Abstract

본 발명은 Nectin-2에 결합하는 항체 약물 접합체 및 이의 용도에 관한 것으로서, Nectin-2에 특이적으로 표적화 할 수 있는 마우스 단일클론 항체(m12G1 clone) 및 키메릭 항-Nectin-2 항체(chimeric 12G1; c12G1)를 제작하였다. c12G1 항체는 높은 친화력으로 인간 Nectin-2의 C2 도메인에 특이적으로 결합했지만, 마우스 Nectin-2에는 결합하지 않았다. 이후, DM1에 접합된 c12G1 항체를 포함하는 항체-약물 접합체를 제작하고, 시험관 내 및 생체 내에서 암세포에 대한 세포 독성 효과를 조사한 결과, c12G1-DM1은 Nectin-2 양성 난소암세포의 유사분열 단계에서 세포 주기 정지를 유도했지만, Nectin-2 음성 암세포에서는 그렇지 않았다. c12G1-DM1은 난소암세포에서 정상 IgG-DM1에 비해 0.1nM~7.4nM 범위의 IC50 으로 ~100배의 세포독성을 유도하였고, c12G1-DM1은 OV-90 세포가 이식된 마우스 이종이식 모델에서 ~91%의 종양 성장 억제를 나타냈다. 이러한 결과는 c12G1-DM1이 Nectin-2 양성 난소암에 대한 잠재적인 치료제로서 유용하게 활용될 수 있음을 제시한다.

Description

Nectin-2에 결합하는 항체 약물 접합체 및 이의 용도
본 발명은 Nectin-2에 결합하는 항체 약물 접합체(antibody drug conjugates: ADC) 및 이의 용도에 대한 것이다.
난소암은 여성 생식기의 두 번째로 흔한 악성 종양으로, 2020년 전 세계적으로 210,000명의 사망자가 발생한 것으로 추정된다. 환자의 70% 이상이 진행 단계에서 진단되기 때문에, 난소암 환자의 진단 후 5년 생존율은 47%에 불과하다. 난소암은 세포 기원에 따라 상피암, 성삭기질암, 생식세포암 및 혼합형으로 분류된다. 이 중 약 90%의 환자가 상피성 난소암(epithelial ovarian cancer: EOC)으로 진단되며, 그 조직학적 서브타입에는 장액성, 자궁내막성, 투명 세포, 점액성, 악성 브레너 종양 및 혼합 조직이 포함되며, 60% 이상의 사례가 장액성 조직으로 분류된다. 고-등급 장액성 암종은 잘 분화된 저-등급 장액성 암종에서 발생한다. 최근 분자 특성 분석의 발전으로 인해, EOC는 타입 I 및 타입 II의 2개의 별개 그룹으로 추가 분류될 수 있는 것으로 나타났다. 타입 I EOC의 발병기전은 주로 자궁내막증 또는 나팔관 관련 장액성 경계선에서 발생하며, 이는 KRAS, BRAF, PTEN, PI3K3CA, CTNNB1 및 ARID1A를 포함하는 게놈 변형을 특징으로 한다. 반면에, 타입 II EOC의 발병기전은 대부분 TP53 돌연변이가 있는 나팔관 상피의 전구체 병변에서 유래한다.
난소암 치료에는 일반적으로 수술 및 화학요법이 병행된다. EOC에 대해 PARP 억제제 및 베바시주맙을 포함한 표적 치료법의 통합은 질병 통제에 상당한 영향을 미친다. 이러한 진전에도 불구하고 진행성 EOC 진단을 받은 대부분의 여성은 궁극적으로 백금-내성 재발이 발생한다. 최근에는 혈관 내피 성장 인자 및 엽산 수용체 알파와 관련된 표적 치료법이 무진행 생존율을 향상시키는 데 주목할 만한 진전을 보였다. 그럼에도 불구하고, 난소암 환자를 효율적으로 치료하기 위한 다른 치료법을 개발해야 하는 의학적 요구가 여전히 충족되지 않고 있다.
Nectin-2 는 Ca2+에 비의존적이면서 면역글로불린과 유사한 구조를 가지는 세포접합 분자이다. 이는 세포접합부(Adherens junction)에서 작용을 하며 최근 연구에 따르면 다양한 암세포에서 과발현됨이 밝혀져 암 치료를 위한 잠재적인 표적으로 고려된다. 특히, Nectin-2는 CD34+ 조혈 및 내피 세포, 종양 세포 및 면역 세포에서 고도로 발현된다는 것이 알려졌다. 예를 들어, 다양한 정상 조직과 암 조직에서 Nectin-2의 발현 수준을 비교하면 암 조직, 특히 유방암, 난소암, 췌장암 및 전립선암 조직에서 높은 수준이 나타난다. 또한, Nectin-2는 자가면역질환의 원인 요소인 성숙한 수지상 세포에서 과발현된다고 보고되어 자가면역질환 치료제로서 가능성도 존재한다.
Nectin-2의 mRNA 및 단백질의 발현은 인접한 비-종양 조직에 비해 유방암, 난소암, 전립선암을 포함한 다양한 암 조직에서 상향 조절된다. 이는 조직학적 서브타입, 등급 또는 원격 전이와 관계없이 난소 종양 생검에서 과발현된다. 난소암 환자의 약 48%에서 Nectin-2 양성인 것으로 나타났다. 놀랍게도, 림프절 음성 종양에 비해 림프절 전이가 있는 종양에서 Nectin-2의 유전자 발현이 더 높은 것으로 확인되었다. 최근 연구에 따르면, 항체 의존성 세포 식작용이 감소된 Nectin-2 항체가 난소암 치료제로 개발될 수 있다고 보고되기도 하였다. 따라서, Nectin-2에 결합하는 항체-약물 접합체는 난소암 치료제로 개발될 가능성이 있다.
본 발명의 목적은 Nectin-2에 특이적으로 결합하는 항체 또는 그의 항원 결합 단편; 및 약물이 결합된 항체 약물 접합체(antibody drug conjugates: ADC)을 제공하는 데에 있다.
본 발명의 다른 목적은 상기 항체 약물 접합체를 유효성분으로 포함하는 Nectin-2가 과발현되는 암 예방 또는 치료용 약학조성물을 제공하는 데에 있다.
상기 목적을 달성하기 위하여, 본 발명은 서열번호 1로 표시되는 아미노산 서열로 이루어진 중쇄 CDR1, 서열번호 2로 표시되는 아미노산 서열로 이루어진 중쇄 CDR2 및 서열번호 3으로 표시되는 아미노산 서열로 이루어진 중쇄 CDR3을 포함하는 중쇄 가변 영역; 및 서열번호 4로 표시되는 아미노산 서열로 이루어진 경쇄 CDR1, 서열번호 5로 표시되는 아미노산 서열로 이루어진 경쇄 CDR2 및 서열번호 6으로 표시되는 아미노산 서열로 이루어진 경쇄 CDR3을 포함하는 경쇄 가변 영역을 포함하는 Nectin-2에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편; 및 약물이 결합된 항체 약물 접합체를 제공한다.
또한, 본 발명은 상기 항체 약물 접합체를 유효성분으로 포함하는 Nectin-2가 과발현되는 암 예방 또는 치료용 약학조성물을 제공한다.

본 발명에서 용어, "항체 약물 접합체(antibody drug conjugates; ADC)"는 표적 암세포로 항암 약물을 전달하기 전까지 항암 약물이 항체에 안정적으로 결합되어 있어야 한다. 표적으로 전달된 약물은 항체로부터 유리되어 표적 세포의 사멸을 유도해야 한다. 이를 위해서는 약물이 항체에 안정적으로 결합함과 동시에 표적 세포에서 유리될 때는 표적 세포의 사멸을 유도할 충분한 세포독성을 가져야 한다.
한편, 상기 항체는 링커를 통하여 약물에 결합될 수 있다. 상기 링커는 본 발명의 항체와 약물 사이를 연결하는 부위로, 세포 내 환경에서 항체에서 약물이 방출될 수 있도록 하며, 항체의 긴 반감기를 반영하여 항체가 전신 순환 중에는 안정하고, 링커와 약물의 결합이 항체의 안정성 및 약물 동태에 영향을 주지 않아야 한다. 상기 링커는 예를 들어 절단성 링커 또는 비절단성 링커를 포함할 수 있다. 절단성 링커의 경우 펩타이드 링커와 같이 세포 내 펩티다아제 또는 프로테아제 효소 예를 들어 리소좀 또는 엔도좀 프로테아제에 의해 절단될 수 있고, 비절단성 링커의 경우 예를 들어 티오에테르 링커는 항체가 세포내 가수분해에 의해 비선택적으로 분해된 후에 약물이 방출될 수 있다.
본 발명에서 용어, "항체"는 면역학적으로 특정 항원과 반응성을 갖는 면역글로불린 분자를 포함하는, 항원을 특이적으로 인식하는 수용체 역할을 하는 단백질 분자를 의미하며, 그 예로, 단일 클론 항체, 다클론 항체, 전장항체(full-length antibody) 및 항체 단편을 모두 포함할 수 있다. 또한 상기 용어, 항체는 이가(bivalent) 또는 이중 특이성 분자(예컨대, 이중특이성 항체), 디아바디, 트리아바디 또는 테트라바디를 포함할 수 있다.
