Abstract

善玉菌への影響が少ないと考えられる方法で、フソバクテリウム(Fusobacterium)属菌を制御することを課題とする。消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、消化管内、特に口腔や腸内菌叢におけるFusobacterium属に属する菌の制御用の組成物を提供する。

Description

Fusobacterium属菌の制御用組成物及びその利用
 本発明は、Fusobacterium属菌の制御用組成物及びその利用に関する。
 フソバクテリウム(Fusobacterium)属菌は、ヒトの口腔や腸内に存在する細菌であり、歯周病の原因菌であることが知られている。またFusobacterium nucleatumは、日和見感染症の原因となることが分かっており、最近では大腸がんへの関与が指摘されている(非特許文献1等)。さらにF. nucleatumは虫垂炎などの消化器感染症から分離され(非特許文献2)、最近では炎症性腸疾患とも関連していることが示唆されている(非特許文献3、4、5)。口腔内におけるFusobacterium nucleatumは、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、アルツハイマー型認知症の原因となることが知られている(非特許文献6、7、8、9、10)。
 一方、フラボノイドの一種であるフラバノン類に分類されるヘスペリジンには、いくつかの作用が知られている。例えば特許文献1には、ヘスペリジン配糖体を含有する、癌抑制剤が記載されている。また特許文献2には、少なくとも1つの抗炎症剤と、少なくとも1つの高甘味度甘味料と、少なくとも1つの甘味改善組成物と、を含む機能性甘味料組成物が記載されており、抗炎症剤の例の一つとして、ヘスペリジンが記載されている。さらに特許文献3には、柑橘類抽出物又はフラバノン若しくはその配糖体(第二成分)を含む、緑茶抽出物又はカテキン(第一成分)の抗がん作用、抗筋萎縮作用、抗肥満作用、コレステロール低下作用、血栓予防作用及び免疫増強作用からなる群から選ばれる少なくとも1つの作用の増強用食品組成物が記載されており、第二成分として、エリオジクチオール、ナリンゲニン及びヘスペレチン、並びにこれらの配糖体からなる群から選ばれる少なくとも1種が記載されている。非特許文献11には、ヘスペリジン、ヘスペレチン、ナリゲニンの、腸のバリア機能及び炎症に対する効果が記載されている。
 また、非特許文献12には、歯周炎の原因菌の一つであるF. nucleatum に対する、ヘスペリジン、ヘスペレチン、ナリンゲニンの最小発育阻止濃度と最小殺菌濃度の実測値が示されている。特許文献4には、(A)クエン酸及びその塩からなる群より選択される少なくとも1種と、(B)ヘスペリジン、ノミリン及びリモニンからなる群より選択される少なくとも1種と、を含有する、口臭抑制用組成物が記載され、クエン酸、ヘスペリジン及びその他の成分を含むレモン果汁の添加によって、水の添加と比較して唾液中のF. nucleatumの菌数が減少したとの実験結果が記載されている。特許文献5には、ポリフェノール及びポリフェノール酸化酵素を有効成分として含む、腸内細菌叢改善剤が記載されており、実施例として、コーヒー抽出物(ポリフェノール含有植物抽出物)とゴボウ凍結乾燥粉末(ポリフェノール酸化酵素含有植物凍結乾燥物)を含む試料Aを摂取した場合に、糞便中のフソバクテリウム属細菌の占有率が低減したとの実験結果が示されている。
 最近では、がんにおけるヘスペリジンの標的を特定するための研究がおこなわれており(非特許文献13)、心血管疾患リスクファクターに対するヘスペリジンの効果や、ヘスペリジンのバイオアベイラビリティに対する腸内細菌叢の役割等が報告されている(非特許文献14)。

 消化管内、特に口腔内や腸内では、Fusobacterium属菌と他の細菌との複雑な共生系が保たれているために、Fusobacterium属菌を制御することが難しいと考えられる。例えば、殺菌剤や抗生物質の投与はFusobacterium属細菌だけでなく、善玉菌の菌数も低減させると考えられる。したがって、善玉菌に影響を及ぼさないプロバイオティクスやプレバイオティクスによる制御が望まれている。
 Fusobacterium属菌を制御可能な有効な素材は見つかっていない。
 本発明は、以下を提供する。
[1] 消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、消化管内におけるフソバクテリウム(Fusobacterium)属に属する菌の制御用の組成物。
(式中、R1~R4は、以下の組み合わせである。)。
[2] グルコシダーゼによる加水分解により式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、フソバクテリウム(Fusobacterium)属に属する菌の制御用の組成物。
(式中、R1~R4は、以下の組み合わせである。)。
[3] R1~R4が、以下の組み合わせである、1又は2に記載の組成物。
[4] 式Iで表される化合物が、ヘスペレチンであり、ヘスペレチンを遊離するフラボノイドが、ヘスペリジンであり、ヘスペリジン、ヘスペリジンの糖転移体、及びヘスペレチンからなる群より選択されるいずれかを含む、1から3のいずれか1項に記載の組成物。
[5] ヘスペリジンの糖転移体が、グルコシルヘスペリジンである、1から4のいずれか1項に記載の組成物。
[6] ヘスペレチンを含む、Fusobacterium属に属する菌の制御用の組成物。
[7] Fusobacterium属に属する菌の制御が、Fusobacterium属に属する菌の菌数の低減である、1から6のいずれか1項に記載の組成物。
[8] プレバイオティクス、及びシンバイオティクスからなる群より選択されるいずれかとして使用するための、1から7のいずれか1項に記載の組成物。
[9] Fusobacterium属に属する菌の制御を介した、炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置用の、1から8のいずれか1項に記載の組成物、又は腸管内及び糞便の少なくとも一方にFusobacterium属菌が検出された対象における、炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置用の、請求項1から8のいずれか1項に記載の組成物。
[10] 炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置用の、1から8のいずれか1項に記載の組成物。
[11] 消化管が口腔である、1から9のいずれか1項に記載の組成物、又はヘスペレチン、及びヘスペリジンの糖転移体からなる群より選択されるいずれかを有効成分として含み、消化管が口腔である、1に記載の組成物。
[12] 消化管が腸である、1から9のいずれか1項に記載の組成物。
[13] 1に定義した、消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれか(ただし、糖転移ヘスペリジンを除く。)を含む、機能性表示食品、特定保健用食品、発酵乳、乳飲料、及び粉末食品から選択されるいずれかである、食品組成物。
[14] 1に定義した、消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれか、及び医薬として許容される添加物を含む、医薬組成物。
[15] 1に定義した、消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、腸溶性製剤。
[16] 前記フラボノイド及びその糖転移体、並びに前記化合物から選択されるいずれかを、0.01 g以上7 g以下で含有する、13若しくは14に記載の組成物、又は15に記載の製剤。
[21] 以下の工程を含む、対象の腸内のFusobacterium属に属する菌の制御方法、又は対象における炎症性腸疾患、及び大腸がんの少なくとも一方のリスクの低減方法:
 対象の腸内のFusobacterium属に属する菌の有無又は量に基づき、対象に、腸内で上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)を遊離するフラボノイド及びその糖転移体、並びに上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む、組成物又は食品素材を摂取させる工程。
[22] 以下の工程を含む、対象に組成物の摂取を提案する方法、対象の食生活をサポートする方法、又は対象の健康をサポートする方法:
 対象の腸内のFusobacterium属に属する菌の有無又は量に基づき、腸内で上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)を遊離するフラボノイド及びその糖転移体、並びに上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む、組成物又は食品素材の摂取を提案する工程。
[23] 以下の工程を含む、対象の口腔内のFusobacterium属に属する菌の制御方法、又は対象における歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかのリスクの低減方法:
 対象の口腔内のFusobacterium属に属する菌の有無又は量に基づき、対象に、口腔内で上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)を遊離するフラボノイド及びその糖転移体、並びに上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む、組成物又は食品素材を摂取又は含漱させる工程。
[24] 以下の工程を含む、対象に組成物の摂取又は含漱を提案する方法、対象の食生活をサポートする方法、又は対象の健康をサポートする方法:
 対象の口腔内のFusobacterium属に属する菌の有無又は量に基づき、口腔内で上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)を遊離するフラボノイド及びその糖転移体、並びに上で定義した式Iであらわされる化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む、組成物又は食品素材の摂取を提案する工程。
[31] 消化管内でFusobacterium属に属する菌を制御する方法において使用するための、消化管内で上で定義した式Iで表される化合物を遊離するフラボノイド(好ましくはヘスペレチン)及びその糖転移体、並びに上で定義した式Iで表される化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む、組成物。消化管内で上で定義した式Iで表される化合物を遊離するフラボノイド(好ましくはヘスペレチン)及びその糖転移体、並びに上で定義した式Iで表される化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかの、消化管内でのFusobacterium属に属する菌の制御のための組成物の製造における、使用。消化管内で上で定義した式Iで表される化合物を遊離するフラボノイド(好ましくはヘスペレチン)及びその糖転移体、並びに上で定義した式Iで表される化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む組成物を対象に投与する工程を含む、消化管内でFusobacterium属に属する菌を制御するための、方法又は非治療的方法。消化管内でFusobacterium属に属する菌を制御するための、消化管内で上で定義した式Iで表される化合物を遊離するフラボノイド(好ましくはヘスペレチン)及びその糖転移体、並びに上で定義した式Iで表される化合物(好ましくはヘスペレチン)からなる群より選択されるいずれかを含む組成物の、使用又は非治療的使用。

