Description
본 발명은 HLA-G에 특이적으로 결합하는 항원 결합 도메인을 포함하는 키메라 항원 수용체, 상기 수용체가 도입된 면역세포 및 이들의 용도에 관한 것이다.
암을 치료하기 위한 방법은 꾸준한 발전과 변화 과정을 거쳐 왔으며, 외과적 수술, 화학요법, 방사선 요법 등의 방법이 현재까지 계속 이용되고 있다. 그러나 이러한 기존의 암 치료 방법들은 암이 전이되지 않은 초기 상태에만 효과가 있는 경우가 대부분이며, 이미 전이가 진행된 상태에서는 예컨대 외과적 수술을 하더라도 추후 재발의 가능성이 높다는 문제점이 있다. 이에, 최근 암을 치료하기 위해 면역 반응을 이용하는 방법에 대한 연구가 계속되어 오고 있다.
그 중에서도 면역세포를 이용하여 면역 반응을 강화시키거나 면역세포를 유전공학적으로 변형하여 다시 환자에게 주입하는 세포치료 방법에 대한 관심이 높아지고 있다. 후자의 방법에는 키메라 항원 수용체(chimeric antigen receptor, CAR)-T 세포가 많이 연구되고 있다. CAR-T 세포는 T 세포에 인공적으로 설계된 CAR 분자를 도입하여 T 세포에 표적 기능을 부여한 세포로 CAR 분자가 특정 종양 항원에 대해 특이성을 보이므로 종양 세포만을 제거할 수 있다.
일반적으로 키메라 항원 수용체 (CAR)는 세포외 항원 결합 도메인, 막관통 도메인 및 세포내 신호전달 도메인을 포함한다. 세포외 항원 결합 도메인은 확인된 종양 항원을 표적으로 하는 단일 사슬 가변 단편(single-chain variable fragment; scFv)을 포함할 수 있다.
한편, 인간 주요 조직적합성 항원, 클래스 I, G (histocompatibility antigen, class I, G)는 인간 백혈구 항원 G (Human leukocyte antigen-G, HLA-G)로도 알려져 있는 면역관문인자로 T 세포, NK 세포, 세포독성 (cytotoxic) T 세포 등의 활성 및 분열을 억제한다. HLA-G의 면역세포 수용체는 ILT2 (Ig-like transcript 2), ILT4, KIR2DL4 (Killer cell immunoglobulin like receptor, two Ig domains and long cytoplasmic tail 4) 등으로 알려져 있으며, 이 수용체들은 모두 ITIM 모티프 (immunoreceptor tyrosine-based inhibitory motif)를 가지고 있다. HLA-G가 수용체에 결합하면 이 모티프로 인해 면역세포를 억제하는 신호전달체계가 활성화된다.
일반세포에서 HLA-G의 발현은 제한적이나, 많은 종류의 암세포에서 HLA-G가 발현되는 것으로 보고되었다 (Lin A, Yan WH. Human Leukocyte Antigen-G (HLA-G) Expression in Cancers: Roles in Immune Evasion, Metastasis and Target for Therapy. Mol. Med. 21(1), 782-791 (2015)). 또한, 다수 암환자의 암세포에서 HLA-G의 발현량과 암 질병의 기수가 연관성이 있다는 보고가 있으며, HLA-G 발현 환자는 생존률이 낮다는 보고도 있다 (Ye, Sr., Yang, H., Li, K. et al. Human leukocyte antigen G expression: as a significant prognostic indicator for patients with colorectal cancer. Mod. Pathol. 20, 375-383 (2007)).
본 발명자들은 HLA-G (Human leukocyte antigen-G) 발현과 관련된 질환을 효과적으로 치료할 수 있는 키메라 항원 수용체 및 이를 포함하는 면역세포를 개발하고자 예의 연구 노력하였다. 그 결과 HLA-G에 특이적으로 결합하는 항-HLA-G 키메라 항원 수용체 및 상기 키메라 항원 수용체를 발현하는 T 세포와 NK 세포를 제작하였다. 또한, 해당 T 세포 및 NK 세포가 HLA-G를 발현하는 세포주에 대해 높은 항암 활성을 나타내는 것을 확인하여 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 항-HLA-G 키메라 항원 수용체 및 상기 수용체를 발현하는 면역세포를 제공하는 것이다.
본 발명의 다른 목적은 항-HLA-G 키메라 항원 수용체를 암호화하는 뉴클레오티드를 포함하는 핵산 및 상기 핵산을 포함하는 벡터를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 면역세포 및 약학적으로 허용되는 담체를 포함하는 약학적 조성물을 제공하는 것이다.

본 발명에서, 용어 "키메라 항원 수용체"는 면역세포 (예를 들어, T 세포)가 특정한 단백질을 인식할 수 있도록 면역세포의 수용체 부위와 암세포 표면에서 발현되는 특정한 항원의 인식부위를 조합한 수용체 단백질이다. 이 수용체들은 키메라의 성질을 띠는데 일반적으로 종양이 발현하는 항원을 인식할 수 있는 항원 결합 부위와 T 세포를 활성화시킬 수 있는 신호 전달 부위를 하나로 합치기 때문이다.
본 발명의 일 구체예에서, 상기 키메라 항원 수용체는 항원 결합 도메인 막관통 도메인 및 세포내 신호전달 도메인을 포함한다. 상기 항원 결합 도메인은 타깃 세포의 HLA-G를 인식하며, 항체 또는 항체 단편일 수 있다. 또한, 상기 항원 결합 단편은 scFv일 수 있으나 이에 제한되지 않는다.
본 발명의 일 구체예에서, 상기 항원 결합 부위는 중쇄 가변 영역 서열과 경쇄 가변 영역 서열을 포함할 수 있으며, 두 서열은 서열번호 24의 아미노산 서열을 포함하는 링커에 의해 연결될 수 있다. 따라서, 상기 항원 결합 부위는 scFv (single-chain fragment variable) 형태일 수 있다.