본 발명에서 용어, "단일 클론 항체"는 실질적으로 동일한 항체 집단에서 수득한 단일 분자 조성의 항체 분자를 지칭하고, 이러한 단일 클론 항체는 다클론 항체가 여러 개의 에피토프에 결합할 수 있는 것과 달리, 특정 에피토프에 대해 단일 결합성 및 친화도를 나타낸다. 본 발명에서 용어, "전장항체"는 2 개의 전체 길이의 경쇄 및 2 개의 전체 길이의 중쇄를 가지는 구조이며, 각각의 경쇄는 중쇄와 다이설파이드 결합으로 연결되어 있다. 중쇄 불변영역은 감마(γ), 뮤(μ), 알파(α), 델타(δ) 및 엡실론(ε) 타입을 가지고 서브클래스로 감마1(γ1), 감마2(γ2), 감마3(γ3), 감마4(γ4), 알파1(α1) 및 알파2(α2)를 가진다. 경쇄의 불변영역은 카파(κ) 및 람다(λ) 타입을 가진다. IgG는 서브타입(subtype)으로, IgG1, IgG2, IgG3 및 IgG4를 포함한다.
본 발명에서 용어, "키메라 항체"는 마우스 항체의 가변영역 및 인간 항체의 불변영역을 재조합시킨 항체로서, 마우스 항체에 비하여 면역 반응이 크게 개선된 항체이다.
본 발명에서 용어, "중쇄"는 항원에 특이성을 부여하기 위한 충분한 가변영역 서열을 갖는 아미노산 서열을 포함하는 가변 영역 VH 및 3 개의 불변 영역 CH1, CH2 및 CH3를 포함하는 전체길이 중쇄 및 이의 단편을 모두 포함할 수 있다. 또한, 본 발명에서 용어, "경쇄"는 항원에 특이성을 부여하기 위한 충분한 가변영역 서열을 갖는 아미노산 서열을 포함하는 가변 영역 VL 및 불변 영역 CL을 포함하는 전체길이 경쇄 및 이의 단편을 모두 포함할 수 있다.
본 발명에서 용어, "단편", "항체 단편" 및 "항원 결합 단편"은 항체의 항원결합 기능을 보유하는 본 발명의 항체의 임의의 단편을 지칭하는 것으로 호환적으로 사용된다. 예시적인 항원 결합 단편은 Fab, Fab', F(ab')2 및 Fv 등을 포함하나, 이에 제한되지 않는다.
본 발명의 항체 또는 그의 항원 결합 단편은 Nectin-2에 특이적으로 결합하는 능력을 나타낼 수 있는 범위 내에서, 본 명세서에 기재된 항체의 서열뿐만 아니라, 이의 생물학적 균등물도 포함할 수 있다. 예를 들면, 항체의 결합 친화도 및/또는 기타 생물학적 특성을 보다 더 개선시키기 위하여 항체의 아미노산 서열에 추가적인 변화를 줄 수 있다. 이러한 변형은 예를 들어, 항체의 아미노산 서열 잔기의 결실, 삽입 및/또는 치환을 포함한다. 이러한 아미노산 변이는 아미노산 곁사슬 치환체의 상대적 유사성, 예컨대, 소수성, 친수성, 전하, 크기 등에 기초하여 이루어진다. 아미노산 곁사슬 치환체의 크기, 모양 및 종류에 대한 분석에 의하여, 아르기닌, 리신과 히스티딘은 모두 양전하를 띈 잔기이고; 알라닌, 글리신과 세린은 유사한 크기를 가지며; 페닐알라닌, 트립토판과 티로신은 유사한 모양을 갖는다는 것을 알 수 있다. 따라서, 이점에 기초하여, 아르기닌, 라신과 히스티딘; 알라닌, 글리신과 세린; 그리고 페닐알라닌, 트립토판과 티로신은 생물학적으로 기능 균등물이라 할 수 있다.
또한, 본 발명은 상기 항체 약물 접합체를 유효성분으로 포함하는 Nectin-2가 과발현되는 암 예방 또는 치료용 약학조성물을 제공한다.
바람직하게는, 상기 암은 유방암, 난소암, 췌장암, 폐암, 전립선암, 식도암, 담낭암 또는 급성골수성백혈병일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 약학조성물은 유효 성분 이외에 약제학적으로 적합하고 생리학적으로 허용되는 보조제를 사용하여 제조될 수 있으며, 상기 보조제로는 부형제, 붕해제, 감미제, 결합제, 피복제, 팽창제, 윤활제, 활택제 또는 향미제 등의 가용화제를 사용할 수 있다. 본 발명의 약학 조성물은 투여를 위해서 유효 성분 이외에 추가로 약제학적으로 허용 가능한 담체를 1 종 이상 포함하여 약학 조성물로 바람직하게 제제화할 수 있다. 액상 용액으로 제제화되는 조성물에 있어서 허용 가능한 약제학적 담체로는, 멸균 및 생체에 적합한 것으로서, 식염수, 멸균수, 링거액, 완충 식염수, 알부민 주사용액, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다.
본 발명의 약학조성물의 약제 제제 형태는 과립제, 산제, 피복정, 정제, 캡슐제, 좌제, 시럽, 즙, 현탁제, 유제, 점적제 또는 주사 가능한 액제 및 활성 화합물의 서방출형 제제 등이 될 수 있다. 본 발명의 약학조성물은 정맥내, 동맥내, 복강내, 근육내, 흉골내, 경피, 비측내, 흡입, 국소, 직장, 경구, 안구내 또는 피내 경로를 통해 통상적인 방식으로 투여할 수 있다. 본 발명의 약학 조성물의 유효성분의 유효량은 질환의 예방 또는 치료 요구되는 양을 의미한다. 따라서, 질환의 종류, 질환의 중증도, 조성물에 함유된 유효 성분 및 다른 성분의 종류 및 함량, 제형의 종류 및 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료 기간, 동시 사용되는 약물을 비롯한 다양한 인자에 따라 조절될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
<실험예>
하기의 실험예들은 본 발명에 따른 각각의 실시예에 공통적으로 적용되는 실험예를 제공하기 위한 것이다.
1. 세포 배양 및 세포주
본 발명에서, 인간 배아신장세포로 HEK-293, 유방암 세포주로 MCF-7, 췌장암 세포주로 PANC-1, AsPC-1 및 BxPC-3와, 난소암 세포주로 Caov-3, ES-2, OVCAR-3, OV-90, SK-OV-3, TOV-112D 및 UWB1.289를 사용하였다. HEK-293, MCF-7, PANC-1 및 Caov-3은 Dulbecco's high glucose modified Eagle's medium (DMEM, Hyclone, Logan, USA)에서 배양하였다. AsPC-1, BxPC-3, SK-OV-3, OVCAR-3 및 UWB1.289는 Roswell Park Memorial Institute (RPMI) 1640 medium (Hyclone, Logan, USA)에서 배양하였고, ES-2는 McCoy's 5A medium (Hyclone, Logan, USA)에서 배양하였다. OV-90 및 TOV-112D는 MCDB-105 medium (Sigma-Aldrich, St. Louis, USA) 및 Medium 199 (M199, Hyclone, Logan, USA)를 1:1 혼합하여 배양하였다. MCDB-105 medium 및 Media 199 medium의 혼합물에 15% FBS 및 1% 페니실린/스트렙토마이신이 첨가된 것을 제외하고는, 세포 배양 배지는 모두 10% 우태아혈청 (FBS, Hyclone, Logan, USA) 및 1% 페니실린/스트렙토마이신 (Hyclone, Logan, USA)이 첨가되었다. 세포주에 대한 자세한 정보는 표 1에 기재하였다. 세포는 5% CO2 배양기에서 가습 조건으로 37℃에서 배양하였다.

2. 항체 제작
Nectin-2에 대한 단일클론항체를 제작하기 위해서, 재조합 Nectin-2 단백질 (50 μg, Sino biological Inc., Beijing, China)을 complete Freund's adjuvant (Sigma-Aldrich, St. Louis, USA)와 함께, 6주령 암컷 Balb/c 마우스의 피하로 주사하였다. 이후 면역화는 incomplete Freund's adjuvant (Sigma-Aldrich, St. Louis, USA)가 에멀젼화된 동량의 면역원을 이용하여 1주일 간격으로 1회 더 반복하였다. 2차 주사 후, 간접 ELISA를 사용하여 혈청 내 높은 역가의 항체를 측정하였다. 하이브리도마 구축을 위해, 최종 추가접종 3일 후, 비장세포를 수확하였고, 50% 폴리에틸렌글리콜 (PEG, Sigma-Aldrich, St. Louis, USA)을 사용하여 SP2/0 세포와 융합시켰다. 융합된 하이브리도마를 DMEM 배지에 HAT 첨가제 (GIBCO, Grand Island, USA)가 포함된 하이포잔틴, 아미노프테린 및 티미딘 (HAT) 배지에서 스크리닝하였다. 선별된 하이브리도마는 세포 배양 상등액을 이용하여 간접 ELISA에서 측정하였다. 항체를 증폭하기 위해서, pristane (Sigma-Aldrich, St. Louis, USA)을 Balb/c 마우스에 접종시킨 일주일 후에 하이브리도마를 칼슘 및 마그네슘이 없는 Dulbecco's phosphate buffered saline (DPBS, Hyclone, Logan, USA)에 희석하여, 10주령 Balb/c 마우스의 복강 내로 주사하였다(5×106 cells/mice). 2주 후 면역화된 마우스로부터 복수를 수확하였고, 상등액을 얻기 위해 842×g로 15분 동안 원심분리하였다. 면역화로 인한 복수는 간접 ELISA, 웨스턴 블롯 및 소듐 도데실 설페이트 폴리아크릴아미드 겔 전기영동(SDS-PAGE)을 통해 확인되었다. protein A/G plus beads (Santa Cruz Biotechnology, Dallas, USA)를 사용하여, 복수로부터 단일클론항체를 정제하였다. 항체의 이소타입은 Beadlyte-Mouse Immunoglobulin Isotyping Kit (Upstate Co., Temecula, USA)를 통해 확인하였다.
