータ113が動作し、高さ走査が行われていることを示す。タイミングチャート806は、780nmの光信号データ取得の動作タイミングを示すタイミングチャートである。6角形で示した時刻で光電子増倍管713および受信回路106が動作し、780nmの一次光に由来する光信号データ取得が行われていることを示す。
 タイミングチャート807は、670nmの光信号データ取得の動作タイミングを示すタイミングチャートである。6角形で示した時刻で光電子増倍管726および受信回路106が動作し、670nmの一次光に由来する光信号データ取得が行われていることを示す。808は光源102の中の780nmのレーザダイオード701の動作タイミングを示すタイミングチャートである。6角形で示した時刻で780nmの一次光が発光し、対物光学系104を介してアレイプレート101に照射されることを示す。
 タイミングチャート809は、光源102の中の670nmのレーザダイオード714の動作タイミングを示すタイミングチャートである。6角形で示した時刻で670nmの一次光が発光し、対物光学系104を介してアレイプレート101に照射されることを示す。
 このように、本実施例では対物光学系104のX座標が往路方向に移動している時には780nmの一次光をアレイプレート101に照射し、780nmの一次光によって発生した光信号データを取得する。一方、対物光学系104のX座標が復路方向に移動している時には670nmの一次光をアレイプレート101に照射し、670nmの一次光によって発生した光信号データを取得する。このように、往路方向と復路方向で励起波長が異なる光信号データを取得することにより、ピストンクランク機構120の機械的誤差や受信回路106の遅延時間などにより発生する往路と復路の画像の位置ずれの影響を低減することができる。
 往路方向の移動中にエンコーダ108によって取得される主走査のX座標がX1に到達すると、780nmの光信号データの取得が開始される。
 復路方向の移動中にエンコーダ108によって取得される主走査のX座標がX2に到達すると670nmの光信号データの取得が開始される。復路方向の移動中に主走査のX座標がX1に到達すると、復路方向の光信号データ取得が完了したと判断し、副走査トリガが出力され、次の行に移動するための副走査が行われる。
 往路方向の移動中にエンコーダ108によって取得される主走査のX座標がX3に到達すると、波長切り替えトリガ信号により780nmの一次光の照射が開始される。また、高さ走査トリガ信号により現在のY座標に対する波長780nmの目標高さへの高さ走査が行われる。往路方向の移動中に主走査のX座標がX4に到達すると、780nmの光信号データ取得が完了したと判断し、波長切り替えトリガ信号により780nmの一次光の照射が停止される。
 復路方向の移動中にエンコーダ108によって取得される主走査のX座標がX4に到達すると、波長切り替えトリガ信号により670nmの一次光の照射が開始される。また、高さ走査トリガ信号により現在のY座標に対する波長670nmの目標高さへの高さ走査が行われる。復路方向の移動中に主走査のX座標がX3に到達すると、670nmの光信号データ取得が完了したと判断し、波長切り替えトリガ信号により670nmの一次光の照射が停止される。
 図9A~図9Cは高さ情報取得動作におけるアレイプレートの高さ走査の位置関係を説明する図である。図9Aおよび図9Bは、アレイプレート101を側面からみた図である。図9Cは、高さ走査中に光センサ105で取得した反射光の光量を、高さごとにプロットしたものである。横軸が光量の大きさ、縦軸が取得した高さを示す。
 図10は本実施例の撮像装置でアレイプレートの高さ情報取得を行う際の動作フローである。高さ情報取得動作の、撮像動作と異なる点は、高さ情報取得時には副走査を行わず、等ピッチで高さ走査を行いながら反射光信号データを取得し、解析し、アレイプレート表面の高さを算出する点である。高さ情報取得においては光センサ105内のフォトダイオード712およびフォトダイオード725を用いる。
 本発明の撮像装置において、前記射出波長制御部は、前記副走査ステップを介さずに、前記第1の走査ステップおよび前記第2の走査ステップの一方から、前記第1の走査ステップおよび前記第2の走査ステップの他方に波長を切り替える期間を含むように、前記光源と前記走査部とを制御することが好ましい。以下に、この場合の説明をする。
