Definitions
- the present invention relates to a method for calibrating a peracetic acid measuring device and a peracetic acid meter calibration system.
- the titration method is susceptible to variability between operators, and requires expert skills in reagent preparation and titration.
- differences in the performance of the peracetic acid measuring device have a significant effect on the titration accuracy, so an expensive peracetic acid concentration measuring device is required for accurate calibration.
- the reagents used must be adjusted for each absorbance measurement, requiring operators to perform complex operations and have high skill.
- the present invention was made in consideration of the above-mentioned problems, and its main objective is to provide a method for accurately calibrating a peracetic acid meter without requiring expensive equipment, advanced operator skills, or complicated operations.
- the peracetic acid meter calibration method of the present invention is characterized in that the peracetic acid meter is calibrated based on the expected change in peracetic acid concentration when peracetic acid in a sample solution is decomposed by adding a peracetic acid decomposition agent whose reaction constant for the peracetic acid decomposition reaction is known, and the change in the actual value when the sample solution is measured with the peracetic acid meter before and after the addition.
- the actual value may be a detection value (current value, voltage value, count value, absorbance, etc.) detected by the peracetic acid meter to measure the peracetic acid concentration, or a calculated value calculated based on these (peracetic acid concentration or a value during calculation to calculate the peracetic acid concentration, etc.).
- the peracetic acid meter calibration method configured in this manner, a peracetic acid decomposition agent with a known reaction constant for the peracetic acid decomposition reaction is added, and the actual values in the sample solution before and after the addition are measured by the peracetic acid meter, which is the object of calibration. Therefore, the peracetic acid meter can be calibrated based on the change in the actual values corresponding to the change in the peracetic acid concentration caused by the addition of the peracetic acid decomposition agent and the theoretical change in the peracetic acid concentration.
- the peracetic acid meter can be calibrated using a sample solution with an unknown peracetic acid concentration, eliminating the need to calibrate the peracetic acid meter by preparing a peracetic acid formulation with a known concentration as in the conventional method.
- the peracetic acid decomposition agent can cause a large change in hydrogen peroxide concentration
- air bubbles are generated when hydrogen peroxide is decomposed, the effect of air bubbles on the measurement by the peracetic acid meter can be reduced by decomposing the hydrogen peroxide first.
- the decomposition of hydrogen peroxide is carried out using a hydrogen peroxide decomposer
- the hydrogen peroxide decomposer may include one or more enzymes selected from the group consisting of catalase, peroxidase, and glutathione peroxidase.
- peracetic acid decomposition agent described above includes one or more selected from the group consisting of thioglycolic acid, glyceryl thioglycolate, methionine, cysteine, methionine derivatives, cysteine derivatives, methionine residue-containing peptides, cysteine residue-containing peptides, cysteamine, thioglycerin, 2-mercapto-4-butanolide, flavonoids, nitrous acid, ascorbic acid, sulfur, platinum, palladium, and halogen elements.
- the peracetic acid meter may be, for example, a diaphragm type that measures current and/or voltage values.
- the effects of the present invention are particularly pronounced in applications that require highly accurate calibration of peracetic acid meters, such as medical cleaning and disinfection equipment, food cleaning equipment, or wastewater monitoring equipment.
- a specific embodiment of the present invention is an endoscope cleaning and disinfecting apparatus that includes a mixture generating means for generating a mixture by mixing a peracetic acid decomposition agent, the reaction constant of which is known for the decomposition reaction of peracetic acid, with a sample solution (cleaning or disinfecting solution) containing peracetic acid, a measuring means for contacting the mixture and measuring the concentration of peracetic acid in the mixture, and a calibration means for calibrating a peracetic acid meter based on the amount of peracetic acid expected to be decomposed by the peracetic acid decomposition agent and the actual value measured by the measuring unit.
- the endoscope cleaning and disinfecting apparatus may further include a sample solution measuring means for holding a fixed amount of the sample solution, collecting a fixed amount of the sample solution, or measuring the amount of the sample solution, and a peracetic acid decomposition agent measuring means for measuring the amount of the peracetic acid decomposition agent added to the sample solution.
- the present invention also includes the following inventions.
- a calculation unit that calculates an expected change in the concentration of peracetic acid when peracetic acid in a sample solution is decomposed by adding a peracetic acid decomposition agent having a known reaction constant for the peracetic acid decomposition reaction, and an amount of change in an actual measurement value when the sample solution is measured with a peracetic acid meter before and after the addition, and a calibration unit that calibrates the peracetic acid meter based on the amount of change in the concentration of peracetic acid and the amount of change in the actual measurement value;
- a peracetic acid meter calibration program for calibrating a peracetic acid meter based on an expected change in peracetic acid concentration when peracetic acid in a sample solution is decomposed by adding a peracetic acid decomposition agent having a known reaction constant for the peracetic acid decomposition reaction, and an actual change in the value when the sample solution is measured with the peracetic acid meter before and after the addition; and a perace
- a peracetic acid decomposition agent with a known reaction constant for the peracetic acid decomposition reaction is added, and actual values such as current and/or voltage in the sample solution before and after the addition are measured using a peracetic acid meter, which is the object of calibration. Therefore, the peracetic acid meter can be calibrated based on the change in the actual value corresponding to the change in the peracetic acid concentration caused by the addition of the peracetic acid decomposition agent and the change in the theoretical peracetic acid concentration.