12G1 클론의 가변 도메인의 아미노산 서열은 핵산 서열분석을 통해 결정하였고, 마우스 12G1 항체의 가변 도메인을 인간 IgG1에 접목시켜 키메릭 12G1(c12G1) 항체를 생성하였다. c12G1은 pCHO 1.0 vector (Thermo Fisher Scientific, Waltham, MA, USA)에 서브클로닝하였고, OptiPRO™ SFM 및 FreeStyle™ MAX Reagent(Thermo Fisher Scientific)를 사용하여 제조사의 지시에 따라, 재조합 플라스미드는 CHO-S cells (Thermo Fisher Scientific)로 일시적으로 형질감염시켰다. 항체는 Protein A Sepharose and SP Sepharose columns (Invitrogen, Waltham, MA, USA)을 사용하여 정제하였다.
3. 간접 ELISA
96-웰 플레이트 (SPL Life Sciences, Pocheon, Korea)는 PBS에 희석된 재조합 Nectin-2 단백질 (100 ng/well)로 코팅되었고, 4℃에서 밤새도록 배양하였다. 이후, 상기 플레이트는 PBS로 3번 씻어냈고, 1% 소혈청알부민 (BSA, Bovogen Biologicals, East Keilor, Australia)이 포함된 PBS로 1시간 동안 차단시켰으며, 3번 씻어냈다. 혈청, 하이브리도마 배양 상등액, 복수 및 정제된 항체는 1차 항체로 사용되었고, 각 웰에 다양한 희석제와 함께 첨가하였으며, 1시간 동안 상온(RT)에서 반응시켜, Nectin-2 단백질에 대한 특이성을 스크리닝하였다. 상기 플레이트는 0.05% Tween-20이 포함된 PBS (PBST, Sigma-Aldrich, St. Louis, USA)로 3번 씻어냈다. Horseradish peroxidase (HRP)-conjugated anti-mouse IgG (1:10,000, Thermo Fisher Scientific, Waltham, USA)는 차단 완충액 (100 μl/well)이 포함된 각 웰에서 1시간 동안 RT로 반응시켰다. 상기 플레이트는 PBST로 3번 씻어냈고, 3,3′,5,5′-tetramethylbenzidine (TMB, Sigma-Aldrich, St. Louis, USA) 퍼록시다제 기질 (100 μl/well)을 첨가하였고, 광노출 없이 RT에서 15분 동안 반응시켰다. 상기 반응은 50 μl의 H2SO4를 5분 동안 RT에서 첨가하여 중단시켰다. 광학밀도 (Optical density; O.D.)는 450 nm 파장에서 Perkinelmer Victor X3 (Perkin Elmer, Waltham, USA)를 이용하여 측정하였다.
4. RNA 추출 및 cDNA 합성
TRIzol reagent (Ambion®, Carlsbad, USA)를 이용하여 제조사의 지시에 따라, HEK-293 또는 하이브리도마 세포로부터 총 RNA를 분리하였다. RNA 농도는 SPECTROstar Nano (BMG LABTECH, Ortenberg, Germany)에 의해 측정하였고, 분리된 RNA의 순도는 ethidium bromide (EtBr, Sigma-Aldrich, St. Louis, USA)로 염색된 1% 아가로스 젤 상에서 측정하였다. 1차-가닥 상보성 DNA (cDNA)는 500 ng 총 RNA 100 μM random hexamer (Enzynomics, Daejeon, Korea) 또는 80 μM oligo dT (Enzynomics, Daejeon, Korea) 및 RNase free water로 합성되었다. RNA는 72℃에서 5분 동안 변성시켰고, 4℃에서 식혔다. 이후, 튜브에 10 mM dNTP, 200 U/μl M-MLV reverse transcriptase (Beamsbio, Seongnam, Korea) 및 5X reaction buffer (50 mM Tris-HCl [pH 7.6], 1 mM dithiothreitol (DTT), 0.1% Nonidet P-40, 0.1 mM ethylenediaminetetraacetica acid (EDTA), 150 mM NaCl 및 50% glycerol)를 첨가하였다. cDNA 합성을 위한 중합효소 연쇄반응 (PCR) 조건은 37℃에서 5분 동안 전-반응, 37℃에서 60분 동안 합성, 72℃에서 5분 동안 역전사효소의 불활성화 및 사용 전까지 4℃에서 보관으로 구성된다. PCR 분석은 MultiGene OptiMax (Labnet, NJ, USA)로 수행하였다. cDNA는 RNase free water와 1:2로 희석되었고, 이후 역전사 중합효소 연쇄반응 (RT-PCR) 및 정량적 실시간 중합효소 연쇄반응 (qRT-PCR)에 사용되었다.
5. 단일클론항체에서 가변 영역 클로닝
합성된 cDNA는 5 U/μl GS-Taq DNA polymerase (Beamsbio, Seongnam, Korea), 10 mM dNTP, 10 μM 특정 프라이머 (표 2) 및 10X GS-Taq reaction buffer (200 mM Tris-HCl [pH 8.8], 100 mM NaCl, 0.1 mM EDTA, 2 mM DTT, 1.0% Triton X-100, and 50% glycerol)로 증폭되었다. 항체 내 중쇄의 열 사이클링은 다음의 조건 하에서 수행되었다; 94℃에서 5분 동안 전-변성, 이어서 94℃에서 1분 동안 변성, 61℃로부터 58℃까지 각 온도 당 5 사이클씩 반복하며 한 사이클마다 1분 동안 결합 (예외적으로 MHV7 또는 11 프라이머는 57℃로부터 53℃까지 결합으로 결합함) 및 72℃에서 1분 동안 신장으로 25 사이클, 72℃에서 7분 동안 최종 신장 및 사용 전까지 4℃에서 보관. 또한, 항체 내 경쇄의 증폭 조건은 51℃로부터 52℃까지 각 온도 당 12 사이클씩 반복하는 결합 온도를 갖는다(MKV 10 프라이머의 경우, 62℃로부터 54℃까지 각 온도 당 5 사이클). 상기 산물은 EDTA로 염색된 1% 아가로스 젤로 확인되었다. 양성 PCR 산물은 TOPclonerTM TA kit (Enzynomics, Daejeon, Korea)로 제조사의 지시에 따라 클로닝되었다. 플라스미드는 DH5α (Enzynomics, Daejeon, Korea)로 형질전화되었고, 양성 클론들은 Fast DNA-spinTM (iNtRON Biothechnology, Seongnam, Korea)로 정제되었으며, EtBr로 염색된 0.8% 아가로스 젤로 확인되었다. 정제된 플라스미드의 서열을 스크리닝 하기 위해 Enzynomics Co., Ltd.의 분석 서비스를 이용하였다.

6. 표면 플라즈몬 공명 분석
표면 플라즈몬 공명 (Surface plasmon resonance; SPR) 분석은 SR7500DC (Reichert Technologies, Depew, USA)을 사용하여 수행되었다. 20 mM 소듐 아세테이트 완충액 (pH 4.5)에 녹인 재조합 Nectin-2 (1 μg, Sinobio, Beijing, China)는 PEG chips (Reichert Technologies, Depew, USA)인 금 칩 상에 제조사의 지시에 따라 고정화시켰다. 결합 친화도 분석을 위한, 1X PBS (pH 7.4)에 녹인 다양한 농도의 12G1 항체의 유속은 30 μl/min이었다. KD 수치는 평형 해리상수 수치이고, 결합 상수 (Ka) 및 해리 상수 (Kd) 수치에 의해 Scrubber 2 software (Reichert Technologies, Depew, USA)를 사용하여 계산되었다.
7. FACS 분석
세포 표면상 Nectin-2의 발현을 측정하기 위해서, enzyme-free cell dissociation buffer (Merck-Millipore, Darmstadt, Germany)를 사용하여 5-7분 동안 37℃에서 반응시켜, 컨플루언스가 70-80%가 되었을 때 다양한 세포주를 수확하였다. 변형된 DPBS에서 1% BSA가 포함된 아이스-콜드 세척 완충액으로 세포를 씻어냈다. 칼슘 및 마그네슘 없이 4℃에서 3분 동안 200 ×g로 원심분리하였다. 12G1 및 정상 마우스 IgG (Santa Cruz Biotechnology, Dallas, USA) 항체는 아이스-콜드 세척 완충액에서 4℃ 교반기를 사용하여 1시간 동안 반응시켰다. 정상 마우스 IgG는 이소타입 대조군으로 사용되었다. 반응 후, 세포를 아이스-콜드 세척 완충액으로 2번 씻어냈다. 이후, 2차 항체인 mouse fluorescein isothiocyanate (FITC)-conjugated Alexa 488 (Thermo Fisher Scientific, Waltham, USA)를 반응시켰고, 아이스-콜드 세척 완충액에서 4℃로 교반시켰다. 세포를 아이스-콜드 세척 완충액으로 2번 씻어냈고, 단일 세포군을 확인하였다. 데이터는 CyFlow cube 6 (Sysmex Partec GmbH, Gφrlitz, Germany)로 획득하였고, FCS express 4 flow research edition (De Novo Software™, Los Angeles, USA)를 사용하여 분석하였다.