[39] 組成物が、Fusobacterium属に属する菌の制御を介した、炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置のためのものである、31から38のいずれか1項に記載の組成物、製造における使用、方法又は非治療的方法、又は使用又は非治療的使用、又は組成物が、腸管内及び糞便の少なくとも一方にFusobacterium属菌が検出された対象における、炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置のためのものである、31から38のいずれか1項に記載の組成物、製造における使用、方法又は非治療的方法、又は使用又は非治療的使用。
[40] 炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置のためのものである、31から38のいずれか1項に記載の組成物、製造における使用、方法又は非治療的方法、又は使用又は非治療的使用。
[41] 消化管が口腔である、31~40のいずれか1項に記載の組成物、製造における使用、方法又は非治療的方法、又は使用又は非治療的使用、又は組成物がヘスペレチン、及びヘスペリジンの糖転移体からなる群より選択されるいずれかを有効成分として含み、消化管が口腔である、31~40のいずれか1項に記載の組成物、製造における使用、方法又は非治療的方法、又は使用又は非治療的使用。
[42] 消化管が腸である、31~40のいずれか1項に記載の組成物、製造における使用、方法又は非治療的方法、又は使用又は非治療的使用。
 消化管で式Iで表される化合物を遊離するフラボノイド及びその糖転移体 、並びに式Iで表される化合物からなる群より選択されるいずれかを用いることで、消化管内菌叢におけるFusobacterium属に属する菌を制御する手段を提供することができる。
 腸内菌叢におけるFusobacterium属に属する菌を制御することで、大腸がんや炎症性腸疾患の処置が期待できる。口腔内菌叢における、Fusobacterium属に属する菌を制御することで、歯周病、口臭、食道がん、誤嚥性肺炎、アルツハイマー型認知症の処置が期待できる。

[有効成分]

 本発明は、消化管内、好ましくは口腔内又は腸内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを有効成分として含む、Fusobacterium属菌の制御用の組成物に関する。また本発明は、グルコシダーゼによる加水分解により式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、フソバクテリウム(Fusobacterium)属に属する菌の制御用の、より特定すると、グルコシダーゼの存在下でのFusobacterium属に属する菌の制御用の、組成物に関する。いずれかとは、数も種類も任意であることをいう。組成物は、有効成分を1種のみ含んでいてもよく、複数種類含んでいてもよい。有効成分として含むとは、その組成物において、目的の機能を発揮するための有効量で用いられていること、目的に資する成分であることが表示により特定されて用いられていること等を指す。有効成分は、機能性表示食品においては、機能性関与成分(特定の保健の目的(疾病リスクの低減に係るものを除く。)に資する成分をいう。)ということがある。
(式Iで表される化合物、それを遊離するフラボノイド)

 一態様では、組成物は、消化管内、好ましくは口腔内又は腸内で式Iで表される化合物を遊離するフラボノイド、及び式Iで表される化合物からなる群より選択されるいずれかを、有効成分として含む。フラボノイドは、植物によって合成されるポリフェノール化合物の総称である。フラボノイドは、配糖体である場合がある。式Iで表される化合物は、アグリコンである場合がある。
 式中、R1~R4は、以下の組み合わせである。
 消化管内で式Iで表される化合物を遊離するとは、消化管内の細菌が産生する酵素や、消化管内に分泌される酵素の働きにより、式Iで表される化合物を生じることをいう。酵素は、酸化還元酵素、転移酵素、加水分解酵素、リアーゼ、異性化酵素、リガーゼ、又はABC輸送体であり得る。
 一態様では、R1~R4は、以下の組み合わせである。すなわち式Iで表される化合物は、ヘスペレチン、ナリンゲニン、エリオジクチオールであり得る。
 特に好ましい態様では、式Iで表される化合物はヘスペレチンであり、ヘスペレチンを遊離するフラボノイドはヘスペリジンである。
 ヘスペリジン (Hesperidin、分子式 : C28H34O15、分子量 : 610.56) は、ビタミンPとして知られるポリフェノールであり、ヘスペレチン(Hesperetin、分子式 : C16H14O6、分子量 : 302.28)の配糖体である。水への溶解性が非常に低い(約0.002g/100mL-水)。ヘスペリジンの糖部分はβ-ルチノース(6-O-α-L-ラムノシル-D-β-グルコース)である。
 なお、以下では、本発明及びその実施態様等について、ヘスペレチン及びヘスペリジンを例に説明することがあるが、その説明は、当業者であれば、式Iで表される化合物、及びそれを遊離するフラボノイドに当てはめて理解することができる。
 腸内においては、ヘスペリジンにβ-グルコシダーゼが作用すると、ヘスペレチンが遊離する(非特許文献14)。ヒトだけでなく、多くの腸内細菌がβ-グルコシダーゼを保有しているため、たいていのヒトが腸内にβ-グルコシダーゼを有する菌を保有していると考えられる(Marta Dabek et al. Deglycosylation of flavonoid and isoflavonoid glycosides by human small intestine and liver beta-glucosidase activity. FEMS Microbiol Ecol. 2008 Dec;66(3):487-95. doi: 10.1111/j.1574-6941.2008.00520.x.)。
 口腔内においても、ヘスペリジンに口腔内細菌が産生するβ-グルコシダーゼが作用すると、ヘスペレチンが遊離する。多くの口腔細菌がβ-グルコシダーゼを保有しているため、たいていのヒトが口腔内にβ-グルコシダーゼを有する菌を保有していると考えられる。また、ヒト唾液中のβ-グルコシダーゼ酵素活性が報告されている(Lucimari Teixeira Essenfelder et al. Salivary β-glucosidase as a direct factor influencing the occurrence of halitosis. Biochem Biophys Rep. 2021 Mar 6;26:100965.)。
 各種の式Iで表される化合物を遊離するフラボノイド、及び式Iで表される化合物は、植物から抽出することにより、得ることができる。ヘスペリジン、及びヘスペレチンを得るための植物の好ましい例は、柑橘類であり、より好ましい例は、みかん、温州みかん、柚子、オレンジ、タンカン、ポンカン、スダチ、シークワーサー、及びレモンである。これらの果実、果皮、及び未熟果から得ることが好ましい。組成物は、式Iで表される化合物を遊離するフラボノイド、及び式Iで表される化合物を植物抽出物として含んでもよい。
(糖転移体、モノグルコシルヘスペリジン)

