Abstract
본 발명은 미산성 차아염소산수 및 피트산(phytic acid)을 혼합하여 제조한 것으로, 기존 차아염소산만이 함유된 항균제는 pH 5~6의 범위에서 2분간 처리하여야 99.9% 이상의 내생포자를 사멸시킬 수 있지만, 본 발명의 내생포자 사멸용 조성물을 pH 4~7의 범위에서 2분간 처리하면 99.9% 이상의 내생포자를 사멸시킬 수 있다.
Description
본 발명은 미산성 차아염소산수 및 피트산(phytic acid)을 혼합하여 제조한 것으로, 차아염소산(HOCl)을 10~80mg/L, 피트산(phytic acid)을 4~500mg/L 농도로 포함하여 살균력이 우수한 내생포자 사멸용 조성물에 관한 것이다.
최근 위생에 대한 관심의 증가로 인해 항균제에 대한 수요가 증가하고 있으며, 항균제에 주로 사용되는 항균물질은 차아염소산(HOCl)을 포함하는 차아염소산수가 있다.
일반적으로 차아염소산수라 함은, 차아염소산을 포함하는 살균 소독수로서 일반적으로 인체에 무해한 물질(GRAS: GenerallyRecognized As Safe)로 미국 FDA로 부터 분류된 목록에 기재되어 있는 물질이며, 염소계 살균소독제로서 강력한 산화제이다.
차아염소산수의 살균, 소독력은 유리잔류염소에 의해 결정된다. 유리잔류염소는 Cl2(aq), 차아염소산(HOCl)과 염소산이온(OCl-)으로 존재하며, 유리잔류염소의 존재 형태는 pH에 의해 그 존재 형태가 달라진다. pH4.0 이하에서는 Cl2(aq)의 형태로 약산 및 중성영역에서는 차아염소산(HOCl)형태로, 알카리영역에서는 염소산이온(OCl-)의 형태로 존재하며, 차아염소산(HOCl)의 살균력이 염소산이온(OCl-)보다 상당히 우수한 것으로 알려져 있다.
이에 차아염소산을 포함하는 항균제는 pH에 따라 급격하게 소독효율이 달라지게 되는데, 특히 포자류를 제어하기에는 한계를 가지고 있다. 따라서, pH에 따른 소독 효율 변화가 없는 항균제의 개발이 필요한 실정이다.
본 발명에서는 더욱 넓은 범위의 pH에서 우수한 내생포자 살균력을 보이는 내생포자 사멸용 조성물을 제공하고자 한다.
본 발명은 미산성 차아염소산수 및 피트산(phytic acid)을 혼합하여 제조한 것으로, 차아염소산(HOCl)을 10~80mg/L, 피트산(phytic acid)을 4~500mg/L 농도로 포함하는 것을 특징으로 하는 내생포자 사멸용 조성물을 제공한다.
본 발명의 내생포자 사멸용 조성물에 있어서, 상기 피트산은 바람직하게 피트산을 45~55%(w/w) 함유하는 수용액 상태로 혼합하는 것이 좋다.
본 발명의 내생포자 사멸용 조성물은 바람직하게 사과산과 구연산이 더욱 혼합된 것이 좋다. 이때, 본 발명의 내생포자 사멸용 조성물은 바람직하게 사과산을 0.0001~1000mg/L, 상기 구연산을 0.0001~1000mg/L 농도로 포함하는 것이 좋다.
본 발명에서는 미산성 차아염소산수에 피트산을 혼합하면, 차아염소산(HOCl)과 피트산의 시너지 효과로 내생포자 살균력이 향상되고, 더욱 넓은 범위의 pH에서 높은 내생포자 제어효율을 보임을 확인하였다.

도 3은 미산성 차아염소산수(Free chlorine + HCl)와 본 발명 내생포자 사멸용 조성물(Free chlorine + 유기산)을 내생포자에 2분간 처리한 후 얻은 pH 변화에 따른 내생포자 제어효율을 보여준다.
본 발명은 미산성 차아염소산수 및 피트산(phytic acid)을 혼합하여 제조한 것으로, 차아염소산(HOCl)을 10~80mg/L, 피트산(phytic acid)을 4~500mg/L 농도로 포함하는 것을 특징으로 하는 내생포자 사멸용 조성물을 제공한다.