8. 면역침전 분석 및 웨스턴 블랏
HEK-293는 70-80% 컨플루언시에서 수확하였고, 20 mM Tris-HCl (pH 7.6), 150 mM NaCl, 1 mM Na2EDTA, 1 mM EGTA, 1% NP-40, 0.5% sodium deoxycholate, 0.1% SDS, 10 mM β-glycerophosphate, 1 mM Na3VO4, 10 mM NaF, 1 mM phenylmethylsulfonyl fluoride (PMSF, Sigma-Aldrich, St. Louis, USA) 및 2 mM 2-mercaptoethanol, protease inhibitor cocktail (PIC, Calbiochem, Darmstadt, Germany) 및 100 mM PMSF를 포함하는 차가운 radioimmunoprecipitation assay buffer (RIPA) 용해 완충액으로 균질화시켰다. 재현탁된 세포는 얼음에서 20분 동안 반응시켰고, 25,000 ×g로 20분 동안 4℃에서 원심분리시켰다.
IP 분석을 위해, 대조군으로서 1 μg의 정상 마우스 IgG 또는 12G1 항체를 A/G plus agarose bead (Santa Cruz Biotechnology, Dallas, USA) 존재하에서, 전체 세포 용해물 (200 μg)과 반응시켰고, 상기 혼합물은 4℃에서 2시간 동안 교반시켰다. 상기 비드는 842 ×g로 1분 동안 4℃에서 원심분리하여 침전시켰고, RIPA 용해 완충액으로 3번 씻어냈다. 침전된 비드는 2X SDS-PAGE sample buffer로 재현탁시켰고, 7분 동안 끓였다. 상기 상등액은 7% SDS-PAGE gel에 적용하였고, 이후 polyvinylidene fluoride (PVDF) microporous membrane (Millipore, MA, USA)로 옮겼다.
웨스턴 블랏을 위해, 상기 멤브레인은 5% skim milk 및 0.2% Tween-20이 포함된 tris-based saline (TBS, 20 mM Tris-HCl [pH 7.6], 150 mM HCl)으로 1시간 동안 RT에서 차단시켰다. 이후, 상기 멤브레인은 anti-rabbit Nectin-2 antibody (1:5000, Abcam, Cambridge, UK) 또는 12G1과 같은 anti-mouse purified Nectin-2 IgG antibody (1:1000 에서 1:10,000까지 희석함), 1:100 희석시킨 복수를 1% BSA 및 0.2% Tween-20가 포함된 TBS에 녹인 1차 항체 용액으로 밤새도록 4℃에서 반응시켰다. 반응 후, 상기 멤브레인은 TBST로 5분씩 3번 씻어냈다. 본 발명에서 사용된 2차 항체는 HRP-conjugated anti-mouse IgG (1:20,000, Thermo Scientific, Waltham, USA) 및 HRP-conjugated anti-rabbit IgG (1:3,000, Thermo Scientific, Waltham, USA)이었고, 1% skim milk가 포함된 TBST에 상기 멤브레인을 RT에서 1시간 동안 반응시켰다. 상기 멤브레인은 TBST로 3번 씻어냈고, enhanced chemiluminescence (ECL) solution (Santa Cruz Biotechnology, Dallas, USA)을 사용하여 특정 시그널을 검출하였다.
9. RNA 간섭
Nectin-2의 녹다운 실험을 위해, 다음의 Nectin-2 siRNA 서열을 사용하였다. #1 sense; 5'-UGACCUGGCUCAGAGUCAUAGCCAA-3' (SEQ. 34) 및 antisense; 5'-UUGGCUAUGACUCUGAGCCAGGUCA-3' (SEQ. 35), #2 sense; 5'-CCUGAUACCUGUGACCCUCUCUGUA-3' (SEQ. 36) 및 antisense; 5'-UACAGAGAGGGUCACAGGUAUCAGG-3' (SEQ. 37), #3 sense; 5'-CACCUUCGUCUGCACAGUCACCAAU-3' (SEQ. 38) 및 antisense; 5'-AUUGGUGACUGUGCAGACGAAGGUG-3' (SEQ. 39). Nectin-2 siRNAs는 센스 및 안티센스 올리고머 (Bioneer, Daejeon, Korea)로 합성되었고, 음성 siRNA는 Bioneer로부터 구입하였다. HEK-293 세포 (2 × 105 cells/well)는 6-웰 플레이트 (Thermo Fisher Scientific, Waltham, USA)에 접종되었고, 12-18 시간 동안 배양되었다. 이후, K2 reagent (Biontex Laboratories GmbH, Munchen, Germany)를 사용하여 제조사의 지시에 따라, 40 nM siRNA를 HEK-293 세포로 72시간 동안 형질감염시켰다. 녹다운 효율은 RT-PCR 또는 qRT-PCR을 사용하여 대조군 siRNA와 비교하였다.
10. RT-PCR 및 qRT-PCR
표적 유전자들은 RT-PCR 및 qRT-PCR을 통해 증폭되었다. RT-PCR은 주형으로 cDNA와 10 μM 특정 프라이머(표 3)를 사용하여 AccuPower PCR PreMix (Bioneer, Daejeon, Korea) 상에서 수행하였다. 증폭 조건은 다음과 같다; 94℃에서 5분 동안 전-변성, 이어서 94℃에서 30초 동안 변성, 60℃에서 30초 동안 결합 및 72℃에서 30초 동안 신장으로 30 사이클, 최종 사이클에서 72℃에서 10분 동안 최종 신장 및 사용 전까지 4℃에서 보관. 상기 PCR 산물은 EtBr로 염색된 3% 아가로스 젤에서 밴드를 분석하였다. qRT-PCR은 유전자 발현 분석을 위해, cDNA, 10 μM 프라이머 및 StepOne Real-Time PCR System (Applied Biosystems, Foster City, USA) 상의 TOPreal qPCR 2X PreMIX with SYBR Green (Enzynomics, Daejeon, Korea)으로 구성된 반응 혼합물을 사용하여 수행하였다. 열 사이클링 조건은 94℃에서 10분 동안 단일 변성, 이어서 95℃에서 15초 및 60℃에서 1분으로 40 사이클, 95℃에서 15초, 60℃에서 1분, 95℃에서 15초의 최종 사이클로 구성된다. 각 실험에서, Nectin-2 발현 수준은 주어진 실험 조건 하에서 GAPDH의 Ct 수치로 정규화되었고, 배수 변화의 수치 (RQ, 2-△△Ct)로 계산되었다. GAPDH는 로딩 대조군으로 사용되었다.

11. ELISA
인간 재조합 넥틴-2 단백질은 1×PBS(실험실 제작)를 사용하여 20ng/100μL/웰로 희석하였고, 96-웰 Immuno Clear Standard Module (Thermo Fisher Scientific) 상에 4℃로 밤새도록 코팅하였다. RT로 2시간 동안 300μL/well의 차단 완충액 (5% BSA를 함유하는 PBS)으로 차단시킨 후, 상기 플레이트를 다음의 일차 항체 (100μL/well)로 반응시켰다: 마우스 항-12G1 또는 항-c12G1은 0.1% Tween-20 및 1% BSA가 포함된 PBS에 RT에서 1시간 30분 동안 희석하였다. 상기 플레이트는 PBST로 4번 씻어냈고, 2차 항체(100 μL/well)로 RT에서 1시간 동안 반응시켰다. 상기 2차 항체는 PBST에 희석된 HRP-conjugated anti-mouse IgG (1:1500 희석, Thermo Fisher Scientific) 및 anti-human IgG (1:1500 희석, Thermo Fisher Scientific)이었다. 이후, 상기 플레이트를 PBST로 4번 씻어냈고, 상기 샘플은 100 μL/well의 1-Step™ Ultra TMB-ELISA Substrate Solution (Thermo Fisher Scientific)으로 3분 동안 반응시켰다. 상기 반응은 1N H2SO4 (50 μL/well)로 종결시킨 이후, SPECTROstar  Nano microplate reader (BMG Labtech, Ortenberg, Germany)를 사용하여 450 nm의 파장에서 흡광도 측정을 수행하였다.
c12G1이 PVRIG에 대한 Nectin-2의 결합을 방해할 수 있는지 확인하기 위해서, 경쟁적 ELISA를 수행하였다. 인간 재조합 PVRIG 단백질(100ng/100μL/well; Sino Biological Inc.)은 ELISA 플레이트 상에 코팅되었고, 각 웰은 상기와 같이 차단시켰다. 인간 재조합 Nectin-2 단백질 (100ng)은 12G1 또는 hIgG 이소타입을 포함 또는 미포함하여 RT에서 1시간 동안 반응시켰다. 차단 후, 상기 플레이트는 전-반응시킨 혼합물 (100 μL/well)로 RT에서 1시간 30분 동안 반응시켰다. 상기 플레이트는 PBST로 4번 씻어냈고, 이후 100 μL/well의 rabbit polyclonal anti-Nectin-2 antibody (1:500, Abcam)로 RT에서 1시간 동안 반응시켰다. HRP-conjugated anti-rabbit IgG(100 μL/well, 1:1500 희석, Thermo Fisher Scientific)를 RT에서 1시간 동안 각 웰에 첨가하였다. 이후, 상기 플레이트는 PBST로 4번 씻어냈고, HRP 시그널을 측정하였다.
12. 이펙터 기능 분석
ADCC 분석을 위해, 표적 세포(T)로서 사용된 OV-90 세포는 세포 해리 완충액을 사용하여 수확하였고, 10 μM Hoechst 33,342 (Invitrogen)로 20분 동안 염색시켰다. 염색된 세포는 DPBS로 3번 씻어냈고, 96-well black cell culture microplate (Greiner Bio-One, Kremsmunster, Austria) 상에 웰 당 5×103 세포 밀도로 접종하였다. 이후, 상기 세포는 각각 표시된 농도의 c12G1로 처리하였고, 30분 동안 반응시켰다. 반응 동안, 이펙터 세포(E)로 사용된 인간 말초 혈액 단핵구 세포 (PBMCs; Cellular Technology Ltd., Cleveland, OH, USA)를 저농도 글루코스 (HyClone)와 함께, 10% FBS가 포함된 DMEM에서 녹였고, DPBS로 3번 씻어냈다. 혈구계를 사용하여 측정시, PBMCs의 생존율은 90% 이상이었다. PBMCs는 c12G1의 존재 유무에 따라 표적 세포 (E/T 비율, 30:1)로 반응시켰다. OV-90 세포는 37℃에서 6시간 동안 반응시켰고, Celigo Imaging Cytometer (Nexelom Bioscience, Lawrence, MA, USA)를 이용하여 계수하였다.
