Description
본 발명은 위양성 신호가 제거된 항원 검출방법 및 키트에 관한 것으로, 더욱 상세하게는 포획항체에 부착되는 포획스트랜드와 검출항체에 부착되는 검출스트랜드 사이에 프렛효과를 이용하여 바이오마커를 용이하게 검출할 수 있는 위양성 신호가 제거된 항원 검출방법 및 키트에 관한 것이다.
종래 면역분석(immunoassay)에 있어서, 포획항체를 이용하여 항원을 기판 표면에 고정하고, 겨자무 과산화효소(horseradish peroxidase, HRP) 또는 형광물질 등이 표지되어 있는 검출항체를 항원과 결합시킨 후 HRP에 의한 버퍼의 색깔 변화 또는 형광물질의 밝기 등을 이용해 항원의 양을 측정하는 방법이 자주 사용되어 왔다.
하지만 검출항체는 항원으로 사용되는 바이오마커에 특이적으로 결합하는 것이 대부분이지만, 일부 검출항체의 경우 세척에 의하여 완전히 기판에서 제거되지 않거나 비특이적 결합에 의해 기판 표면과 같이 항원이 아닌 다른 곳에 결합할 수 있으며, 그 결과 위양성(false positive) 신호로 나타나 측정된 항원의 양을 부정확하게 만들 수 있다.
이런 위양성 신호는 시료 내에 항원(바이오마커)의 양이 많은 경우는 무시할 수 있지만, 바이러스 항원 검출 등 항원의 양이 적은 경우에는 큰 오차를 발생시킬 수 있는 문제가 있다. 도 1은 종래 면역분석에 있어서 발생할 수 있는 위양성 문제를 나타내는 도면이다. 특히 이러한 위양성 신호의 경우 바이러스성 질환 또는 암진단과 같이 항원의 양이 극히 적은 질병을 진단할 때 진단의 정확도를 떨어트리는 요인으로 작용할 수 있다.
따라서 항원의 양이 적을 경우 다량의 채혈에 따른 피검자의 심리적, 신체적 부담을 줄이고 진단의 정확성을 높이기 위해 높은 민감도를 가지면서 위양성 신호가 없는 신규한 기술의 개발이 요구된다.
전술한 문제를 해결하기 위하여, 본 발명은 포획항체에 부착되는 포획스트랜드와 검출항체에 부착되는 검출스트랜드 사이에 프렛효과를 이용하여 바이오마커를 용이하게 검출할 수 있는 위양성 신호가 제거된 항원 검출방법 및 키트를 제공하고자 한다.
상술한 문제를 해결하기 위해, 본 발명은 기판; 상기 기판 위에 부착될 수 있으며, 형광물질이 표지된 포획스트랜드를 구비한 포획항체; 상기 포획스트랜드의 염기서열 일부 또는 전부와 상보적이며, 형광물질이 표지된 검출스트랜드를 구비한 검출항체; 및 버퍼를 포함하되, 상기 버퍼는 상기 포획스트랜드와 상기 검출스트랜드가 상보적 결합을 수행할 수 있는 농도 미만의 이온농도를 가지는 것인 위양성 신호가 제거된 항원 검출용 키트를 제공한다.

또한 본 발명은 검출스트랜드와 포획스트랜드 사이의 상보적 결합을 형광관찰 직전에 수행하는 것으로 항원 없이 검출스트랜드와 포획스트랜드가 결합하여 나타내는 위양성 신호를 최소화할 수 있는 항원 검출용 키트를 제공할 수 있다.

이하에서는 본 발명의 바람직한 실시예를 상세하게 설명한다. 본 발명을 설명함에 있어서 관련된 공지 기술에 대한 구체적인 설명이 본 발명의 요지를 흐리게 할 수 있다고 판단되는 경우 그 상세한 설명을 생략하기로 한다. 명세서 전체에서, 단수의 표현은 문맥상 명백하게 다르게 뜻하지 않는 한 복수의 표현을 포함하는 것으로 이해되어야 하고, “포함하다” 또는 “가지다”등의 용어는 기술되는 특징, 숫자, 단계, 동작, 구성요소, 부분품 또는 이들을 조합한 것이 존재함을 지정하려는 것이지, 하나 또는 그 이상의 다른 특징들이나 숫자, 단계, 동작, 구성요소, 부분품 또는 이들을 조합한 것들의 존재 또는 부가 가능성을 미리 배제하지 않는 것으로 이해되어야 한다. 또, 방법 또는 제조 방법을 수행함에 있어서, 상기 방법을 이루는 각 과정들은 문맥상 명백하게 특정 순서를 기재하지 않은 이상 명기된 순서와 다르게 일어날 수 있다. 즉, 각 과정들은 명기된 순서와 동일하게 일어날 수도 있고 실질적으로 동시에 수행될 수도 있으며 반대의 순서대로 수행될 수도 있다.
본 명세서에 개시된 기술은 여기서 설명되는 구현예들에 한정되지 않고 다른 형태로 구체화될 수도 있다. 단지, 여기서 소개되는 구현예들은 개시된 내용이 철저하고 완전해질 수 있도록 그리고 당업자에게 본 기술의 기술적 사상이 충분히 전달될 수 있도록 하기 위해 제공되는 것이다. 도면에서 각 장치의 구성요소를 명확하게 표현하기 위하여 상기 구성요소의 폭이나 두께 등의 크기를 다소 확대하여 나타내었다. 전체적으로 도면 설명시 관찰자 시점에서 설명하였고, 일 요소가 다른 요소 위에 위치하는 것으로 언급되는 경우, 이는 상기 일 요소가 다른 요소 위에 바로 위치하거나 또는 그들 요소들 사이에 추가적인 요소가 개재될 수 있다는 의미를 모두 포함한다. 또한, 해당 분야에서 통상의 지식을 가진 자라면 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 본 발명의 사상을 다양한 다른 형태로 구현할 수 있을 것이다. 그리고 복수의 도면들 상에서 동일 부호는 실질적으로 서로 동일한 요소를 지칭한다.