하기 실시예에 따르면, HCl을 사용하여 pH를 조절한 미산성 차아염소산수는 미세한 pH 변화에 의해서도 내생포자 제어능이 급격하게 변하는데, pH 5.5에서 최대 제어능을 보이다가 pH 5.0 또는 pH 6.0 정도만의 변화가 생겨도 내생포자 제어효율(조성물을 바실러스 포자에 2분간 처리하여 얻은 사멸율)이 급격히 줄어드는 것을 확인할 수 있었다 (도 3 참조). 반면, 미산성 차아염소산수에 피트산을 첨가하여 pH를 조절한 본 발명의 내생포자 사멸용 조성물은 pH 4.0~7.0의 넓은 pH 스펙트럼에서 균일한 내생포자 제어 효율을 보임을 확인할 수 있고, 이 범위 내에서 99.9% 이상의 내생포자 제어효율을 보였다.
넓은 pH 스펙트럼에서 균일한 내생포자 제어능을 보이는 본 발명의 특징은 본 발명의 내생포자 사멸용 조성물을 제조할 때, pH를 예민하게 조절할 필요가 없음 또는 현장에서 사용할 때 pH를 예민하게 조절할 필요가 없음을 의미하는데, 이는 본 발명의 제조 적성 또는 사용 적성을 매우 향상시키는 매우 유용한 기술적 특징이다. 즉, 본 발명의 미산성 차아염소산수의 제조를 위해 사용하는 원수(지하수 또는 수돗물) 또는 제조한 미산성 차아염소산수의 pH가 날씨, 계절, 제조과정 또는 유통과정 중의 여러 요인으로 비교적 넓은 범위에서 변동하더라도 본 발명 조성물이 제조스펙 상에서 제공하는 유효한 내생포자 사멸능을 비교적 넓게 유지할 수 있음을 의미하는 것이다.
한편, 본 발명의 내생포자 사멸용 조성물은 미산성 차아염소산수에 피트산을 혼합하여 제조하는 것에 특징이 있는데, 바람직하게, 차아염소산(HOCl)은 10~80mg/L 농도로 포함하고, 피트산은 바람직하게 4~500mg/L 농도로 포함하는 것이 좋다. 더욱 바람직하게는 차아염소산(HOCl)을 20~30mg/L, 피트산을 9~15mg/L의 농도로 포함하는 것이 더욱 좋다.
하기 실시예에 따르면, 20~30mg/L 차아염소산(HOCl) 함유 미산성 차아염소산수에 피트산을 9~15mg/L의 농도로 첨가시키는 경우 pH 4.0~7.0의 범위에서 99.9% 이상의 내생포자 제어효율을 보임을 확인할 수 있었다.
한편, 본 발명에서 미산성 차아염소산수는 바람직하게 3~10%(w/w) 농도의 식품첨가물 염산(식염산, HCl) 용액을 사용하여 무격막 전해조를 통해 전기분해하여 얻어진 것일 수 있다.
식염산 수용액을 전기분해하면 차아염소산(HOCl)을 함유하는 용액을 얻을 수가 있는데, HOCl은 pH의 변화에 따라 Cl2 또는 OCl- 로 변화하는 특징이 있다. 그런데, HOCl의 살균능이 가장 우수하기 때문에, 염산 등의 pH 조절제를 첨가해서 용액 중 HOCl의 농도가 가장 높은 pH 4.0~6.0으로 조정하여 미생물(균체) 살균제로 사용하고 있다 (도 1 참조).
그런데, 본 발명에서는 내생포자를 대상으로 하여 사멸능을 테스트한 결과, 기존처럼 염산을 사용하여 pH를 조절할 경우, pH 변화에 따라 내생포자 사멸능(제어능)이 매우 예민하게 크게 변화하는 것을 확인하고 본 발명에서는 이와 같은 예민한 변화를 방지할 수 있는 기술을 개발하고자 예의 노력한 것이다. 그 결과, 유기산의 일종인 피트산을 첨가하여 pH를 조절할 경우, 내생포자의 사멸능이 pH 변화에 따라 예민하게 변화하지 않음을 확인하고 본 발명을 완성한 것이다.