본 명세서에서 사용되는 용어, "핵산"은 DNA 및 RNA 분자를 포괄적으로 포함하는 의미를 가지며, 핵산 분자에서 기본 구성단위인 뉴클레오티드는 자연의 뉴클레오티드뿐만 아니라, 당 또는 염기 부위가 변형된 유사체(analogue)도 포함한다 (Scheit, Nucleotide Analogs, John Wiley, New York(1980); Uhlman 및 Peyman, Chemical Reviews(1990) 90:543-584). 본 발명의 항-HLA-G 키메라 항원 수용체를 암호화하는 핵산 분자의 서열은 변형될 수 있으며, 상기 변형은 뉴클레오티드의 추가, 결실, 또는 비보존적 치환 또는 보존적 치환을 포함한다.

본 발명의 또 다른 양태는 상기 핵산 분자를 포함하는 재조합 벡터이다.
본 발명에서 사용되는 벡터는 당 분야에 공지된 벡터를 다양하게 사용할 수 있고, 상기 항원 수용체를 생산하고자 하는 숙주세포의 종류에 따라 프로모터 (promoter), 종결자 (terminator), 인핸서 (enhancer) 등과 같은 발현조절 서열, 막 표적화 또는 분비를 위한 서열 등을 적절히 선택하고 목적에 따라 다양하게 조합할 수 있다.
본 발명의 벡터를 세포 내로 도입하고 발현시키는 방법은 관련 기술분야에 잘 알려져 있다. 벡터는 당업계에 공지된 방법에 의해 숙주 세포, 예를 들어 포유동물, 박테리아, 효모, 또는 곤충 세포 내로 쉽게 도입될 수 있다. 예를 들면, 벡터는 물리적, 화학적, 또는 생물학적 수단에 의해 숙주 세포 내로 전달될 수 있다. 상기 물리적 수단은 인산칼슘 침전, 리포펙션, 입자 폭격(particle bombardment), 미세주입, 전기천공(electroporation) 등을 포함한다. 상기 화학적 수단은 콜로이드 분산액 시스템, 예컨대 거대분자 복합체, 나노 캡슐, 마이크로스피어, 비드, 및 수중유 에멀젼, 마이셀(micelle), 혼합된 마이셀, 및 리포좀을 포함하는 지질-기반 시스템을 포함한다. 또한, 상기 생물학적 수단은 상술한 렌티바이러스, 레트로바이러스 등, DNA 또는 RNA 벡터의 사용을 포함한다.
본 발명의 벡터는 플라스미드 벡터, 코즈미드 벡터, 박테리오 파아지 벡터 및 바이러스 벡터 등을 포함하나 이에 제한되지 않는다. 적합한 벡터는 프로모터, 오퍼레이터, 개시코돈, 종결코돈, 폴리아데닐화 시그널 및 인핸서 같은 발현 조절 요소 외에도 막 표적화 또는 분비를 위한 시그널 서열 또는 리더 서열을 포함하며 목적에 따라 다양하게 제조될 수 있다.
본 발명의 일 구체예에서는 렌티-바이러스 벡터를 사용하였다. 본 발명의 구체적인 일 구현예에 있어서, 상기 벡터는 프로모터를 추가로 포함한다. 상기 프로모터는 예컨대 EF-1 프로모터일 수 있으나, 이에 한정되는 것은 아니다.
본 발명은 또한 전술한 항-HLA-G 키메라 항원 수용체를 세포 표면에 발현하는 면역세포를 제공한다.
본 발명에서 상기 면역세포는 자연 살해 세포 (natural killer cell), T 세포, 자연 살해 T 세포, B 세포, 대식세포, 수지상 세포, 자연 살해 수지상 세포, 비만세포 및 이들의 전구세포로 이루어진 군으로부터 선택될 수 있다.
상기 항-HLA-G 키메라 항원 수용체는 전술한 벡터를 통해 면역세포에 도입될 수 있으며, 도입 방법은 당 분야에서 공지된 바와 같이 숙주 세포에 따라 적합한 표준 기술을 선택할 수 있다.
본 발명의 일 구체예에서는 항-HLA-G 키메라 항원 수용체를 암호화할 수 있는 폴리뉴클레오티드를 포함하는 재조합 벡터를 T 세포 및 NK 세포에 감염시켜 키메릭 항원 수용체를 발현하는 T 세포 및 NK 세포를 제작하였다. 구체적으로 재조합 벡터에 포함된 항-HLA-G 키메라 항원 수용체 암호화 서열이 mRNA로 전사되고, 상기 mRNA로부터 항-HLA-G 키메라 항원 수용체 폴리펩티드가 번역되어 T 세포 및 NK 세포의 표면에 발현된다. 필요한 경우 재조합 벡터가 감염된 T 세포 및 NK 세포를 배양할 수 있다.
항-HLA-G 키메라 항원 수용체를 발현하는 면역세포는 HLA-G를 항원으로 인식하여 HLA-G에 특이적으로 결합할 수 있으며, 결합 후 세포내 신호전달 도메인을 통해 면역세포의 활성화를 유도하고 종양 특이적 사멸을 유도할 수 있다.
따라서, 본 발명의 또 다른 양태는 면역세포 및 약학적으로 허용되는 담체를 포함하는 암의 예방 또는 치료에서의 사용을 위한 약학적 조성물을 제공한다.
본 발명에서, 상기 약학적 조성물은 HLA-G를 과발현하는 암의 치료에 사용될 수 있으나 이에 제한되지 아니한다. 상기 암은 췌장암, 유방암, 난소암, 신경교종, 자궁경부암, 자궁내막암, 식도암, 위암, 간암, 폐암, 대장암, 비인두암, 구강암, 갑상선암, 전립선암, 신장암, 담낭암, 담관암, 혈액암 및 흑색종 등으로 이루어진 군에서 선택될 수 있다.
