本発明の一実施形態に係る魚類の生物の飼育方法は、プランタリシンおよびナイシンから選択される少なくとも1つを魚類の生物に投与することを含む。本発明の一実施形態に係る魚類の生物の飼育方法は、プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を魚類の生物に投与することを含む。投与は、経口投与、浸漬投与または注射投与であってよい。
本発明の一実施形態に係るラクトコッカス・ガルビエ(レンサ球菌)による魚類の病気に対する予防または治療用組成物は、プランタリシンおよびナイシンから選択される少なくとも1つを含む。本発明の一実施形態に係るラクトコッカス・ガルビエによる魚類の病気に対する予防または治療用組成物は、プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を含む。プランタリシン、ナイシンおよび上記乳酸菌の菌体調製物を用いて、ラクトコッカス・ガルビエによる魚類感染症を予防または治療することができる。本明細書において、治療には、症状の緩和、症状の好転および完治を含む。予防または治療用組成物は、動物用医薬品であってよい。
本発明の一実施形態に係るラクトコッカス・ガルビエによる魚類の病気に対する予防または治療方法は、プランタリシンおよびナイシンから選択される少なくとも1つを魚類の生物に投与することを含む。本発明の一実施形態に係るラクトコッカス・ガルビエによる魚類の病気に対する予防または治療方法は、プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を魚類の生物に投与することを含む。投与は、経口投与、浸漬投与または注射投与であってよい。
本発明の一実施形態に係るラクトコッカス・ガルビエの増殖抑制剤または殺菌剤は、プランタリシンおよびナイシンから選択される少なくとも1つを含む。本発明の一実施形態に係るラクトコッカス・ガルビエの増殖抑制剤または殺菌剤は、プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を含む。プランタリシン、ナイシンおよび上記乳酸菌の菌体調製物は、ラクトコッカス・ガルビエの増殖を抑制すること、またはラクトコッカス・ガルビエを殺菌することができる。増殖抑制剤または殺菌剤は、魚類の生物の飼育用組成物と同じ形態であってよい。増殖抑制剤または殺菌剤は、ラクトコッカス・ガルビエに対する抗菌活性を有する物質をさらに含んでもよい。
分離源(イカ塩辛)を滅菌水と共に摩砕した。摩砕液を適宜希釈して1/2MRS液体培地に添加し、集積培養した。集積培養液を炭酸カルシウム含有のMRS寒天培地に塗抹し、ハローを形成した微生物を分離した。以下、この分離株を分離株Aという。過酸化水素に分離株Aの培養液を懸濁したところ、気泡が発生しなかった。分離株Aは、カタラーゼ活性を有しないことから、乳酸菌であることが確認された。
(1)16S rRNA遺伝子解析
分離株AからゲノムDNAを抽出し、得られたゲノムDNAを鋳型として、クローニング用フォワードプライマー9Fおよびクローニング用リバースプライマー1510R(中川恭好他:遺伝子解析法 16S rRNA遺伝子の塩基配列決定法、日本放線菌学会編、放線菌の分類と同定、88-117pp.日本学会事務センター、2001)を用いて16S rRNA遺伝子のPCR増幅を行った。PCR増幅はTks Gflex DNA ポリメラーゼ(タカラバイオ社製)を用いて行い、PCR後の増幅産物を精製した。
MRS寒天培地に分離菌Aを塗布し、温度30℃で48時間好気培養し、以下の方法で細胞形態、グラム染色性、運動性およびコロニー形態を観察した。細胞形態は、光学顕微鏡BX50F4(オリンパス社製)によって観察した。グラム染色ではフェイバーG「ニッスイ」(日水製薬社製)を用いた。コロニー形態は、実体顕微鏡SMZ800N(ニコン社製)によって観察した。Barrow&Feltham(Cowan and Steel’s Manual for the Identification of Medical Bacteria,3rd ed.Cambridge:Cambridge University Press;1993.)に記載の方法に基づき、カタラーゼ反応、オキシダーゼ反応、ブドウ糖からの酸/ガス産生およびブドウ糖の酸化/発酵(O/F)について試験を行った。API50CHBキット(bioMerieux社製、フランス)を用いて、細菌の生理・生化学的性状反応を調べた。