 一態様では、組成物は、消化管内、好ましくは口腔内又は腸内で式Iで表される化合物を遊離するフラボノイドの糖転移体を含む。フラボノイドの糖転移体とは、フラボノイドに単糖が1から20個付加したものをいう。フラボノイドの糖転移体は、通常、元のフラボノイド自体よりも水溶性が高い。
 一態様では、組成物は、消化管内、好ましくは口腔内又は腸内でヘスペレチンを遊離するフラボノイドの糖転移体として、ヘスペリジンの糖転移体(糖転移ヘスペリジンということもある。)を含む。糖転移ヘスペリジンの例は、α-グルコシルヘスペリジン(モノグルコシルヘスペリジンということもある。)、α-ジグルコシルヘスペリジン、α-トリグルコシルヘスペリジン、α-テトラグルコシルヘスペリジン、α-ペンタグルコシルヘスペリジンが挙げられる。
 好ましい態様においては、組成物は、消化管内、好ましくは口腔内又は腸内でヘスペレチンを遊離するフラボノイドの糖転移体として、モノグルコシルへスぺリジンを含む。
 モノグルコシルへスぺリジン(Monoglucosyl hesperidin、 分子量 :772.70)は、ヘスペリジンより水への溶解度が大きい(約197g/100mL-水)。モノグルコシルヘスペリジンは、α-グルコシダーゼ、及びβ-グルコシダーゼによって分解され、ヘスペリジンを生じる。α-グルコシダーゼは、唾液に含まれ、また腸内細菌が分泌する。β-グルコシダーゼは、ヒト、腸内細菌、及び口腔細菌のものが同定されている(A J Day at al. Deglycosylation of flavonoid and isoflavonoid glycosides by human small intestine and liver beta-glucosidase activity. FEBS Lett. 25;436(1):71-5, 2019、Matsumoto S. et al. Glucosyl hesperidin: safety studies. Fundam Toxicol Sci 8:299-317, 2019)。
 フラボノイドの糖転移体は、通常、フラボノイドに酵素の働きにより糖供与体から糖を転移することにより製造される。ヘスペリジン、エリオシトリン、ナリンギン、ネオヘスペリジン、ジオスミン、リナリン、ポンシリン、プルニン、又はヘスペレチン-7-グルコシドの糖転移体、及びその製造方法については、特開平7-107972号公報を参考にすることができる。またヘスペリジン糖転移体、及びその製造方法については、特開平3-7593号公報を参考にすることができる。
[用途]
(機能・作用)

 本発明の組成物は、Fusobacterium属に属する菌(単に、Fusobacterium属菌ということもある。)を制御するために用いることができる。好ましい態様では、組成物は、消化管内、好ましくは口腔又は腸内菌叢でのFusobacterium属菌を制御するために用いることができる。口腔又は腸内菌叢でFusobacterium属菌制御するとは、口腔又は腸内に棲む様々な菌が存在する状態において、Fusobacterium属に属する菌を制御することを指す。菌を制御するとは、菌数を制御(コントロール)すること、Fusobacterium属菌を含む腸菌群を調節すること、Fusobacterium属菌を含む腸菌群を調節することを助けることを指す。好ましい態様では、組成物はFusobacterium属菌の菌数を低減するためのもの、Fusobacterium属菌の菌数を低減することを助けるためのものである。
 口腔は、歯、歯周組織、舌、及び唾液腺を含む。腸とは、ヒトや動物において、細菌が常在し、摂取した食物を消化・吸収する消化管を指す。腸(腸管ということもある。)は、小腸及び大腸を含み、好ましくは大腸である。