본 명세서에서, '및/또는' 이라는 용어는 복수의 기재된 항목들의 조합 또는 복수의 기재된 항목들 중의 어느 항목을 포함한다. 본 명세서에서, 'A 또는 B'는, 'A', 'B', 또는 'A와 B 모두'를 포함할 수 있다.
본 명세서에서 "바이오마커"는 생명체의 정상 또는 병리적인 상태를 확인할 수 있는 생체물질을 모두 포함한다.
본 명세서에서 "프렛"은 인접한 2개의 형광물질의 공명에 의해 에너지가 전달되는 메커니즘을 의미한다. 구체적으로는, 빛에 의해 여기(excited)된 형광물질의 에너지가 인접한 다른 형광물질에 전달되는 현상을 일컫는 용어로, 당업자가 인식하는 통상적인 프렛의 의미를 모두 포함하는 것일 수 있다.
본 명세서에서 "프렛 효율"은 빛에 의해 여기(excited)된 형광물질의 에너지가 인접한 다른 형광물질에 전달되는 현상을 이용하여, 형광물질 사이의 거리에 따라 상이한 에너지 전달 효율을 값으로 나타낸 것을 의미하고, 당업자가 인식하는 통상적인 프렛 효율의 의미를 모두 포함하는 것일 수 있다.
본 명세서에서 "도너"는 빛을 받아 여기되면 에너지를 전달하는 형광물질을 의미하고, 2 이상의 형광물질이 인접하는 경우, 상대적으로 짧은 파장의 빛을 흡수 또는 방출하는 형광물질을 의미한다.
본 명세서에서 "억셉터"는 여기상태의 도너로부터 에너지를 전달받는 형광물질을 의미하고, 2 이상의 형광물질이 인접하는 경우, 상대적으로 긴 파장의 빛을 흡수 또는 방출하는 형광물질을 의미한다.
본 명세서에서 "스트랜드"는 생체고분자의 사슬 또는 생체고분자를 모사하여 합성한 고분자의 사슬로서, 상기 생체고분자는 단일가닥 또는 이중가닥 구조를 가질 수 있으며, 핵산 또는 핵산 유사체를 포함한다. 상기 핵산은 디옥시리보핵산(DNA) 또는 리보핵산(RNA)을 포함하고, 상기 핵산 유사체는 펩타이드 핵산(PNA), 잠금 핵산(LNA), 모폴리노 핵산(MNA), 글리콜 핵산(GNA), 트레오스 핵산(TNA)을 포함한다.
최근 단분자 형광현미경 기술의 개발로 분자를 하나씩 관찰할 수 있게 되었고 이를 바이오마커 검출에 적용함으로써 민감도는 fg/ml 수준으로 1000배 가량 증가하게 되었다(Nature 473, 484-488, 2011). 통상적으로 사용하는 형광 이미징에서는 관찰하려는 분자에 형광물질을 고정하여, 서로 다른 종류의 바이오마커를 형광신호를 이용해 구분하는 방법을 사용한다. 이때 각각 서로 다른 색의 형광물질을 고정하고 색깔 차이를 이용해 바이오마커의 종류를 구분하는 방법을 사용하나, 통상의 이미지 센서로 구분하여 관찰할 수 있는 형광물질의 종류는 3~4가지 정도이다.
따라서, 단분자 형광현미경 기술을 사용할 경우, 민감도는 fg/ml로 기존 방법에 비해 1000배 가량 증가하나 검출할 수 있는 바이오마커의 종류는 여전히 3~4 가지 정도로 제한된다.
한편, 단분자 형광현미경의 높은 민감도를 유지하면서 다양한 타겟 물질을 동시에 검출하는 방법으로서, FRET(Fluorescence Resonance Energy Transfer)에 기초한 DNA-PAINT 기술인 FRET-PAINT 기술이 제공된다. FRET이란 최초에 여기된 도너(donor)의 여기된 상태의 에너지가 억셉터(acceptor)로 전달되는 현상을 의미한다. 도너 물질은 일반적으로 억셉터 물질에 비해 더 짧은 파장의 빛을 방출하게 되는데, 이때의 방출 파장은 억셉터의 흡수 파장과 중첩(spectral overlap)된다. 에너지 전달 속도와 효율은 도너의 방출 파장과 억셉터의 흡수 파장의 중첩의 정도, 도너의 양자 효율, 도너와 억셉터의 전이 쌍극자의 상대적인 배열 정도, 그리고 도너와 억셉터 사이의 거리 등에 의존하게 된다.
예를 들어 FRET-PAINT 기술에 따르면, 도킹스트랜드에 두 가지 DNA 결합 자리들을 가질 수 있으며, 각각 도너스트랜드 및 억셉터스트랜드를 위한 자리가 될 수 있다. 단일 분자 국지화(single-molecule localization)를 위해서 억셉터의 FRET 신호가 사용된다. FRET-PAINT 기술을 사용하면 억셉터가 직접 여기되지 않고 FRET에 의해 여기되어 이미저(도너 및 억셉터) 농도를 수십~수백배 높일 수 있으므로 DNA-PAINT에 비해 이미징 스피드가 수십~수백 배 향상될 수 있다. (대한민국 등록특허 제2195625호))
본 명세서에 개시된 기술 역시 FRET-PAINT 기술을 기반으로 하여 높은 민감도와 정확도를 갖는 항원 검출방법 및 키트를 제공하고자 한다. 하지만 배경기술에서 소개한 바와 같이 FRET-PAINT 기술을 사용하여 항원을 검출할 경우에도 위양성 신호의 문제는 여전히 존재할 수 있으므로 이러한 문제를 개선하기 위해 예를 들어 포획항체와 검출항체 각각에 서로 프렛쌍(FRET pair)을 형성할 수 있는 형광물질이 도입된 스트랜드를 결합시키는 방법을 사용할 수 있다. 즉 항원과 결합된 위치에 검출항체와 포획항체가 둘 다 있어야 프렛이 검출될 수 있어 위양성 신호가 발생하지 않는다고 기대할 수 있다. 하지만 이 경우에도 또 다른 위양성 문제가 발생할 수 있다.