分離源として、レンブを用いた以外は、実験1と同じ方法により、分離株Bを分離した。分離株Bの同定を実験1と同じ方法によって行なった。
分離源として、アダンを用いた以外は、実験1と同じ方法により、分離株Cを分離した。分離株Cの同定を実験1と同じ方法によって行なった。
分離源として、ギランイヌビワを用いた以外は、実験1と同じ方法により分離株Dを分離した。分離株Dの同定を実験1と同じ方法によって行なった。
分離源として、マツボックリを用いた以外は、実験1と同じ方法により分離株Eを分離した。分離株Eの同定を実験1と同じ方法によって行なった。
分離源として、ギランイヌビワを用いた以外は、実験1と同じ方法により分離株Fを分離した。分離株Fの同定を実験1と同じ方法によって行なった。
NITE BP-03198、NITE BP-03199、NITE BP-03200、NITE BP-03202、ATCC-11454およびNITE ABP-03481の菌体調製物が、ラクトコッカス・ガルビエI型(TUMSAT-K9408株)およびII型(TUMSAT-LgN1株)の増殖を抑制するかを検証した。図7を参照して実験方法を説明する。270μLのTodd-Hewitt(TH)培地10に乳酸菌の菌体調製物11を30μL添加した。さらに、ラクトコッカス・ガルビエを含む菌液12(約105cfu/mL)を3μL添加し、25℃で8時間震盪培養した。5μLの菌培養液13をTH寒天培地14に滴下し、25℃で12時間培養し、ラクトコッカス・ガルビエのコロニー15が形成されるかを観察した。観察されたコロニーが5個以下である場合、抗菌活性があると判断した。乳酸菌の菌体調製物は、以下の方法で調製した。NITE BP-03198、NITE BP-03199、NITE BP-03200およびNITE BP-03202については、まずそれぞれの乳酸菌をMRS Brothで温度30℃で培養し、培養液を得た。その後、上記培養液を遠心分離することで菌体を沈殿させ、培養上製を回収した。回収した培養上清を限外ろ過膜(メルクミリポア社、AMICON ULTRA-15 3 KDa cutoff)を用いて10倍に濃縮することで、菌体調製物を得た。ATCC-11454およびNITE ABP-03481については、上述のNITE BP-03198等と同様の方法で、菌体調製物を得た。ネガティブコントロールとして乳酸菌用培地(MRS Broth)をラクトコッカス・ガルビエを含む菌液に添加した。各乳酸菌株が有するプランタリシンおよびナイシンの種類、ならびにラクトコッカス・ガルビエに対する抗菌活性の結果を表13に示す。
合成したプランタリシンがラクトコッカス・ガルビエI型(TUMSAT-K9408株)およびII型(TUMSAT-LgN1株)の増殖を抑制するかを検証した。実験8の方法は、乳酸菌の菌体調製物の代わりに合成プランタリシンを用いたこと以外、実験7と同じである。各合成プランタリシンは、一般的な有機合成の手法を用いて合成した。プランタリシンAを含む溶液(プランタリシンA)、プランタリシンEとプランタリシンFとを1:1の比率(モル比)で混合した溶液(プランタリシンEF)、プランタリシンJとプランタリシンKとを1:1の比率(モル比)で混合した溶液(プランタリシンJK)またはプランタリシンNC8αとプランタリシンNC8βとを1:1の比率(モル比)で混合した溶液(プランタリシンNC8αβ)を、それぞれラクトコッカス・ガルビエを含む菌液に添加し、25℃で8時間震盪培養した。培養後のラクトコッカス・ガルビエを含む菌液を寒天培地に滴下し、ラクトコッカス・ガルビエのコロニーが形成されるかを観察した。結果を表15に示す。