 本発明に関し、Fusobacterium属菌というときは、16S rRNA遺伝子に基づく分子系統解析により、Fusobacterium属であると同定される菌をいう。16S rRNA遺伝子に基づく分子系統解析による属の判断基準は当業者にはよく知られている(Stackebrandt E, Ebers J. Taxonomic parameters revisited: tarnished gold standards. Microbiol Today 2006;33:152-155.)。
 ある成分が、Fusobacterium属菌を制御するか否かは、次のように評価することができる。詳細には、評価したい成分を含むGAM寒天培地と含まないGAM寒天培地に適切なFusobacterium(たとえば、Fusobacterium nucleatum JCM JCM8532T)を播種し、適切な条件(たとえば、37 °C、嫌気条件下、48時間)で培養し、コロニー数を測定し、比較する。そして、測定結果を、評価したい成分の代わりに対照(例えば、滅菌水)を加えた点でのみ異なるが他の点では同一の条件で培養した寒天培地についての測定結果と比較し、判断することができる。
 ある成分が、腸内菌叢においてFusobacterium属菌を制御するか否かは、次のように評価することができる。健常人から提供された糞便を適切な培地に添加し、必要に応じ一定期間培養した後、評価したい成分を加え、適切な条件で培養(例えば、腸管内の条件に類似の、37 °C、嫌気条件下、16時間の培養)する。このとき、糞便がFusobacterium属菌を含まない場合は、培養開始時に、入手可能ないずれかのFusobacterium属菌を添加して培養を行う。添加するFusobacterium属菌は、例えばF. nucleatum JCM8532Tである。添加量は、実験の条件に応じて適宜とすることができるが、例えば、0.1~10×104 cfu/mLである。そして、培養物中の菌叢におけるFusobacterium属菌の菌数を測定する。そして、測定結果を、評価したい成分の代わりに対照(例えば、滅菌水)を加えた点でのみ異なるが他の点では同一の条件で培養した培養物についての測定結果と比較し、判断することができる。
  菌叢に含まれるFusobacterium属菌の菌数は、公知の方法により求めることができる。好ましい方法の一つは、培養物から抽出されたDNAについて16Sメタゲノム解析(16S rRNA遺伝子アンプリコンのシーケンス解析)を行うことである。16Sメタゲノム解析を実施する場合、DNAの抽出は市販のキットを用いて行うことができる。解析されるゲノム領域は、細菌を判定できる限り特に制限されないが、16S rRNA遺伝子のV3-V4領域を用いることができる。細菌の解析を行うためのプライマー、増幅条件、アンプリコンの精製等の方法も、当業者にはよく知られた方法を用いることができる。配列の解読は性能がより高い次世代シーケンサーで行うことが好ましい。得られたデータの解析にはQIIME 2TM等の次世代マイクロバイオーム・バイオインフォマティクス・プラットフォームを利用することができる。16Sメタゲノム解析について、当業者はSanschagrin S, Yergeau E. Next-generation sequencing of 16S ribosomal RNA gene amplicons. J Vis Exp. 2014;(90):51709. Published 2014 Aug 29. doi:10.3791/51709等の情報を参照することができる。
 本発明に係るFusobacterium属菌は、ヒトの口腔や腸管内に存在する細菌であり、歯周病の原因菌であることが知られている。またFusobacterium nucleatumは、日和見感染症の原因となることが分かっており、最近では大腸がんへの関与が指摘されている(非特許文献1等)。さらにF. nucleatumは虫垂炎などの消化器感染症から分離され(非特許文献2)、最近では炎症性腸疾患とも関連していることが示唆されている(非特許文献3、4、5)。したがって、一態様では、組成物はFusobacterium属菌の制御を介した炎症性腸疾患、及び大腸がんからなる群より選択されるいずれかの処置のために、用いることができる。
 炎症性腸疾患は、潰瘍性大腸炎、及びクローン病を含む。
 口腔内におけるFusobacterium nucleatumは、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、アルツハイマー型認知症の原因となることが知られている(非特許文献6、7、8、9、10)。したがって、一態様では、組成物は、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、アルツハイマー型認知症に、用いることができる。より特定すると組成物は、Fusobacterium属菌の制御を介した歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、アルツハイマー型認知症に、用いることができる。
 また、組成物は、腸管内におけるFusobacterium属菌の増殖制御により改善される疾患又は状態の処置のために用いることができる。このような疾患又は状態としては、様々なものがあり得る。
 本発明に関し、疾患又は状態について処置というときは、発症リスクの低減、発症の遅延、予防、治療、進行の停止、遅延を含む。また、疾患又は状態からの改善を助ける、疾患又は状態を緩和する、疾患又は状態となるリスクを低減ことを含む。治療は、根治療法(原因を取り除く治療)、及び対症療法(症状を改善する治療)を含む。改善又は処置のための行為には、医師及び医師の指示を受けた看護師、助産師などが行う医療行為と、医師以外の者、例えば薬剤師、栄養士(管理栄養士、スポーツ栄養士を含む)、保健師、助産師、看護師、臨床検査技師、スポーツ指導員、医薬品製造者、医薬品販売者、食品製造者、食品販売者等が行う、非治療的行為を含む。さらに予防又は発症リスクの低減は、特定の食品の摂取の推奨、栄養指導(傷病者に対する療養のため必要な栄養の指導、及び健康の保持増進のための栄養の指導を含む)を含む。
(対象)

 本発明の組成物は、消化管内、好ましくは口腔内又は腸内菌叢におけるFusobacterium属菌を制御する、特に、菌数を低減することが望ましい又は必要な対象;炎症性腸疾患、又は大腸がんからなる群より選択されるいずれかである対象;並びに腸管内におけるFusobacterium属菌の増殖を制御することにより改善される疾患又は状態である対象;歯周病、又は口臭からなる群より選択されるいずれかである対象;並びに口腔内におけるFusobacterium属菌の増殖を制御することにより改善される疾患又は状態である対象に、投与するのに適している。なお本発明に関し、投与は、医薬品を対象に投与する場合のほか、医薬品以外の食品等を対象に摂取させる意味でも用いられる。投与(する)は、摂取)(させる)と読み替えることができ、また投摂(させる)は投与(する)と読み替えることができる。
 腸管内におけるFusobacterium属菌を制御することが望ましい又は必要な対象は、腸管内及び糞便の少なくとも一方にFusobacterium属菌が検出された対象、腸管内及び糞便の少なくとも一方にFusobacterium属菌の菌数が比較多く検出された対象を含む。また、腸管内におけるFusobacterium属菌を制御することが望ましい又は必要な対象は、腸内菌叢(フローラ)検査、糞便中の物質検査、及び糞便メタボローム解析等の任意の検査・解析により、Fusobacterium属菌の数が比較多いと認められた対象、Fusobacterium属菌が検出された対象、炎症性腸疾患リスクが高いと認められた対象、大腸がんリスクが高いと認められた対象、検査・解析結果に基づき医師、看護師、薬剤師、栄養士等、又はコンピュータプログラムによる判断により、Fusobacterium属菌を制御可能な飲食品や医薬の摂取を提案された対象を含む。
 口腔内におけるFusobacterium属菌を制御することが望ましい又は必要な対象は、口腔内(たとえば、唾液、舌苔、及び歯垢から選択されるいずれか)にFusobacterium属菌が検出された者、唾液、舌苔、及び歯垢から選択されるいずれかにFusobacterium属菌の菌数が比較的多く検出された者を含む。また、口腔内におけるFusobacterium属菌を制御することが望ましい又は必要な対象は、口腔菌叢(フローラ)検査、唾液、舌苔、及び歯垢から選択されるいずれかのサンプル中に含まれる物質検査、及び唾液、舌苔、及び歯垢から選択されるいずれかのサンプルにおけるメタボローム解析等の任意の検査・解析により、Fusobacterium属菌の数が比較多いと認められた対象、Fusobacterium属菌が検出された対象、歯周病リスクが高いと認められた対象、口臭発生リスクが高いと認められた対象、口臭に悩んでいる者、検査・解析結果に基づき医師、看護師、薬剤師、栄養士等、又はコンピュータプログラムによる判断により、Fusobacterium属菌を制御可能な飲食品や医薬の摂取を提案された対象を含む。また、腸内及び糞便の少なくとも一方のFusobacterium属菌の数を減らしたい対象、大腸がんリスクを減らしたい対象、及び炎症性腸疾患リスクを減らしたい対象を含む。一態様では、対象は、腸内及び糞便の少なくとも一方に、Fusobacterium属菌が検出された対象、又はFusobacterium属菌の数が比較多いと認められた対象である。腸内及び糞便の少なくとも一方にFusobacterium属菌の数が検出された、又は比較多いとは、例えば対象から採取した便を0.1%(w/v)の濃度で含む培養液を37℃、嫌気条件下で16時間培養したときに、培養液中のFusobacterium属菌の菌数が、1×103cells/mL以上、好ましくは1×104cells/mL以上、より好ましくは1×104cells/mL以上、さらに好ましくは5×105cells/mL以上である場合を指す。あるいは対象から採取した糞便中のFusobacterium属菌の菌数が、1×104cells/g以上、好ましくは5×104cells/g以上、より好ましくは1×105cells/g以上、さらに好ましくは5×105cells/g以上である場合を指す。菌数の測定は、例えば定量的PCRで行う。
 一態様では、対象は、健常(医師により病気とは診断されていない)である。対象は、ヒトであってもよく、ヒト以外の動物であってもよい。
 対象の年齢は特に制限されず、対象は、例えば、新生児(生後28日以内);乳児(生後1年未満);幼児(生後1~6年);小児(7 歳以上、15 歳未満);成人(15歳以上);65歳以上の者であり得る。好ましい態様では、対象は。比較的高齢の者、例えば60歳以上の者、65歳以上の者、70歳以上の者、75歳以上の者である。腸管内のFusobacterium属菌は、加齢と共に増えることが多く報告されている(例えば、Odamaki et al. BMC Microbiology (2016) 16:90
DOI 10.1186/s12866-016-0708-5)。また、口腔内におけるFusobacterium属菌を含有する唾液が気管を通じて肺に到達することで、肺に感染し、誤嚥性肺炎を引き起こすことが明らかとなっている。
[組成物]
(食品組成物等)