도 2는 FRET-PAINT 기술을 사용하여 항원을 검출하는 과정에서 위양성 문제가 발생하는 경우를 나타낸다. 도 2의 (a)에서, 검출항체에는 한 종 이상의 형광물질이 한 개 이상 표지되어 있는 한 개 이상의 검출스트랜드가 결합되어 있고, 포획항체에는 한 종 이상의 형광물질이 한 개 이상 표지되어 있는 한 개 이상의 포획스트랜드가 결합되어 있다.
이때 검출스트랜드와 포획스트랜드의 염기서열 일부 또는 전부는 상보적이며, 검출스트랜드와 포획스트랜드가 상보적으로 결합할 경우 검출스트랜드에 표지되어 있는 형광물질과 포획스트랜드에 표지되어 있는 형광물질은 한 개 이상의 프렛쌍(FRET pair)을 형성한다.
따라서 프렛 신호가 검출될 경우 해당 위치에는 검출항체와 포획항체가 모두 존재함을 알 수 있다. 도 2의 (b) 및 (c)는 검출스트랜드와 포획스트랜드가 상보적으로 결합함으로써 형성되는 프렛쌍의 예들을 나타낸다. 도 2의 (b) 및 (c)와 같이 검출스트랜드에는 도너 형광물질이 표지되고 포획스트랜드에는 억셉터 형광물질이 표지되는 것이 바람직하나 이에 한정되는 것은 아니다.
한편 도 2의 (b)와 같이 도너와 억셉터로 각각 한 종의 형광물질이 사용될 수도 있지만, 도 2의 (c)와 같이 도너와 억셉터로 각각 2종 이상의 형광물질이 사용될 수도 있다.
하지만 포획스트랜드와 검출스트랜드 간의 프렛 신호를 관찰하여 항원을 검출하는 방법을 사용할 경우에도 도 2의 (d)와 같이 검출스트랜드와 포획스트랜드의 염기서열은 상보적이므로 검출항체와 포획항체 사이에 항원이 존재하지 않아도 자발적 결합이 가능하며, 이는 위양성 신호의 원인이 될 수 있다.
이에 본 발명의 경우 상기 포획스트랜드와 상기 검출스트랜드 사이의 결합을 형광관찰 시점 직전에 수행하는 것으로 항원 없이 포획스트랜드와 검출스트랜드가 상보적으로 결합하는 것을 최소화하여 위양성 신호의 발생을 최소화할 수 있는 항원 검출용 키트를 제공할 수 있다.
본 발명은 기판; 상기 기판 위에 부착될 수 있으며, 형광물질이 표지된 포획스트랜드를 구비한 포획항체; 상기 포획스트랜드의 염기서열 일부 또는 전부와 상보적이며, 형광물질이 표지된 검출스트랜드를 구비한 검출항체; 및 버퍼를 포함하되, 상기 버퍼는 상기 포획스트랜드와 상기 검출스트랜드가 상보적 결합을 수행할 수 있는 농도 미만의 이온농도를 가지는 것인 위양성 신호가 제거된 항원 검출용 키트에 관한 것이다.
상기 기판은 타겟 항원을 포획하고 관찰하기 위한 영역으로 그 형태는 특별히 제한되지 않으나, 평판, 구형 입자, 막대형 구조, 및 기타 비정형 구조의 형태를 가질 수 있으며 그 재질도 유리, 석영, 플라스틱 등으로 이루어질 수 있다. 상기 기판은 통상 유리 재질로 된 평판 형태의 슬라이드 글라스나 커버슬립일 수 있다. 바람직하게는 상기 기판은 #1 또는 #1.5 커버슬립일 수 있다. 상기 기판은 상기 포획항체를 부착할 수 있도록 폴리에틸렌글리콜-비오틴(PEG-Biotin)과 같은 유기물로 표면처리된 것일 수 있다.
상기 기판은 비특이적 결합은 방지하되 포획항체는 특이적으로 결합할 수 있도록 처리되어 있을 수 있다, 이때 비특이적 결합 방지를 위하여 상기 기판의 표면은 폴리에틸렌글리콜, BSA 등이 코팅될 수 있으며, 포획항체의 특이적 결합 유도하기 위하여 비오틴-폴리에틸렌글리콜, 비오틴-BSA, Anti-DIG 항체 등을 코팅할 수 있다.
상기 포획항체는 바이러스나 바이오마커와 같은 항원을 특이적으로 결합하여 포획할 수 있는 항체로서, 상기 포획항체의 표면에 비오틴(biotin)을 도입하고 상기 기판에는 비오틴과 결합하는 아비딘(avidin), 뉴트라비딘(neutravidin) 또는 스트렙타비딘(strepavidin)을 도입하여 상기 포획항체를 상기 기판에 부착시킬 수 있다. 상기 검출항체는 상기 포획항체에 결합되어 있는 항원과 특이적으로 결합할 수 있는 항체이다.
본 명세서에서 상기 포획항체와 검출항체로서 사용되는 '항체'는 구체적으로 단일클론 항체(모노클로날 항체, 완전 길이 단일클론항체 포함), 다클론 항체(폴리클로날 항체), 다중특이 항체(예를 들어 이중특이 항체), 및 항체 단편(예를 들어, 가변 영역 및 목적하는 생물 활성을 나타내는 항체의 다른 부분)을 포함할 수 있다. 또한 본 명세서에서의 항체는 모노클로날 항체 및 폴리클로날 항체를 모두 포함하고, 키메라 항체, 인간화된 항체, 인간항체를 모두 포함할 수 있다. 바람직하게는 Fab, F(ab)2, Fab', F(ab')2, Fv, 디아바디(diabody), 나노바디(nanobody) 또는 scFv를 포함할 수 있으며, 보다 바람직하게는 scFv, Fab, 나노바디(nanobody) 및 면역글로불린 분자일 수 있다. 본 명세서에서 “항체”는 면역글로불린 외에도 타겟 물질과 특이적 결합을 할 수 있어 상기 항체와 동일한 기능을 할 수 있는 핵산, 핵산 유사체, 펩타이드로 구성된 압타머(aptamer)를 포함한다.
