Abstract
본 발명은 고정화된 이산화탄소 환원 효소를 이용한 연속적인 개미산 제조 장치 및 방법에 관한 것으로, 환원된 전자전달체가 생산되는 전기화학 반응기와 이산화탄소를 개미산으로 전환하는 효소 반응기를 서로 분리함으로써 향후 스케일업을 하는데 문제가 없어 개미산 생산 스케일을 크게 하는데 유리하다. 또한, 이온 교환수지탑을 이용하여 생산된 개미산을 분리 회수함으로써 생산물 저해 현상을 막을 수 있어 개미산 생산이 지속적으로 이루어질 수 있으며, 별도의 pH를 조절할 필요가 없는 장점이 있다.
Description
본 발명은 고정화된 이산화탄소 환원 효소를 이용하여 이산화탄소로부터 개미산을 연속적으로 제조할 수 있는 장치 및 방법에 관한 것이다.
화석 연료를 이용하는 산업 전반의 빠른 발전은 대기 중 이산화탄소의 농도를 급속하게 증가시키고 있다. 게다가, 대기 중 이산화탄소의 증가가 온실효과를 통한 지구온난화를 발생시키는 결정적인 원인임이 다수의 논문들에 의해 확인된 바 있다. 따라서 이러한 이산화탄소를 고부가가치의 화학물질로 전환하는 것은 대기 중 이산화탄소 증가의 속도를 늦추는데 필수적인 기술로 상정되고 있다. 다행히도, 이산화탄소는 개미산, 디메틸 카보네이트, 고분자 등 환경 친화적인 화학 플랫폼을 생산할 수 있는 유망한 재생 가능한 자원이다.
이산화탄소로부터 생산할 수 있는 많은 후보 물질들 중에서 개미산은 경제적 및 환경적 이익 측면에서 가장 이상적인 화학 원료이다. 특히, 개미산을 전기에너지로 전환시키는 직접적 개미산 연료셀(direct formate fuel cell; DFFC)은 풍력, 태양력, 수력 등 다양한 재생 에너지원에서 생산된 에너지를 개미산으로 쉽게 저장할 수 있기 때문에 재생 에너지 저장의 해결책으로 제시되고 있다. 또한, 개미산은 환경에서 불연성, 무독성, 및 불활성이므로 수소 가스보다 안전하게 수송이 가능한 장점이 있다.
이처럼 유망한 잠재력에도 불구하고, 이산화탄소로부터 고부가가치의 화합물을 생산하는 기존의 기술은 가혹한 반응 조건과 희귀한 귀금속 촉매 및 수소 가스, 수소화물 등과 같은 값 비싼 환원제를 요구하는 치명적인 한계를 갖고 있다. 상대적으로 낮은 전기 에너지 비용을 고려할 때, 이산화탄소의 전기 촉매 반응이 유망한 대안일지라도, 일부 화학 전기 촉매는 이산화탄소의 개미산 전환 중에 높은 비율로 부산물인 수소 가스를 생산하기 때문에 불충분한 반응선택성을 보여주고 있는 실정이다. 따라서, 효소 기반의 전기화학반응에 의한 이산화탄소로부터의 개미산 생산은 탁월한 개미산 선택성으로 인하여 더 많은 관심을 받고 있다.
그러나, 효소 기반의 전기화학반응의 경우 이러한 장점에도 불구하고 여러가지 문제점으로 인하여 실용화되지 못하고 있다. 그 중 가장 큰 요인은 이산화탄소 환원반응속도가 큰 금속 함유 효소의 경우, 산소에 극히 불안정하여 쉽게 활성을 잃는다는 것이다. 또한, 이산화탄소를 환원시키는 반응과 생성된 개미산을 다시 산화시키는 반응은 모두 효소가 촉매하므로 결과적으로 반응 평형이 존재하고 이로 인하여 고농도의 개미산 생산이 어려운 단점이 존재한다.
이에, 본 발명에서는 이산화탄소를 개미산으로 전환하는 효소 반응기와 환원된 전자전달체가 생산되는 전기화학 반응기의 음극 부위를 서로 분리하고/하거나 이온 교환수지탑을 도입함으로써 장시간 연속적으로 고농도의 개미산을 생산할 수 있는 장치 및 방법을 개발하기에 이르렀다.
본 발명의 일 양상은 고정화된 이산화탄소 환원 효소를 이용하여 이산화탄소로부터 개미산을 연속적으로 제조할 수 있는 장치를 제공하는 것이다.
본 발명의 다른 양상은 고정화된 이산화탄소 환원 효소를 이용하여 이산화탄소로부터 개미산을 연속적으로 제조할 수 있는 방법을 제공하는 것이다.
본 발명의 일 양상은
(i) 캐소드(cathode)부 및 애노드(anode)부를 포함하며, 캐소드 및 애노드는 전기적으로 전원(electrical power source)에 연결되어 있는 전기화학 반응기로서, 상기 캐소드부는 전자전달체를 포함하며, 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치가 상기 캐소드부에 구비되어 있는, 전기화학 반응기;
(ii) 고정화된 이산화탄소 환원 효소를 포함하는 효소 반응기; 및
(iii) 상기 캐소드부에 용해된 이산화탄소와 환원된 전자전달체를 상기 효소 반응기로 전달하거나, 효소 반응기에서 생산된 개미산과 산화된 전자전달체를 전기화학 반응기로 전달하기 위한 펌프를 포함하는, 개미산 제조 장치를 제공한다.