 本発明の組成物は、食品組成物又は医薬組成物とすることができる。食品及び医薬品は、特に記載した場合を除き、ヒトのためのもののみならず、ヒト以外の動物のためのものを含む。食品は、特に記載した場合を除き、一般食品、機能性食品、栄養組成物を含み、また治療食(治療の目的を果たすもの。医師が食事箋を出し、それに従い栄養士等が作成した献立に基づいて調理されたもの。)、食事療法食、成分調整食、介護食、治療支援用食品を含む。食品は、特に記載した場合を除き、固形物のみならず、液状のもの、例えば飲料、ドリンク剤、流動食、及びスープを含む。機能性食品とは、生体に所定の機能性を付与できる食品をいい、例えば、特定保健用食品(条件付きトクホ[特定保健用食品]を含む)、機能性表示食品、栄養機能食品を含む保健機能食品、特別用途食品、栄養補助食品、健康補助食品、サプリメント(例えば、錠剤、被覆錠、糖衣錠、腸溶性コーティング等による腸溶処理剤、腸溶性製剤、腸溶性製剤カプセル((腸溶性ソフトカプセル、腸溶性ハードカプセル、大腸デリバリーカプセル等))、液剤等の各種の剤型のもの)、ダイエタリーサプリメント、フードサプリメント、メディカルフード(アメリカ食品医薬品局(Food and Drug Administration: FDA)の定義に従う)、美容食品(例えば、ダイエット食品)等の、健康食品の全般を包含している。また、本発明において「機能性食品」とは、コーデックス(FAO/WHO合同食品規格委員会)の食品規格に基づく健康強調表示(Health claim)が適用される健康食品を包含している。フードサプリメントとは、通常の食事を補充し、栄養学的又は生理学的な効果を持つ栄養素又はその他の物質を単独又は組み合わせて濃縮したものであり、製品に Food Supplementと記載されているものである。ダイエタリーサプリメントとは、食事を補充することを目的とした製品(タバコを除く)で、対象とされている成分を 1 種類以上含み、製品に Dietary supplement と記載されているものである。
 組成物は、歯科用、又は口腔用のものであってもよい。歯科用、又は口腔用の組成物は、医薬組成物であってもよく、医薬部外品等の医薬品以外のものであってもよい。
 一態様では、食品組成物に代えて、食品素材を用いることができる。本発明に関し、食品素材とは、加工していない植物の全部又は一部をいう。食品素材の好ましい例は、柑橘類(ミカン科ミカン亜科ミカン連(カンキツ連))、より好ましくは、みかん、温州みかん、オレンジ、タンカン、ポンカン、スダチ、シークワーサー、レモン、カボス、キンカン等の、果実、果皮、及び未熟果である。
(投与経路等)

  本発明の組成物は、経口的に投与してもよく、非経口的に、例えば経管的(胃瘻、腸瘻)に投与してもよいし、経鼻的に投与してもよいが、経口的に投与することが好ましい。
 組成物は、繰り返し対象に投与することができ、また長期間にわたり継続して対象に投与することができる。期間は特に限定されないが、効果が十分に認められるためには、比較的長い期間、例えば3日以上、1週間以上、2週間以上、1カ月以上、3カ月以上、6カ月以上、1年以上で継続して投与するとよい。
 組成物は、日常的に投与してもよく、リスクが高いときなどに前もって投与してもよく、又は必要が生じた場合に投与してもよい。組成物は、食事として投与してもよく、食前、食後、食間に投与してもよく、組成物により改善したい疾患又は状態が生じたとき、アプリや電子機器による通知を受けたときに投与してもよい。
(用量、含有量)
 本発明の組成物の投与量は、目的の効果が発揮される量であればよい。投与量は、対象の年齢、体重、症状等の種々の要因を考慮して、適宜設定することができる。
 組成物の1日あたりの投与量は、ヘスペレチンの量として(ヘスペレチン以外の有効成分を用いる場合は、遊離するヘスペレチンの量として。また、複数の有効成分を含む場合は遊離するヘスペレチンの合計量として)、0.01 g以上となるようにすることができ、0.05 g以上となるようにすることができ、0.1 g以上となるようにすることができ、0.15 g以上とすることが好ましく、0.2 g以上とすることがより好ましく、0.4 g以上とすることがさらに好ましく、0.8 g以上とすることがさらに好ましく、1 g以上とすることがさらに好ましい。ポリフェノール摂取による菌叢変化の試験では100mg/day程度で変化がみられている(Melanie Le Sayec et al. Clinical Nutrition 41 (2022) 2549e2561. https://doi.org/10.1016/j.clnu.2022.08.024)ことから、このような量であれば、目的の効果が得られると考えられるからである。一日あたりの有効成分の上限値は、下限値がいずれの場合であっても、10g以下とすることができ、7 g以下とすることができ、5 g以下とすることができ、4 g以下としてもよく、3 g以下としてもよく、2 g以下としてもよい。
 投与は、一日1回でもよく、1日複数回、例えば2~10回に分けてもよい。1回あたりの有効成分の投与量は、0.003 g以上となるようにすることができ、0.015 g以上となるようにすることができ、0.01 g以上となるようにすることができ、0.015 g以上とすることが好ましく、0.02 g以上とすることがより好ましく、0.04 g以上とすることがさらに好ましく、0.08 g以上とすることがさらに好ましく、0.1 g以上とすることがさらに好ましい。一回あたりの有効成分の上限値は、下限値がいずれの場合であっても1 g以下とすることができ、0.7 g以下とすることができ、0.5 g以下とすることができ、0.4 g以下としてもよく、0.3 g以下としてもよく、0.2 g以下としてもよい。
 組成物中の有効成分の含有量は、組成物の形態に応じて適宜とすることができる。例えば、組成物の固体分あたりの有効成分の含有量は、5 %以上とすることができ、10 %以上とすることが好ましく、20 %以上とすることがより好ましく、25 %以上とすることがさらに好ましい。固体分あたりの有効成分の上限値は、下限値がいずれの場合であっても、90 %以下とすることができ、80 %以下とすることができ、70 %以下としてもよく、60 %以下としてもよく、50 %以下としてもよい。なお本発明に関し、%は、特に記載した場合を除き、質量%の意である。
 組成物が液体である場合、有効成分の含有量は、0.01 %以上とすることができ、0.03 %以上とすることが好ましく、0.06 %以上とすることがより好ましく、0.1 %以上とすることがさらに好ましい。液体である場合の有効成分の含有量の上限値は、下限値がいずれの場合であっても、10 %以下とすることができ、8 %以下とすることができ、6 %以下としてもよく、4 %以下としてもよく、3 %以下としてもよい。
(他の成分、添加剤)