본 명세서에서, "스트랜드"는 일반적인 형태, 형광물질 부착이 가능하도록 기능기가 도입된 형태, 또는 형광물질이 이미 부착된 형태 등 다양한 형태를 가질 수 있다. 상기 스트랜드에 기능기(functional group)를 도입하면 원하는 위치에 원하는 형광물질을 부착할 수 있으며 하나의 스트랜드에 동일 또는 상이한 기능기를 여러 개 부착시킬 수도 있다. 스트랜드의 어느 위치에 도너와 억셉터를 부착하느냐에 따라 도너와 억셉터 사이의 거리가 달라지고 FRET 효율 또한 달라질 수 있다. 이러한 성질을 이용하면 타겟 항원의 다중 검출이 가능하다.
상기 형광물질은 자외선, 전기에너지, 열에너지 등의 외부 에너지에 의해 여기되어 그 에너지를 빛으로 바꾸는 물질로서, 유기 형광체 또는 무기 형광체를 포함할 수 있다. 상기 유기 형광체의 예는 로다민(Rhodamine), 알렉사(Alexa) Fluor dye, 플루오레세인(fluorescein), FITC(fluorescein isothiocyanate), FAM(5-carboxy fluorescein), ATTO dye, BODIPY, CF dye, Cyanine(Cy) dye, DyLight Fluor 및 텍사스 레드(Texas Red)일 수 있고, 상기 무기 형광체의 예는 양자점을 들 수 있다.
상기 포획스트랜드와 상기 검출스트랜드가 상보적으로 결합할 때 상기 포획스트랜드의 형광물질과 상기 검출스트랜드의 형광물질이 서로 프렛쌍(FRET pair)을 형성하게 된다. 상기 포획스트랜드의 형광물질과 상기 검출스트랜드의 형광물질은 상기 포획스트랜드와 상기 검출스트랜드의 특정 염기(base), 3'- 또는 5'-말단(end), 또는 기본골격(backbone) 상에서 서로 도너(donor) 또는 억셉터(acceptor)로 작용할 수 있다. 이때 상기 프렛쌍으로부터 발생하는 형광 신호로부터 하기 식으로 정의되는 FRET 효율을 측정할 수 있다.
FRET 효율 = (억셉터가 내는 빛의 세기)/(도너와 억셉터가 내는 빛의 세기 합)
또한 상기 포획스트랜드와 상기 검출스트랜드는 한쌍의 프렛쌍을 형성할 수도 있지만, 두쌍 이상의 프렛쌍을 형성할 수 있다. 이를 통하여 상기 FRET에 의한 신호의 세기를 강하게 하거나 도너 또는 억셉터의 종류를 상이하게 하는 것으로 다종의 바이오마커를 동시에 구분하는 것도 가능하다(도 5 참조).
한편 상기 포획스트랜드, 검출스트랜드는 바람직하게는, DNA, RNA, PNA, LNA, MNA, GNA, TNA와 같은 핵산 또는 이의 유사체일 수 있다.
상기 버퍼는 상기 포획항체 및 상기 검출항체의 혼합을 용이하도록 할 뿐만 아니라 상기 항체들의 보관을 용이하게 할 수 있도록 첨가되는 물질로서 상기 버퍼는 PBS버퍼, 트리스(Tris)버퍼, HEPES버퍼, MOPS버퍼, 바이카보네이트버퍼, 또는 MES버퍼를 사용할 수 있다. 이때 버퍼는 항체를 안정화시키는 역할을 수행하므로 항체를 불안정하게 하지 않는 어떠한 버퍼를 사용해도 무방하다.
일반적으로 상기 스트랜드를 구성하는 DNA의 경우 상보적으로 형성되는 다른 DNA가닥과 수소결합을 통하여 DNA 쌍을 형성할 수 있지만, 각 DNA가닥은 전기적으로 음성을 가지고 있어 각 DNA가닥 사이는 척력을 형성하게 된다. 기존의 FRET-PAINT와 같은 형광 관찰법에 사용되는 버퍼의 경우 이러한 DNA가닥 사이의 척력을 극복하고 수소결합을 수행하기 위하여 다량의 양이온을 포함하게 된다.
하지만 위에서 살펴본 바와 같이 상기 버퍼에 다량의 양이온이 포함되는 경우 상기 포획스트랜드와 상기 검출스트랜드 사이에 상보적인 결합이 수행되어 항원 없이도 FRET쌍을 형성할 수 있으며, 이는 위양성 신호를 나타낼 수 있다(도 2의 (d) 참조)
따라서 본 발명에서는 이러한 스트랜드 사이의 결합을 최소화 할 수 있도록 상기 버퍼는 상기 포획스트랜드와 상기 검출스트랜드가 상보적 결합을 수행할 수 있는 농도 미만의 이온농도를 가지는 것이 바람직하다. 이를 통하여 상기 포획 스트랜드와 상기 검출스트랜드 사이의 상보적 결합은 추가적인 양이온 공급이전 까지 억제될 수 있다(도 3의 (a)). 즉 상기 포획항체와 상기 검출항체를 포함하는 항원 검출용 키트에 바이오마커를 포함하는 항원을 투입하고 항원-항체 반응을 통하여 상기 포획항체-항원-검출항체의 결합이 완료되는 시점까지 상기 검출스트랜드와 상기 포획스트랜드 사이의 상보적인 결합이 억제될 수 있다. 이후 세척과정을 통하여 미결합된 검출항체를 제거할 수 있으며, 적절한 시점에 양이온을 공급하는 것으로 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적 결합을 유도할 수 있다(도 3의 (b)). 이를 통하여 상기 항원에 부착된 포획항체 및 검출항체 사이에서만 FRET 발광이 나타날 수 있다.