CDC 분석을 위해, OV-90 세포(5 × 103 /well)는 96-well black cell culture microplate (Greiner Bio-One) 상에 접종하였고, 밤새도록 반응시켰다. 이후, c12G1를 상기 플레이트에 첨가하였고, 세포를 30분 동안 반응시켰다. 다음으로, 20% (v/v) 인간 혈청 보체 (Quidel, San Diego, CA, USA)를 플레이트에 첨가하였고, 37℃에서 6시간 동안 반응시켰다. 세포는 10 μM Hoechst 33342로 20분 동안 염색시켰고, Celigo Imaging Cytometer를 사용하여 계수하였다.
13. 내재화 분석
세포는 5% BSA를 함유한 DPBS에서 Human BD Fc Block™(BD Biosciences, San Diego, CA, USA)으로 RT에서 10분 동안 차단시켰다. 이후, 2% BSA 및 75 μg/mL 사이클로헥시미드를 포함하는 차가운 무혈청 배지에 희석된 c12G1(1 μg/mL)을 세포에 첨가하고, 상기 세포를 얼음에서 1시간 동안 반응시켰다.
두 번 씻어낸 후, 세포는 2% BSA 및 75 μg/mL 사이클로헥시미드가 포함된 차갑거나 예열된 무혈청 배지에 재현탁시킨 후, 60-mm 접시에 접종하였다. 각 샘플은 4℃ 또는 37℃에서 1시간 또는 3시간 동안 반응시켰다. 상기 세포를 수확하였고, 세척 완충액(2% BSA를 함유한 차가운 DPBS)에 희석한 플루오레세인 이소티오시아네이트-접합 항-인간 IgG (0.3 μg/mL; Thermo Fisher Scientific)로 얼음에서 1시간 동안 염색하였다. 세 번 씻어낸 후, CyFlow Cube 6을 사용하여 형광 신호를 검출하였고, FCS Express™ 6 flow cytometry software를 사용하여 분석하였다. pH 민감성 Zenon™ 염료를 사용하여 추가적인 내재화 분석을 수행하였다. OV-90 세포, SK-OV-3 세포 또는 Caov-3 세포(1 × 105 세포)를 48-웰 플레이트에 접종하였고, 37℃에서 1시간 동안 반응시켰다. 이후, 세포를 c12G1 항체/Zenon™ 염료 혼합물(10:100 nM)로 RT에서 5분간 처리하였고, 추가로 37℃에서 15 시간 동안 반응시켰다. 그 후, 0.25% Trypsin/1mM EDTA (Welgene Inc., Gyeongsan, Korea)를 사용하여 세포를 수확하였고, 1000× g 에서 3분 동안 4℃에서 원심분리하였다. 세포는 차가운 DPBS로 씻어냈고, 0.1% BSA를 함유한 차가운 DPBS에 재현탁시켰다. CytoFLEX(Beckman Coulter Inc., Brea, CA, USA)를 사용하여 형광 신호를 검출하였고, CytExpert software(Beckman Coulter Inc.)를 사용하여 분석하였다.
14. ADCs 제작
항체는 BupH™ PBS Packs(Thermo Fisher Scientific)으로 투석하였고, smcc-DM1 (MedChemExpress, Monmouth Junction, NJ, USA)으로 RT에서 1:20의 몰비로 1.5시간 동안 접합시켰다. 상기 혼합물은 20℃에서 20분 동안 21,000 × g 로 원심분리하여 응집물을 제거하였다. ADCs는 Sephadex G-25 PD-10 desalting column (GE Healthcare, Chicago, IL, USA)을 사용하여 정제하였고, 10 mM sodium succinate, 6% (v/v) sucrose 및 0.05% (v/v) Tween-20 pH 5.0 에 보관하였다. ADCs는 SDS-PAGE 및 자외선 분광법을 사용하여 추가 분석하였다. DAR 비율 분석은 이전에 기재된 대로 수행하였다.
15. 세포 주기 및 시험관 내 세포독성 분석
세포는 96-웰 흑색 세포 배양 마이크로플레이트(부착 세포의 경우, 3-8 × 103 cells/well 밀도) 또는 6-웰 플레이트(현탁 세포의 경우, 1 × 105 세포 cells/well 밀도)에 접종하였고, 가습된 5% CO2-포함 챔버에서 밤새도록 반응시켰다. 이후, 세포는 24시간 또는 48시간 동안 1 μg/mL의 항체 또는 ADCs와 함께 반응시켰다. 세포는 70% 아이스-콜드 에탄올로 4℃에서 30분간 고정하였고, 차가운 DPBS로 두 번 씻어냈다. 또한, 세포는 propidium iodide solution (50 μg/mL, 0.1 mg/mL RNase 및 0.05% Triton™ X-100 포함)으로 37℃에서 1시간 동안 염색하였다. 이후, 세포는 Celigo Imaging Cytometer를 사용하여 문서화되었고, FCS Express™ 6 flow cytometry software를 사용하여 분석하였다.
세포독성 분석을 위해, 세포는 96-웰 흑색 세포 배양 마이크로플레이트(2-5 × 103 cells/well)에 접종하였다. 이후, 0-400μg/mL 범위의 농도로 항체 또는 ADC로 세포를 처리하였고, 세포 성장 속도에 따라 플레이트를 3~4일 동안 추가로 배양하였다. 세포 컨플루언시가 80-90%에 도달하면, 세포를 10 μM Hoechst 33342 또는 1 μg/mL Calcein AM (Invitrogen)으로 20분간 염색하였고, Celigo Imaging Cytometer를 사용하여 계수하였다. IC50 수치는 Prism 5 (GraphPad Software Inc., San Diego, CA, USA)를 사용하여 계산되었다.
16. 생체 내 실험
동물 연구는 아주대학교 동물기관관리위원회의 승인을 받았다(IACUC 승인 번호: 2021-0019). 실험은 미국 국립보건원(National Institutes of Health)에서 발행한 실험실 동물의 관리 및 사용에 대한 지침을 준수하였다. 모든 실험은 관련 지침 및 규정에 따라 수행되었다. c12G1-DM1의 독성을 평가하기 위해, C57BL/6 마우스(Koatech, Pyeongtaek, Korea)에 비히클 또는 c12G1-DM1 20mg/kg을 정맥 투여하였고, 15일 동안 3일마다 체중 변화를 관측하였다.
이종이식 분석을 위해, OV-90(5 × 106/100 μL), SK-OV-3(5 × 106 /100 μL) 및 Caov-3(5 × 106 /100 μL) 세포를 동등 부피의 Matrigel  ( Corning, Corning, NY, USA)와 혼합하였고, 5주령 암컷 CB-17 중증 복합 면역결핍 마우스(Orient Bio Inc., Seongnam, Korea)에 피하 주사하였다. 평균 종양 부피가 100-200 mm3에 도달하면, 마우스를 효능 연구를 위해 처리군으로 임의 추출하였다. DPBS에 희석된 비히클, c12G1 또는 ADCs를 지정된 농도로 일주일에 1회, 3번 마우스에 정맥 투여하였다. 종양 크기는 Vernier caliper를 사용하여 측정하였고, 종양 부피는 다음과 같이 계산하였다.

종양 성장 억제율은 다음과 같이 계산되었다.

여기서 RTV는 상대적인 종양 부피이다. 실험이 끝나면, 마우스는 CO2를 사용하여 안락사시켰다 .
17. 통계 분석
통계 분석은 Prism 5를 사용하여 수행하였다. 데이터는 평균 ± 평균의 표준 오차(S.E.M.) 또는 ± 표준 편차(S.D.)로 표시하였고, 통계적 유의성을 결정하기 위해 one-way ANOVA 및 unpaired Student's t-test를 사용하여 분석하였다. p < 0.05는 통계적 유의성이 있는 것으로 간주하였다.
<실시예 1> 단일클론 Nectin-2 항체의 제작
마우스 단일클론항체를 제작하기 위해서, 재조합 성숙 인간 Nectin-2를 Balb/c 마우스에 주사하였다. His-tag(340 a.a.)를 가진 Nectin-2 단백질은 구입하였다. 단백질의 순도는 95% 이상인 것으로 확인되었다. Nectin-2 단백질은 환원 SDS-PAGE에서 48 kDa의 분자량을 갖는 것으로 확인되었다. 상기 질량은 글리코실화 때문에 예측된 분자량인 36.2 kDa 보다 약간 더 높았다.