 ステップS1001にて、CPU301は高さ情報取得用のパラメータを同期回路311に設定する。パラメータとして、高さ情報取得を行う位置のY座標Yh、主走査方向の画素ピッチXp、高さ走査方向の画素ピッチZp、XZ平面上の高さ走査範囲を示す点901(X5,Z5)、点902(X6,Z6)、主走査方向の回転速度Xsを設定する。本発明において、XZ平面上の点901、点902を対角とする矩形の領域903を高さ走査領域という。主走査方向の画素数Nx=(X6-X5)/Xp、高さ走査方向の画素数Nz=(Z2-Z1)/Zpを算出しておく。
 本実施例ではX5=500、X6=22500、Z5=2000、Z6=6000、Xp=10[μm]、Zp=10[μm]、Xs=1200[rpm]とする。この場合には、Nx=2200、Nz=400となる。
 高さ情報取得処理においては撮像処理と異なり、光信号データの取得間隔は大きくてよい。そのため、高さ情報取得にかかる時間短縮およびデータ容量削減を行うことができる。
 ステップS1002にて、CPU301は、モータ制御回路307、308、309に指示を出し、アレイプレート101および対物光学系104を高さ情報取得開始位置に移動させる。
 高さ情報取得開始位置のX座標はピストンクランク機構120の走査範囲の端部であり、X座標値は-2500とする。高さ情報取得開始位置のY座標はパラメータで指定されたYhとする。また、高さ情報取得開始位置のZ座標はパラメータで指定されたZ5とする。ステップS1003にて、CPU301は、撮像処理のステップS405と同様にして主走査を開始させ、ステップS1004に進む。
 ステップS1004にて、CPU301は、780nmおよび670nmの光照射を開始させ、ステップS1005に進む。
 ステップS1005にて、同期回路311は対物光学系104が改行位置に到達したか否かを判定する。座標算出回路310から出力される対物光学系104のX座標が撮像領域内から撮像領域外に移行した時に改行位置に到達したと判断する。現在の主走査方向が往路方向(X座標が増加する方向)の場合には、対物光学系104のX座標がX6を超えたところで改行位置に到達したと判断する。
 往路方向の走査は、図9Bの軌跡904であらわされる。現在の主走査方向が復路方向(X座標が減少する方向)の場合には、対物光学系104のX座標がX5よりも小さくなったところで改行位置に到達したと判断する。復路方向の走査は、図9Bの軌跡906であらわされる。主走査方向の初期値は往路方向であり、その後ストローク端に到達するたびに復路方向、往路方向を交互に繰り返す。
 高さ情報取得処理においては撮像処理と異なり、往路と復路両方で両方の波長のデータを取得する。そのため、往路と復路の両方で改行を行うことができ、高さ情報取得にかかる時間を短縮することができる。
 改行位置に到達した場合には、同期回路311は高さ走査トリガ信号を出力し、ステップS1014に進む。改行位置に到達していない場合には同期回路311は高さ走査トリガ信号を出力せずにステップS1006に進む。
 ステップS1006にて、同期回路311は撮像処理のステップS409と同様にして対物光学系104が目標位置に到達したか否かを判定する。目標位置は図9Bの908のような、軌跡上の複数の点であり、X方向のピッチがXp、Z方向のピッチはZpである。
 目標位置に到達した場合には、同期回路311はデータ取得トリガ信号を出力し、ステップS1007に進む。まだ目標位置に到達していない場合には、同期回路311はデータ取得トリガ信号を出力せずにステップS1009に進む。
 ステップS1007にてデータ取得制御回路312は撮像処理のステップS410と同様にして光信号データの取得を行い、ステップS1008に進む。
 ただし、データ取得回路はADコンバータ736およびADコンバータ738で取得した各波長の反射光量に対応するNx×Nz個の光信号データをメモリ303に保存した後には内部のデータ取得完了レジスタを1にする。データ取得回路は各波長の反射光量に対応するNx×Nz個の光信号データをメモリ303に保存していない時には内部のデータ取得完了レジスタを0にする。
 ステップS1008にて同期回路311は、撮像処理のステップS411と同様にして内部に保持する目標位置を更新し、ステップS1009に進む。
 ステップS1009にてCPU301は、撮像処理のステップS412と同様にして光信号データ取得が終了しているか否かを判定する。光信号データ取得が終了したと判断した場合には、ステップS1010に進む。光信号データ取得が終了していないと判断した場合には、ステップS1005に進む。
 ステップS1010にて、CPU301は、撮像処理のステップS413と同様にして光照射を停止させ、ステップS1011に進む。
 ステップS1011にて、CPU301は、撮像処理のステップS414と同様にして主走査を停止させ、ステップS1012に進む。