ナイシンA精製品がラクトコッカス・ガルビエI型(TUMSAT-K9408株)およびII型(TUMSAT-LgN1株)の増殖を抑制するかを検証した。実験9の方法は、乳酸菌の菌体調製物や合成プランタリシンの代わりにナイシンA精製品を用いたこと以外、実験7、8と同じである。ナイシンA精製品(シグマアルドリッチ社製)を水に溶解し、遠心により不溶成分を除去することにより、ナイシンA溶液を得た。ナイシンA溶液をラクトコッカス・ガルビエを含む菌液に添加し、25℃で8時間震盪培養した。培養後のラクトコッカス・ガルビエを含む菌液を寒天培地に滴下し、ラクトコッカス・ガルビエのコロニーが形成されるかを観察した。結果を表16に示す。
ブリに乳酸菌の菌体調製物を経口投与し、レンサ球菌症による死亡が抑制されるかを検証した。ブリの稚魚(もじゃこ)を4つの試験区にわけ、下記の飼料を与えた。
(1)コントロール:通常の飼料(株式会社ヒガシマルの錦江3号)
(2)NITE BP-03198の培養上清の膜ろ過濃縮液に飼料を浸漬したもの(10倍濃縮液0.3mL/g)
(3)NITE ABP-03481の培養上清の塩析濃縮液に飼料を浸漬したもの(10倍濃縮液0.3mL/g)
(4)NITE BP-03198の生菌懸濁液に飼料を浸漬したもの(1.2×109cfu/g)
NITE BP-03198の培養上清の膜ろ過濃縮液は、次のように調製した。まず、当該乳酸菌をMRS Brothで温度30℃で培養後、遠心分離によって菌体を沈殿させて培養上清を得た。得られた培養上清を精密ろ過膜(Synder社、PES30)を用いて10倍に濃縮して当該培養上清の膜ろ過濃縮液を得た。NITE ABP-03481の培養上清の塩析濃縮液は、次のように調製した。まず、当該培養上清に終濃度5.5Mとなるように塩化ナトリウムを加え、4℃で一晩攪拌した。その後、当該培養上清を遠心分離することで沈殿物を取得した。取得した沈殿物を10μM塩酸水溶液(pH5)に溶解して当該培養上清の塩析濃縮液を得た。上記塩析濃縮液は、塩化ナトリウムを加える前の培養上清を基準として10倍に濃縮した液に相当する。生菌懸濁液は、次のように調製した。NITE BP-03198をMRS液体培地で温度30℃下で培養し、凍結グリセロールストック(4×109cfu/mL、3mL/tube)を作製した。凍結グリセロールストックを解凍し、遠心分離によって上清を除いた後、3mLのPBSで再懸濁して生菌懸濁液を調製した。

(a)配列番号1~7のいずれかに記載のアミノ酸配列、
(b)配列番号1~7のいずれかに記載のアミノ酸配列に対して、85%以上の同一性を有するアミノ酸配列、または
(c)配列番号1~7のいずれかに記載のアミノ酸配列に対して、1以上15以下のアミノ酸残基が欠損、置換、挿入および/または付加されたアミノ酸配列。 - プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を含む、魚類の生物の飼育用組成物。
- プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を含む、ラクトコッカス・ガルビエによる魚類の病気に対する予防または治療用組成物。
- 前記プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌は、NITE-BP-03198、NITE-BP-03199、NITE-BP-03200、NITE-BP-03201、NITE-BP-03202およびNITE-ABP-03481から選択される少なくとも1種である、請求項5または6に記載の組成物。
- 前記プランタリシン遺伝子は、プランタリシンA遺伝子、プランタリシンE遺伝子、プランタリシンF遺伝子、プランタリシンJ遺伝子、プランタリシンK遺伝子、プランタリシンNC8α遺伝子およびプランタリシンNC8β遺伝子から選択される少なくとも1つであり、
前記ナイシン遺伝子は、ナイシンA遺伝子およびナイシンZ遺伝子から選択される少なくとも1つである、請求項5または6に記載の組成物。 - 前記魚類は、スズキ目またはサケ目である、請求項1、2、5及び6のいずれか1項に記載の組成物。