본 발명의 다른 양상은
(1) 전기화학 반응기의 캐소드부에서 전자전달체를 환원시키고 이산화탄소를 용해시키는 단계로서, 상기 전자전달체는 캐소드부에 포함되어 있고, 상기 이산화탄소는 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치로부터 제공되는 단계;
(2) 상기 캐소드부에서 환원된 전자전달체와 용해된 이산화탄소를, 고정화된 이산화탄소 환원 효소를 포함하는 효소 반응기로 전달하는 단계; 및
(3) 상기 효소 반응기에서 이산화탄소 환원 효소에 의해 이산화탄소를 환원시켜 개미산을 생산하는 단계를 포함하는, 개미산의 제조 방법을 제공한다.
본 발명의 또 다른 양상은, 상기 개미산의 제조 방법에서 (4) 개미산 생산 반응으로 인해 산화된 전자전달체를 상기 효소 반응기에서 상기 전기화학 반응기의 캐소드부로 전달하는 단계를 추가로 포함하며, 상기 단계 (1) 내지 (4)가 반복적으로 수행되는, 개미산의 제조 방법을 제공한다.
본 발명에서 따른 개미산 제조 장치 및 방법은 이산화탄소를 개미산으로 전환하는 효소 반응기와 환원된 전자전달체가 생산되는 전기화학 반응기의 음극 부위가 서로 분리되어 있기 때문에 향후 스케일업을 하는데 문제가 없어 개미산 생산 규모를 크게 할 수 있으므로 상업화하는데 유리하다. 또한, 생산된 개미산이 연속적으로 이온 교환수지탑에 의하여 회수되므로 생산된 개미산에 의하여 유발되는 생산물 저해 현상을 막을 수 있어 개미산 생산이 지속적으로 이루어질 수 있다. 뿐만 아니라, 생산된 개미산에 의하여 해리되어 생산된 수소 이온 농도와, 이온 교환수지탑에 의하여 회수될 때 해리된 수산화 이온 농도가 일치하여 별도로 pH를 조절할 필요가 없는 장점이 있다. 따라서, 본 발명에 따라 이산화탄소로부터 개미산을 제조하는 장치는 배치형이지만 중간에 반응을 중단할 필요없이 연속적으로 개미산을 생산하는 것이 가능하다.

본 발명의 실시 형태는 여러 가지 다른 형태로 변형될 수 있으며, 본 발명의 범위가 이하 설명하는 실시 형태로 한정되는 것은 아니다. 또한, 본 발명의 실시 형태는 당해 기술분야에서 평균적인 지식을 가진 자에게 본 발명을 더욱 완전하게 설명하기 위해서 제공되는 것이다.
나아가, 명세서 전체에서 어떤 구성요소를 "포함"한다는 것은 특별히 반대되는 기재가 없는 한 다른 구성요소를 제외하는 것이 아니라 다른 구성요소를 더 포함할 수 있다는 것을 의미한다.
본 발명의 일 양상은,
(i) 캐소드부 및 애노드부를 포함하며, 캐소드 및 애노드는 전기적으로 전원에 연결되어 있는 전기화학 반응기로서, 상기 캐소드부는 전자전달체를 포함하며, 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치가 상기 캐소드부에 구비되어 있는, 전기화학 반응기;

(iii) 상기 캐소드부에 용해된 이산화탄소와 환원된 전자전달체를 상기 효소 반응기로 전달하거나, 효소 반응기에서 생산된 개미산과 산화된 전자전달체를 전기화학 반응기로 전달하기 위한 펌프를 포함하는, 개미산 제조 장치를 제공한다.
상기 개미산 제조 장치에서 전기화학 반응기는 캐소드부 및 애노드부를 포함하며, 캐소드 및 애노드는 전기적으로 전원에 연결되고 서로 이격될 수 있다. 이때 상기 캐소드부는 전자전달체를 포함하며, 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치에 의해 이산화탄소가 캐소드부로 공급된다.
상기 전기화학 반응기의 캐소드부는 전자전달체를 포함함으로써 전자전달체를 통해 개미산 제조 효율을 증가시킬 수 있다. 전자전달체는 처음에 전기화학 반응기의 캐소드부에 한 번 넣어주면 캐소드부에서 환원되었다가 효소 반응기에서 개미산 생산 후 산화되어 다시 전기화학 반응기로 순환된다.
상기 전자전달체는 천연 전자전달체 또는 인공 전자전달체일 수 있다. 또한, 상기 전자전달체는 바이올로젠(viologen)기를 갖는 전자전달체 및 아데닌 디뉴클레오티드(adenine dinucleotide)기를 갖는 전자전달체로 이루어진 군에서 선택되는 적어도 하나의 전자전달체일 수 있다.
구체적으로, 상기 전자전달체는 바이올로젠(viologen)기를 갖는 화합물일 수 있으며, 예를 들어, 알킬 바이올로젠(alkyl viologen), 벤질 바이올로젠(benzyl viologen), NAD(nicotinamide adeninedi nucleotide) 및 FAD(Flavin adenine dinucleotide)로 이루어진 군에서 선택되는 적어도 하나의 전자전달체일 수 있으며, 상기 알킬 바이올로젠(alkyl viologen)은 메틸 바이올로젠(methyl viologen), 에틸 바이올로젠(ethyl viologen), 프로필 바이올로젠(propyl viologen) 등을 포함할 수 있으며, 이에 한정되는 것은 아니다.