 ステップS1012にて、CPU301は、撮像処理のステップS415と同様にしてアレイプレート101および対物光学系104を停止位置に移動させ、ステップS1013に進む。
 ステップS1013にて、CPU301はメモリ303に保存されたADコンバータ736およびADコンバータ738で取得した各波長の反射光量に対応するNx×Nz個の光信号データを読み出して、解析し、アレイプレートの高さ情報を波長ごとに算出する。具体的には、同じ高さで取得したNx個の光信号データを加算平均し、高さごとの平均光量を求める。平均光量をZ座標ごとに並べるとアレイプレート101の表面と裏面にあたる2つのピークが存在する。図9Cにおいて、ピーク909がアレイプレート101の表面からの反射光によるピーク、ピーク910が、アレイプレート101の裏面からの反射光によるピークを示す。そのうち、よりZ座標が大きいピーク909、すなわち表面に相当するピークを示すZ座標911を位置Yhにあたる高さ情報とする。
 なお、反射光量に対応する光信号データを用いて求めた高さ情報は、反射光にたいしてフォーカスが合う高さである。蛍光と反射光でフォーカス位置にずれがある場合には、その分を補正してもよい。例えば、反射光量に対応する光信号データを用いて求めた高さ情報に対し既知のフォーカス位置ずれ量を加算して、蛍光検出で用いる高さ情報としてもよい。
 本実施例では波長670nmと波長780nmを光源、光学系、光センサ、受信回路をそれぞれ備えていることにより、各波長の光信号データを同時に取得することができ、それぞれの波長に対する高さ情報を短時間で取得することができる。
 ステップS1014では、CPU301はモータ制御回路309に指示を出し、アレイプレート101を高さ走査方向にZpだけ移動させる。モータドライバ114へ電圧パルス信号を1パルス送った際のアレイプレート101の移動量をMzとすると、モータ制御回路309がモータドライバ114へ出力するパルス数はZp/Mzとなる。高さ走査は図9Bの高さ走査905および高さ走査907であらわされ、Z方向の移動距離はZpである。
 モータ制御回路309は、CPU301からの指示に基づき、対物光学系104が主走査方向の撮像領域外にいる間に、高さ走査が完了するような速度でモータドライバ114にパルス信号を出力する。
 ステップS1015において、同期回路311は現在のZ座標をそれまでのZ座標からZpだけインクリメントして、ステップS1009に進む。
 撮像処理におけるステップS402の傾き情報取得の際には、図10で示した高さ情報取得処理を位置Y3および位置Y4の2回実施し、Z3_670、Z3_780、Z4_670、Z4_780を求める。
 ADコンバータ736で取得した光信号データから求めたY3に対する高さ情報がZ3_780であり、ADコンバータ738で取得した光信号データから求めたY3に対する高さ情報がZ3_670である。
 同様に、ADコンバータ736で取得した光信号データから求めたY4に対する高さ情報がZ4_780であり、ADコンバータ738で取得した光信号データから求めたY4に対する高さ情報がZ4_670である。
 続いて、各高さ情報から式1および式2を用いて傾き情報K_670およびK_780を求める。
 以上説明してきたように、本実施例の撮像装置では、同期回路を用いることにより、対物光学系の位置と同期して、副走査、波長切り替え、高さ走査を行う。波長ごとに求めた高さ情報および傾き情報をもとに、波長ごとに高さを補正しながら二次元走査を行う。これにより、波長間で光学系のずれなどによりフォーカス位置に差がある場合にも、アレイプレート全面でピントの合う、複数の波長の蛍光画像を取得することが可能になる。
 これにより、2つの波長それぞれでのフォーカス位置を最適化できるので、例えばアレイプレート個体間のタンパク質量の違いを蛍光輝度の日で補正したりする場合に、より信頼性が向上する、といった利点がある。
 対物光学系が撮像範囲外にいる間に副走査と高さ走査を行うことにより、撮像範囲内ではアレイプレートを動かす必要がない。そのため、アレイプレート駆動に伴う振動の影響が少なくすることができる。さらに高性能なフォーカスセンサや低振動なアクチュエータなどからなる高速サーボ制御系をもたないため、簡便な構成で、アレイプレート全面でピントの合う蛍光画像を取得することが可能になる。
 また、本実施例での高さ情報取得方法では、高さ走査だけでなく、主走査も行いNx個のデータを平均化することにより、アレイプレート上に部分的に汚れや液体がついていた場合でも安定してピークを検出することができる。そのため、アレイプレート上の1点に対して高さ方法を行うよりも、高さ情報取得の安定性を向上させることができる。