Nectin-2 단백질의 면역화 전과 후에 마우스 혈청을 수확하였고, 비특이적(도 1A) 및 특이적 항체(도 1B)의 존재에 대해 간접 ELISA로 평가하였다. 전-면역화된 혈청의 O.D. 수치는 1:100으로 희석하여, 0.069, 0.064, 0.06 및 0.071이었는데, 이는 전-면역화된 혈청이 임의의 비특이적 시그널을 나타내지 않는다는 것을 의미한다(도 1A). 상기 결과는 항원의 주입이 적절하다는 것을 나타낸다. 최종 주입 전에도, 마우스 혈청을 수확하였고, 간접 ELISA를 사용하여 특이 항체의 존재를 확인하였다. 면역화된 혈청은 O.D. 수치가 1:10,000으로 희석해도 1.0 이상인 것으로 나타났다(도 1B). 높은 항체 역가가 형성되는 것을 확인하여, 최종 투여 후에 비장을 채취하였고, 비장세포를 SP2/0 마우스 골수종 세포와 융합하여, 하이브리도마를 제작하였다. 융합된 세포들을 HAT 배지에서 선별하였고, 최종적으로 간접 ELISA로 시험시에 높은 항체 역가를 나타낸 하이브리도마 클론인 12G1을 선택하였다. 상기 하이브리도마 클론은 제한 희석을 통해 서브클로닝하였고, 이후 단일 콜로니를 수확하기 위해 스크리닝하였다. 상기 클론의 확장된 세포로부터 얻은 세포 배양 상등액은 간접 ELISA 및 웨스턴 블랏으로 검증하였다(도 1C 및 도 1D). 각 세포 배양 상등액의 O.D. 수치는 1.0 이상이었다(도 1C). 상기 결과는 높은 역가의 항체가 존재한다는 것을 나타낸다. 상기 클론인 12G1이 Nectin-2 단백질과 결합하는지 확인하기 위해서, 웨스턴 블랏을 수행하였다. 상기 항체들은 50 ng 이상의 Nectin-2 단백질을 검출할 수 있었다(도 1D).
anti-Nectin-2 항체를 분비하는 12G1 단일클론은 10주령 Balb/c 마우스 복수에 하이브리도마를 주입하여 증폭되었다. 하이브리도마 주입 2주 후에 복수를 수확하였다. 주입에 의해 생산된 복수는 간접 ELISA, 웨스턴 블랏 및 SDS-PAGE 분석 결과, 항원에 대해 양성이었다(도 2A, 도 2B 및 도 2C). 항체의 존재는 SDS-PAGE를 사용하여 복수에서 확인하였다(도 2A). 12G1 복수의 O.D. 수치는 1:100,000 희석에서도 1.18이었다(도 2B). 웨스턴 블랏 분석 결과에서도 12G1은 최소 20 ng의 Nectin-2 단백질을 인지하는 민감성을 나타냈다(도 2C). 항체의 이소타입은 카파 사슬로 이루어진 IgG1 서브클래스로 확인되었다(표 4). IgG 항체는 protein A/G plus beads를 사용하여 정제되었고, 클론의 농도를 측정하였다. 정제된 항체는 다양한 희석 인자에 대해 간접 ELISA로 분석하였고, O.D. 수치의 변화는 상기 ELISA 결과와 유사하였다(도 2D). coomassie blue R-250으로 염색한 SDS-PAGE 젤 분석 결과, 12G1 항체의 순도는 중쇄 (~50 kDa) 및 경쇄 (~25 kDa)로 검출되었다(도 2E).
Nectin-2 및 이의 항체인 12G1 사이의 결합 친화도를 측정하기 위해, SPR 분석을 수행하였다(도 3). KD 수치는 결합 상수 (Ka) 및 해리 상수 (Kd)로 계산하였다. 12G1 항체는 499 pM이었다(도 3, 하단 패널).
추가적으로 항원-결합 사이트를 확인하기 위해서, 하이브리도마의 총 RNA를 추출하였고, cDNA를 만들기 위해 합성하였는데, 이는 PCR을 이용하여 항체의 가변 영역에 대한 특이적인 프라이머로 증폭되었다. 산물 크기는 12G1 항체에서 중쇄 가변 영역 (VH)이 약 500 bp이었고, 경쇄 가변 영역 (VL)이 약 450 bp이었다(도 4). 상보성-결정 영역 (complementarity-determining regions; CDRs)이 다양성 항원 특이성에 기여하기 때문에, 각 항체에서 서열분석을 통해 가변 영역 내의 CDRs을 찾기 위해, PCR 산물은 TA 벡터로 클로닝되었고, 결찰된 플라스미드를 분석하였다(도 5). VH의 모든 PCR 산물은 각 클론에서 하나였다(도 4, 왼쪽). 12G1 항체는 MHV6 프라이머를 사용하여 양성 산물을 획득하였다. 12G1 항체에서, VL의 PCR 산물은 3개 (MKV2, MKV3 및 MKV6 프라이머)이었다. MKV2 프라이머를 사용한 PCR 산물의 경우, MKV2 프라이머가 항체에서의 유사유전자가 아닌 하이브리도마에서의 유사유전자를 증폭시켰기에 비특이적인 반응의 결과였다(도 4, 오른쪽). MKV3 및 MKV6 프라이머를 이용한 증폭은 12G1 항체에서 각각 동일한 서열분석 결과가 나타났다(도 5 및 표 5).

< 실시예 2> Nectin -2 마우스 단일클론항체의 특성 분석
FACS 및 IP 분석을 사용하여, 제작된 Nectin-2 마우스 단일클론항체의 잠재적 적용성을 확인하였다. FACS 분석에서는 HEK-293 세포를 사용하였고, 세포가 고정된 조건에서 반응을 하는지 또는 하지 않는지를 결정하기 위해 항체의 농도를 최적화하였다. 세포를 4% 파라포름알데히드로 고정시에, 12G1 항체와 반응하지 않았다. 따라서, 모든 세포는 살아있는 세포를 이용하여 검사하였다.
FACS 분석을 위해 FITC-접합 2차 항체를 최적화하기 위해서, 2 μg의 FITC-접합 2차 항체에 비해 4 μg의 FITC가 더 높은 비-특이적 배경 염색을 나타내는 것을 확인하였다(도 6A, 중간 및 오른쪽). 또한, 1 μg의 FITC-접합 2차 항체는 더 낮은 비-특이적 배경 염색을 나타냈으나, 양성 염색이 감소하는 것으로 나타났다. 이에 FITC-접합 2차 항체는 2 μg를 사용하였다.
FACS 분석을 위한 1차 항체를 최적화하기 위해서, 이소타입 대조군으로서 정상 마우스 IgG 항체를 다양한 농도 (0.05-10 μg)에서 시험하였다(도 6B). 1차 항체를 위한 정상 마우스 IgG의 비-특이적 염색은 용량-의존적 방식으로 증가하였다. 다만, 정상 마우스 IgG를 0.05 μg 사용하면, Nectin-2 양성 염색도 비특이적 시그널과 함께 감소한다는 점에서 해당 농도는 Nectin-2 염색에 적용할 수 없었다. 따라서, FACS 분석을 위해 0.1 μg의 1차 항체를 선택하였다. Nectin-2 항체인 12G1에 대한 FACS 분석은 최적화된 실험 조건에서 수행되었고, 결과는 12G1 항체가 양성 염색되는 것으로 나타났다(도 6C). 이에, 12G1 항체를 추가적인 특성 분석에 적용하였다.
IP 분석을 위해, 단백질을 HEK-293 세포로부터 추출하였다. 단백질 추출물 (200 μg)은 protein A/G plus beads 존재하에서, 1 μg의 정상 마우스 IgG 또는 12G1 항체로 반응시켰다. 상기 혼합물은 2시간 동안 4℃에서 반응시켰고 교반시켰다. 용촐된 단백질은 7% SDS-PAGE에 로딩하였고, 1차 항체로 항-토끼 Nectin-2 항체로 블랏하였다(도 7). 항-토끼 Nectin-2 항체와의 내재성 IP에서, 12G1 항체와의 침전은 ~75 kDa에서 검출되었다. 또한, 항-토끼 Nectin-2 항체를 사용한, 정상 마우스 IgG 항체와의 침전은 검출되지 않았는데, 이는 Nectin-2의 침전이 Nectin-2 항체에 의존적이라는 것을 나타낸다(도 7, 상단 패널). 추가적으로, 각 샘플의 IgG 중쇄는 coomassie blue 염료로 젤을 염색하거나, HRP-접합 항-마우스 IgG에 의해 검출되었다(도 7, 중간 패널). 상기 결과는 12G1 항체가 IP 분석에 사용될 수 있음을 뒷받침한다.
<실시예 3> 세포주에서 Nectin-2의 발현
HEK-293에서 Nectin-2 발현을 확인하기 위해서, RT-PCR 및 qRT-PCR 분석을 수행하여, GAPDH, Nectin-2, Nectin-2α Nectin-2δ를 증폭하였다. 내재성 대조군으로서 GAPDH를 사용하였다. Nectin-2는 이의 변이체의 공통 영역 (895-995 bp)을 증폭하였다. RT-PCR에 의한 GAPDH, Nectin-2, Nectin-2α Nectin-2δ의 증폭 산물 크기는 각각 122, 102, 56 및 139 bp이었다(도 8A). 상기 결과는 HEK-293이 Nectin-2α 및 δ를 모두 발현하였고, Nectin-2δ 발현 수준은 Nectin-2α 보다 더 높다는 것을 나타냈다(도 8A, 도 8B 및 도 8C). 항체 특이성을 추가적으로 확인하기 위해서, Nectin-2의 발현을 녹다운시키기 위한 특이적인 siRNA를 디자인하였는데, 이는 Nectin-2의 세포외 도메인 내 공통 영역을 표적으로 한다(도 8D). 이후, 각각의 siRNA는 HEK-293 세포로 형질감염시켰고, Nectin-2의 녹다운은 RT-PCR 및 qRT-PCR을 이용하여 확인하였다(도 8E 및 도 8F). 상기 결과는 Nectin-2α 및 δ의 발현은 모두 하향 조절되었고, 모든 siRNA 형질감염의 효율은 약 50%로 상당히 녹다운되었다는 것을 나타낸다(도 8E 및 도 8F).
siRNA (#1 + #2 + #3) 또는 음성대조군 siRNA를 혼합하여 Nectin-2로 형질감염된 HEK-293 세포는 FACS에 의해 12G1 항체로 평가하였다(도 9). 대조군 siRNA로 형질감염된 세포를 12G1 항체로 염색한 후의 평균 형광 세기 (mean fluorescence intensity; MFI)는 138이었는데(도 9, 하단 패널 내 왼쪽 바), Nectin-2 siRNA를 혼합하여 형질감염된 것과 비교하면 66.6이었다(도 9, 하단 패널 내 오른쪽 바). Nectin-2 siRNA가 HEK-293 세포로 형질감염되면, 염색된 Nectin-2 개체가 극적으로 감소한 것과 마찬가지로 12G1에 의해 염색되는 정도도 감소함을 확인하였다. 상기 결과는 12G1 항체가 Nectin-2 단백질에 특이적으로 결합함을 보여준다.
