Condensation of IIIa-J with reagents NH 4 Cl in the presence of a base such as DIPEA in a solvent such as DMF results in IIIa-K, which can be further converted to IIIa by the removal of the protective group Boc.

Next day, the supernatant was replaced with medium containing serially diluted compounds (6 points, 4-fold dilutions in duplicate) and 2 ⁇ M CP-100356. Cells were then infected with SARS-CoV-2 ⁇ CoV/Isolate Hong Kong/VM20001061/2020 strain at a multiplicity of infection (MOI) of 0.005. The final concentration of DMSO in the cell culture was 0.5%.