이를 위하여 상기 버퍼는 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적 결합이 수행될 수 있는 농도 미만의 이온농도를 가질 수 있으며, 구체적으로는 상기 버퍼는 100mM 미만의 양이온 농도를 가질 수 있다. 일반적으로 DNA와 같은 핵산 또는 핵산유도체 사이의 상보적인 결합을 위해서는 100mM 이상의 양이온 농도가 필요한 것으로 알려져 있다. 따라서 상기 버퍼의 경우 100mM 미만의 양이온 농도를 가짐에 따라 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적인 결합이 억제될 수 있다. 이때 상기 버퍼의 양이온 농도는 상기 포획스트랜드와 상기 검출스트랜드로 사용되는 핵산 또는 핵산유도체의 종류, 스트랜드의 길이, 염기서열, 버퍼의 온도에 따라 상이한 농도를 가질 수 있다.
상기 항원 검출용 키트는 항원이 공급되어 상기 포획항체 및 상기 검출항체와 결합된 이후, 양이온을 포함하는 버퍼가 공급되어 상기 포획스트랜드와 상기 검출스트랜드가 상보적으로 결합할 수 있다. 상기와 같이 검출스트랜드와 포획스트랜드 사이의 상보적 결합은 상기 버퍼의 낮은 이온 농도로 인하여 억제될 수 있다(도 3의 (a)). 하지만 이러한 억제가 지속되는 경우 상기 포획스트랜드와 상기 검출스트랜드 사이의 FRET 발광이 수행되지 않을 수 있으므로, 적절한 시점에 양이온을 포함하는 버퍼를 공급하여 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적인 결합을 유도할 수 있다(도 3의 (b)).
이를 구체적으로 살펴보면 상기 포획항체 및 상기 검출항체를 포함하는 기판에 상기 항원이 공급되면 항원-항체반응을 통하여 검출항체-항원-포획항체로 구성되는 복합체를 형성할 수 있다. 이 시점에서는 상기 버퍼내의 이온농도가 낮게 유지됨에 따라 상기 포획스트랜드와 상기 검출스트랜드의 상보적인 결합이 억제될 수 있다.
이후 상기 기판을 세척하게 되면 미결합된 검출항체가 제거될 수 있으며, 이때 기존의 형광 분석법과는 달리 상기 포획스트랜드와 상기 검출스트랜드가 상보적인 결합을 수행하지 않으므로, 항원 없이 상기 포획스트랜드와 상기 검출스트랜드가 결합된 복합체의 형성은 최소화될 수 있다.
상기와 같은 세척이 완료된 이후 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적인 결합을 위하여 양이온을 공급할 수 있으며, 이때 공급되는 양이온은 양이온 단독으로 공급되거나 버퍼와 혼합되어 상기 버퍼를 대체하는 방식으로 공급될 수 있다. 이때 상기 포획스트랜드와 상기 검출스트랜드는 상보적인 결합을 수행할 수 있으며, 여기광을 공급하는 경우 FRET발광을 수행할 수 있다. 이때 사용되는 양이온은 상기 포획스트랜드 및 검출스트랜드 사이의 상보적인 결합을 유도할 수 있는 양이온이라면 제한 없이 사용될 수 있지만, 나트륨이온, 칼륨이온 또는 마그네슘이온이 사용될 수 있다. 아울러 이러한 양이온의 공급을 위하여 염화나트륨, 염화칼륨 또는 염화마그네슘이 사용될 수 있다.
물론 이 경우에도 미결합된 포획항체 및 검출항체가 상기 기판에 일부 남아 있을 수 있지만, 포획항체에서 발산되는 형광의 경우 필터를 이용하여 간단히 제거할 수 있으며, 미결합된 검출항체의 경우 FRET를 이용한 발광을 수행하지 못하므로, 위양성 신호를 최소화 하는 것이 가능하다(도 4 참조).
아울러 본 발명의 경우 위에서 살펴본 바와 같이 원하는 시점까지 FRET발광을 지연시킬 수 있으므로 양이온 농도를 순차적으로 조절하는 것으로 각 FRET 쌍의 발광시점을 조절할 수 있는 장점을 가질 수 있다. 기존의 형광분석법의 경우 대부분 각 형광이 동시에 발광을 수행하지만 본 발명의 경우 양이온 농도를 조절하는 것으로 각 형광의 발광시점을 시간에 따라 조절할 수 있다. 이를 통하여 각 항원의 종류를 더욱 용이하게 구별하는 것이 가능하다.
본 발명은 또한 (a) 형광물질이 표지된 포획스트랜드를 구비한 포획항체를 준비하는 단계; (b) 상기 포획스트랜드의 염기서열 일부 또는 전부와 상보적이며, 형광물질이 표지된 검출스트랜드를 구비한 검출항체를 준비하는 단계; (c) 상기 포획항체 및 상기 검출항체를 제1 버퍼와 혼합하는 단계; (d) 항원을 포함하는 시료를 도입하여, 상기 항원, 상기 포획항체 및 상기 검출항체 간에 항원-항체 반응을 유도하는 단계; (e) 제2 버퍼를 도입하여 상기 포획스트랜드와 상기 검출스트랜드를 상보적으로 결합시키는 단계; 및 (f) 상기 포획스트랜드와 상기 검출스트랜드의 결합에 의해 생성되는 형광 신호를 측정하는 단계를 포함하는 위양성 신호가 제거된 항원 검출방법에 관한 것이다.
포획항체나 검출항체에 스트랜드를 부착하는 방법은 당업계에 공지된 통상적인 단백질 분자와 핵산 분자를 결합시키는 방법에 의해서 수행될 수 있다. 예를 들어, 두 개의 서로 다른 반응기를 동시에 가지고 있는 화합물을 사용하여 핵산 분자와 단백질 분자 간의 결합 반응을 수행하여 부착할 수 있다. 일례로 SMCC (succinimidyl 4-(N-maleimidomethyl)cyclohexane-1-carboxylate)는 아민(amine)에 반응하는 NHS-ester와 싸이올(thiol)에 반응하는 말레이미드(maleimide) 그룹을 가지고 있는데, 싸이올기가 부착되어 있는 스트랜드를 SMCC와 반응시킨 후 그 결과물을 검출항체와 반응시키면 검출항체의 N-말단 또는 라이신에 있는 아민기와 반응하여 검출항체에 스트랜드를 붙일 수 있다.