 本発明に関し、組成物は、食品又は医薬品として許容可能な他の有効成分や栄養成分を含んでいてもよい。そのような成分の例は、脂質(例えば、乳脂肪、植物油脂、中鎖脂肪酸含有油脂)、たんぱく質(例えば、乳たんぱく質、乳たんぱく質濃縮物(MPC)、乳清たんぱく質濃縮物(WPC)、乳清たんぱく質単離物(WPI)、α-ラクトアルブミン(α-La)、β-ラクトグロブリン(β-Lg)、熱変性ホエイたんぱく質及び酵素処理ホエイたんぱく質)、アミノ酸類(例えば、リジン、アルギニン、グリシン、アラニン、グルタミン酸、ロイシン、イソロイシン、バリン)、糖質(グルコース、ショ糖、果糖、麦芽糖、トレハロース、エリスリトール、マルチトール、パラチノース、キシリトール、デキストリン)、電解質(例えば、ナトリウム、カリウム、カルシウム、マグネシウム)、ビタミン(例えば、ビタミンA、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンB12、ビタミンC、ビタミンD、ビタミンE、ビタミンK、ビオチン、葉酸、パントテン酸及びニコチン酸類)、ミネラル(例えば、銅、亜鉛、鉄、コバルト、マンガン)、抗生物質、食物繊維等である。
 本発明に関し、組成物は有効成分以外のプレバイオティクスを含んでいてもよい。プレバイオティクスとは、大腸管内の特定の細菌の増殖及び活性を選択的に変化させることより、宿主に有利な影響を与え、宿主の健康を改善する難消化性食品成分ということができる。組成物には、有効成分以外のプレバイオティクスとして、一種を用いてもよく、二種以上を用いてもよい。
  有効成分以外のプレバイオティクスは、組成物に含まれる有効成分の効果を妨げない限り、特に制限されない。有効成分以外のプレバイオティクスの例として、ガラクトオリゴ糖、フラクトオリゴ糖、キシロオリゴ糖、イソマルトオリゴ糖、ラフィノース、ラクチュロース、ラクトスクロース、大豆オリゴ糖、コーヒーオリゴ糖、食物繊維、グルコン酸が挙げられる。
  また組成物は、食品又は医薬品として許容される添加物をさらに含んでいてもよい。そのような添加物の例は、不活性担体(固体や液体担体)、賦形剤、界面活性剤、結合剤、崩壊剤、滑沢剤、溶解補助剤、懸濁化剤、コーティング剤、着色剤、保存剤、緩衝剤、pH調整剤、乳化剤、安定剤、甘味料、酸化防止剤、香料、酸味料、天然物である。より具体的には、水、他の水性溶媒、製薬上で許容される有機溶媒、コラーゲン、ポリビニルアルコール、ポリビニルピロリドン、カルボキシビニルポリマー、アルギン酸ナトリウム、水溶性デキストラン、水溶性デキストリン、カルボキシメチルスターチナトリウム、ペクチン、キサンタンガム、アラビアゴム、カゼイン、ゼラチン、寒天、グリセリン、プロピレングリコール、ポリエチレングリコール、ワセリン、パラフィン、ステアリルアルコール、ステアリン酸、ヒト血清アルブミン、マンニトール、ソルビトール、ラクトース、スクラロース、ステビア、アスパルテーム、アセスルファムカリウム、クエン酸、乳酸、りんご酸、酒石酸、リン酸、酢酸、果汁、野菜汁等である。
(剤型・形態)

  食品組成物は、固体、液体、混合物、懸濁液、粉末、顆粒、ペースト、ゼリー、ゲル、カプセル等の任意の形態に調製されたものであってよい。また、本発明に係る食品組成物は、乳製品、サプリメント、菓子、飲料、ドリンク剤、調味料、加工食品、惣菜、スープ等の任意の形態にすることができる。より具体的には、本発明の組成物は、流動食、半流動食、ゼリー、ゲル、粉末、調製粉乳、調製液状乳、妊産婦・授乳婦用粉乳・液状乳、発酵乳、バー、ムース、チョコレート、ビスケット、アイスクリーム、発酵乳、乳酸菌飲料、乳性飲料、乳飲料、清涼飲料、タブレット、チーズ、パン、ビスケット、クラッカー、ピッツァクラスト、病者用食品、栄養食品、冷凍食品、加工食品等の形態とすることができ、また飲料や食品に混合して投与するための、顆粒、粉末、ペースト、濃厚液等の形態とすることができる。顆粒、粉末は、キューブ状、又はスティック状(1回分の量を分包したもの)とすることができる。本発明に関し、調製粉乳とは、「乳及び乳製品の成分規格等に関する省令(以下「乳等省令」と略する。)」で定義されているように、生乳、牛乳、特別牛乳若しくは生水牛乳又はこれらを原料として製造した食品を加工し、又は主要原料とし、これに乳幼児に必要な栄養素を加え粉末状にしたものをいう。本発明に関し、調製液状乳とは、乳等省令で定義されているように、生乳、牛乳、特別牛乳若しくは生水牛乳又はこれらを原料として製造した食品を加工し、又は主要原料とし、これに乳幼児に必要な栄養素を加え液状にしたものをいう。
  調製粉乳又は調製液状乳(これらをまとめて調製粉乳類ということがある。また調製粉乳類の説明において、調製粉乳を例に説明することがあるが、その説明は、調製液状乳にも当てはまる。)は、乳児用、フォローアップ用(離乳食の補完用)、低出生体重児用、小児用、成人用、妊産婦用、授乳婦用、高齢者用、アレルギーを有する者用、乳糖不耐症の者用、先天代謝異常の者用とすることができる。調製粉乳類の好ましい例は、乳児用、フォローアップ用、低出生体重児用、小児用のものである。
  腸溶性製剤、及び医薬組成物は、経口投与に適した、錠剤、顆粒剤、散剤、丸剤、カプセル剤等の固形製剤、液剤、懸濁剤、シロップ剤等の液体製剤、ジェル剤、エアロゾル剤等の任意の剤型にすることができる。
 組成物は、口腔内におけるFusobacterium属に属する菌の制御用のものである場合、口腔内に比較的長時間含まれる形態が好ましい。このような形態の例は、含嗽剤、キャンデー、グミ、ガム、トローチ剤、舌下剤、口腔用貼付剤、軟膏剤、パスタ剤、ゼリー、口腔用錠剤を含む。口腔用のものである場合、歯磨用組成物、洗口用組成物であってもよい。
(その他)