구체적으로, 상기 전자전달체는 메틸 바이올로젠(methyl viologen), 에틸 바이올로젠(ethyl viologen), 벤질 바이올로젠(Benzyl viologen), NAD(nicotinamide adenine dinucleotide) 및 FAD(Flavin adenine dinucleotide)로 이루어진 군에서 선택되는 적어도 하나의 전자전달체일 수 있고, 보다 구체적으로, 메틸 바이올로젠(methyl viologen) 및 에틸 바이올로젠(ethyl viologen)로 이루어진 군에서 선택되는 적어도 하나의 전자전달체일 수 있다.
상기 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치는 본 발명의 일 실시예에 따라 캐소드부에 설치될 수 있으며, 캐소드에 이산화탄소 또는 이산화탄소를 포함하는 기체를 주입할 수 있는 장치라면 특별히 제한되지 않는다.
상기 개미산 제조 장치에서 효소 반응기는 고정화된 이산화탄소 환원 효소를 포함한다. 상기 이산화탄소 환원 효소는 포름산 탈수소효소(formate dehydrogenase)일 수 있으며, 상기 포름산 탈수소효소는 이산화탄소와 접촉하여 이산화탄소를 개미산으로 전환시킬 수 있다. 구체적으로, 포름산 탈수소효소는 하기 반응식 1의 반응을 촉매하여 개미산을 제조할 수 있다:

상기 포름산 탈수소효소는 천연으로부터 유래될 수도 있고, 당해 분야에서 널리 공지된 다양한 단백질 합성 방법으로 획득할 수 있다. 일례로서, 유전자 재조합과 단백질 발현 시스템을 이용하여 제조하거나 단백질 합성과 같은 화학적 합성을 통하여 시험관 내에서 합성하는 방법 및 무세포 단백질 합성법 등으로 제조될 수 있다. 또한, 일례로서, 상기 포름산 탈수소효소는 펩티드, 식물 유래 조직이나 세포의 추출물, 미생물(예를 들어 세균류 또는 진균류, 그리고 특히 효모)의 배양으로 얻어진 생산물일 수 있다.
용어 "단백질(Protein)"은 아마이드 결합(또는 펩티드 결합)으로 연결된 2개 이상의 아미노산으로 이루어진 폴리머를 의미한다.
용어 "발현(expression)"은 폴리펩티드(polypeptide)가 구조 유전자로부터 생산되는 과정을 지칭한다. 상기 과정은 유전자(폴리뉴클레오티드)의 mRNA로의 전사, 및 이러한 mRNA의 폴리펩티드(단백질)(들)로의 해독을 포함한다.
용어 "재조합"은 세포가 이종의 핵산을 복제하거나, 상기 핵산을 발현하거나 또는 펩티드, 이종의 펩티드 또는 이종의 핵산에 의해 코딩된 단백질을 발현하는 세포를 지칭하는 것이다. 재조합 세포는 상기 세포의 천연 형태에서는 발견되지 않는 유전자 또는 유전자 절편을, 센스 또는 안티센스 형태 중 하나로 발현할 수 있다. 또한 재조합 세포는 천연 상태의 세포에서 발견되는 유전자를 발현할 수 있으나, 상기 유전자는 변형된 것으로써 인위적인 수단에 의해 세포 내 재도입된 것이다.
상기 포름산 탈수소효소는 포름산 탈수소효소의 아미노산 서열과 각각 약 70% 이상, 약 75% 이상, 약 80% 이상, 약 85%이상, 약 90% 이상, 약 92% 이상, 약 95% 이상, 약 97% 이상, 약 98% 이상, 또는 약 99% 이상의 서열 상동성을 갖는 단백질을 포함할 수 있다.
용어 "상동성(Homology)"은 야생형 아미노산 서열과의 유사한 정도를 나타내기 위한 것으로서, 이러한 상동성의 비교는 당업계에서 널리 알려진 비교 프로그램을 이용하여 수행할 수 있으며, 2개 이상의 서열간 상동성을 백분율(%)로 계산할 수 있다.
상기 포름산 탈수소효소는 포름산 탈수소효소의 동질효소일 수 있고, 예를 들어, 상기 포름산 탈수소효소는 메틸로박테리움속(Methylobacterium sp.), 칸디다속(Candida sp.), 싸이오바실러스속(Thiobacillus sp.), 랄스토니아속(Ralstonia sp.) 및 로도박터속(Rhodobacter sp.)으로 이루어진 군에서 선택되는 적어도 하나의 속의 미생물에서 유래할 수 있다.
구체적으로, 상기 포름산 탈수소효소는 메틸로박테리움 엑스토쿠엔스(Methylobacterium extorquens), 칸디다 보이디니(Candida boidinii), 싸이오바실러스속(Thiobacillus sp.) KNK65MA, 랄스토니아 유트로파(Ralstonia eutropha), 및 로도박터 캡슐레이터스(Rhodobacter capsulatus)로 이루어진 군에서 선택되는 적어도 하나의 미생물에서 유래할 수 있다.
일 구체예로서, 상기 포름산 탈수소효소는 텅스텐을 함유하는 메틸로박테리움 엑스토르쿠엔스(Methylobacterium extorquens) AM1 유래 FDH1(formate dehydrogenase 1)일 수 있으며, 유전자 재조합으로 제조된 것일 수 있다. 구체적으로, 상기 메틸로박테리움 엑스토르쿠엔스 AM1은 FDH1알파(formate dehydrogenase 1 alpha subunit)를 코딩하는 내인성 유전자 및 FDH1베타(formate dehydrogenase 1 beta subunit)를 코딩하는 내인성 유전자가 결실되고, 메틸로박테리움 엑스토르쿠엔스 AM1 유래 FDH1 및 히스티딘 표지를 코딩하는 외인성 유전자가 도입된 재조합 미생물일 수 있다.