또한, 본 발명에서 미산성 차아염소산수는 염산(식염산, HCl) 외 염화나트륨(NaCl)을 첨가하여 제조한 것일 수 있다. 염산만을 사용하여 미산성 차아염소산수를 제조할 경우, 차아염소산(HOCl)을 최대 30mg/L 농도까지만 제조할 수 있다. 하지만, 염화나트륨(NaCl)을 첨가할 경우, 차아염소산을 80mg/L의 농도까지 제조할 수 있다.
한편, 본 발명에서 사용하는 피트산은 일 예로 피트산을 45~55%(w/w) 함유하는 수용액 상태로 내생포자 사멸용 조성물에 첨가한 것일 수 있다.
한편, 본 발명의 내생포자 사멸용 조성물은 사과산(malic acid)과 구연산(citric acid)을 더욱 포함하는 것이 좋다. 이때 사과산은 0.0001~1000mg/L, 상기 구연산은 0.0001~1000mg/L 농도로 포함하는 것이 좋다. 사과산 또는 구연산을 첨가함으로써 내생포자 사멸능을 더욱 향상시킬 수 있다.
한편, 본 발명의 내생포자 사멸용 조성물은 기타 보조제를 더욱 포함할 수도 있다.
이하, 본 발명의 내용을 하기 실시예를 통하여 보다 상세하게 설명하고자 한다. 다만, 본 발명의 권리범위가 하기 실시예에만 한정되는 것은 아니고 그와 등가의 기술적 사상의 변형까지를 포함한다.
[실시예 1: 차아염소산(HOCl)의 피트산 첨가에 따른 바실러스 포자의 불활성화 증가 확인]
(1) pH 범위에 따른 염소의 분포
식염산 수용액을 전기분해하면 유리염소(Free chlorine=HOCl, OCl-)는 pH에 따라 Cl2, HOCl, OCl-로 존재하게 되는데, 차아염소산(HOCl)의 소독 효율이 차아염소산이온(OCl-) 보다 수십 배에서 수백 배 높은 것으로 보고되고 있다 (도 1).
하지만, 도 1에서 보는 바와 같이 용액의 pH가 4.0~6.0인 상태에서는 유리염소가 차아염소산 형태로 유지되고, pH 8.0 이상에서는 OCl-로 존재한다. 또한, pH 2.0 이하에서는 Cl2 형태로 존재한다. 따라서, 차아염소산을 포함하는 항균제의 항균효율은 pH 범위에 따라 급속하게 변하게 되는 것이다.
(2) 차아염소산나트륨 (NaOCl) 단독 처리군
한편, 내생포자(바실러스 포자)를 대상으로 차아염소산나트륨(NaOCl) 30ppm함유 수용액을 처리한 결과, 2분 경과 시, 약 27% 제어효율을 나타내었고, 10분 경과 시, 약 59% 미만의 제어효율이 관찰되었다 (도 2). 이는 차아염소산나트륨 (NaOCl) 수용액의 경우 pH가 높아 대부분 차아염소산이온(OCl-) 형태로 존재하기 때문에 제어효율이 낮은 것을 의미한다.
(3) 미산성 차아염소산수 단독 처리군
식염산 또는 식염산 및 염화나트륨 수용액의 전기분해를 통해 제조한 30ppm HOCl 함유 '미산성 차아염소산수'의 pH를, HCl을 첨가해서 pH가 4.0~8.0 범위를 나타내도록 하였다. 이후, 내생포자(바실러스포자)를 대상으로 하여 2분간 처리하였는데, pH 5.0~6.5 범위에서 96.3~99.97%의 수준으로 제어하는 것을 확인하였다 (도 3). 다만, pH 5.5를 기점으로 해서 좌우의 pH 변화에 매우 예민하게 내생포자 제어능이 감소함을 확인할 수 있었다.