상기와 같이 스트랜드가 부착된 항체(포획항체 및 검출항체)가 준비된 이후 제1 버퍼를 공급하여 혼합할 수 있다. 이때 상기 제1 버퍼의 경우 위에서 살펴본 바와 같이 포획스트랜드와 검출스트랜드 사이의 상보적인 결합을 억제하기 위하여 낮은 이온농도를 가지는 버퍼를 사용할 수 있다. 즉 본 단계에서는 상기 포획 스트랜드와 상기 검출스트랜드 사이의 상보적인 결합이 억제되어 항원 없이 상기 포획항체와 상기 검출항체가 결합되는 것이 최소화 될 수 있다.
상기 시료는 검출 대상이 되는 타겟 물질을 함유한 것이라면 그 종류가 특별히 제한되지 않으나, 조직, 혈액, 혈청, 혈장, 타액, 점막액, 뇨 등일 수 있으며, 예를 들어 질환이 의심되는 동물 또는 인간 개체로부터 분리 수득된 것일 수 있다.
상기 항원은 바이러스, 박테리아, 핵산, 펩타이드, 단백질, 소포체, miRNA, 엑소좀, 순환종양세포, 바이오마커 등일 수 있다. 바람직하게는 상기 항원은 바이오마커일 수 있다.
본 명세서에 개시된 기술은 세포, 조직 및 기관 등을 관찰하는 데 사용될 수 있다. 특히 본 명세서에 개시된 기술이 한 번의 검사에서 여러 종의 생체물질을 검출하여 신속하게 질병을 조기진단할 수 있다는 점에서 바이오마커 검출용으로 매우 유용하게 사용될 수 있다.
상기 바이오마커는 생물처리과정, 병원성을 일으키는 과정, 치료를 위한 약리학적 과정의 측정 또는 평가 등의 통상적인 과학 또는 의료분야에서 사용되는 것이면 제한 없이 이용가능하고, 특별히 제한되는 것은 아니다.
상기 바이오마커는 예를 들어 혈액, 타액, 소변 등의 생체액에서 검출할 수 있는 폴리펩티드, 펩티드, 핵산, 단백질 또는 대사물질일 수 있으며, 구체적으로는 AFP(alpha fetoprotein), CA15-3, CA27-29, CA19-9, CA-125, 칼시토닌(calcitonin), 칼레티닌(calretinin), CEA(carcinoembryonic antigen), CD34, CD99, MIC-2, CD117, 크라모그라닌(chromogranin), 사이토케라틴(cytokeratin, various types: TPA, TPS, Cyfra21-1), 데스민(desmin), EMA(epithelial membrane antigen), Factor VIII, CD31, FL1, GFAP(glial fibrillary acidic protein), GCDFP-15(gross cystic disease fluid protein), HMB-45, hCG(human chorionic gonadotropin), 면역글로불린(immunoglobulin), 인히빈(inhibin), 케라틴(keratin, various types of keratin), 백혈구 마커(lymphocyte marker), MART-1(Melan-A), Myo D1, MSA (muscle-specific actin), 뉴로필라멘트(neurofilament), NSE(neuron-specific enolase), PLAP(placental alkaline phosphatase), PSA(prostate-specific antigen), PTPRC(CD45), S100 protein, SMA(smooth muscle actin), 시냅토파이신(synaptophysin), TK(thymidine kinase), Tg(thyroglobulin), TTF-1(thyroid transcription factor-1), M2-PK, 바이멘틴(vimentin), 인터루킨(interleukin), CD24, CD40, 인테그린(integrin), 시스타틴(cystatin), 인터페론(interferon), TNF(tumor necrosis factor), MCP, VEGF, GLP, ICA, HLA-DR, ICAM, EGFR, FGF, BRAF, GFEB, FRS, LZTS, CCN, 뮤신(mucin), 렉틴(lectin), 아포리포단백질, 티로신, 신경세포 접착분자 유사 단백질, 파이브로넥틴, 포도당, 요산, 탄산탈수효소 또는 콜레스테롤 등일 수 있다.
또한 상기 시료는 상기 바이오마커가 포함되지 않은 용액으로 희석될 수 있으며, 상기 시료에 포함된 상기 바이오마커의 농도는 종류에 따라 fg/ml부터 mg/ml까지 매우 크게 차이가 날 수 있다. 바람직하게는 이미지 센서에 적당한 수의 바이오마커 분자가 검출되도록 다양한 비율로 희석될 수 있다. 바람직하게는 서로 다른 비율로 희석된 동일한 시료에 대해서 수행될 수 있으며, 가장 바람직하게는 1:10 내지 1,000의 중량비로 수 차례 희석하여 사용할 수 있다.
상기와 같이 항원이 공급되면 항원-항체반응을 통하여 포획항체-항원-검출항체의 복합체를 구성할 수 있다. 이때 상기 포획항체는 상기 항원과 결합되기 이전 상기 기판에 부착되어 있거나 상기 항원과 결합된 이후 상기 기판에 부착될 수 있다. 즉 상기 기판에 부착된 상기 포획항체에 상기 항원 및 상기 검출항체가 부착될 수 있으며, 이와는 별도로 포획항체-항원-검출항체의 복합체를 구성한 다음 상기 기판에 부착되는 것도 가능하다.
상기 포획항체-항원-검출항체의 복합체가 상기 기판상에 형성된 이후 미결합된 포획항체 및 검출항체를 제거하기 위하여 상기 기판은 1~10회 세척될 수 있다. 이때 사용되는 세척액으로는 물 또는 상기 제1 버퍼를 함유하는 용액일 수 있다.
상기와 세척이 완료된 이후 제2 버퍼를 공급하여 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적인 결합을 수행할 수 있다. 상기 제2 버퍼의 경우 상기 포획스트랜드와 상기 검출스트랜드 사이의 상보적인 결합을 유도할 수 있는 충분한 양의 양이온이 포함될 수 있으며, 이를 통하여 상기 포획스트랜드와 상기 검출스트랜드사이의 FRET 쌍을 형성하는 것이 가능하다.