보다 구체적으로 상기 포름산 탈수소효소는 메틸로박테리움 엑스토르쿠엔스(Methylobacterium extorquens) AM1(ATCC 14718, GenBank accession No. CP001510.1) 유래 포름산 탈수소효소 I일 수 있다. 상기 메틸로박테리움 엑스토르쿠엔스 AM1 유래 포름산 탈수소효소 I은 MeFDH I 알파 subunit(GenBank accession No. ACS42636.1) 및 MeFDH I 베타 subunit(GenBank accession No. ACS42635.1)으로 이루어질 수 있고, 상기 메틸로박테리움 엑스토르쿠엔스 유래 포름산 탈수소효소 I은 fdh1A(GenBank accession, CP001510.1:5169596-5172565, Methylobacterium extorquens AM1, complete genome) 및 fdh1B(GenBank accession, CP001510.1:5167825-5169543, Methylobacterium extorquens AM1, complete genome)가 코딩하는 폴리펩티드일 수 있다.
또한, 보다 나은 화학적 안정성, 강화된 약리 특성(반감기, 흡수성, 역가, 효능 등), 변경된 특이성(예를 들어, 광범위한 생물학적 활성 스펙트럼), 감소된 항원성을 획득하기 위하여, 상기 포름산 탈수소효소의 N- 또는 C-말단에 보호기가 결합되어 있을 수 있다. 상기 보호기는 아세틸기, 플루오레닐 메톡시 카르보닐기, 포르밀기, 팔미토일기, 미리스틸기, 스테아릴기 또는 폴리에틸렌글리콜(PEG)일 수 있으나, 상기 포름산 탈수소효소의 개질, 특히 포름산 탈수소효소의 안정성을 증진시킬 수 있는 성분이라면, 제한없이 포함될 수 있다.
상기 포름산 탈수소효소는 표적화 서열, 태그(tag), 표지된 잔기를 위한 특정 목적으로 제조된 아미노산 서열을 추가적으로 포함할 수 있으며, 특별히 이에 제한되지 않는다. 구체적으로, 상기 포름산 탈수소효소는 히스티딘 표지(histidine tag)된 포름산 탈수소효소일 수 있다.
상기 효소 반응기는 고정화된 이산화탄소 환원 효소를 포함하며, 이산화탄소 환원 효소는 물리적 흡착 또는 화학적 결합에 의해 고정화할 수 있다. 고정화된 이산화탄소 환원 효소를 사용함으로써 이산화탄소 환원 효소의 재사용이 가능하여 연속 공정에 사용하기 유리하다.
구체적으로, 상기 히스티딘 표지된 포름산 탈수소효소는 Ni-NTA(nickel-nitrilotriacetic acid)에 의해 고정화된 포름산 탈수소효소일 수 있다. 히스티딘 표지된 포름산 탈수소효소는 Ni-NTA에 고정화된 경우, 고정화되지 않은 히스티딘 표지된 포름산 탈수소효소에 비해 장기간 동안 효율이 떨어지지 않고 개미산 제조가 가능하며, 재사용이 가능할 뿐만 아니라, 용존 산소 농도에 따라 활성 감소가 적을 수 있다.
상기 Ni-NTA는 Ni-NTA(nickel-nitrilotriacetic acid)로서, Ni-NTA를 포함한다면 제한되지 않으나, 예를 들어, Ni-NTA resin(수지), Ni-NTA affinity resin, Ni-NTA column(컬럼), Ni-NTA affinity column, Ni-NTA agarose, Ni-NTA bead, Ni-NTA 아가로즈 비드, Ni-NTA magnetic bead 등일 수 있고, 구체적으로, 상기 Ni-NTA는 Ni-NTA 아가로즈 비드일 수 있다.
본 발명의 일 구체예에서는 텅스텐을 포함하는 메틸로박테리움 엑스토르쿠엔스 AM1 유래 포름산 탈수소효소가 히스티딘 표지를 가지도록 재조합 효소를 생산하고 이를 Ni-NTA 아가로즈 비드에 고정화시켜 분리와 동시에 효소 촉매로 손쉽게 준비할 수 있도록 한다. 예컨대, FDH1 alpha 서브유닛 C-말단에 히스티딘 표지를 부가적으로 코딩하고 FDH1 beta 서브유닛을 코딩하는 외인성 유전자가 도입된 플라스미드를 이용하여 텅스텐을 포함하는 재조합 FDH1을 제조하고, 이렇게 제조된 FDH1 효소는 Ni-NTA 아가로즈 비드를 이용하여 FDH1-His tag와의 친화도를 바탕으로 손쉽게 Ni-NTA 아가로즈 비드에 고정화한다. 이렇게 고정화된 효소를 이용하여 전기화학 반응기에서 장기간 운전을 통하여 고농도의 개미산을 생산하고 동시에 연속적인 재사용이 가능하게 한다.
상기 개미산을 제조하는 장치에서, 이산화탄소를 용해시키고 전자전달체를 환원시키는 캐소드부를 포함하는 전기화학 반응기와, 이산화탄소를 개미산으로 전환하는 반응이 진행되는 효소 반응기는 서로 독립적으로 분리하여 설치될 수 있다. 이전에는 전기화학 반응기에 이산화탄소 환원 효소를 직접 추가하였는데, 전극은 면적이 제한되므로 개미산 생산 스케일을 키우는데 제약이 있었다. 본 발명에 따라 전기화학 반응기와 이산화탄소를 개미산으로 전환하는 효소 반응기를 분리하여 설치하는 경우, 전극 면적에 제한을 받지 않으므로 개미산 생산 규모를 키우는데 문제가 없어 개미산을 대량 생산하기에 유리할 수 있다.