- 飼料または飼料添加剤である、請求項1、2、5及び6のいずれか1項に記載の組成物。
- 飼育水または飼育水添加剤である、請求項1、2、5及び6のいずれか1項に記載の組成物。
- 注射剤である、請求項1、2、5及び6のいずれか1項に記載の組成物。
- 請求項1、2、5及び6のいずれか1項に記載の組成物を、魚類の生物に与えることを含む、魚類の生物の飼育方法。
- プランタリシンおよびナイシンから選択される少なくとも1つを魚類の生物に投与することを含む、魚類の生物の飼育方法。
- プランタリシンおよびナイシンから選択される少なくとも1つを魚類の生物に投与することを含む、ラクトコッカス・ガルビエによる魚類の病気に対する予防または治療方法。
- 前記プランタリシンは、プランタリシンA、プランタリシンE、プランタリシンF、プランタリシンJ、プランタリシンK、プランタリシンNC8αおよびプランタリシンNC8βから選択される少なくとも1つであり、
前記ナイシンは、ナイシンAおよびナイシンZから選択される少なくとも1つである、請求項14または15に記載の方法。 - 前記プランタリシンは、(a)~(c)のいずれかの配列を含み、かつ抗菌活性を有するペプチドを含む、請求項14または15に記載の方法;
(a)配列番号1~7のいずれかに記載のアミノ酸配列、
(b)配列番号1~7のいずれかに記載のアミノ酸配列に対して、85%以上の同一性を有するアミノ酸配列、または
(c)配列番号1~7のいずれかに記載のアミノ酸配列に対して、1以上15以下のアミノ酸残基が欠損、置換、挿入および/または付加されたアミノ酸配列。 - プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を魚類の生物に投与することを含む、魚類の生物の飼育方法。
- プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を含む魚類の生物に投与することを含む、ラクトコッカス・ガルビエによる魚類の病気に対する予防または治療方法。
- 前記プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌は、NITE-BP-03198、NITE-BP-03199、NITE-BP-03200、NITE-BP-03201、NITE-BP-03202およびNITE-ABP-03481から選択される少なくとも1種である、請求項18または19に記載の方法。
- 前記プランタリシン遺伝子は、プランタリシンA遺伝子、プランタリシンE遺伝子、プランタリシンF遺伝子、プランタリシンJ遺伝子、プランタリシンK遺伝子、プランタリシンNC8α遺伝子およびプランタリシンNC8β遺伝子から選択される少なくとも1つであり、
前記ナイシン遺伝子は、ナイシンA遺伝子およびナイシンZ遺伝子から選択される少なくとも1つである、請求項18または19に記載の方法。 - 前記投与は、経口投与、浸漬投与または注射投与である、請求項14、15、18及び19のいずれか1項に記載の方法。
- 前記魚類は、スズキ目またはサケ目である、請求項14、15、18及び19のいずれか1項に記載の方法。
- プランタリシンおよびナイシンから選択される少なくとも1つを含む、ラクトコッカス・ガルビエに対する増殖抑制剤または殺菌剤。
- プランタリシンおよびナイシンから選択される少なくとも1つをラクトコッカス・ガルビエに接触させることを含む、ラクトコッカス・ガルビエの増殖抑制または殺菌方法。
- プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物を含む、ラクトコッカス・ガルビエに対する増殖抑制剤または殺菌剤。
- プランタリシン遺伝子およびナイシン遺伝子から選択される少なくとも1つを有する細菌の菌体もしくは菌体培養物またはこれらの抽出物をラクトコッカス・ガルビエに接触させることを含む、ラクトコッカス・ガルビエの増殖抑制または殺菌方法。
- 受領番号がNITE-ABP-03481である細菌。
- 請求項28に記載の細菌の菌体もしくは菌体培養物またはこれらの抽出物。