 なお、本実施例では、スライドガラス表面からの反射光のピーク位置を用いて高さ情報を取得する例を用いて説明したが、高さ情報の取得方法はこれに限定されない。例えば、アレイプレート上の一部のスポットからの蛍光信号の輝度のピーク位置を用いて高さ情報を取得してもよい。この場合には、高さ情報取得のために一部のスポットに光を当てる必要があるが、光センサで反射光を取得しなくてよいため、光学系の部品点数を削減することができる。
 なお、本実施例ではアレイプレートの短手方向を主走査方向、長手方向を副走査方向としているが、走査方向はこれに限らない。例えば長手方向に主走査を行い、短手方向に副走査を行ってもよい。その場合、ピストンクランク機構120のストロークを長くする必要があるが、副走査の回数は少なくなるので撮像時間を短縮できる可能性がある。
[実施例1の変形例]
 なお、本実施例では、Y3とY4の2点で測定した高さ情報で線形補間して求まる直線を目標高さとしたが、本発明における目標高さは直線には限らない。枠内領域508の外により多くの測定点を設け、目標高さを高次関数などであらわすこともできる。それにより、より複雑なフォーカス位置ずれも精度よく補正することができる。また、枠内領域508の右側と左側で個別の目標高さを用い、副走査方向だけでなく、主走査方向の傾きを補正してもよい。
[実施例2]
 以下に本発明の実施例2を説明する。
 本発明の実施例2が、実施例1と異なる点は、波長間のフォーカス位置ずれが大きく、副走査期間内に高さ調整できる範囲を超えている場合への対応の点である。
 本発明の実施例1では、副走査期間内にフォーカス位置の高さの差を補正するための高さ走査を行うが、高さ走査用の高さ走査モータ113およびZ方向リニアステージ112にも速度や加速度の制約があるため、副走査期間Ty内に高さ走査可能な距離Zdに制約がある。本発明の実施例2では高さ走査可能範囲を加味して各波長の目標位置を調整する。
 本実施例での装置構成、アレイプレート、コントローラの内部構成、装置の詳細構成については、それぞれ実施例1と同じであるため説明を省略する。
 本実施例の動作フローの多くは実施例1と同じであるが、図4のステップS403で実施している各行の目標高さ算出処理が異なる。図11を用いて本実施例の目標高さの算出について説明する。
 なお、図11において図6Aおよび図6Bと同じ符号は同じ意味を示すため説明を省略する。直線1101は波長780nmに対する目標高さ602基準での、副走査期間での移動可能な高さの最大値である。副走査期間を8ms、ステージの最大移動速度を10mm/sとすると、移動可能な距離Zdは80μmとなる。直線1101は波長780nmに対する目標高さ602に80μmを加算したものであり、Z_780(Y)+Zdであらわされる。
 直線1102は波長780nmに対する目標高さ602基準での、副走査期間での移動可能な高さの最小値である。
 直線1102は波長780nmに対する目標高さ602から80μmを減算したものであり、Z_780(Y)―Zdであらわされる。
 直線1101と直線1102の間の範囲が、780nmから670nmへ波長切り替えする際に、副走査期間内に移動可能な範囲となる。
 本実施例では図4のステップS403にて、各波長の目標高さを求める際に、移動可能な範囲内になるように調整を行う。
 すなわち、Z_780(Y)は式4に従って求めるがZ_670(Y)は、直線1101と直線1102の間の範囲に収まるように、以下の式のように調整を行う。

 MINは各引数の最小値を返す関数、MAXは各引数の最大値を返す関数である。
 図11の例では直線1101と波長670nmに対する目標高さ601の交点1103よりもY座標が小さい範囲では波長670nmに対する目標高さ601がZ_670(Y)となり、交点1103よりもY座標が大きい範囲では直線1101がZ_670(Y)となる。
 このように移動可能距離を加味して目標高さを調整することで、フォーカス位置ずれが大きい場合にも、確実に副走査期間内に高さ走査を完了させつつ、ある程度までフォーカスを合わせることができる。撮像範囲中では撮像範囲内ではアレイプレートを動かさないため、振動による影響を低減することができる。
 なお、本実施例では波長780nmの目標高さZ_780(Y)を基準とし、それに移動可能距離を加減算して移動可能範囲を求めたが、基準の取り方はこれに限らない。逆に波長670nmの目標高さZ_670(Y)を基準としてもよい。また、Z_780(Y)とZ_670(Y)の中間の値を基準としてもよい。