 本発明の組成物の製造において、有効成分の配合の段階は、適宜選択することができる。有効成分の特性を著しく損なわない限り配合の段階は特に制限されない。例えば、有効成分を原材料に混合して配合することができる。あるいは、有効成分を製造の最終段階で添加し、有効成分を含む組成物を製造することができる。
 本発明の組成物には、使用目的(用途)を表示することができ、また特定の対象に対して投与を薦める旨を表示することができる。
 本発明の組成物には、使用目的(用途)を表示することができ、一態様では、特定の対象に対して投与を薦める旨を表示する。表示する対象の例は、上で対象について説明したとおりである。また、本発明の組成物には、一態様では、組成物又は有効成分による機能や機能に基づく用い方を表示する。例えば、腸管内におけるFusobacterium属菌の増殖制御ができること、腸管内におけるFusobacterium属菌の増殖抑制ができること、腸管内におけるFusobacterium属菌の増殖制御(好ましくは増殖抑制)により改善される疾患又は状態を処置することができること、プレバイオティクスとして用い得ること、シンバイオティクス(プロバイオティクスとプレバイオティクスを組み合わせたもの)として用い得ること、又は炎症性腸疾患、大腸がんを処置できる旨を表示することができる。なお、それぞれの文言の先頭に、「一時的な」、「長期的な」といった期間を適宜表示してもよい。
 表示は、明示的に又は非明示的にすることができ、明示的な表示の例は、製品自体、パッケージ、容器、ラベル、タグ等の有体物への直接的な記載であり、非明示的な表示(黙示ということもできる。)の例は、ウェブサイト、店頭、パンフレット、展示会、メディアセミナー等のセミナー、書籍、新聞、雑誌、テレビ、ラジオ、郵送物、電子メール、音声等の、場所又は手段による、広告・宣伝活動を含む。
 一態様では、組成物の摂取を薦める旨の表示は、パーソナルに行う。このような表示は、対象あての文書(書面であるか、電子的であるかを問わない。)、対象のタブレット端末、スマートフォン、及びパーソナルコンピュータ、対象のSNS等を利用して行うことができる。また、このような表示は、対象についての腸内菌叢(フローラ)検査、糞便中の物質検査、及び糞便メタボローム解析、口腔菌叢(フローラ)検査、口腔中(たとえば、唾液、舌苔、歯垢中)の物質検査、並びに唾液、舌苔、及び歯垢中から選択されるいずれかのメタボローム解析等の任意の検査・解析の結果の表示と共に行うことができる。
 一態様では、対象の腸管内のFusobacterium属に属する菌の有無又は量に基づき、対象に、腸内でヘスペレチンを遊離するフラボノイド及びその糖転移体、並びにヘスペレチンからなる群より選択されるいずれかを含む、組成物又は食品素材を摂取させる工程を含む、対象の腸管内のFusobacterium属に属する菌の制御方法、又は対象における炎症性腸疾患、及び大腸がんの少なくとも一方のリスクの低減方法が提供される。また一態様では、対象の腸管内のFusobacterium属に属する菌の有無又は量に基づき、腸内でヘスペレチンを遊離するフラボノイド及びその糖転移体、並びにヘスペレチンからなる群より選択されるいずれかを含む、組成物又は食品素材の摂取を提案する工程を含む、対象に組成物の摂取を提案する方法、対象の食生活をサポートする方法、又は対象の健康をサポートする方法が提供される。これらの方法は、他の工程、例えば以下のいずれかを含んでいてもよい:
 対象者から糞便試料を採取する工程、又は対象者に試料を採取させる工程;試料中のFusobacterium属菌の有無又は量を分析する工程;分析結果を表示する工程;分析結果に基づき、対象がFusobacterium属菌数を低減することが望ましい又は必要か否かを判定する工程;判定結果を表示する工程;腸内でヘスペレチンを遊離するフラボノイド及びその糖転移体、並びにヘスペレチンからなる群より選択されるいずれかを含む、組成物又は食品素材である製品を表示する工程。
 一態様では、対象の口腔内のFusobacterium属に属する菌の有無又は量に基づき、対象に、口腔内でヘスペレチンを遊離するフラボノイド及びその糖転移体、並びにヘスペレチンからなる群より選択されるいずれかを含む、組成物又は食品素材を摂取させる工程、又は組成物により含漱させる工程を含む、対象の口腔内のFusobacterium属に属する菌の制御方法、又は対象における歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかのリスクの低減方法が提供される。また一態様では、対象の口腔内のFusobacterium属に属する菌の有無又は量に基づき、口腔内でヘスペレチンを遊離するフラボノイド及びその糖転移体、並びにヘスペレチンからなる群より選択されるいずれかを含む、組成物又は食品素材の摂取を提案する工程、又は組成物による含漱を提案する工程を含む、対象に組成物の摂取を提案する方法、対象に組成物による含漱を提案する方法、対象の食生活をサポートする方法、又は対象の健康をサポートする方法が提供される。これらの方法は、他の工程、例えば以下のいずれかを含んでいてもよい:
 対象から口腔内から得られる試料を採取する工程、又は対象に試料を採取させる工程;試料中のFusobacterium属菌の有無又は量を分析する工程;分析結果を表示する工程;分析結果に基づき、対象がFusobacterium属菌数を低減することが望ましい又は必要か否かを判定する工程;判定結果を表示する工程;口腔内でヘスペレチンを遊離するフラボノイド及びその糖転移体、並びにヘスペレチンからなる群より選択されるいずれかを含む、組成物又は食品素材である製品を表示する工程。
  以下、実施例を用いて、本発明をさらに具体的に説明する。但し、本発明の技術的範囲は、これら実施例に限定されるものではない。 
[実施例1]

 糞便を、ヘスペリジン (富士フイルム和光純薬)を含む培地で培養し、Fusobacteriumを定量することでヘスペリジンによりFusobacteriumの菌数が低減することを確認した。
 詳細には、6人の糞便を0.3%(w/v)のヘスペリジン(ヘスペレチンとしては、0.15%(w/v))を含むmYCFA培地で37 °C、嫌気条件下で16時間培養し、各培養液からMaxwell(登録商標)RSC Instrument (プロメガ社)によりDNAを抽出した。
 菌数の測定は次のように行った。
 予めFusobacterium属菌の菌数を測定した培養液から抽出したDNAに対して定量的PCRを行い、検量線を作成した。上記で得られたDNAに対して定量的PCRを行い、検量線に基づいてFusobacterium属菌の菌数を算出した。その後、培養液1mLあたりの菌数に補正した。定量的PCRに用いたプライマー配列は表1に示される通りであった。スタンダードはFusobacterium nucleatum subsp. nucleatum JCM 8532T(Fusobacterium属菌のType species)から抽出したDNAを使用した。
 培養前の段階からFusobacteriumが検出されない便については、培養開始時にFusobacterium nucleatum JCM8532Tを1×104 cfu/ml添加した。
 その結果、コントロール(ヘスペリジンの代わりにPurewaterを添加)に比べ、ヘスペリジンを添加したすべての培養物でFusobacterium菌数が減少していた(図1)。
 以上の結果から、ヘスペリジンはFusobacteriumの菌数を低減させている可能性が示唆された。
[実施例2]

 次に、ヘスペレチン(富士フイルム和光純薬)を含む寒天培地でFusobacteriumを培養し、ヘスペレチンを含む培地ではFusobacteriumが生育しないことを確認した。詳細には、ヘスペレチンを0.3%(w/v)含むGAM寒天培地と含まないGAM寒天培地にFusobacterium nucleatum JCM JCM8532T を播種し、37 °C、嫌気条件下で48時間培養し、コロニー数を計測した。
その結果、ヘスペレチンを含まない培地ではコロニーが検出された一方で、ヘスペレチンを含む培地ではコロニーが出現しなかった(図2)。
 以上の結果からヘスペレチンはFusobacteriumの生育を阻害することが示された。
[小括]