상기 혈액암은 급성 골수성 백혈병 (Acute Myeloid Leukemia, AML), 급성 림프모구 백혈병 (Acute Lymphoblastic Leukemia, ALL), 만성 골수성 백혈병 (Chronic Myelogenous Leukemia, CML), 다발골수종 (Multiple Myeloma, MM), 또는 림프종 (Lymphoma)일 수 있다.
본 명세서에서 사용되는 용어, "예방"은 본 발명의 조성물의 투여로 질환을 억제시키거나 진행을 지연시키는 모든 행위를 의미하며, "치료"는 질환의 발전의 억제, 경감 또는 제거를 의미한다.
본 발명의 약학 조성물은 약제학적으로 허용되는 담체를 추가로 포함할 수 있으며, 상기 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산칼슘, 알기네이트, 젤라틴, 규산칼슘, 미세결정성 셀룰로오스, 폴리비닐피롤리돈, 셀룰로오스, 물, 시럽, 메틸셀룰로오스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 암 예방 또는 치료용 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 문헌[Remington's Pharmaceutical Sciences(19th ed., 1995)]에 상세히 기재되어 있다.
또한, 본 발명에 따른 약학적 조성물은 경구투여, 주입(infusion), 정맥내 투여 (intravenous injection), 근육내 투여 (intramuscular injection), 피하 투여 (subcutaneous injection), 복강내 투여 (intraperitoneal injectoon), 직장내 투여 (Intrarectal administration), 국소 투여(topical administration), 비내 투여(intranasal injection) 등으로 투여될 수 있지만, 이에 한정되는 것은 아니다. 경구 투여시, 단백질 또는 펩타이드는 소화가 되기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화될 수 있으며, 본 발명의 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.

본 발명의 일 구체예에 따르면, 본 발명의 약학적 조성물은 1회 투여량 내에 상기 면역세포 (예컨대, T 세포 또는 자연 살해 세포)의 수가 치료 대상 내의 상기 종양세포, 예를 들어 췌장암 세포, 유방암 세포, 난소암 세포 또는 신경교종 세포 수의 1배 내지 10배로 포함된 것일 수 있다.
본 명세서에서 용어 "약제학적 유효량"은 질환의 치료, 예방 및 진단하는 데 충분한 양을 의미한다.
본 발명의 약학 조성물은 당해 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화하여 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 산제, 좌제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다.
본 발명의 또 다른 양태는 본 발명의 항-HLA-G 키메라 항원 수용체 또는 상기 키메라 항원 수용체가 발현된 면역세포 또는 상기 약학적 조성물을 약제학적 유효량으로 대상체, 예컨대 인간 또는 인간을 제외한 포유류에게 투여하는 것을 포함하는, HLA-G 과발현 관련 질환, 예를 들어 암의 진단, 예방 또는 치료 방법이다. 본 발명은 또한, 본 발명의 항-HLA-G 키메라 항원 수용체 또는 상기 키메라 항원 수용체가 발현된 면역세포를 이를 필요로 하는 개체, 예컨대 인간 또는 인간을 제외한 포유류에게 투여하는 단계를 포함하는, HLA-G 과발현 관련 질환, 예를 들어 암의 진단 방법을 제공한다.
본 발명에서, 용어 "개체"란, HLA-G 과발현 관련 질환이 발병하였거나 발병할 수 있는 인간을 포함한 원숭이, 소, 말, 양, 돼지, 닭, 칠면조, 메추라기, 고양이, 개, 마우스, 쥐, 토끼 또는 기니아 피그를 포함한 모든 동물을 의미한다.
본 발명에 따른 키메라 항원 수용체는 HLA-G에 대한 특이성 및 친화성이 우수하므로, 항-HLA-G 키메라 항원 수용체를 발현하는 면역세포는 암세포에 대한 세포독성이 우수하여 HLA-G의 고발현과 관련된 다양한 암 등의 치료에 사용될 수 있다.
도 1a 내지 도 1c는 본 발명의 HLA-G CAR(항체단편 1 CAR), HLA-G CAR(항체단편 2 CAR), 및 HLA-G CAR(항체단편 3 CAR) 컨스트럭트의 구조를 나타낸 도이다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예 1: 세포주 및 배양
인간 난소암 세포주 SK-OV-3와 인간 삼중 음성 유방암 세포주 MDA-MB-231은 한국세포주은행(Korea)으로부터 공급받아 사용하였다. SK-OV-3와 MDA-MB-231은 10% FBS(Gibco, USA)와 1% Antibiotic-Antimycotic(Gibco, USA)를 포함하는 RPMI 1640(Gibco, USA) 배지에서 배양하였다.
실시예 2: HLA-G 과발현 세포주 제작 및 배양
2-1. B2M-HLA-G-EGFP-PuroR 유전자 발현 렌티바이러스의 제조
B2M-HLA-G-EGFP-PuroR 유전자의 전달을 위한 렌티바이러스는 플라스미드 DNA 형질전환법을 이용하여 제작하였다. TransIT-293 Transfection Reagent(Mirus Bio LLC, USA)을 이용하였고, 제조사의 프로토콜에 따라 수행하였다. 실험 전날 100 ㎜ 접시에 Lenti-XTM 293T 세포주(Clontech, USA)를 5Х106개 파종하고, 다음날 pLV-EF1A-B2M-IRES-HLA-G-CMV-EGFP-T2A-PuroR 렌티바이러스 벡터(도 3), gag-pol 발현 벡터 및 VSV-G 외피(envelope) 발현 벡터를 형질전환시켰다. 세포를 약 72시간 동안 배양한 후 세포 상층액(supernatant)을 모두 수확하였다. 상층액을 0.45 um 필터(Millipore, USA)로 여과하여 세포 조각(cell debris)을 제거하고, B2M-HLA-G-EGFP-PuroR 렌티바이러스가 포함된 여과액은 사용 전까지 -80℃에 냉동 보관하였다.