[実施例2の変形例]
 本発明の実施例2では波長間のフォーカス位置ずれが大きく、副走査期間内に高さ調整できる範囲を超えている場合に、目標高さを調整していたが、主走査速度を調整してもよい。例えば、主走査速度を遅くして副走査期間Tyを2倍にすれば、高さ走査可能な距離Zdを2倍にすることができる。このようにして波長670nmの目標高さ601が直線1101と直線1102の間におさまるようにする。
 この方法では主走査速度を遅くすることにより撮像時間がかかるが、波長間のフォーカス位置ずれが大きい場合にも両方の波長で最適な値にフォーカスを合わせることができる。
[実施例3]
 以下に本発明の実施例3を説明する。
 本発明の実施例3が、実施例1と異なる点は、目標高さの算出をよりシンプルにしたことである。
 実施例1では波長780nmに対する目標高さ602と波長670nmに対する目標高さ601をそれぞれフォトダイオードで個別の波長の反射光を測定して、別々に求めたが、本実施例では、一方の波長の目標高さに一定値を加えたものを他方の波長の目標高さとする。これにより、実施例1より簡便な方法で波長間フォーカスずれを補正する。本実施例での装置構成、アレイプレート、コントローラの内部構成、装置の詳細構成については、それぞれ実施例1と同じであるため説明を省略する。
 本実施例の動作フローの多くは実施例1と同じであるが、図4のステップS403で実施している各行の目標高さ算出処理が異なる。
 本実施例では図4のステップS403にて、780nmの目標高さに一定値Zoを加えたものを680nmの目標高さとする。本実施例ではアレイプレート全体にわたって波長間のフォーカスずれのばらつきが小さく一定値Zoであらわされる。
 すなわち、Z_780(Y)は式4に従って求めるがZ_670(Y)は以下の式に従う。

 Zoの値は高さ情報取得時に、各波長の高さ情報から測定される。以下の式であらわされる。なお、以下の式では位置Y3での高さ情報を用いているが、位置Y3以外の位置での高さ情報の差分を用いてZoを算出してもよい。Zoの値は正負の値を取りうる。

 この方法によれば、図12のように波長670nmに対する目標高さ601と波長780nmに対する目標高さ602の差分値はアレイプレート全体にわたって一定値になるため、ステップS418での高さ走査時の移動量の算出を簡略化することができる。
 なお、本実施例ではZoの値は高さ情報取得時に測定によって求めていたが、Zoを求める方法はこれに限らない。例えば、校正時に撮像装置外部の治具を用いて測定してもよい。あるいは、既知の複数のZoの値を用いて校正用のアレイプレートを測定し、670nmの輝度が最も高くなるZoの値を採用してもよい。これらの方法によって670nmの反射光測定を省略すれば、光センサ、受信回路、共焦点光学系の部品点数を削減することができる。
 なお、本実施例では波長780nmの目標高さZ_780(Y)を基準とし、それに一定値を加算してZ_670(Y)を求めたが、基準の取り方はこれに限らない。逆に波長670nmの目標高さZ_670(Y)を基準とし、それに一定値を加算してZ_780(Y)を求めてもよい。
 以上説明してきたように、本発明の実施例3では、より簡便な方法で波長間のフォーカス位置ずれを補正することができる。アレイプレートの厚みや傾きの位置によるばらつきが少ない場合にはアレイプレート全面でピントの合う、複数の波長の蛍光画像を取得することが可能になる。
[実施例3の変形例]
 本発明の実施例3では、Z_780(Y)に一定値を加えてZ_670(Y)を求めたが、さらに簡略化し、波長780nmと波長670nmそれぞれアレイプレート全面にわたって1つの値を目標高さとしてもよい。例えば、位置Y3での高さ情報取得処理を行い、以下の式に従ってZ_780(Y)およびZ_670(Y)を求める。

 位置Y4での高さ情報取得処理は省略する。
 この方法によれば、図13のように波長670nmに対する目標高さ601と波長780nmに対する目標高さ602はアレイプレート全体にわたって一定値になるため、ステップS418での高さ走査時の移動量の算出を簡略化することができる。
 この方法では、さらに簡便な方法で波長間のフォーカス位置ずれを補正することができる。アレイプレート傾きが少ない場合にはアレイプレート全面で、ある程度ピントの合う複数の波長の蛍光画像を取得することが可能になる。
[実施例4]
 以下に本発明の実施例4を説明する。
 本発明の実施例4が、実施例1と異なる点は、複数の波長の目標高さの中間の高さを両波長共通の目標高さとして用いる点である。