FACS 분석에 기반하여, 유방암, 난소암 및 췌장암 세포주를 포함한 다양한 암 세포주에서 Nectin-2 발현이 높은 세포의 발현 수준을 12G1 항체를 통해 검증하였다(도 10). Nectin-2의 발현 수준은 OV-90 세포에서 가장 높은 반면, Caov-3 세포에서 가장 낮은 발현 수준을 나타냈다. 상기 결과를 바탕으로, 12G1 항체는 생화학적 분석에서의 적용 가능성을 특별히 인정받을 수 있다는 것을 뒷받침한다.
FACS 분석에서, Nectin-2 발현 수준은 세포 표면 상의 단백질 수준을 나타낸다. mRNA 및 단백질 수준 사이의 연관성을 확인하기 위해서, 얼마나 많은 Nectin-2 mRNAs가 전사되는지 확인하기 위한 qRT-PCR을 수행하였다(도 11). OV-90 및 Caov-3 세포에서 mRNA-단백질 발현 수준은 FACS 및 qRT-PCR 분석 사이에 연관성이 있는 것으로 관측되었다. 그 결과, OV-90 세포는 Nectin-2을 높게 발현했으나, Caov-3 세포는 낮은 발현 수준을 나타냈다. 하지만, 다른 세포들은 발현 수준의 불일치가 나타났으며, 다양한 세포에서 Nectin-2의 발현 비율에 차이가 없었다(도 11).
<실시예 4> 키메릭 12G1(c12G1) 항체의 특성 분석
본 발명자들은 m12G1 항체의 가변 도메인을 인간 IgG1에 접목시켜 키메릭 12G1 항체 (c12G1)를 제작하였다. FACS 분석 결과, 난소암 세포주에 대한 c12G1 항체의 결합은 m12G1의 결합과 유사한 것으로 나타났다(도 12A). 또한, 추가적인 녹-다운 실험 결과, c12G1 항체가 Nectin-2에 특이적으로 결합한다는 것을 확인하였다(도 12B). 또한, SPR 분석 결과, 인간 Nectin-2 단백질 (KD = 2.90 × 10-10 M)에 대한 결합 친화도에는 차이가 없는 것으로 나타났다(도 12C).
최근에, 암세포에서 과발현되는 Nectin-2 단백질이 NK 및 세포독성 T 세포의 Nectin-2 단백질 수용체(PVRIG)와 결합하여, 면역 체크포인트로 기능하는 것으로 밝혀졌다. 면역세포의 사이토카인 분비 및 세포독성 활성은 Nectin-2의 V 도메인이 PVRIG와 결합하면 억제되었다. c12G1 항체가 Nectin-2 및 PVRIG의 상호작용을 억제하여, 면역 세포 활성자로 적용될 수 있는지 확인하기 위해서, 본 발명자들은 경쟁적 효소-결합 면역흡착 분석 (ELISA)을 수행하였다. 도 13A에서 나타낸 바와 같이, c12G1 항체는 PVRIG에 대한 Nectin-2의 결합을 방해하지 않았는데, 이는 c12G1 항체가 Nectin-2의 V 도메인 이외의 부위에 결합한다는 것을 뒷받침한다. c12G1 항체의 결합 도메인을 확인하기 위해서, 본 발명자들은 인간 Nectin-2의 여러 결핍 돌연변이들을 제작하였다(도 13B). 야생형 (WT), Δ1 또는 Δ2 돌연변이의 형질감염 후의 면역침전 분석 결과, c12G1 항체는 WT 및 Δ1 돌연변이에는 결합할 수 있었으나, Δ2 돌연변이에는 결합하지 않았는데, 이는 c12G1 항체가 Nectin-2의 첫번째 C2 도메인에 결합한다는 것을 나타낸다(도 13C). 또한, 인위적인 과발현의 경우, Nectin-2 단백질이 내재성 Nectin-2 단백질과 구조적으로 상이할 수 있으므로, 본 발명자들은 c12G1 항체가 내재성 Nectin-2 단백질에 결합할 수 있는지 확인하였다. HEK293 세포 용해물을 사용한 면역침전 분석 결과, c12G1 항체는 내재성 Nectin-2 단백질에 결합할 수 있는 것으로 나타났다(도 13D).
또한, 종양학에서 네이키드 항체의 치료 효율은 보체 의존성-세포독성(CDC) 및 항체-의존성 세포-매개 세포독성(ADCC)을 포함하는 이펙터 기능을 동시에 매개하면서 항원에 결합하는 능력에 따라 결정된다. OV-90 세포주를 사용한 시험관 내 이펙터 기능 분석 결과, c12G1 항체의 CDC 및 ADCC 활성은 20 μg/mL 농도를 넘지 못하는 것으로 관측되었다. 상기 결과는 네이키드 c12G1 항체가 이펙터 기능에 의한 항암 효과를 나타낼 것으로 기대할 수는 없고, 즉, 암 치료를 위한 면역항암제로서의 적용은 어렵다는 것을 나타낸다.
<실시예 5> 시험관 내 및 생체 내에서 항-종양 활성을 나타내는 c12G1 ADC
비록 OV-90 세포주에서 Nectin-2가 높게 과발현되지만, c12G1 항체는 항체 자체의 형태로는 항-종양제로 사용될 수 없다. 따라서 c12G1 항체를 이용한 ADC는 난소암을 효과적으로 치료하기 위한 합리적인 선택이 될 수 있다. ADC로서의 적용 가능성을 확인하기 위해, 본 발명자들은 난소암 세포에서 c12G1 항체의 내재화 효율을 조사하였다. FACS 분석에 따르면, c12G1 항체의 내재화 효율은 54.9% (OV-90), 59.1% (SK-OV-3) 및 57.9% (Caov-3)인 것으로 나타났다(도 14A). 리소좀 내의 pH는 약산성인 것으로 알려져 있다. 따라서, 본 발명자들은 pH 민감성 형광 염료인 Zenon™과 접합된 항-Fc FAB를 사용하여, c12G1 항체/넥틴-2 복합체의 리소좀으로의 이동을 추가 분석하였다. 도 14B에 나타낸 바와 같이, c12G1 항체의 내재화 효율은 OV-90 세포에서 81.4%, SK-OV-3 세포에서 45%, Caov-3 세포에서 28%였다. 종합해 보면, 상기 결과는 c12G1 항체가 난소암 치료를 위해 독소의 특이적인 전달을 위한 효율적인 운반체로 사용될 수 있음을 뒷받침한다. 이후, 본 발명자들은 비절단성 링커 및 미세소관 억제제로 구성된 smcc-DM1을 사용하여 ADC를 제작하였다. 소듐 도데실 설페이트-폴리아크릴아미드 겔 전기영동(SDS-PAGE) 분석 결과, smcc-DM1과 c12G1 항체의 접합이 약간의 크기 변화를 유도하는 것으로 나타났다(도 15A). 또한, DM1 및 항체는 각각 252 nm 및 280 nm에서 자외선을 흡수하므로, 본 발명자들은 252 nm 및 280 nm에서 네이키드 항체(c12G1) 및 ADC(c12G1-DM1)의 흡광도를 비교 분석하였다. DM1 및 c12G1의 접합은 252 nm에서 흡광도를 증가시켰다(도 15B). 약물-항체 비율(drug-to-antibody ratio; DAR)은 이전에 설명한 대로 측정하였으며, 약 5.07인 것으로 나타났다. ADC의 표적-결합 친화도는 smcc 링커의 N-하이드록시석시니미드 에스테르와 항체 내 리신의 1차 아민과의 접합을 통한 형태 변화에 의해 감소될 수 있으므로, 본 발명자들은 12G1 항체 및 12G1 ADC의 결합 친화도를 비교하였다. ELISA 및 FACS 분석 결과, c12G1 항체와 c12G1 ADC의 Nectin-2에 대한 결합 친화도는 유사한 것으로 나타났다(도 15C 및 도 15D). 상기 결과는 smcc-DM1과 c12G1 항체의 접합이 Nectin-2에 대한 결합 친화도에 영향을 미치지 않는다는 것을 나타낸다.