 このように、ヘスペリジンのような、β-グルコシダーゼによりヘスペレチンを遊離するフラボノイドとその糖転移体、及びヘスペレチンは、β-グルコシダーゼを産生する細菌が存在する消化管内(口腔や腸管)において、Fusobacterium属菌の菌数の低減のために用いることができるといえる。口腔内や腸内で腸内細菌により産生されるβ-グルコシダーゼによりヘスペレチンを遊離するフラボノイドとその糖転移体、及びヘスペレチンは、口腔内や腸内菌叢におけるFusobacterium属菌の菌数の低減のために用いることができるといえる。
また、ヘスペレチンはそれ自体で、Fusobacterium属菌の菌数の低減のために用いることができるといえる。Fusobacterium属菌は、最近では大腸がんへの関与(非特許文献1等)が示唆されており、F. nucleatumは虫垂炎などの消化器感染症から分離され(非特許文献2)、最近では炎症性腸疾患とも関連していることが示唆されている(非特許文献3、4、5)。口腔内におけるFusobacterium属菌は、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症の原因となることが知られている(非特許文献6、7、8、9、10)。したがって消化管内で、βグルコシダーゼにより、ヘスペレチンを遊離するフラボノイド及びその糖転移体 、並びにヘスペレチンは、いずれもこれらの疾患の処置のために用いることができるといえる。
[製造例1:発酵乳]

 牛乳と乳製品(牛乳由来)と水を、最終製品の無脂乳固形分が9.5%、乳脂肪分が3.0%、モノグルコシルヘスペリジンが0.2%となるように混合して、ヨーグルトベースミックスを調製する。調製したヨーグルトベースミックスを均質化後、95℃、5分間加熱殺菌し、その後、約40℃まで冷却する。この殺菌済ヨーグルトベースミックスに、Lactobacillaceae科乳酸菌を接種し、Lactobacillaceae科乳酸菌が100gあたり5×109個以上となるまで発酵させ、腸内菌叢におけるFusobacterium属菌の制御のためのヨーグルトを製造する。
[製造例2:健康食品]

 各原料(酸味料、甘味料、安定剤、果汁、香料、モノグリコシルヘスペリジン、水)を混合して、清涼飲料を得、約500mLずつをボトル詰めし、Fusobacterium属菌の制御用の飲料を調製する。モノグリコシルヘスペリジンは、500mLあたり、400mg配合する。
[製造例3:タブレット]

 各原料(賦形剤、酸味料、甘味料、安定剤、果汁、香料、モノグリコシルヘスペリジン)を混合して、打錠することにより、口腔内におけるFusobacterium属菌の制御用のタブレットを調製する。モノグリコシルヘスペリジンは、タブレット1gあたり、6mg配合する。
[製造例4:ガム]

 各原料(マルチトール、ガムベース、酸味料、甘味料、増粘剤(アラビアガム)、ヘスペリジン、光沢剤、着色料(クチナシ、紅花黄))を混合して、口腔内におけるFusobacterium属菌の制御用の粒状ガムを製造する。モノグリコシルヘスペリジンは、ガム1gあたり、4mg配合する。
[製造例5:カプセル剤]

 林原ヘスペリジン(登録商標)S(糖転移ヘスペリジン)を20mgずつ腸溶性カプセルに粉末充填して、腸内におけるFusobacterium属菌の制御用、腸疾患用、整腸(便通を整える) 用、又は軟便 ・便秘 ・腹部膨満感改善用の医薬品を製造する。
[製造例6:腸溶性サプリメント(錠剤)]

 ヘスペレチンを含む原料の混合粉体を、打錠し、コーティングし、1錠当たり10~40mgのヘスペレチンを含む錠剤を得る。Lactobacillaceae科乳酸菌以外の添加物は、トウモロコシデンプン、デキストリン、乳糖水和物、沈降炭酸カルシウム、アメ粉、白糖、タルク、ステアリン酸マグネシウムである。
[製造例7:腸放出サプリメント(カプセル剤)]

 胃腸管の内容物と接触して約8時間後にその内容物を放出するようにデザインされたパルスカプセル(Pulsincap(登録商標)、Scherer DDS, Ltd.)中に、ヘスペレチンを1カプセル当たり10~40mg充填する。
 本発明は、Fusobacterium属菌の制御用の組成物により、人々の健康維持・改善をサポートする。また本発明によれば、人々の健康維持・改善をサポートする食品組成物と、食品の製造方法を提供することができる。さらに本発明により、様々な人々の栄養の改善が実現され、健康的な生活が確保され、福祉が促進されうる。

Claims (16)

  1.  消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、消化管内におけるフソバクテリウム(Fusobacterium)属に属する菌の制御用の組成物。
    (式中、R1~R4は、以下の組み合わせである。)。
  2.  グルコシダーゼによる加水分解により式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、フソバクテリウム(Fusobacterium)属に属する菌の制御用の組成物。
    (式中、R1~R4は、以下の組み合わせである。)。
  3.  R1~R4が、以下の組み合わせである、請求項1又は2に記載の組成物。
  4.  式Iで表される化合物が、ヘスペレチンであり、ヘスペレチンを遊離するフラボノイドが、ヘスペリジンであり、
     ヘスペリジン、ヘスペリジンの糖転移体、及びヘスペレチンからなる群より選択されるいずれかを含む、請求項1又は2に記載の組成物。
  5.  ヘスペリジンの糖転移体が、グルコシルヘスペリジンである、請求項4に記載の組成物。
  6.  ヘスペレチンを含む、Fusobacterium属に属する菌の制御用の組成物。
  7.  Fusobacterium属に属する菌の制御が、Fusobacterium属に属する菌の菌数の低減である、請求項1、2、又は6に記載の組成物。
  8.  プレバイオティクス、及びシンバイオティクスからなる群より選択されるいずれかとして使用するための、請求項1、2、又は6に記載の組成物。
  9.  腸管内及び糞便の少なくとも一方にFusobacterium属菌が検出された対象における、炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置用の、請求項1、2、又は6に記載の組成物。
  10.  炎症性腸疾患、大腸がん、歯周病、口臭、口腔がん、食道がん、誤嚥性肺炎、及びアルツハイマー型認知症から選択されるいずれかの処置用の、請求項1、2、又は6に記載の組成物。
  11.  ヘスペレチン、及びヘスペリジンの糖転移体からなる群より選択されるいずれかを有効成分として含み、消化管が口腔である、請求項1に記載の組成物。
  12.  消化管が腸である、請求項1に記載の組成物。
  13.  請求項1に定義した、消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれか(ただし、糖転移ヘスペリジンを除く。)を含む、機能性表示食品、特定保健用食品、発酵乳、乳飲料、及び粉末食品から選択されるいずれかである、食品組成物。
  14.  請求項1に定義した、消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、医薬組成物。
  15.  請求項1に定義した、消化管内で式Iで表される化合物を遊離するフラボノイド及びその糖転移体、並びに式Iで表される化合物からなる群より選択されるいずれかを含む、腸溶性製剤。
  16.  前記フラボノイド及びその糖転移体、並びに前記化合物から選択されるいずれかを、0.01 g以上7 g以下で含有する、請求項13若しくは14に記載の組成物、又は請求項15に記載の製剤。