2-2. B2M-HLA-G-EGFP-PuroR 유전자 발현 SK-OV-3 및 MDA-MB-231 세포주 제작 및 배양
실험 전날 100 ㎜ 접시에 SK-OV-3 및 MDA-MB-231 세포주 각각을 1Х106개씩 파종하고, 앞서 제작한 B2M-HLA-G-EGFP-PuroR 렌티바이러스를 접시 하나당 3 mL씩 새로운 배지 7 mL과 섞어서 접시에 넣어주었다. 추가로 배지에 폴리브렌(polybrene)을 8 ug/mL 농도로 첨가하였다. 그리고 24시간 후 배지를 2 ㎍/㎖ 농도의 퓨로마이신(puromycin; Sigma-aldrich, USA)이 포함된 새로운 배지로 교체하였다. 퓨로마이신에 의해 선별이 어느 정도 진행되면 HLA-G 과발현 세포를 단일 세포로 분리하기 위하여 96 웰 플레이트에서 세포를 0.5개 세포/웰 농도로 계대 배양하였다. 최종 선별이 완료된 후 배지에 퓨로마이신을 0.5 ug/mL 농도로 첨가하였다. 형광현미경으로 관찰하여 EGFP 형광 신호가 높은 몇 개의 웰을 선정하고, 해당 웰에 세포가 가득 차면 세포를 회수하여 차례대로 48웰, 24웰, 12웰, 6웰, T25 및 T75 플라스크에서 배양하였다. 최종적으로 세포를 T175 플라스크에서 배양하여 세포 스탁(stock)을 제작하였다.
2-3. B2M-HLA-G-EGFP-PuroR 유전자 발현 SK-OV-3 및 MDA-MB-231 세포주에서의 HLA-G 발현 확인
상기 2-2에서 제작한 HLA-G 과발현 세포주에서 HLA-G의 발현을 다음과 같이 확인하였다.
B2M-HLA-G-EGFP-PuroR 유전자 발현 SK-OV-3 및 MDA-MB-231 세포주 1Х105개 세포를 100 uL D-PBS(Gibco, USA)에 부유시켜 세포 샘플을 준비하였다. 상기 세포 샘플에 항-HLA-G(TANKIO17) 항체 1 ug을 첨가하여 4℃에서 30분 동안 반응시켰다. 반응이 끝나면 세포를 D-PBS로 2회 세척한 후 다시 부유시켰다. 세포 샘플에 R-Phycoerythrin AffiniPure F(ab')2 Fragment Goat Anti-Human IgG (H+L)(Jackson ImmunoResearch, USA) 0.25 ug을 첨가하여 4℃에서 30분 동안 반응시켰다. 반응 후 세포를 D-PBS로 2회 세척하고, 유세포분석법으로 HLA-G의 발현을 확인하였다. 그 결과, B2M-HLA-G-EGFP-PuroR 렌티바이러스를 전달한 SK-OV-3 세포의 표면에서는 HLA-G가 약 99.8%, MDA-MB-231 세포의 표면에서는 HLA-G가 약 98.9% 발현되고 있음을 확인하였다(도 4).
실시예 3: 키메라 항원 수용체 구축
본 발명의 키메라 항원 수용체(chimeric antigen receptor, CAR)는 인공적으로 합성되었다.
좀 더 상세하게는, 하기의 세포 외 도메인과 세포 내 도메인 코딩 서열을 SOE-PCR(Splicing by overlap extension PCR)을 통해 인공적으로 합성하여 CAR를 제작하였다 (도 1a 내지 1c). PCR 산물을 MluI 및 XbaI으로 절단한 후 3세대 자가-불활성화 렌티바이러스 발현 벡터인 pLV-EF1A-MCS 벡터의 MluI 및 XbaI 사이트에 삽입하였다 (도 2a 내지 2c).
- 세포 외 도메인: CD8α 유래 신호 서열(leader sequence, LS), HLA-G 결합 도메인 (항체단편 1, 항체단편 2 또는 항체단편 3; scFv), CD8α 유래 힌지(hinge) 및 CD8α 유래 막관통(transmembrane, TM) 도메인

본 발명의 일 예에 따른 키메라 항원 수용체를 하기 표 1에 정리하였다. 키메라 항원 수용체의 도메인들은 서로 직렬로(in tandem) 연결되며, 또한 인프레임(in frame)으로 연결된다. 상세하게는, CD8α 유래 신호 서열, HLA-G 결합 도메인(scFv 형태), CD8α 유래 힌지, CD8α 유래 막관통 도메인, 4-1BB 유래 세포 내 신호전달 도메인, CD3ζ 유래 세포 내 신호전달 도메인과 종결 코돈 (stop codon)인 TAA가 연결된 것이다.

실시예 4: 항-HLA-G CAR-T 세포의 제작
4-1. 항-HLA-G CAR 유전자 발현 렌티바이러스의 제조
항-HLA-G CAR 유전자의 전달을 위한 렌티바이러스는 플라스미드 DNA 형질전환법으로 제작하였다. TransIT-293 Transfection Reagent(Mirus Bio LLC, USA)을 이용하였고, 제조사의 프로토콜에 따라 수행하였다. 실험 전날 100 mm 접시에 Lenti-XTM 293T 세포주(Clontech, USA)를 5x106개로 파종하고, 다음날 pLV-EF1A-HLA-G CAR 렌티바이러스 벡터, gag-pol 발현 벡터 및 VSV-G 외피 발현 벡터를 형질전환시켰다. HLA-G CAR 렌티바이러스 벡터는 다음과 같다; 항체단편 1, 항체단편 2 또는 항체단편 3. 형질전환 후 세포를 약 72시간 배양하고 배양 완료 후 세포 상층액을 모두 수확하였다. 상층액을 0.45 um 필터(Millipore, USA)로 여과 세포 조각을 제거하였다. Lenti-XTM Concentrator(Clontech, USA)를 이용하여 제조사의 프로토콜에 따라 렌티바이러스를 약 100배 농축하고 사용 전까지 -80℃에 냉동 보관하였다.