상기 개미산 제조 장치는 전기화학 반응기의 캐소드부에 용해된 이산화탄소와 환원된 전자전달체를 효소 반응기로 전달하거나, 효소 반응기에서 생산된 개미산과 산화된 전자전달체를 전기화학 반응기로 전달하기 위한 펌프를 포함한다.
상기 펌프는 이산화탄소 및 환원된 전자전달체를 포함하는 수용액을 전기화학 반응기의 캐소드부에서 효소 반응기로 전달하거나, 효소 반응기에서 생산된 개미산과 산화된 전자전달체를 포함하는 수용액을 전기화학 반응기로 전달하는 등 액체를 이송 위한 수단으로, 이러한 기능을 수행할 수 있는 장치라면 특별히 제한되지 않는다. 구체적으로 펌프는 액체 이송용 펌프로서, 예컨대 일정량의 물질을 전달할 수 있는 정량 펌프(peristatic pump)를 사용할 수 있다. 상기 정량 펌프는 일정량의 환원된 전자전달체와 용해된 이산화탄소를 캐소드부에서 효소 반응기로 공급하게 된다.
한편, 상기 개미산 제조 장치는 이온 교환수지탑을 추가로 포함할 수 있다. 구체적으로, 개미산은 전해질에서 음전하를 갖기 때문에, 음이온 교환수지를 이용하여 개미산을 선택적으로 흡착할 수 있다.
상기 이온 교환수지탑은 효소 반응기와 전기화학 반응기 사이에 위치할 수 있다. 이렇게 되면, 고정화된 이산화탄소 환원 효소에 의해 이산화탄소로부터 생산된 개미산 및 산화된 전자전달체를 포함하는 수용액을 음이온 교환수지가 충진된 충진탑에 주입함으로써 개미산을 음이온 교환수지에 부착시키고 부착되지 않은 산화 상태의 전자전달체가 포함된 수용액은 전기화학 반응기의 캐소드부로 보내지게 된다.
상기와 같이 이온 교환수지탑을 추가로 포함하는 경우, 생산된 개미산이 연속적으로 이온 교환수지탑에 의하여 회수되므로 생산된 개미산에 의하여 유발되는 생산물 저해 현상을 막을 수 있어 개미산을 계속 연속적으로 생산할 수 있다. 또한, 생산된 개미산에 의하여 해리되어 생산된 수소 이온 농도와 이온 교환수지탑에 의하여 회수될 때 해리된 수산화 이온 농도가 일치하여 별도의 pH를 조절할 필요가 없게 된다.
본 발명의 다른 양상에서,
(1) 전기화학 반응기의 캐소드부에서 전자전달체를 환원시키고 이산화탄소를 용해시키는 단계로서, 상기 전자전달체는 캐소드부에 포함되어 있고, 상기 이산화탄소는 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치로부터 제공되는 단계;
(2) 상기 캐소드부에서 환원된 전자전달체와 용해된 이산화탄소를, 고정화된 이산화탄소 환원 효소를 포함하는 효소 반응기로 전달하는 단계; 및
(3) 상기 효소 반응기에서 이산화탄소 환원 효소에 의해 이산화탄소를 환원시켜 개미산을 생산하는 단계를 포함하는, 개미산의 제조 방법을 제공한다.
단계 (1)은 개미산 제조 효율을 증가시키기 위하여 전기화학 반응기의 캐소드부에서 전자전달체를 환원시키고, 이산화탄소를 용해시키는 단계이다. 이때, 상기 전자전달체는 캐소드부에 포함되어 있고, 상기 이산화탄소는 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치로부터 제공된다.
상기 전자전달체는 바이올로젠(viologen)기를 가지는 전자전달체 및 아데닌 디뉴클레오티드(adenine dinucleotide)기를 가지는 전자전달체로 이루어진 군에서 선택되는 적어도 하나의 전자전달체일 수 있고, 상기 “전자전달체”는 전술한 범위 내일 수 있다. 예컨대, 상기 전자전달체는 바이올로젠(viologen)기를 갖는 화합물일 수 있다.
상기 이산화탄소는 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치로부터 제공될 수 있으며, 상기 장치는 전술한 범위 내일 수 있다.
상기 효소 반응기는 이산화탄소를 개미산으로 전환하는데 필요한, 고정화된 이산화탄소 환원 효소를 포함하고 있다. 상기 이산화탄소 환원 효소는 포름산 탈수소효소일 수 있으며, 일 구체예에서, 상기 포름산 탈수소효소는 텅스텐을 함유하는 메틸로박테리움 엑스토르쿠엔스 AM1 유래 FDH1일 수 있다. 상기 포름산 탈수소효소는 히스티딘 표지된 포름산 탈수소효소일 수 있거나/있고 Ni-NTA(Nickel-Nitrilotriacetic Acid)에 고정화될 수 있다.
상기 “포름산 탈수소효소”, “히스티딘 표지”, “Ni-NTA” 등은 전술한 범위 내일 수 있다.
단계 (2)는 상기 캐소드부에서 환원된 전자전달체와 용해된 이산화탄소를 효소 반응기로 전달하는 단계이며, 단계 (3)는 상기 효소 반응기에서 이산화탄소 환원 효소에 의해 이산화탄소를 환원시켜 개미산을 생산하는 단계이다.