- the peracetic acid meter can be calibrated using a sample solution with an unknown peracetic acid concentration, eliminating the need to prepare a peracetic acid preparation with a known concentration by titration or absorbance using a peracetic acid concentration measurement device separate from the peracetic acid meter to be calibrated, as in the past.
- This makes it possible to provide a method for accurately calibrating a peracetic acid meter without requiring expensive equipment, advanced operator skills, or complicated operations.

- the cleaning disinfectant W recovered in the cleaning disinfectant tank 21 after being supplied to the cleaning disinfectant tank 1 and used for cleaning and/or disinfecting medical instruments and the like can be reused if the peracetic acid concentration is within a predetermined concentration range.
- the peracetic acid concentration of the cleaning disinfectant W recovered in the cleaning disinfectant tank 21 is measured by the peracetic acid meter 251 to confirm whether it can be reused in the next or subsequent cleaning and disinfecting process. If the peracetic acid concentration in the cleaning disinfectant W recovered in the cleaning disinfectant tank 21 falls outside the predetermined concentration range, the cleaning disinfectant W is discharged from the discharge flow path 222 to the outside.
- the decomposition agent flow control unit 432 adjusts the amount of the cleaning disinfectant W in the cleaning disinfectant tank 21 to a predetermined amount.
- the liquid level sensor (sample solution measuring means) 252 detects the water level of the cleaning disinfectant tank, and if the water level is lower than a predetermined water level (for example, the minimum water level set in the cleaning disinfectant tank 21), the decomposition agent flow control unit 432 controls the valve 24 to supply water from the water supply pipe 224 via the cleaning disinfection tank 1 and the return flow path 223 to the cleaning disinfectant tank 21 so that the water level in the cleaning disinfectant tank 21 becomes the predetermined water level.

- the peracetic acid meter (measuring means) 251 measures the peracetic acid concentration of the cleaning disinfectant W in the cleaning disinfectant tank 21.
- a peracetic acid decomposition agent is added to the cleaning and disinfecting liquid tank 21, and the peracetic acid decomposition reaction is started simultaneously with the addition.
- the user adds a predetermined amount of peracetic acid decomposition agent to the cleaning and disinfection tank 1, and the decomposition agent flow control unit 432 controls the valve 24 to introduce the peracetic acid decomposition agent added to the cleaning and disinfection tank 1 into the cleaning and disinfection liquid tank 21 via the return path 223.

- the decomposer addition mechanism 43 corresponds to the mixed solution generating means.
- the peracetic acid meter 251 measures the peracetic acid concentration in the mixture obtained by mixing the cleaning disinfectant W with the peracetic acid decomposition agent.
- the measurement conditions at this time are preferably as similar as possible to the measurement conditions used when the peracetic acid meter 251 measures the peracetic acid concentration of the cleaning disinfectant W immediately before the peracetic acid decomposition agent is added.
- the calculation unit 41 calculates the difference between the constant current values in the measurements of the cleaning disinfectant solution W and the mixed solution by the peracetic acid meter 251, and the theoretical amount of peracetic acid decomposed that is expected to be decomposed between these two measurements.
- the theoretical amount of peracetic acid decomposed is calculated based on, for example, the reaction time and reaction temperature, the amount of cleaning disinfectant solution W in the cleaning disinfectant solution tank 21, the concentration, amount and reaction constant of the added peracetic acid decomposer, etc.
- the user may input necessary information from this information to the calibration system 4 in advance.
- the calibration process is completed when the calibration unit (calibration means) 42 calibrates the peracetic acid meter 251 based on the change in the actual current value calculated in this manner and the change in the theoretical current value calculated from the theoretical amount of peracetic acid decomposition.
- the calibration mode may be automatically selected periodically, or the calibration mode may be selected by the user at any time.
- the specific method of this calibration is as follows.
- the current value difference can be converted into the peracetic acid concentration by the following formula (1).
- C0 represents the reference peracetic acid concentration
- C represents the peracetic acid concentration of the measurement target solution
- ⁇ i represents the difference in current value between C0 and C. If the value of C0 and the current value detected when the concentration is C0 are known, the amount of change in the concentration of peracetic acid in the sample solution corresponding to the amount of change in the current value can be calculated from this formula.