4-2. 항-HLA-G CAR 발현 T 세포 제작 및 배양
공여자를 모집하고 백혈구성분채혈(leukapheresis)을 통해 백혈구를 얻었다. 상기 백혈구에서 SepMateTM-50(STEMCELL technology, Canada)과 Ficoll-Paque PLUS(GE healthcare, Sweden)를 이용하여 말초혈 단핵세포(peripheral blood mononuclear cell, PBMC)를 획득하였다. 그 후 QuadroMACSTM Separator(Miltenyi Biotec, Germany)와 LS 컬럼(Miltenyi Biotec, Germany), 인간 CD4 MicroBeads(Miltenyi Biotec, Germany) 및 인간 CD8 MicroBeads,(Miltenyi Biotec, Germany)을 이용하여 양성 선택(positive selection) 방법으로 인간 T 세포를 분리하였다. 인간 T 세포는 TexMACSTM 배지(Miltenyi Biotec, Germany)를 배양액으로 사용하고, T Cell TransActTM, human(Miltenyi Biotec, Germany)을 첨가하여 활성화시켰다. T 세포의 성장을 위해 배양액에 인간 IL-2(R&D systems, USA)를 100 U/mL로 첨가하여 배양하였다. 48시간 배양 후 활성화된 T 세포를 수확하여 렌티바이러스 전달에 사용하였다.
MOI(Multiplicity of infection, 감염 배수) 3 정도의 렌티바이러스와 함께 활성화된 인간 T 세포를 12 웰 플레이트에 웰 당 1x106개를 넣어주고 배양액이 2.5 mL이 되도록 하였다. 1,000 xg로 15분간 원심분리하여 T 세포에 렌티바이러스를 전달하였다. T 세포를 48시간 동안 정치 배양한 후 새로운 배지로 교체하였다. 형질도입된 인간 T 세포는 2~3일 간격으로 mL당 5x105개로 계대 배양하고, 세포 수가 mL당 2x106개를 넘지 않도록 유지하였다. 특별한 언급이 없는 한, 인간 T 세포는 배양액에 IL-2(R&D systems, USA)를 100 U/mL로 첨가하여 배양하였다.
4-3. 항-HLA-G CAR 발현 T 세포에서 CAR 발현 확인
3명의 공여자로부터 얻은 백혈구로 제작한 항-HLA-G CAR 발현 T 세포에서 세포 표면에 항-HLA-G CAR가 발현하는지 확인하였다.
1x105개의 세포를 수확한 후 100 uL D-PBS에 부유시켜 세포 부유액을 준비하였다. PE conjugation kit(Abcam, USA)를 사용하여 항-HLA-G 항체 제작에 사용한 B2M-HLA-G 항원에 PE 형광을 접합시켰다. 상기 B2M-HLA-G 항원-PE 접합체를 세포 부유액에 첨가하여 4℃에서 30분 동안 반응시켰다. 반응 종료 후 세포를 D-PBS로 2회 세척하고, 유세포분석법으로 항-HLA-G CAR의 발현 수준을 확인하였다.
그 결과, 공여자 1에서는 항-HLA-G CAR(항체단편 1 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 34.9%, 항-HLA-G CAR(항체단편 2 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 45.8%, 항-HLA-G CAR(항체단편 3 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 43.3% 발현되고 있음을 확인하였다(도 5a).
그 결과, 공여자 2에서는 항-HLA-G CAR(항체단편 1 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 37.6%, 항-HLA-G CAR(항체단편 2 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 57.3%, 항-HLA-G CAR(항체단편 3 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 38.1% 발현되고 있음을 확인하였다(도 5b).
그 결과, 공여자 5에서는 항-HLA-G CAR(항체단편 1 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 30.4%, 항-HLA-G CAR(항체단편 2 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 46.2%, 항-HLA-G CAR(항체단편 3 CAR) 유전자를 전달한 T 세포의 표면에서 항-HLA-G CAR가 약 37.4% 발현되고 있음을 확인하였다(도 5c).
실시예 5: 생체 외(
in vitro
)에서 항-HLA-G CAR 발현 T 세포의 항암 효능 확인
다음의 방법을 통해 타깃 세포(target cell, T)에 대한 항-HLA-G CAR 발현 T 세포(효과기 (effector) 세포, E)의 항암 효능을 확인하였다.
(i) 타깃 세포들은 모두 EGFP를 과발현할 수 있도록 제작되었다.
(ii) 타깃 세포들을 48 웰 플레이트에 웰 당 1x105 세포/150 uL로 첨가하였다.
(iii) 항-HLA-G CAR 발현 T 세포 및 대조군 T 세포를 웰 당 1x106 세포/150 uL, 5x105 세포/150 uL, 1x105 세포/150 uL(E:T 비율=10, 5, 1)로 첨가하여 타깃 세포와 24시간 반응시켰다.
(iv) 타깃 세포가 죽게 되면 EGFP 형광 신호가 사라지기 때문에 24시간 후에 유세포분석법을 이용하여 EGFP를 발현하는 세포의 수를 카운팅하였다.

실험 결과, 공여자 1의 백혈구로부터 제작한 3종 항-HLA-G CAR(항체단편 1 CAR), 항-HLA-G CAR(항체단편 2 CAR), 항-HLA-G CAR(항체단편 3 CAR) 발현 T 세포의 HLA-G 과발현 SK-OV-3-HLA-G 세포주에 대한 항암 효능은 항-HLA-G CAR를 발현하지 않는 대조군 T 세포에 비해 상당히 높았다. 그러나 HLA-G를 발현하지 않는 SK-OV-3 세포주에 대해서는 3종의 항-HLA-G CAR 발현 T 세포와 대조군 T 세포의 살상능에 차이가 없었다 (도 6a).