또한, 도 3을 보면, 내생포자의 제어효율이 99.9% (3log) 이상으로 관찰되는 범위는 약 pH 5.5 주변으로 매우 제한적 범위인 것으로 나타났고, 이 pH 범위를 살짝만 벗어나더라도, 내생포자 제어효율이 큰 편차로 떨어지는 것을 확인할 수 있었다. 특히 pH 4.0~7.0의 범위를 넘어가게 되면 90% 미만으로 제어효율이 크게 감소함을 알 수 있었다. 이와 같은 pH 민감성은 소독제 제조 또는 실제 현장에서 소독제 사용시에 다양한 요인에 의한 pH 변화에 대응할 수 없어 큰 부담으로 작용할 수 있다.
(4) 미산성 차아염소산수에 유기산 복합 처리군
상기와 같은 pH 민감성을 해결하고자, 본 발명에서는 유기산을 첨가해 보았다. 식염산 또는 식염산 및 염화나트륨 수용액의 전기분해를 통해 제조한 30ppm 차아염소산(HOCl) 함유 '미산성 차아염소산수'의 pH를, 유기산의 일종인 피트산(phytic acid) 50%(w/w) 함유 수용액을 미량 첨가하여 pH를 다양하게 조절하여 보았다. 이후, 내생포자(바실러스 포자)를 대상으로 하여 2분간 처리하였다. 그런데, 놀랍게도 유기산(피트산 수용액)을 첨가하여 pH를 조절한 경우, pH 4.0~7.0의 넓은 범위에서 내생포자 제어능이 균일하게 유지됨을 알 수 있었고, 특히나 제어능에 있어서도 99.9%(3log) 이상의 내생포자 제어효율을 보임을 확인할 수 있었다 (도 3). 이는 종래의 방법인 HCl을 첨가해서 pH를 조절한 경우에 비해, 1,000배(3log)의 제어 활성을 보이는 pH 스펙트럼이 현저히 넓어진 것을 의미하는 것이고, 이는 제조 과정 또는 실제 현장에서 pH 변화에 대해 매우 능동적으로 대응할 수 있음을 의미하는 것이다. 즉, 매우 좁은 범위의 pH를 유지해야 하는 민감성을 크게 완화시켜 주는 것이다.
[실시예 2: 차아염소산(HOCl) 및 피트산을 포함하는 본 발명 내생포자 사멸용 조성물 제조]
식염산 수용액의 전기분해를 통해 제조한 30ppm 차아염소산(HOCl) 함유 '미산성 차아염소산수' 1L에 피트산(phytic acid) 50%(w/w) 함유 수용액을 넣어 피트산을 14.32ppm 농도로 함유하는 본 발명의 내생포자 사멸용 조성물을 제조하였다.
[실시예 3: 차아염소산(HOCl), 피트산, 사과산 및 구연산을 포함하는 본 발명 내생포자 사멸용 조성물 제조]
식염산 수용액의 전기분해를 통해 제조한 30ppm 차아염소산(HOCl) 함유 '미산성 차아염소산수' 1L에 피트산(phytic acid) 50%(w/w) 함유 수용액, 사과산 및 구연산 분말을 넣어 피트산 14.32ppm, 사과산 1.79ppb, 구연산 1.79ppb 농도로 함유하는 본 발명의 내생포자 사멸용 조성물을 제조하였다.
Claims (4)
- 미산성 차아염소산수 및 피트산(phytic acid)을 혼합하여 제조한 것으로, 차아염소산(HOCl)을 10~80mg/L, 피트산(phytic acid)을 4~500mg/L 농도로 포함하는 것을 특징으로 하는 내생포자 사멸용 조성물.
- 제1항에 있어서,상기 피트산은,피트산을 45~55%(w/w) 함유하는 수용액 상태로 혼합하는 것을 특징으로 하는 내생포자 사멸용 조성물.
- 제1항에 있어서,상기 내생포자 사멸용 조성물은,사과산과 구연산이 더욱 혼합된 것을 특징으로 하는 내생포자 사멸용 조성물.
- 제3항에 있어서,상기 내생포자 사멸용 조성물은,상기 사과산을 0.0001~1000mg/L, 상기 구연산을 0.0001~1000mg/L 농도로 포함하는 것을 특징으로 하는 내생포자 사멸용 조성물.