본 발명에 따른 상기 개미산의 제조 방법은 (4) 개미산 생산 반응으로 인해 산화된 전자전달체를 상기 효소 반응기에서 상기 전기화학 반응기의 캐소드부로 전달하는 단계를 추가로 포함하며, 상기 단계 (1) 내지 (4)는 반복적으로 수행될 수 있다. 상기 단계 (4)는 단계 (3)에서 산화된 전자전달체를 캐소드부로 전달하는 단계이다. 본 발명에 따른 상기 개미산의 제조 방법은 상기 단계 (1) 내지 (4)를 반복적으로 수행함으로써 개미산을 연속적으로 생산할 수 있다.
상기 단계 (2) 및 단계 (4)에서 이산화탄소, 전자전달체, 생성된 개미산 등의 전달은 펌프에 의해 수행될 수 있으며, 상기 “펌프”는 전술한 바와 같다.
본 발명에 따른 개미산 제조 방법에서 상기 단계 (1)과 단계 (3)이 서로 독립적으로 분리되어 수행된다. 이와 같이 전기화학 반응과 이산화탄소를 개미산으로 전환하는 효소 반응을 분리하여 수행하는 경우, 전기화학 반응기의 전극 면적에 제한을 받지 않으므로 개미산 생산 규모를 키우는데 문제가 없어 개미산을 대량 생산하는 것이 가능하게 된다.
또한, 본 발명에 따른 개미산의 제조 방법은 상기 단계 (3) 이후 단계 (4) 전에, 생성된 개미산을 이온 교환수지탑에 흡착시켜 회수하는 단계를 추가로 포함할 수 있다. 구체적으로, 개미산은 전해질에서 음전하를 갖기 때문에, 음이온 교환수지탑에 개미산을 흡착시켜 회수할 수 있다. 따라서, 효소 반응기에서 이산화탄소 환원 효소에 의해 이산화탄소를 환원시켜 제조된 개미산이 음이온 교환수지탑에서 흡착에 의해 제거되므로, 개미산으로 인한 생산물 저해 현상이 생기지 않을 수 있다. 또한, 생산된 개미산에서 해리되는 수소 이온이 음이온 교환수지탑에서 해리되는 수산화 이온에 의해 중화되어 별도로 pH를 조절할 필요가 없게 되는 효과가 있어, 개미산을 장시간 연속적으로 제조하는 것이 가능하다.
일 구체예에서, (a) Ni-NTA 아가로즈 비드에 고정화된 텅스텐을 포함하는 재조합 FDH1를 효소 반응기에 충진하고, (b) 전기화학 반응기의 캐소드부에 이산화탄소를 주입하여 수중에 용해시키는 한편, 캐소드부에 용해된 전자전달체(예: 에틸 바이올로젠)을 환원시키고, (c) 액체 이송용 펌프(예: 정량 펌프)로 캐소드부의 이산화탄소와 전자전달체가 포함된 수용액을 효소 반응기에 공급하고, (d) 효소 반응기 내에서 고정화된 포름산 탈수소효소에 의하여 이산화탄소와 환원된 전자전달체를 소모하여 개미산을 생산하고, (e) 생산된 개미산 및 산화된 전자전달체를 포함한 수용액은 액체 이송용 펌프로 음이온 교환수지가 충진된 충진탑에 주입하여 개미산을 음이온 교환수지에 부착시키고 (f) 부착되지 않은 산화 상태의 전자전달체가 포함된 수용액은 전기화학 반응기의 캐소드부로 다시 주입된다.
이와 같이, 본 발명에 따라 전기화학 반응기의 캐소드부에서 이산화탄소를 용해시키면서 동시에 전자전달체를 환원시켜서 연속적으로 효소 반응기에 공급함과 동시에 효소 반응기에서 생산된 개미산을 이온 교환수지를 이용하여 연속적으로 분리하는 것에 의해 전기화학 반응기에서의 생산성을 증가시킬 수 있다.
상기 제조 방법은 하나의 예시로서 제시된 본 발명의 일 실시예에 한정되는 것은 아니며, 모순되지 않는 한 상기 개미산 제조 장치에 대한 설명이 적용될 수 있다.
이하, 본 발명을 제조예 및 실시예를 통해 상세히 설명한다.
단, 후술하는 제조예 및 실시예는 본 발명을 일 측면에서 구체적으로 예시하는 것일 뿐, 본 발명이 이에 한정되는 것은 아니다.
<제조예> FDH1-His tag 재조합 효소의 제조
pCM110의 메탄올-유도성 발현을 적용하여 재조합 MeFDH1의 발현 시간 및 수준을 제어하였다. MeFDH1알파/베타암호화 유전자(GenBank 수탁 번호 ACS42636.1(fdh1a), ACS42635.1(fdh1b))는 알파 서브유닛의 C-말단에 His6 태그로 증폭되었다. PCR 산물은 멀티클로닝 사이트(Spe1 및 Kpn1)를 통해 pCM110에 삽입되었다. 모든 플라스미드 및 돌연변이 균주는 하기 표 1에 설명되어 있다.