 実施例1では波長780nmに対する目標高さ602と波長670nmに対する目標高さ601を求め、波長切り替え後に各波長の目標高さに合わせるように高さ走査を行ったが、本実施例では、波長780nmに対する目標高さ602と波長670nmに対する目標高さ601を平均した高さを求め、波長780nmおよび波長670nmの両方で同じ高さになるように高さ走査を行う。
 本実施例での装置構成、アレイプレート、コントローラの内部構成、装置の詳細構成については、それぞれ実施例1と同じであるため説明を省略する。
 本実施例では図4のステップS403にて、以下の式によって各波長の目標高さを求める。

 これらはいずれも実施例1の式3、式4で求めた波長670nm、波長780nmの目標高さの平均の高さになり、図14の直線1401であらわされる。
 本発明の実施例4の方法では、Z_670(Y)とZ_780(Y)は同じであるため、波長切り替え時の高さ走査は省略することができ、副走査が行われる時に高さ走査を行えばよい。
 図8で示すように往路で波長780nmのデータを取得し、復路で波長670nmのデータを取得したのちに副走査が行われる場合には、往路から復路の間のタイミングチャート805を省略することができる。
 以上説明してきたように本発明の実施例4では、各波長に合わせた最適な高さには調整できないが、両方の波長で、ある程度ピントが合った蛍光画像を取得することができる。この方法では、波長間のフォーカスずれ量によらず高さ走査の移動量が少なくなり、ステージの移動距離の制約が大きい場合にも適用できる。
[実施例4の変形例]
 本変形例では、両方の波長で同じ高さを用いるため、波長切り替えをせずに同時に2つの波長の光を照射する場合にも適用可能である。本発明の実施例1では2つの波長の発光を時分割で行っていたが、レーザダイオード701とレーザダイオード714を同時に発光させ、アレイプレート101からの蛍光をダイクロイックミラー727で分離して光電子増倍管713および光電子増倍管726で取得することにより、2つの波長による蛍光データを同時に取得することができる。
 往路と復路で両方の波長によるデータを取得し、往路と復路両方で副走査を行うことで、撮像時間を短縮することができる。
 図15は本変形例での主走査、副走査、高さ走査、光照射、光信号データ取得の動作タイミングを示すタイミングチャートである。これらのタイミングチャートの横軸は時間である。タイミングチャート801、802、806、807は、本発明の実施例1の図8で示した内容と同じであるため説明を省略する。
 1501は副走査の動作タイミングを示すタイミングチャートである。6角形で示した時刻で副走査モータ110が動作し、副走査が行われていることを示す。
 1502は高さ走査の動作タイミングを示すタイミングチャートである。
 6角形で示した時刻で高さ走査モータ113が動作し、高さ走査が行われていることを示す。
 復路方向の移動中に主走査のX座標がX1に到達すると、往路方向の光信号データ取得が完了したと判断し、副走査トリガが出力され、次の行に移動するための副走査が行われる。
 往路方向の移動中に主走査のX座標がX2に到達すると、復路方向の光信号データ取得が完了したと判断し、副走査トリガが出力され、次の行に移動するための副走査が行われる。
 1503は光源102の中の780nmのレーザダイオード701の動作タイミングを示すタイミングチャートである。6角形で示した時刻で780nmの一次光が発光し、対物光学系104を介してアレイプレート101に照射されることを示す。
 1504は光源102の中の670nmのレーザダイオード714の動作タイミングを示すタイミングチャートである。6角形で示した時刻で670nmの一次光が発光し、対物光学系104を介してアレイプレート101に照射されることを示す。
 波長切り替えトリガ信号は用いず、780nmの一次光および670nmの一次光は撮像中、常時照射される。
 このように、本変形例では、780nmと780nmの一次光を同時にアレイプレート101に照射し、往路と復路両方で両方の波長によって発生した光信号データを取得する。これにより、往路方向と復路方向で励起波長が異なる光信号データを取得する場合に比べ、効率よくデータを取得することが可能になり撮像時間を短縮することができる。
 その際に、各波長の目標高さの平均の位置に合わせて高さ走査を行うことで、ある程度ピントの合う、複数の波長の蛍光画像を取得することが可能になる。
 本発明は上記実施の形態に制限されるものではなく、本発明の精神及び範囲から離脱することなく、様々な変更及び変形が可能である。従って、本発明の範囲を公にするために以下の請求項を添付する。