즉 본 발명의 경우 상기 제2 버퍼의 주입시점까지 상기 스트랜드 사이의 상보적인 결합이 억제될 수 있으므로, 항원 없이 상기 검출항체와 상기 포획항체가 결합되어 위양성 신호가 형성되는 것을 최소화 할 수 있다. 또한 위에서 살펴본 바와 같이 원하는 시점에 FRET 쌍을 형성할 수 있으므로, 양이온 농도를 조절하여 각 FRET 쌍의 발광시점을 조절하는 것 역시 가능하다.
또한 상기 제2 버퍼는 상기와 같은 양이온을 공급하기 위하여 염화나트륨, 염화칼륨 또는 염화마그네슘을 포함할 수 있다.
한편, 상기 형광 신호의 측정은 단일분자 현미경을 이용하여 이미징함으로써 수행될 수 있다. 서로 다른 두 종류의 형광물질이 매우 가까이 위치할 때 한 형광물질(도너)의 에너지가 다른 형광물질(억셉터)로 전달되는 현상을 FRET이라 하는데, 상기 포획스트랜드와 상기 검출스트랜드가 상보적으로 결합할 때 상기 포획스트랜드의 형광물질과 상기 검출스트랜드의 형광물질이 서로 프렛쌍(FRET pair)을 형성할 수 있다. 따라서 상술한 프렛쌍의 형성에 의해 형광 신호가 나타나는 곳에 타겟 항원이 위치함을 알 수 있다.
결론적으로 상술한 위양성 신호가 제거된 항원 검출용 키트 및 항원검출방법에 따르면, 다중진단이 용이하고 민감도가 높은 FRET-PAINT 기술을 이용하면서도 버퍼 내의 양이온 농도를 조절하는 것에 의해 포획스트랜드와 검출스트랜드 사이의 상보적인 결합을 원하는 시점까지 억제할 수 있다. 따라서 본 발명의 경우 용이하게 위양성신호를 제거할 수 있어 검체 내 미량의 각종 바이오마커나 바이러스 등의 항원을 검출할 경우에 발생할 수 있는 오차가 대폭 감소된다.
이하, 본 발명의 바람직한 실시예를 첨부한 도면을 참조하여 당해 분야의 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 설명하기로 한다. 또한, 본 발명을 설명함에 있어 관련된 공지의 기능 또는 공지의 구성에 대한 구체적인 설명이 본 발명의 요지를 불필요하게 흐릴 수 있다고 판단되는 경우에는 그 상세한 설명을 생략하기로 한다. 그리고 도면에 제시된 어떤 특징들은 설명의 용이함을 위해 확대 또는 축소 또는 단순화된 것이고, 도면 및 그 구성요소들이 반드시 적절한 비율로 도시되어 있지는 않다. 그러나 당업자라면 이러한 상세 사항들을 쉽게 이해할 것이다.
실시예 1. 포획항체 및 검출항체 구성
CEA(암태아성항원, Carcino-Embryonic Antigen)을 검출하기 위해 억셉터인 Cy5가 표지되어 있는 포획스트랜드를 포획항체에 표지했고, 도너인 Alexa Fluor 488이 표지되어 있는 검출스트랜드를 검출항체에 표지했다. 포획스트랜드와 검출스트랜드는 각각 20개의 뉴클레오티드로 구성되며, 15개의 뉴클레오티드가 상보적으로 결합되고, 15개 중 5개가 C 또는 G이며, 나머지 5개는 TTTTT가 되어 상보적으로 결합되지 못하도록 구성했다. 포획스트랜드와 검출스트랜드가 상보적으로 결합할 경우 Alexa Fluor 488과 Cy5 사이의 거리는 6 염기쌍이 되도록 구성했다. 동시에 여러 종류의 항원을 검출하기 위해서는 다양한 도너와 억셉터를 사용할 수도 있고, 도너와 억셉터 모두를 관찰하여 프렛효율을 계산하고 이용할 수도 있으나 본 실시예에서는 한 종의 항원만을 검출하므로 한 종류의 도너와 억셉터를 사용했고, 프렛효율을 이용하지는 않았다.
실시예 2. 제1 버퍼와 제2 버퍼의 양이온 농도 확인
제1 버퍼와 제2 버퍼 모두 포획항체와 항원, 항원과 검출항체 사이의 면역반응에 의한 결합은 일어나되, 제1 버퍼에서는 포획스트랜드와 검출스트랜드 사이의 상보적 결합은 일어나지 않도록 하고, 제2 버퍼에서는 포획스트랜드와 검출스트랜드 사이의 상보적 결합은 일어나도록 하는 양이온 농도를 찾기 위한 실험을 수행했다. 제1 버퍼와 제2 버퍼 모두 트리스(Tris·HCl, pH 8.0) 버퍼를 기본으로 하되, 염화나트륨(NaCl)의 농도를 조절했다. 실험시 버퍼에 항원을 혼합하지 않았으므로 포획항체와 검출항체는 면역반응을 통해 서로 결합할 수 없다. 다만 포획항체에 표지되어 있는 포획스트랜드와 검출항체에 표지되어 있는 검출스트랜드가 서로 상보적이므로 염화나트륨의 농도가 충분히 높으면 도2 (d)와 같이 항원이 없이도 검출항체가 기판에 고정될 수 있다. 포획항체와 검출항체를 혼합한 후 포획항체를 기판에 고정하고, 검출스트랜드에 표지되어 있는 도너의 유무를 관찰함으로써 포획스트랜드와 검출스트랜드의 결합 여부를 파악할 수 있다.
다양한 염화나트륨 농도(도 6에서 (a) 60mM, (b) 40mM, (c) 20mM, (d) 10mM)를 갖는 트리스 버퍼에 항원은 제외하고, 포획항체와 검출항체만 혼합한 후 포획항체를 기판에 고정하고, 검출항체의 검출스트랜드에 표지되어 있는 도너 Alexa Fluor 488과 포획항체의 포획스트랜드에 표지되어 있는 억셉터 Cy5를 관찰하였다.