세포 배양을 위해 pCM110-MeFDH1-His를 함유하는 녹아웃 돌연변이체(MeAM1(Δ를 광학 밀도(OD600)가 0.5 내지 0.8에 도달할 때까지 30℃에서 20시간 동안 성장시킨 다음 메탄올을 최종 첨가하였다. 0.5%의 농도로 재조합 발현을 유도하였다. 배양 48시간 후에 배양된 세포를 수확하고 원심분리(7000rpm, 4℃, 15분)에 의해 얻은 세포 펠릿의 분취량을 사용할 때까지 -70℃에서 보관했다. 상기 미생물들의 기본적인 배양 배지는 탄소공급원으로 16 g/L 숙신산 및 최소염 배지(minimal salt medium) (1.62 g/L NH4Cl, 0.2 g/L MgSO4, 2.21 g/L K2HPO4, 및 1.25 g/L NaH2PO4·2H2O)를 포함한다. 재조합 미생물을 선별하기 위한 선택적 항생제로는 50 ㎍/mL 리파미신 (Rif), 50 ㎍/mL 카나미신 (Kan), 또는 10 ㎍/mL 테트라시클린 (Tet)을 사용하였다. 상기 미생물들은 모두 1 L 에를렌마이어 쉐이크 플라스크 중에 200 mL의 부피로 하여 26℃, 200 rpm에서 배양하였다. 세포 농도가 OD600 기준으로 0.5에서 0.8이 될 때 0.5% 메탄올을 첨가하여 목표로 하는 FDH1-his tag 효소를 발현하였다. 세포는 총 48시간을 배양하였으며, 7,000 rpm에서 15분간 원심 분리하여 배양된 세포를 획득하였다. 이후 이를 세포 파쇄기를 이용하여 분쇄한 후 4,611 g 30분간 원심 분리하여 상등액을 회수한 후 이를 Ni-NTA 아가로즈 비드 (Bio-Rad, Cat No. 7321010)을 이용하여 FDH1-his tag만을 선택적으로 고정화시켰다.

<실시예 1> 순환형 전기화학 반응기 제작 및 실험
산업에 적용하기 위하여 전기화학 반응기에 순환 시스템을 적용하고 각 반응 요소들을 모듈화하여 연속 공정을 가능하게 하였다. 순환 시스템은 정량 펌프를 이용하여 캐소드의 전해질에서 환원된 에틸 바이올로겐(EV)와 포화 용해된 이산화탄소를 효소 반응기로 전달하고, 전자를 전달한 산화 EV와 개미산은 다시 캐소드 전해질로 전달되어 EV는 전자를 얻고 이산화탄소가 용해되어 고정화 효소에 공급된다. 도 2에 순환 시스템을 이용한 전기화학 반응기 모식도를 나타내었다.
상기 순환형 전기화학 반응기를 이용하여 개미산을 생산하였으며, 그 결과를 도 3a 및 도 3b에 나타내었다. 순환형 전기화학 반응기는 500시간 동안 1.74 M의 개미산을 생산하였으며, 패러데이 효율이 100.3%로서 모든 공급된 전자가 개미산을 만드는데 이용되는 것을 확인하였다. 그러나 이 실험에서 500시간 이후에 개미산의 농도가 더 이상 늘어나지 않았다. 이는 두 가지 원인으로 1) 경쟁적 생산물 저해 기작과 2) 효소의 장기간 반응에 따른 비활성화를 유추할 수 있다. 이에 따라, 500시간 반응 후 개미산이 없는 새로운 전해질로 교체하며 재반응을 진행하였다. 그 결과, 1.74 M에서 더 이상 늘지 않았던 개미산이 교체된 후에 생산성이 회복되는 것을 확인하였다. 이로부터 장기간 반응 시에 개미산의 생산성이 감소하는 주된 원인이 생산물에 의한 경쟁적인 저해 현상임을 확인하였다.
<실시예 2> 이온 교환수지탑을 이용한 개미산의 실시간 분리 실험
상기 실시예 1에서 확인한 개미산 생산성 감소의 주된 원인에 따라, 순환형 전기화학 반응기에서 생성된 개미산의 실시간 분리 공정이 추가로 요구되었다. 개미산은 전해질에서 음전하를 갖기 때문에, 음이온 교환수지를 이용하는 경우 개미산의 선택적인 흡착이 예상되었다.
이에 따라 순환 시의 개미산 흡착 관련 실험을 진행하였으며, 모식도 및 조건을 도 4에 나타내었다. 음이온 교환수지를 이용하여 개미산을 분리하는 경우 배출된 시료의 개미산 농도 및 pH 변화를 측정한 결과를 도 5에 나타내었다. 전기화학적 개미산 생산 실험에서 총 68 mmol의 개미산이 생산되었으며, 이와 대비하여 45 g의 음이온 교환수지(Ambersep 900)는 총 70 mmol의 개미산을 흡착하였다. 이로부터 충진된 음이온 교환수지탑이 생산되는 개미산을 모두 흡착할 수 있을 것으로 추측되었다.
하기 표 2는 전기화학 반응기의 개미산 생산량과 음이온 교환수지의 개미산 흡착량을 비교한 것이다.

<실시예 3> 이온 교환수지탑을 이용한 연속적인 개미산 회수 및 생산성 향상 성능 평가
순환형 전기화학 반응기에서 고정화 효소 뒤로 음이온 교환수지탑을 장착하였다. 이와 더불어 고정화 효소 입구와 출구에 순환용 큐벳을 장착하여 실시간으로 환원된 EV의 농도를 측정하였다. 도 6은 음이온 교환수지를 이용한 순환형 전기화학적 CO2 환원반응 시스템의 모식도이다.
음이온 교환수지탑 장착 유무에 따른 전류의 변화를 도 7에 나타내었다. 도 7에 따르면, 음이온 교환수지탑을 장착하지 않은 기존의 순환형 전기화학 반응기에서 전류가 감소하는 추세를 나타내었던 것과는 달리, 이온 교환수지탑을 장착하였을 때 전류가 400 시간 동안 유지되었다.