공여자 2의 백혈구로부터 제작한 3종 항-HLA-G CAR(항체단편 1 CAR), 항-HLA-G CAR(항체단편 2 CAR), 항-HLA-G CAR(항체단편 3 CAR) 발현 T 세포의 HLA-G 과발현 MDA-MB-231-HLA-G 세포주에 대한 항암 효능은 항-HLA-G CAR를 발현하지 않는 대조군 T 세포에 비해 상당히 높았다. 반면, HLA-G를 발현하지 않는 MDA-MB-231 세포주에 대해서는 3종의 항-HLA-G CAR 발현 T 세포와 대조군 T 세포의 살상능 차이가 없었다 (도 6b).
공여자 5의 백혈구로부터 제작한 3종 항-HLA-G CAR(항체단편 1 CAR), 항-HLA-G CAR(항체단편 2 CAR), 항-HLA-G CAR(항체단편 3 CAR) 발현 T 세포의 HLA-G 과발현 SK-OV-3-HLA-G 및 MDA-MB-231-HLA-G 세포주에 대한 항암 효능도 항-HLA-G CAR를 발현하지 않는 대조군 T 세포에 비해 상당히 높았다. 그러나 HLA-G를 발현하지 않는 SK-OV-3 및 MDA-MB-231 세포주에 대해서는 3종의 항-HLA-G CAR 발현 T 세포와 대조군 T 세포의 살상능 차이가 없었다 (도 6c).
상기 결과들을 통해 항-HLA-G CAR 발현 T 세포가 HLA-G를 높게 발현하는 세포에 대해서만 특이적으로 높은 살상능을 가짐을 확인하였다.
공여자 5에서 제작한 항-HLA-G CAR 발현 T 세포 및 대조군 T 세포가 타깃 세포와 E:T 비율 1:1로 처리된 실험군에서 세포 상층액을 수확하였다. 수확한 상층액에서 Human IFN-gamma Quantikine ELISA Kit(R&D systems, USA)로 인터페론 감마의 농도를 측정하였다.
그 결과, 3종의 항-HLA-G CAR(항체단편 1 CAR), 항-HLA-G CAR (항체단편 2 CAR), 항-HLA-G CAR (항체단편 3 CAR) 발현 T 세포와 HLA-G를 과발현하는 SK-OV-3-HLA-G 또는 MDA-MB-231-HLA-G 타깃 세포주를 공배양한 경우에만 인터페론 감마의 분비가 유의적으로 증가함을 확인하였다(도 7).
실시예 6: 생체 외(
in vitro
)에서 T 세포 배양 조건에 따른 항-HLA-G CAR 발현 T 세포의 항암 효능 비교
앞서 실시예 4-2에서 언급한 방식으로 항-HLA-G CAR(항체단편 2 CAR) 발현 T 세포를 제작하였다. 이 때, T 세포의 성장을 위해 배양액에 인간 IL-2(R&D systems, USA)를 100 U/mL로 첨가하거나 또는 IL-7 (Miltenyi Biotec, Germany) 및 IL-15 (Miltenyi Biotec, Germany)를 각각 12.5 ng/mL로 첨가하여 배양하였다.
항-HLA-G CAR 발현 T 세포의 세포 표면에서 항-HLA-G CAR가 발현되는지 확인한 결과, 배양액에 IL-2를 넣어 배양한 항-HLA-G CAR 발현 T 세포의 표면에서는 항-HLA-G CAR가 약 48.3% 발현되고 있었으며, 배양액에 IL-7과 IL-15를 넣어 배양한 항-HLA-G CAR 발현 T 세포의 표면에서는 항-HLA-G CAR가 약 47.9% 발현되고 있음을 확인하였다(도 8).
항-HLA-G CAR 발현 T 세포와 타깃 세포를 공배양하여 항암 효능을 확인한 결과, HLA-G를 과발현하는 SK-OV-3-HLA-G 세포주에 대한 항-HLA-G CAR 발현 T 세포(IL-2)와 항-HLA-G CAR 발현 T 세포 (IL-7/IL-15)의 항암 효능은 유사하게 우수하였다. 반면, HLA-G를 발현하지 않는 SK-OV-3 세포주에 대해서는 살상능이 상당히 낮아 2종의 항-HLA-G CAR 발현 T 세포 간에 살상능의 차이가 없었다(도 9).
항-HLA-G CAR 발현 T 세포가 타깃 세포와 E:T 비율 1:1로 처리된 실험군에서 세포 상층액을 수확하여 Human IFN-gamma Quantikine ELISA Kit (R&D systems, USA)로 인터페론 감마의 농도를 측정하였다. 그 결과, 항-HLA-G CAR 발현 T 세포 (IL-2) 또는 항-HLA-G CAR 발현 T 세포 (IL-7/IL-15)와 HLA-G를 과발현하는 SK-OV-3-HLA-G 타깃 세포주를 공배양 했을 경우에만 인터페론 감마의 분비가 유의적으로 증가하였고, 2종 항-HLA-G CAR 발현 T 세포 간에 차이가 거의 없음을 확인하였다(도 10).
상기 결과들을 통해 T 세포의 성장을 위해 배양액에 첨가하는 사이토카인(IL-2 또는 IL-7/IL-15)의 종류에 관계없이 항-HLA-G CAR 발현 T 세포가 유사한 수준의 CAR 발현을 보이고, 생체 외(in vitro)에서 2종 항-HLA-G CAR 발현 T 세포 모두 좋은 항암 효능을 나타내는 것을 확인하였다.