- the relational expression between the amount of change in the current value and the amount of change in the peracetic acid concentration can be determined depending on the type of peracetic acid meter 251 and the environment in which it is used.
- the change in the peracetic acid concentration calculated from the theoretical amount of peracetic acid decomposed is compared with the change in the peracetic acid concentration converted from the actual current values (actual values) measured before and after the addition of the peracetic acid decomposition agent based on the previously obtained formula (1), and the peracetic acid meter 251 can be calibrated by correcting the deviation between them.
- C 0 in FIG. 4 indicates the peracetic acid concentration in the sample solution before the addition of the peracetic acid decomposition agent
- C indicates the peracetic acid concentration after a predetermined time has elapsed since the addition of the peracetic acid decomposition agent.
- the current value after the addition of the peracetic acid decomposition agent here indicates the current value after a predetermined time has elapsed since the addition of the peracetic acid decomposition agent to the sample solution.
- a current value when zero water that does not contain peracetic acid is measured may be used to further perform zero point correction for the peracetic acid meter 251.
- a calibration curve showing the correlation (for example, proportionality) between the amount of change in peracetic acid concentration and the amount of change in current value may be created using the current value when zero water is measured and stored in a storage unit (not shown), and the calibration unit may be configured to calibrate the peracetic acid meter using this calibration curve.
- the peracetic acid meter 251 that measures the peracetic acid concentration in the cleaning and disinfecting solution W can be calibrated using the cleaning and disinfecting solution W whose peracetic acid concentration is unknown. Therefore, there is no need to use a peracetic acid preparation with a known peracetic acid concentration as in the past.
- the medical washer/disinfectant 100 in order to reliably disinfect and sterilize medical instruments and the like with the washer/disinfectant, it is necessary to constantly maintain the concentration of peracetic acid in the washer/disinfectant at a standard value or higher.
- the peracetic acid meter 251 In such a medical cleaning and disinfecting apparatus 100, in order to accurately measure the concentration of peracetic acid contained in the cleaning and disinfecting solution using the peracetic acid meter 251, the peracetic acid meter 251 needs to be calibrated periodically. If the calibration method according to this embodiment is adopted in such a medical cleaning and disinfecting apparatus 100, the burden on the user regarding the maintenance work of the peracetic acid meter 251 can be significantly reduced.
- the medical cleaning and disinfecting apparatus may further include a temperature control mechanism for adjusting the temperature inside the cleaning and disinfecting solution tank.
- a temperature control mechanism for adjusting the temperature inside the cleaning and disinfecting solution tank.
- the cleaning and disinfecting tank 1 is provided with a tray-shaped cleaning and disinfecting tank lower member 11' including a part of its bottom and side surfaces, which prevents medical instruments from being put into the cleaning and disinfecting tank 1, and different cleaning and disinfecting tank lower members 11' may be used for cleaning and disinfecting medical instruments and for calibration.

- a mark 111' such as a protrusion only to the cleaning and disinfecting tank lower member ' dedicated to the calibration mode, so that the medical cleaning and disinfecting device can distinguish the type of the cleaning and disinfecting tank lower member 11 and select the calibration mode.
- a cleaning/disinfecting tank lower part 11' dedicated to calibration a predetermined amount of peracetic acid decomposition agent may be placed in the cleaning/disinfecting tank lower part 11' beforehand.
- the amount of cleaning disinfectant stored in the cleaning disinfectant tank is detected by a liquid level sensor, but for example, as shown in FIG. 6, a temperature sensor 3'' for measuring the temperature of the peracetic acid decomposer may be further provided, and this temperature sensor 3'' may be used to measure the temperature of the peracetic acid decomposer before it is added to the cleaning disinfectant, and the temperature of the mixed solution after the peracetic acid decomposer is added may be measured, thereby calculating the volume of the cleaning disinfectant before the addition of the peracetic acid decomposer from the temperature change of the mixed solution due to the addition of the peracetic acid decomposer.
- the method for calculating the volume of the cleaning and disinfecting solution is not limited to the above-mentioned method, and the volume may be calculated by measuring the absorbance, conductivity, etc. of the cleaning and disinfecting solution after labeling it with a color or the like.
- the step of determining the volume of the cleaning disinfectant may be performed after measuring the peracetic acid concentration of the cleaning disinfectant or the mixed solution.
- a cleaning disinfectant collection section may be provided that collects a predetermined amount of cleaning disinfectant from the cleaning disinfectant tank, and a peracetic acid decomposer may be added to the cleaning disinfectant collected by this cleaning disinfectant collection section.
- a peracetic acid decomposer may be added to the cleaning disinfectant collected by this cleaning disinfectant collection section.
- the cleaning disinfectant may be manually collected from the cleaning disinfectant tank by the user.