또한, 음이온 교환수지탑을 장착한 경우 고정화된 효소의 입구와 출구에서의 환원된 EV의 농도를 도 8에 나타내었다. 환원된 EV의 농도는 음이온 교환수지탑을 장착하였을 때 4 내지 6 mM로 유지되는 것을 확인할 수 있었으나, EV의 농도가 10 mM보다 낮게 유지되고 낮은 전류로 보아 소량의 EV가 음이온 교환수지에 흡착되는 것으로 추정되었다.
도 9는 전해질에서의 개미산 농도 및 전류에 의하여 예상되는 개미산 농도를 비교한 그래프이다. 전류 데이터에서 예상되는 개미산 농도 대비 전해질에서 낮은 개미산 농도가 유지되며, 이 차이만큼 음이온 교환수지탑에 개미산이 흡착되는 것으로 추정되었다.
음이온 교환수지탑을 이용한 전기화학적 개미산 생산 실험 후, 음이온 교환수지탑의 컬럼을 따로 분리하여 개미산 탈착 실험을 진행하였다. 이에 따라 1 M의 NaOH를 이용하였을 때 개미산이 탈착됨을 확인하였다. 전해질에서의 개미산 농도와 수지에서 탈착된 개미산의 총 몰 수는 107.2 mmol로 측정되었으며, 전류에서 예상되는 개미산의 몰수는 107.7 mmol로 최종 99.5%의 패러데이 효율을 갖는 반응 시스템을 확보하였다.
음이온 교환수지탑의 컬럼에서 흡착 분리된 개미산의 농도를 도 10으로 나타내었다.
하기 표 3은 전기화학 반응기에서의 개미산 생산량과 페러데이 효율을 나타낸 것이다.

Claims (22)
- (i) 캐소드(cathode)부 및 애노드(anode)부를 포함하며, 캐소드 및 애노드는 전기적으로 전원(electrical power source)에 연결되어 있는 전기화학 반응기로서, 상기 캐소드부는 전자전달체를 포함하며, 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치가 상기 캐소드부에 구비되어 있는, 전기화학 반응기;(ii) 고정화된 이산화탄소 환원 효소를 포함하는 효소 반응기; 및(iii) 상기 캐소드부에 용해된 이산화탄소와 환원된 전자전달체를 상기 효소 반응기로 전달하거나, 효소 반응기에서 생산된 개미산과 산화된 전자전달체를 전기화학 반응기로 전달하기 위한 펌프를 포함하는, 개미산 제조 장치.
- 제1항에 있어서,상기 전자전달체는 바이올로젠(viologen)기를 갖는 화합물인, 개미산 제조 장치.
- 제1항 또는 제2항에 있어서,상기 이산화탄소 환원 효소는 포름산 탈수소효소(formate dehydrogenase)인, 개미산 제조 장치.
- 제1항 내지 제3항 중 어느 한 항에 있어서,상기 포름산 탈수소효소는 메틸로박테리움 엑스토르쿠엔스(Methylobacterium extorquens) AM1 유래 FDH1(formate dehydrogenase 1)인, 개미산 제조 장치.
- 제1항 내지 제4항 중 어느 한 항에 있어서,상기 포름산 탈수소효소는 히스티딘 표지된 포름산 탈수소효소인, 개미산 제조 장치.
- 제1항 내지 제5항 중 어느 한 항에 있어서,상기 포름산 탈수소효소는 Ni-NTA(nickel-nitrilotriacetic acid)에 고정화된 것인, 개미산 제조 장치.
- 제1항 내지 제6항 중 어느 한 항에 있어서,상기 펌프는 정량 펌프(peristatic pump)인, 개미산 제조 장치.
- 제1항 내지 제7항 중 어느 한 항에 있어서,상기 전기화학 반응기와 효소 반응기는 서로 독립적으로 분리되어 있는, 개미산 제조 장치.
- 제1항 내지 제8항 중 어느 한 항에 있어서,이온 교환수지탑을 추가로 포함하는, 개미산 제조 장치.
- 제1항 내지 제9항 중 어느 한 항에 있어서,상기 이온 교환수지탑은 음이온 교환수지탑인, 개미산 제조 장치.
- (1) 전기화학 반응기의 캐소드부에서 전자전달체를 환원시키고 이산화탄소를 용해시키는 단계로서, 상기 전자전달체는 캐소드부에 포함되어 있고, 상기 이산화탄소는 이산화탄소 또는 이산화탄소를 포함하는 기체를 공급하는 장치로부터 제공되는 단계;(2) 상기 캐소드부에서 환원된 전자전달체와 용해된 이산화탄소를, 고정화된 이산화탄소 환원 효소를 포함하는 효소 반응기로 전달하는 단계; 및(3) 상기 효소 반응기에서 이산화탄소 환원 효소에 의해 이산화탄소를 환원시켜 개미산을 생산하는 단계를 포함하는, 개미산의 제조 방법.
- 제11항에 있어서,(4) 개미산 생산 반응으로 인해 산화된 전자전달체를 상기 효소 반응기에서 상기 전기화학 반응기의 캐소드부로 전달하는 단계를 추가로 포함하며,상기 단계 (1) 내지 (4)가 반복적으로 수행되는, 개미산의 제조 방법.
- 제11항 또는 제12항에 있어서,상기 단계 (1)에서 전자전달체는 바이올로젠기를 갖는 화합물인, 개미산의 제조 방법.
- 제11항 내지 제13항 중 어느 한 항에 있어서,상기 단계 (2)에서 이산화탄소 환원 효소는 포름산 탈수소효소인, 개미산의 제조 방법.
- 제11항 내지 제14항 중 어느 한 항에 있어서,상기 포름산 탈수소효소는 메틸로박테리움 엑스토르쿠엔스 AM1 유래 FDH1인, 개미산의 제조 방법.