Claims (25)

  1.  長手方向と短手方向を有し一方の面にスポットアレイを有するアレイプレートに対して、一次光を照射し二次光を検出する対物光学系を走査し、蛍光像を取得する、撮像装置の制御方法であって、
     第1の波長の一次光を出射する前記対物光学系を用いて前記一方の面を測距し、前記長手方向における複数の位置に対応する前記第1の波長の一次光における複数の第1の距離情報を取得する、第1の測距工程と、
     前記複数の第1の距離情報を用いて、前記長手方向における主走査位置に応じた前記第1の波長の一次光に対応する第1の走査条件を取得する、第1の走査条件取得工程と、
     前記第1の走査条件および前記第1の距離情報の少なくともいずれかを用いて、前記主走査位置に応じた前記第1の波長の一次光とは光の波長が異なる第2の波長の一次光に対応する第2の走査条件を取得する、第2の走査条件取得工程と、
     前記第1の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第1の走査工程と、
     前記第2の走査条件で、前記短手方向に沿って前記対物光学系を走査する、第2の走査工程と、
     前記主走査位置を前記長手方向に沿って変更する、副走査工程と
    を含む、撮像装置の制御方法。
  2.  前記第1の走査工程において、前記対物光学系が検出した蛍光強度情報と走査位置情報とに基づいて、前記第1の波長の一次光を前記スポットアレイに照射することで得られる第1の蛍光像を取得する工程と、
     前記第2の走査工程において、前記対物光学系が検出した蛍光強度情報と走査位置情報とに基づいて、前記第2の波長の一次光を前記スポットアレイに照射することで得られる第2の蛍光像を取得する工程と
    を含む、請求項1に記載の撮像装置の制御方法。
  3.  前記第1の走査工程と前記第2の走査工程は、前記第1の波長の一次光と前記第2の波長の一次光とのそれぞれが、前記長手方向において共通の主走査位置に対応する共通の走査経路を走査されるように行われる、請求項1または2に記載の撮像装置の制御方法。
  4.  前記第1の走査工程と前記第2の走査工程は、前記第1の波長の一次光と前記第2の波長の一次光とが、前記共通の走査経路を逆方向に走査するように行われる、請求項3に記載の撮像装置の制御方法。
  5.  前記第1の走査工程と前記第2の走査工程とをそれぞれ1回以上行う毎に、前記副走査工程を1回行う、請求項1~4のいずれか1項に記載の撮像装置の制御方法。
  6.  前記第1の走査工程と前記第2の走査工程とをそれぞれ1回行う毎に、前記副走査工程を1回行う、請求項1~4のいずれか1項に記載の撮像装置の制御方法。
  7.  前記第1の距離情報に基づいて、前記第2の波長の一次光における第2の距離情報を出力する、距離補正工程をさらに含み、
     前記第2の走査条件取得工程は、前記複数の第1の距離情報を用いて前記距離補正工程において得られた前記第2の距離情報と、前記第1の走査条件とを用いて、前記第2の走査条件を取得する、請求項1~6のいずれか1項に記載の撮像装置の制御方法。
  8.  前記第1の走査条件に対応する前記第2の波長の一次光における第2の走査条件を出力する、走査条件補正工程をさらに含み、
     前記第2の走査条件取得工程は、前記第1の走査条件に基づいて前記走査条件補正工程から前記第2の走査条件を取得する、請求項1~7のいずれか1項に記載の撮像装置の制御方法。
  9.  前記対物光学系を用いて前記一方の面を前記第2の波長の一次光で測距し、前記第2の波長の一次光における第2の距離情報を取得する、第2の測距工程をさらに含む、請求項1~8のいずれか1項に記載の撮像装置の制御方法。
  10.  前記第2の測距工程が、前記長手方向における複数の位置で行われる、請求項9に記載の撮像装置の制御方法。
  11.  前記第2の走査条件取得工程が、前記第2の測距工程で取得した前記第2の距離情報と、前記第1の走査条件とを用いて、前記第2の走査条件を取得する、請求項9または10に記載の撮像装置の制御方法。
  12.  前記スポットアレイの蛍光像を撮像するための一次光は、前記第1の波長の一次光および前記第2の波長の一次光のいずれか一方を含み、
     前記スポットアレイ上の反応生成物を標識する二次抗体蛍光像を撮像するための一次光は、前記第1の波長の一次光および前記第2の波長の一次光のいずれか他方を含む、請求項1~11のいずれか1項に記載の撮像装置の制御方法。
  13.  長手方向と短手方向を有し、一方の面にスポットアレイを有するアレイプレートを、光学的に二次元に撮像する、撮像装置であって、