- the conditions for measuring the peracetic acid concentration in the cleaning and disinfecting solution or mixed solution using a peracetic acid meter be the same before and after the addition of the peracetic acid decomposition agent, but the conditions do not necessarily have to be the same.
- the decomposition agent addition mechanism may further include a peracetic acid decomposition agent measuring section and/or a peracetic acid decomposition agent feeding section that automatically adds a predetermined amount of peracetic acid decomposition agent to the cleaning and disinfecting solution.
- the medical cleaning and disinfecting device may be configured to issue a warning to inform the user that an abnormality has occurred in the peracetic acid meter, etc., if the amount of change in the current value before and after the peracetic acid decomposition agent falls outside a preset threshold value.
- the hydrogen peroxide may be reduced on the surface of the working electrode of the peracetic acid meter, which may result in the detection of not only a change in current value based on a change in the concentration of peracetic acid, but also a change in current value that includes a change in the concentration of hydrogen peroxide.
- a peracetic acid decomposer when a peracetic acid decomposer is added to the sample solution and the change in the concentration of peracetic acid in the sample solution is measured, it is believed that the peracetic acid meter can be calibrated more accurately if the decomposer addition mechanism adds a hydrogen peroxide decomposer to the sample solution, taking into account the theoretical change in the hydrogen peroxide concentration.
- the decomposition agent adding mechanism may further include a hydrogen peroxide decomposition agent measuring unit and/or a hydrogen peroxide decomposition agent feeding unit that adds a hydrogen peroxide decomposition agent that decomposes hydrogen peroxide to the cleaning and disinfecting solution.
- the hydrogen peroxide decomposer may be a solid or liquid, so long as it contains a hydrogen peroxide decomposing substance capable of decomposing hydrogen peroxide in a cleaning and disinfecting solution containing peracetic acid used in medical cleaning and disinfecting equipment and food cleaning equipment.
- the hydrogen peroxide decomposition substance As shown in FIG. 7, it is preferable to use, as the hydrogen peroxide decomposition substance, catalase, peroxidase, glutathione peroxidase, or the like, which does not decompose peracetic acid but can essentially decompose only hydrogen peroxide.
- the hydrogen peroxide decomposition agent may contain only one of these hydrogen peroxide decomposition substances, or may contain two or more of them.
- step 1 When hydrogen peroxide is decomposed, oxygen is generated, which generates bubbles. If the generated bubbles adhere to the electrode surface, the sensitivity of the electrode becomes unstable, which may result in poor measurement accuracy. Therefore, as shown in Figures 7 and 8, in step 1, a hydrogen peroxide decomposer is added to decompose hydrogen peroxide in the cleaning and disinfecting solution to remove the generated bubbles, and then in step 2, the peracetic acid concentration is measured in the same manner as in Figure 4, which is preferable because this allows more accurate detection of changes in the peracetic acid concentration due to the addition of a peracetic acid decomposer.
- the hydrogen peroxide decomposer when hydrogen peroxide is decomposed in advance in this manner, even if the hydrogen peroxide decomposer decomposes some of the peracetic acid in the cleaning and disinfecting solution as shown in FIG. 8, it is perfectly possible to use hydrogen peroxide decomposers other than those mentioned above as long as all of the peracetic acid in the cleaning and disinfecting solution is not decomposed.
- the hydrogen peroxide decomposer may be added automatically by the decomposer addition mechanism as described above, or may be added manually by the user.
- the method for calibrating a peracetic acid meter according to the present invention can be applied not only to the above-mentioned medical cleaning and disinfecting apparatus, but also to a peracetic acid meter provided in a food cleaning apparatus, etc. It goes without saying that the method can also be applied to a peracetic acid meter provided in an apparatus that does not use a cleaning and disinfecting solution, such as a wastewater monitoring apparatus, or a peracetic acid meter that is not built into an apparatus.
- a diaphragm type peracetic acid meter is used has been described, but it goes without saying that the peracetic acid meter does not have to be of the diaphragm type.
- the peracetic acid meter measures a voltage value as an actual measurement value
- the relationship between the voltage value and the peracetic acid concentration such as the above-mentioned formula (1), can be obtained in the same manner as in the case of the current value.
- the effects of the present invention can be achieved even when the actual measured value is not limited to a current value and/or a voltage value, but is an intermediate value used when calculating the peracetic acid concentration based on the current value and/or voltage value.
- a peracetic acid meter when a peracetic acid meter is used that does not measure a current value and/or a voltage value, but measures the peracetic acid concentration of a sample solution by, for example, absorbance or colorimetry, other indicated values such as absorbance or color wavelength that have a proportional or other correlation with the amount of change in the peracetic acid concentration in the sample solution may be used as the actual measured value.
- the present invention provides a method for calibrating a peracetic acid meter with high accuracy without requiring expensive equipment, advanced operator skills, or complicated operations.