DM1은 미세소관 조립을 억제하고 적극적으로 분열하는 세포의 세포사멸을 촉진함으로써 G2/M 단계에서 세포 주기 정지를 유도한다. 세포 주기 분석 결과, c12G1-DM1은 G2/M 단계에서 세포 분열을 억제하는 것으로 나타났는데, 이는 sub-G1 집단 내의 세포사멸 세포의 증가를 유도한다(도 16A). 다만, c12G1-DM1은 Nectin-2 음성 세포주인 Daudi의 세포주기에 영향을 미치지 않았다. 이후, 본 발명자들은 난소암 세포주에서 c12G1-DM1의 세포독성 활성을 분석하였다. c12G1-DM1은 OV-90, SK-OV-3 및 Caov-3 세포에서 시험관 내 세포독성 활성을 나타냈으며, 반수 최대 억제 농도(IC50) 수치는 100 피코몰에서 나노몰 범위였다. Nectin-2 양성 세포주에 대한 c12G1-DM1의 시험관 내 세포 독성 활성은 Nectin-2 음성 세포주에 대한 것보다 5-300배 더 높았다(도 16B). 다음으로, OV-90, SK-OV-3 및 Caov-3 세포를 이종이식한 마우스 모델을 사용하여, c12G1-DM1의 생체 내 효능을 조사하였다. OV-90 세포주에서, 네이키드 c12G1 항체 및 IgG-DM1은 종양 성장을 억제하지 않은 반면, c12G1-DM1은 용량 의존적 방식으로 종양 성장을 억제하였다(도 16C). 추가적으로, 3 mg/kg 및 5 mg/kg의 c12G1-DM1은 약 25일 동안 종양의 완전한 관해를 유도하였다(도 16C). 또한, 알 수 없는 이유로 치료 자체의 초기 단계부터 투여에 반응하지 않은 5mg/kg 치료군 중 한 마리의 마우스를 제외하고, c12G1-DM1은 SK-OV-3에서는 어떠한 항종양 활성도 나타내지 않은 반면, c12G1-DM1은 Caov-3 세포에서는 용량 의존적 방식으로 종양 성장을 억제하였다(도 16C). ADC 처리된 마우스의 체중에는 변화가 없었으며, 정상 C57BL/6 마우스를 사용한 20 mg/kg c12G1-DM1의 단일 독성 연구는 체중 변화에 영향을 미치지 않았다. 종합하면, 상기 결과는 Nectin-2를 표적으로 하는 ADC가 난소암뿐만 아니라 유방암, 결장암, 폐암을 포함한 다른 Nectin-2 양성 암의 치료에 유용할 수 있다는 것을 뒷받침한다.
또한, 본 발명자들은 c12G1-smcc-DM1 및 c12G1-vc-PAB-MMAE를 비교하였다(도 17 및 도 18). 미세소관 억제제인 DM1 및 MMAE는 나노몰과 수십 나노몰 농도 사이에서 유사한 세포독성을 나타냈다. 난소암 세포주를 사용한 분석 결과, smcc-DM1과 vc-MMAE는 유사한 활성을 나타냈다. 비록 c12G1-smcc-DM1 및 c12G1-vc-PAB-MMAE의 DAR과 표적-결합 친화도는 유사하였고(5.07 vs 4.79), smcc-DM1에는 방관자 효과는 나타나지 않았으나, 비절단성 링커 페이로드인 smcc-DM1은 카텝신 B-절단성 링커 페이로드인 vc-PAB-MMAE 보다 훨씬 더 높은 세포독성 활성을 나타냈다. 카텝신 B를 포함한 프로테아제는 EOC에서 높게 발현 및 분비되어, 세포외 기질의 분해를 통한 전이를 일으킨다. 비절단성 링커 페이로드인 smcc-DM1은 리소좀 내 다양한 프로테아제에 의한 항체 분해 시 방출되지만, 절단성 링커는 분비된 카텝신 B에 의해 세포외 환경에서 절단될 수 있으며, 이는 c12G1-vc-PAB-MMAE의 MMAE가 암세포로 내재화되기 전에 세포외 환경으로 방출되어 치료 효율을 감소시킬 수 있다. 만약 ADC에서 비절단성 링커의 시험관 내 활성이 효과적인 치료 효율을 나타낸다면, 이는 항체의 분해된 펩타이드가 암세포 근처의 면역 세포 활성화를 촉진할 수 있기 때문에 이는 생체 내 효능으로 직접 해석될 수 있다. 다만, 이를 위해서는 추가 조사가 필요하다. 또한, smcc-DM1을 사용한 c12G1 항체의 아민 커플링은 비환원 및 환원 SDS-PAGE 모두에서 균일한 밴드 패턴을 나타낸 반면, mc-vc-PAB-MMAE를 가진 항체 내 유리 티올 그룹 연결은 환원 SDS-PAGE에서 관찰된 밴드 패턴은 균일했지만 비환원 SDS-PAGE에서는 불균일한 밴드 패턴을 나타냈다. 상기 결과는 mc-vc-PAB-MMAE의 접합 후에도 항체에 여전히 유리 티올 그룹이 존재하여 비환원 SDS-PAGE에서도 중쇄와 경쇄로 분리되고 용액 내 구조적 불안정성이 있음을 뒷받침한다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

  1. 서열번호 1로 표시되는 아미노산 서열로 이루어진 중쇄 CDR1, 서열번호 2로 표시되는 아미노산 서열로 이루어진 중쇄 CDR2 및 서열번호 3으로 표시되는 아미노산 서열로 이루어진 중쇄 CDR3을 포함하는 중쇄 가변 영역; 및 서열번호 4로 표시되는 아미노산 서열로 이루어진 경쇄 CDR1, 서열번호 5로 표시되는 아미노산 서열로 이루어진 경쇄 CDR2 및 서열번호 6으로 표시되는 아미노산 서열로 이루어진 경쇄 CDR3을 포함하는 경쇄 가변 영역을 포함하는 Nectin-2에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편; 및 약물이 결합된 항체 약물 접합체(antibody drug conjugates; ADC).

  6. 제5항에 있어서, 상기 항암제는 다우노루비신(daunorubicin), 독소루비신(doxorubicin), 에피루비신(epirubicin), 이다루비신(idarubicin), 픽산트론(pixantrone), 사바루비신(sabarubicin), 발루비신(valrubicin), 파클리탁셀(paclitaxel), 도세탁셀(docetaxel), 메클로에타민(mechloethamine), 클로람부실(chlorambucil), 페닐알라닌(phenylalanine), 무스타드(mustard), 사이클로포스파미드(cyclophosphamide), 이포스파미드(ifosfamide), 카르무스틴(carmustine: BCNU), 로무스틴(lomustine: CCNU), 스트렙토조토신(streptozotocin), 부설판(busulfan), 티오테파(thiotepa), 시스플라틴(cisplatin), 카보플라틴(carboplatin), 닥티노마이신(dactinomycin: actinomycin D), 플리카마이신(plicamycin), 마이토마이신 C(mitomycin C), 빈크리스틴(vincristine), 빈블라스틴(vinblastine), 테니포사이드(teniposide), 토포테칸(topotecan), 이리도테칸(iridotecan), 우라무스틴(uramustine), 멜파란(melphalan), 벤다무스틴(bendamustine), 다카바진(dacarbazine), 테모졸로마이드(temozolomide), 알트레타민(altretamine), 듀오카르마이신(duocarmycin), 네다플라틴(nedaplatin), 옥사리플라틴(oxaliplatin), 사트라플라틴(satraplatin), 트리플라틴 테트라나이트레이트(triplatin tetranitrate), 5-플루오로우라실(5-fluorouracil), 6-머캅토퓨린(6-mercaptopurine), 카페시타빈(capecitabine), 클라드리빈(cladribine), 클로파라빈(clofarabine), 시스타르빈(cystarbine), 플록스유리딘(floxuridine), 플루다라빈(fludarabine), 겜시타빈(gemcitabine), 하이드록시우레아(hydroxyurea), 메토트렉세이트(methotrexate), 페메트렉세드(pemetrexed), 펜토스타틴(pentostatin), 티오구아닌(thioguanine), 에토포사이드(etoposide), 미토산트론(mitoxantrone), 이자베필론(izabepilone), 빈데신(vindesine), 비노렐빈(vinorelbine), 에스트라머스틴(estramustine), 메이탄신(maytansine), DM1(mertansine, 메르탄신), DM4, 돌라스타틴(dolastatin), 아우리스타틴 E(auristatin E), 아우리스타틴 F(auristatin F), 모노메틸 아우리스타틴 E(monomethyl auristatin E, MMAE) 및 모노메틸 아우리스타틴 F(monomethyl auristatin F)로 이루어진 군에선 선택된 어느 하나 이상인 것을 특징으로 하는 항체 약물 접합체.
  7. 제1항에 있어서, 상기 항체 또는 이의 항원 결합 단편은 링커를 통하여 약물과 결합되는 것을 특징으로 하는 항체 약물 접합체.
  8. 제7항에 있어서, 상기 링커는 6-말레이미도카프로일(MC), 말레이미도프로파노일(MP), 발린-시트룰린(val-cit), 알라닌-페닐알라닌(ala-phe), p-아미노벤질옥시카르보닐(PAB), N-숙신이미딜 4-(2-피리딜티오)펜타노에이트(SPP), N-숙신이미딜 4-(N-말레이미도메틸)시클로헥산-1 카르복실레이트(SMCC), 발린-시트룰린-p-아미노벤질옥시카르보닐(val-cit-PAB) 또는 N-숙신이미딜(4-요오도-아세틸)아미노벤조에이트(SIAB)인 것을 특징으로 하는 항체 약물 접합체.
  9. 제1항 내지 제8항 중 어느 한 항의 항체 약물 접합체를 유효성분으로 포함하는 Nectin-2가 과발현되는 암 예방 또는 치료용 약학조성물.
  10. 제10항에 있어서, 상기 암은 유방암, 난소암, 췌장암, 폐암, 전립선암, 식도암, 담낭암 또는 급성골수성백혈병인 것을 특징으로 하는 암 예방 또는 치료용 약학조성물.