     前記アレイプレートが載置される、載置部と、
     異なる波長の一次光を切り替えて射出する光源と光学的に結合し、前記アレイプレートに一次光を出射する光射出部と、前記アレイプレートからの二次光を検出する採光部とを含む、対物光学系と、
     前記載置部と前記対物光学系との前記一次光の光軸方向における作動距離を規定する、距離規定部と、
     前記載置部と前記対物光学系との、前記光軸方向と交差する面内における相対位置を変更する、走査部と、
     前記一次光の波長毎の前記面内の複数位置における前記作動距離を走査条件として取得する走査条件取得部と、
     前記一次光の波長に関する情報と、前記採光部からの受光強度に関する情報と、前記相対位置に関する情報とに基づいて、前記一次光に対応する蛍光像を取得する画像化部と、
    を有する撮像装置。
  14.  光源をさらに有する、請求項13に記載の撮像装置。
  15.  距離補正部をさらに有する、請求項13または14に記載の撮像装置。
  16.  走査条件補正部をさらに有する、請求項13~15のいずれか1項に記載の撮像装置。
  17.  長手方向と短手方向を有し、一方の面にスポットアレイを有するアレイプレートを、光学的に二次元に撮像する、撮像装置であって、
     前記アレイプレートが載置される、載置部と、
     異なる波長の一次光を切り替えて射出する光源と光学的に結合し、前記アレイプレートに一次光を出射する光射出部と、前記アレイプレートからの二次光を検出する採光部とを含む、対物光学系と、
     前記載置部と前記対物光学系との光軸方向と交差する面内における相対位置を変更するために、前記短手方向と前記長手方向に沿って、前記相対位置を変更し、主走査を行う主走査部と、副走査を行う副走査部とをそれぞれ含む、走査部と、
     前記短手方向における副走査位置と、前記長手方向における主走査方向と、主走査回数とに応じて、前記光射出部から射出される光の波長を変更するように、前記光源と前記走査部とを制御する、射出波長制御部と、
     前記一次光の波長に関する情報と、前記採光部からの受光強度に関する情報と、前記相対位置に関する情報とに基づいて、前記一次光に対応する蛍光像を取得する、画像化部と
    を有する、撮像装置。
  18.  前記射出波長制御部は、第1の波長の一次光と、前記第1の波長の一次光とは光の波長が異なる第2の波長の一次光と、を切り替えるように前記光射出部から射出される光の波長を変更するように、前記光源と前記走査部とを制御する、請求項17に記載の撮像装置。
  19.  前記射出波長制御部は、共通の走査経路上を前記第1の波長の一次光と前記第2の波長の一次光が主走査するように、前記第1の波長の一次光および前記第2の波長の一次光の一方を主走査した後で、前記第1の波長の一次光および前記第2の波長の一次光の他方に切り替えるように、前記光源と前記走査部とを制御する、請求項18に記載の撮像装置。
  20.  前記射出波長制御部は、共通の走査経路上を前記第1の波長の一次光で走査する第1の走査ステップと前記第2の波長の一次光で走査する第2の走査ステップとをそれぞれ1回以上行う毎に、副走査するステップを1回行うように、前記光源と前記走査部とを制御する、請求項18または19に記載の撮像装置。
  21.  前記射出波長制御部は、共通の走査経路上を前記第1の波長の一次光で走査する第1の走査ステップと前記第2の波長の一次光で走査する第2の走査ステップとをそれぞれ1回行う毎に、前記副走査するステップを1回行うように、前記光源と前記走査部とを制御する、請求項18~20のいずれか1項に記載の撮像装置。
  22.  前記射出波長制御部は、前記副走査するステップを介さずに、前記第1の走査ステップおよび前記第2の走査ステップの一方から、前記第1の走査ステップおよび前記第2の走査ステップの他方に波長を切り替える期間を含むように、前記光源と前記走査部とを制御する、請求項20に記載の撮像装置。
  23.  光源をさらに有する、請求項17~22のいずれか1項に記載の撮像装置。
  24.  前記載置部と前記対物光学系との前記一次光の光軸方向における作動距離を規定する、距離規定部をさらに有し、
     前記射出波長制御部は、前記距離規定部が規定する前記作動距離を前記一次光の波長に応じて制御する、請求項17~23のいずれか1項に記載の撮像装置。
  25.  前記一次光の波長毎の前記面内の複数位置における前記作動距離を走査条件として取得する走査条件取得部をさらに有する、請求項24に記載の撮像装置。