실시예 7: 항-HLA-G CAR-NK 세포의 제작 및 항암 효능 확인
7-1. 항-HLA-G CAR 발현 NK 세포 제작 및 배양
공여자를 모집하여 백혈구성분채혈 (leukapheresis)을 통해 백혈구를 얻었다. 얻은 백혈구로부터 SepMateTM-50 (STEMCELL technology, Canada)과 Ficoll-Paque PLUS (GE healthcare, Sweden)를 이용하여 말초혈 단핵세포 (peripheral blood mononuclear cell, PBMC)를 획득하였다. 그 후 NK cell isolation kit (Miltenyi Biotec, Germany)를 이용하여 음성 선택(negative selection) 방법으로 인간 NK(natural killer, 자연 살해) 세포를 분리하였다. 인간 NK 세포는 NK MACS® Medium (Miltenyi Biotec, Germany)을 배양액으로 사용하였다. NK 세포의 효율적인 성장을 위해 배양액에 GemCellTM Human Serum AB (GeminiBio, USA)를 5%, 인간 IL-2 (R&D systems, USA)와 IL-15 (Miltenyi Biotec, Germany)를 각각 500 U/mL, 140 U/mL 농도로 첨가하여 배양하였다.
NK 세포에 렌티바이러스를 효율적으로 전달하기 위하여 RetroNectin®(Takara, Japan)과 BX-795 (Sigma-aldrich, USA)를 사용하였다. MOI 10 정도의 렌티바이러스를 RetroNectin이 코팅된 24 웰 플레이트에 넣고, 제조사의 프로토콜에 따라 렌티바이러스를 흡착시켰다. 배양 10일차의 NK 세포 5x105개에 2.5 uM의 BX-795를 넣어 30분간 처리하였다. 이후 렌티바이러스가 흡착된 플레이트에 BX-795가 처리된 NK 세포를 넣고 1,000 xg로 15분간 원심분리하여 NK 세포에 렌티바이러스를 전달하였다. 그 후 NK 세포를 24시간 동안 정치 배양 후 새로운 배지로 교체하였다. 형질도입된 인간 NK 세포는 3~4일 간격으로 적절히 계대 배양하였다.
7-2. 항-HLA-G CAR 발현 NK 세포에서의 CAR 발현 확인
우선 항-HLA-G CAR 발현 NK 세포가 제대로 배양됐는지 확인하였다. 1Х105개의 세포를 수확한 후 100 uL D-PBS에 부유시켜 세포 부유액을 준비하였다. VioBlue가 접합된 CD3 항체 (Miltenyi Biotec, Germany)와 FITC가 접합된 CD56 항체 (Miltenyi Biotec, Germany)를 상기 세포 부유액에 첨가하고 4℃에서 30분 동안 반응시켰다. 반응 후 세포를 D-PBS로 2회 세척한 후, 유세포분석법으로 CD3와 CD56의 발현 수준을 확인하였다.
그 결과, CD3-CD56+인 NK 세포의 비율이 대조군 NK 세포에서 99.1%, 항-HLA-G CAR(항체단편 1 CAR) 유전자를 전달한 NK 세포에서 99.1%, 항-HLA-G CAR(항체단편 2 CAR) 유전자를 전달한 NK 세포에서 98.9%, 항-HLA-G CAR(항체단편 3 CAR) 유전자를 전달한 NK 세포에서 99.1%로 나타나 거의 모든 세포가 CD3-CD56+ NK 세포임을 확인하였다 (도 11).
그 다음으로 각 세포의 표면에서 항-HLA-G CAR가 발현하는지 확인하였다. 1Х105개의 세포를 수확한 후 100 uL D-PBS에 부유시켜 세포 부유액을 준비하였다. PE conjugation kit (Abcam, USA)를 사용하여 항-HLA-G 항체 제작에 사용한 B2M-HLA-G 항원에 PE 형광을 접합시켰다. 상기 B2M-HLA-G 항원-PE 접합체를 세포 부유액에 첨가하여 4℃에서 30분 동안 반응시켰다. 반응 종료 후 세포를 D-PBS로 2회 세척하고, 유세포분석법으로 항-HLA-G CAR의 발현을 확인하였다.
그 결과, 항-HLA-G CAR (항체단편 1 CAR) 유전자를 전달한 NK 세포의 표면에서 항-HLA-G CAR가 약 18.8%, 항-HLA-G CAR (항체단편 2 CAR) 유전자를 전달한 NK 세포의 표면에서 항-HLA-G CAR가 약 25.5%, 항-HLA-G CAR (항체단편 3 CAR) 유전자를 전달한 NK 세포의 표면에서 항-HLA-G CAR가 약 22.4% 발현되고 있음을 확인하였다(도 12).
7-3. 생체 외(
in vitro
)에서의 항-HLA-G CAR 발현 NK 세포의 항암 효능 확인
실시예 5와 같은 방법으로 항-HLA-G CAR 발현 NK 세포의 항암 효능을 확인하였다.
그 결과, HLA-G를 과발현하는 SK-OV-3-HLA-G 세포주에 대한 3종의 항-HLA-G CAR (항체단편 1 CAR), 항-HLA-G CAR (항체단편 2 CAR), 항-HLA-G CAR (항체단편 3 CAR) 발현 NK 세포의 항암 효능은 항-HLA-G CAR를 발현하지 않는 대조군 NK 세포에 비해 상당히 높았다. 그러나 SK-OV-3 세포주에서는 항-HLA-G CAR 발현 NK 세포와 대조군 NK 세포의 살상능 차이가 없었다 (도 13).