트리스 버퍼의 염화나트륨의 농도가 낮아질수록 기판에 고정된 Alexa Fluor 488의 수가 적게 관찰되었는데(도 6 참조), 이는 포획스트랜드와 검출스트랜드의 결합이 안정적이지 않아 검출항체가 기판에 잘 고정되지 않음을 의미한다. 염화나트륨의 농도가 일정 수치 이하일 경우(도 6의 (c) 및 (d)참조)에는 농도와 무관하게 Alexa Fluor 488의 수가 일정하게 나오는 것을 확인하였는데, 이는 도 1과 같이 검출항체가 기판에 비특이적으로 결합한 결과이다. 이로부터 가용한 제1 버퍼의 염화나트륨 농도 범위를 확인했다(20mM 이하).
또한 과도한 염화나트륨을 사용하는 경우 형광의 변화를 확인하기 위한 실험을 실시하였다. 도 7에서 (a) 100mM, (b) 200mM, (c) 300mM, (d) 500mM의 염과나트륨을 사용한 경우 형광의 변화를 나타낸 것이다. 염화나트륨의 농도가 높아질수록 Alexa Fluor 488의 수가 많이 관찰되었고(도 7 참조), 일정 농도 이상에서는 일정하게 나오는 것을 확인(도 7의 (c) 및 (d) 참조)하였다. 이는 포획스트랜드와 검출스트랜드가 안정적으로 결합함을 의미한다. 이로부터 가용한 제2 버퍼의 염화나트륨 농도 범위를 확인했다(300mM 이상).
마지막으로 제1 버퍼에 포획항체, 검출항체 및 항원을 모두 혼합한 후 포획항체를 기판에 고정하고, 제2 버퍼를 도입하였다. 이후 포획스트랜드와 검출스트랜드의 결합에 의해 생성되는 프렛에 의한 형광신호를 측정하였다. 동일한 위치에서 도너와 억셉터 모두 존재함을 확인했고, 도너와 억셉터 사이에 프렛이 일어남을 확인했다. 이는 도2 (b)와 같이 온전한 반응이 일어났음을 의미하며, 제1 버퍼와 제2 버퍼의 염화나트륨 농도에서 항원과 항체 사이의 면역반응이 정상적으로 일어남을 의미한다.
실시예 3. 위양성 신호 확인
제1 버퍼에 5 nM 포획항체, 5 nM 검출항체 및 다양한 농도의 항원(500fM, 200fM, 0fM)을 혼합한 후 포획항체를 기판에 고정하고, 제2 버퍼를 도입한 후 검출항체의 검출스트랜드에 표지되어 있는 도너 Alexa Fluor 488과 포획항체의 포획스트랜드에 표지되어 있는 억셉터 Cy5를 각각 관찰했다.
항원의 농도가 낮아질수록 Cy5의 수가 감소(도 8의 (a) 및 (b))했고, 항원을 넣지 않을 경우 Cy5는 관찰되지 않았다(도 8의 (c) 참조). 이를 바탕으로 본 발명에 따른 항원검출민감도는 수 fM 수준임을 알 수 있었다.
반면 항원의 농도와 무관하게 일정한 수의 Alexa Fluor 488이 관찰되었는데(도 9 참조), 이는 도 1과 같이 검출항체가 기판에 비특이적으로 결합한 결과이며, 이를 바탕으로 기존 방법의 항원검출민감도는 수백 fM 수준임을 알 수 있었다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
Claims (12)
- 기판;상기 기판 위에 부착될 수 있으며, 형광물질이 표지된 포획스트랜드를 구비한 포획항체;상기 포획스트랜드의 염기서열 일부 또는 전부와 상보적이며, 형광물질이 표지된 검출스트랜드를 구비한 검출항체; 및버퍼;를 포함하되,상기 버퍼는 상기 포획스트랜드와 상기 검출스트랜드가 상보적 결합을 수행할 수 있는 농도 미만의 이온농도를 가지는 것인 위양성 신호가 제거된 항원 검출용 키트.
- 제1항에 있어서,상기 버퍼는 PBS버퍼, 트리스(Tris)버퍼, HEPES버퍼, MOPS버퍼, 바이카보네이트버퍼, 또는 MES버퍼인 위양성 신호가 제거된 항원 검출용 키트.
- 제1항에 있어서,상기 버퍼는 100mM 미만의 양이온 농도를 가지는 위양성 신호가 제거된 항원 검출용 키트.
- 제1항에 있어서,상기 항원 검출용 키트는 항원이 공급되어 상기 포획항체 및 상기 검출항체와 결합된 이후, 양이온을 포함하는 버퍼가 공급되어 상기 포획스트랜드와 상기 검출스트랜드가 상보적으로 결합하는 것인 위양성 신호가 제거된 항원 검출용 키트.
- 제1항에 있어서,상기 포획스트랜드와 상기 검출스트랜드가 상보적으로 결합할 때 상기 포획스트랜드의 형광물질과 상기 검출스트랜드의 형광물질이 서로 프렛쌍(FRET pair)을 형성하는 것인 위양성 신호가 제거된 항원 검출용 키트.
- 제5항에 있어서,상기 포획스트랜드의 형광물질과 상기 검출스트랜드의 형광물질은 상기 포획스트랜드와 상기 검출스트랜드의 특정 염기(base), 3'- 또는 5'-말단(end), 또는 기본골격(backbone) 상에서 서로 도너(donor) 또는 억셉터(acceptor)로 작용하는 것인 위양성 신호가 제거된 항원 검출용 키트.
- 제5항에 있어서,상기 프렛쌍으로부터 발생하는 형광 신호로부터 하기 식으로 정의되는 FRET 효율을 측정하는 것인 위양성 신호가 제거된 항원 검출용 키트.FRET 효율 = (억셉터가 내는 빛의 세기)/(도너와 억셉터가 내는 빛의 세기 합)