Claims (22)
- HLA-G (human leukocyte antigen-G)에 특이적으로 결합하는 항원 결합 도메인;막관통 도메인; 및세포내 신호전달 도메인;을 포함하고,상기 HLA-G에 특이적으로 결합하는 항원 결합 도메인은 하기에서 선택되는 서열을 포함하는, 항-HLA-G 키메라 항원 수용체(chimeric antigen receptor, CAR):(a) 서열번호 31의 아미노산 서열을 갖는 중쇄 상보성 결정 부위 1 (complementarity determining region 1, CDR1), 서열번호 32의 아미노산 서열을 갖는 중쇄 CDR2 및 서열번호 33의 아미노산 서열을 갖는 중쇄 CDR3를 포함하는 중쇄 가변 영역; 및서열번호 34의 아미노산 서열을 갖는 경쇄 CDR1, 서열번호 35의 아미노산 서열을 갖는 경쇄 CDR2 및 서열번호 36의 아미노산 서열을 갖는 경쇄 CDR3를 포함하는 경쇄 가변 영역;(b) 서열번호 37의 아미노산 서열을 갖는 중쇄 CDR1, 서열번호 38의 아미노산 서열을 갖는 중쇄 CDR2 및 서열번호 39의 아미노산 서열을 갖는 중쇄 CDR3를 포함하는 중쇄 가변 영역; 및서열번호 40의 아미노산 서열을 갖는 경쇄 CDR1, 서열번호 41의 아미노산 서열을 갖는 경쇄 CDR2 및 서열번호 42의 아미노산 서열을 갖는 경쇄 CDR3를 포함하는 경쇄 가변 영역; 또는(c) 서열번호 43의 아미노산 서열을 갖는 중쇄 CDR1, 서열번호 44의 아미노산 서열을 갖는 중쇄 CDR2 및 서열번호 45의 아미노산 서열을 갖는 중쇄 CDR3를 포함하는 중쇄 가변 영역; 및서열번호 46의 아미노산 서열을 갖는 경쇄 CDR1, 서열번호 47의 아미노산 서열을 갖는 경쇄 CDR2 및 서열번호 48의 아미노산 서열을 갖는 경쇄 CDR3를 포함하는 경쇄 가변 영역.
- 제1항에 있어서, 상기 HLA-G에 특이적으로 결합하는 항원 결합 도메인은 항체 또는 항원 결합 단편인, 항-HLA-G 키메라 항원 수용체.
- 제2항에 있어서, 상기 항원 결합 단편은 scFv인, 항-HLA-G 키메라 항원 수용체.
- 제1항에 있어서, 상기 키메라 항원 수용체는 신호 서열을 추가로 포함하는, 항-HLA-G 키메라 항원 수용체.
- 제4항에 있어서, 상기 신호 서열은 CD8 알파, 인간 면역글로불린 중쇄 또는 hGM-CSF 수용체 알파-사슬 유래 신호 서열인, 항-HLA-G 키메라 항원 수용체.
- 제5항에 있어서, 상기 CD8 알파 유래 신호 서열은 서열번호 8의 아미노산 서열을 포함하는 것인, 항-HLA-G 키메라 항원 수용체.
- 제1항에 있어서, 상기 HLA-G에 특이적으로 결합하는 항원 결합 도메인은 힌지에 의해 막관통 도메인과 연결되는, 항-HLA-G 키메라 항원 수용체.
- 제7항에 있어서, 상기 힌지는 CD8 알파의 힌지, IgG1의 힌지, IgG4의 힌지, IgD의 힌지, IgG1 CH3, CD28의 세포외 도메인 또는 이들의 조합으로 이루어진 군에서 선택되는, 항-HLA-G 키메라 항원 수용체.
- 제8항에 있어서, 상기 CD8 알파의 힌지는 서열번호 10의 아미노산 서열을 포함하는, 항-HLA-G 키메라 항원 수용체.
- 제1항에 있어서, 상기 막관통 도메인은 CD8 알파, T 세포 수용체, CD28, CD3 엡실론(epsilon), CD45, CD4, CD5, CD9, CD16, CD22, CD33, CD37, CD64, CD80, CD86, CD99, CD134, CD137 및 CD154의 알파(α), 베타(β) 또는 제타(ζ)로 이루어진 군으로부터 선택되는 단백질의 막관통 도메인인, 항-HLA-G 키메라 항원 수용체.
- 제10항에 있어서, 상기 CD8 알파의 막관통 도메인은 서열번호 12의 아미노산 서열을 포함하는, 항-HLA-G 키메라 항원 수용체.

- 제1항에 있어서, 상기 세포내 신호전달 도메인은 OX40, CD2, CD27, CD28, CD99, ICAM-1, LFA-1 (CD11a/CD18), ICOS (CD278) 및 4-1BB (CD137)로 이루어진 군으로부터 선택된 단백질의 신호전달 도메인을 공동자극 도메인으로 추가적으로 포함하는, 항-HLA-G 키메라 항원 수용체.
- 제14항에 있어서, 상기 4-1BB 유래 공동자극 도메인은 서열번호 14의 아미노산 서열을 포함하는, 항-HLA-G 키메라 항원 수용체.
- 제1항 내지 제15항 중 어느 한 항에 따른 항-HLA-G 키메라 항원 수용체를 암호화하는 뉴클레오티드 서열을 포함하는 단리된 핵산.
- 제16항의 단리된 핵산을 포함하는 벡터.
- 제1항 내지 제15항 중 어느 한 항에 따른 항-HLA-G 키메라 항원 수용체를 표면에 발현하는, 면역세포.
- 제18항에 있어서, 상기 면역세포는 자연 살해 세포 (natural killer cell), T 세포, 자연 살해 T 세포, B 세포, 대식세포, 수지상 세포, 자연 살해 수지상 세포, 비만세포 및 이들의 전구세포로 이루어진 군으로부터 선택되는 것인, 면역세포.
- 제18항 또는 제19항의 면역세포 및 약학적으로 허용되는 담체를 포함하는 암의 예방 또는 치료에서의 사용을 위한 약학적 조성물.
- 제20항에 있어서, 상기 암은 췌장암, 유방암, 난소암, 신경교종, 자궁경부암, 자궁내막암, 식도암, 위암, 간암, 폐암, 대장암, 비인두암, 구강암, 갑상선암, 전립선암, 신장암, 담낭암, 담관암, 혈액암 및 흑색종으로 이루어진 군으로부터 선택되는, 약학적 조성물.
